Evidence for a sequential transfer of iron amongst ferritin, transferrin and transferrin receptor during duodenal absorption of iron in rat and human

AIM: To elucidate the sequential transfer of iron amongst ferritin, transferrin and transferrin receptor under various iron status conditions. METHODS: Incorporation of 59Fe into mucosal and luminal proteins was carried out in control WKY rats. The sequential transfer of iron amongst ferritin, transferrin and transferrin receptor was carried out in iron deficient, control and iron overloaded rats. The duodenal proteins were subjected to immunoprecipitation and quantitation by specific ElISA and in situ localization by microautoradiography and immunohistochemistry in tandem duodenal sections. Human duodenal.biopsy (n = 36) collected from subjects with differing iron status were also stained for these proteins. RESUlTS: Ferritin was identified as the major protein that incorporated iron in a time-dependent manner in the duodenal mucosa. The concentration of mucosal ferritin was significantly higher in the iron excess group compared to control, iron deficient groups (731.5 ± 191.96 vs 308.3 ± 123.36, 731.5 ± 191.96 vs 256.0 ± 1.19, P le that of luminal transferrin which was significantly higher than the mucosal did not differ among the groups (10.9 ± 7.6 vs 0.87 ± 0.79, 11.1 ± 10.3 vs 0.80 ± 1.20, 6.8 ± 4.7 vs 0.61 ± 0.63, P l transfer of iron. This was demonstrated to occur through the initial.binding of iron to luminal transferrin then to absorptive cell surface transferrin receptors. The staining intensity of these proteins variedaccording to the iron nutrition in humans, with intense staining of transferrin receptor observed in iron deficient subjects. CONClUSION: It is concluded that the intestine takes up iron through a sequential transfer involving interaction of luminal transferrin, transferrin-transferrin receptor and ferritin.

Preliminary Report on the Anurans of Mount Hilong-hilong, Agusan Del Norte, Eastern Mindanao, Philippines

Mount Hilong-hilong is a key biodiversity area, spanning several municipalities in the provinces of the Caraga Region （Agusan del Norte, Agusan del Sur, Surigao del Norte and Surigao del Sur）, northeastern Mindanao Island, southern Philippines. The Hilong-hilong massif remains one of the most signiifcant forested areas in Mindanao, threatened with habitat modification （forest removal, degradation） and other anthropogenic disturbances related to renewable resource extraction. Amphibians are key indicator species for environmental quality and are useful focal taxa for conservation efforts. Relying on