Pedigree and genetic analysis of a novel mutation carrier patient suffering from hereditary nonpolyposis colorectal cancer

AIM： To screen a suspected Hungarian HNPCC family to find specific mutations and to evaluate their effect on the presentation of the disease. METHODs： The family was identified by applying the Amsterdam and Bethesda Criteria. Immunohistochemistry was performed, and DNA samples isolated from tumor tissue were evaluated for microsatellite instability. The identification of possible mutations was carried out by sequencing the hMLH1 and hMsH2 genes. REsULTs： Two different mutations were observed in the index patient and in his family members. The first mutation was located in exon 7, codon 422 of hMsH2, and caused a change from Glu to sTOP codon. No other report of such a mutation has been published, as far as we could find in the international databases. The second mutation was found in exon 3 codon 127 of the hMsH2 gene, resulting in Asp→ser substitution. The second mutation was already published, as a non-pathogenic allelic variation. CONCLUsION： The pedigree analysis suggested that the newly detected nonsense mutation in exon 7 of the hMsH2 gene might be responsible for the development of colon cancers. In family members where the exon 7 mutation is not coupled with this missense mutation, colon cancer appears after the age of 40. The association of these two mutations seems to decrease the age of manifestation of the disease into the early thirties.

Plant essential oils combined with organic acids restored lipopolysaccharide-induced leaky intestine via gut microbial modulation in weaned piglets

Intestine derived lipopolysaccharide (LPs) is closely related to systemic inflammation and disorders, yet little is known about its roles in the weanling stress of piglets and its potential as a nutritional intervention target. This study aimed to investigate the potential of essential oils (EO) and organic acids (OA) in mitigating weaning stress in piglets by modulating the circulation of intestine derived LPs. seventytwo weaned piglets at 21 d old with body weight of 8.12 +/- 0.168 kg were randomly divided into a control group (CON) and an experimental group, each consisting of six pens with six piglets per pen, and were fed either a basal diet or a basal diet supplemented with 3 kg/t OA + 500 g/t EO (EO + OA). On the 14th day of the feeding trial, 12 weaned piglets were randomly selected from the CON group, and 6 piglets were selected from the experimental group. Based on diet composition and stress treatment, these 18 piglets were divided into the following three groups: 1) CON group. Piglets were fed a basal diet and received an intraperitoneal injection of saline as a control. 2) LPs group. Piglets were fed a basal diet and received an intraperitoneal injection of LPs (100 mg/kg body weight) to induce stress. 3) EO + OA + LPs group. Piglets were fed a basal diet supplemented with EO and OA and received an intraperitoneal injection of LPs (100 mg/kg body weight) to induce stress. The results showed that EO + OA significantly ameliorated the oxidative imbalance and inflammation disorder induced by LPs in piglets' serum and intestine by inhibiting the activation of the Toll-like receptor 4 (TLR4)/nuclear factor- kappa B (NF-kB)/mitogen-activated protein kinase (MAPK) signaling pathway. Background and Objective: Tick-borne hemoparasitic illnesses pose a serious threat to the well-being and productivity of cattle. This study aimed to investigate the prevalence of tick-borne hemoparasites in Cameroon, with a specific focus on the Noun and Nde Divisions of the West Region. Methodology: A total of 423 cattle from 10 villages in both divisions were included in the study. Blood smears were prepared and subjected to microscopic screening for Babesia and Anaplasma parasites. Results: The prevalence of Anaplasma was found to be 23.4%, while Babesia exhibited a seroprevalence of 17.3%. samples from Institute of Agricultural Research for Development (IRAD) and Koupa Ngangnou demonstrated significantly higher prevalence rates, potentially influenced by climate variations affecting tick populations. Additionally, 17.3% of the animals exhibited low hematocrit levels, indicative of anemia. No significant associations were observed between the presence of hemoparasite infection and cattle characteristics. Conclusion: This study provides fundamental data on the extensive distribution and impact of tick-borne hemoparasites in a significant cattle-producing region of Cameroon.

For the past 2 years,the coronavirus responsible for the COVID-19 infection has become a world pandemic,ruining the lives and economies of several nations in the *** has scaled up research on the virus and the resulting infection with the goal of developing new vaccines and *** products are known to be a rich source of lead compounds for drug discovery,including against infectious diseases caused by microbes(viruses,bacteria and fungi).In this review article,we conducted a literature survey aimed at identifying natural products with inhibitory concentrations against the coronaviruses or their target proteins,which lie below 10μ*** led to the identification of 42 compounds belonging to the alkaloid,flavonoid,terpenoid,phenolic,xanthone and saponin *** cut off concentration of 10μM was to limit the study to the most potent chemical entities,which could be developed into therapies against the viral infection to make a contribution towards limiting the spread of the disease.

study on the multi-targets mechanism of YiQiFuMai powder injection on cardio-cerebral ischemic diseases based on network pharmacology

Aim YiQiFuMai Powder Injection is a well-known traditional Chinese medicine formula that has been used extensively in clinical treatment of cardio-cerebral ischemic diseases in China. However, the mechanisms under-lying its clinical efficacy remain unknown. In this study, a network pharmacology approach was employed to identify the YiQiFuMai Powder Injection＇s potential pathways and targets against cardio-cerebral ischemia. The target-path- way interaction network clustered the signaling pathways based on high degree nodes of the drug-target network. The potential protein targets presented in the highly scored clustered pathways were the key network hubs and concentrated on one or limited functional signaling pathways amenable to experimental verification. Twelve main functional annota- tion clusters and main signaling pathways for YiQiFuMai Powder Injection were established by Biocarta analysis, in- eluding the NF-KB signaling pathway, the MAPKinase signaling pathway and the mTOR-signaling pathway and so on. YiQiFuMai Powder Injection is hypothesized to target multiple proteins with a high degree and betweenness of net- work. In addition, the most related pathways were also confirmed in tumor necrosis factor-alpha （TNF-oL） induced human vascular endothelial cell line EA. hy926 by Western blot. This study elucidates the systematic network and pathway analysis of multi-targets in YiQiFuMai Powder Injection. The results provide the possible mechanisms for its mode of action against cardio-cerebral ischemic diseases and may also reveal new clues for its potential application in the inflammatory diseases or tumors.

Objective: To investigate the phytochemical constituents, antioxidant and anthelmintic activities of the crude methanol extract of Cissus populnea(C. populnea) rhizomes. Methods: Phytochemical screening was performed using standard protocols, and column chromatography of silica gel was used for the compounds isolation. DPPH antiradical scavenging assay was performed in order to evaluate the antioxidant activity. Total phenolic content was evaluated using the Folin–Ciocalteu assay. The anthelmintic activity was screened on the bovine adult male forms of parasitic nematode Onchocerca ochengi, by the in vitro evaluation of the inhibition of adult worm motility and mortality. Worms were incubated in the presence of different concentrations of the plant extract and effects on survival were monitored after 24 and 48 h. Results: The preliminary phytochemical screening revealed the presence of phenolic compounds, saponins, steroids, tannins, and terpenoids. Bergenin and a mixture of phytosterol, β-sitosterol and stigmasterol were isolated from this extract and were identified by nuclear magnetic resonance, mass spectrometry and by comparison with published data. The crude methanol extract of C. populnea rhizomes showed a strong DPPH antiradical activity with a good amount of total phenolic content((20.69±2.13) g gallic acid equivalent/100 g of extract) and significant anthelmintic activity comparable to the standard drug ivermectin. Bergenin was found to be inactive even after 72 h of incubation. Conclusions: This study constitutes the first report on the anthelmintic activity of this plant and supports the traditional use of C. populnea as a natural antioxidant and anthelmintic.

Application of Power Electronics and Control for Dual Battery Packs Management with Voltage Balancing and state of Charge Estimation

Energy storage, such as lead acid batteries, is necessary for renewable energy sources’ autonomy because of their intermittent nature, which makes them more frequently used than traditional energy sources to reduce operating costs. The battery storage system has to be monitored and managed to prevent serious problems such as battery overcharging, over-discharging, overheating, battery unbalancing, thermal runaway, and fire dangers. For voltage balancing between batteries in the pack throughout the charging period and the sOC estimate, a modified lossless switching mechanism is used in this research’s suggested battery management system. The OCV state of charge calculation, in the beginning, was used in conjunction with the coulomb counting approach to estimate the sOC. The results reveal that correlation factor K has an average value of 0.3 volts when VM ≥ 12 V and an average value of 0.825 when VM ≤ 12 V. The battery monitoring system revealed that voltage balancing was accomplished during the charging process in park one after 80 seconds with a sOC difference of 1.4% between Batteries 1 and 2. On the other hand, the system estimates the state of charge during the discharging process in two packs, with a maximum DOD of 10.8 V for all batteries. The project’s objectives were met since the BMs estimated sOC and achieved voltage balance.

地表蒸散发是地球表面水量和热量平衡的重要组成部分,在全球水循环和物质能量循环中扮演着重要角色。因此,如何在干旱区域准确估算蒸散发,这对于了解详细的地表水文过程和提供更有效的水资源管理政策以及生态环境保护至关重要。传统测量蒸散发的方法和仪器局限于点尺度,在区域范围研究中存在诸多问题,如人员仪器成本高和易受环境条件影响等。遥感技术克服了传统方法在区域蒸散发研究中的缺点和不足,在节省成本的同时又保证了计算精度,成为区域蒸散发研究的重要技术手段。本文以干旱区的嘉峪关市为研究区,以Landsat 8遥感影像及其他相关数据为基础,运用sEBAL模型估算出了研究区2017年12月21日、2018年3月11日、7月17日和10月5日的日蒸散发量。并分析了关键地表特征参数、模型中主要热通量和日蒸散发的时空分布变化。使用FAO Penman-Monteith公式对sEBAL模型估算结果进行验证。最后,通过地理探测器定量地分析了相关参数对蒸散发空间分布的影响。本文主要工作和成果如下:(1)通过对Landsat 8卫星影像波段的处理,获得地表特征参数,使用这些参数估算得到sEBAL模型中的热分量,并对关键的地表参数和热通量进行时空分析。结果表明:归一化植被指数、地表反照率、地表温度、地表净辐射通量、土壤热通量和显热通量受人类活动影响和下垫面覆盖情况在空间分布上具有明显差异性;时间上,上述主要参数除地表反照率外,均呈现季节性变化,表现为“先增大后减小”的趋势。(2)采用蒸发比不变的方法对瞬时蒸散发进行时间尺度的拓展,得到日蒸散发并进行时空分析。水域的蒸发量最大,绿洲地区、山区和城市区域蒸散发量次之,戈壁地区日蒸散发最低。7月日均蒸散发最大,达到4.335 mm/d,蒸散发离散程度高;3月和10月的日均蒸散发次之,分别为2.516 mm/d和2.089 mm/d;研究区12月的蒸散发量最低,为0.648 mm/d,离散程度低。(3)通过FAO Penman-Monteith公式对遥感估算结果进行验证。两种方法计算出来的农作物的日蒸散发量相差不大,相对误差为8%,结果证明,基于遥感手段的sEBAL模型能够有效反演研究区的实际蒸散发情况。(4)通过地理探测器探究7种影响因子对蒸散发空间分布的影响。在分异及因子探测器中,影响因子对蒸散发的解释力由高到低依次表现为地表净辐射通量(q=0.911)>地表温度(q=0.723)>地表反照率(q=0.483)>归一化植被指数(q=0.297)>海拔高度(q=0.276)>地表比辐射率(q=0.275)>植被覆盖度(0.213)。交互作用探测器显示,任意两个影响因子共同作用于蒸散发时,其交互作用对蒸散发空间分布的解释力都大于任意单个因子对蒸散发的影响,都呈现出双因子增强的效果。在风险区探测器中,地表反照率与地表温度对平均蒸散发量都呈现抑制作用,植被覆盖度、地表净辐射通量和地表比辐射率与平均蒸散发量成正相关,归一化植被指数和海拔高度与蒸散发的关系比较复杂。生态探测器探测结果显示,除高程和地表比辐射率因子对蒸散发量的影响没有明显差异外,其余任意两个影响因子对蒸散发的影响均存在明显的差异。
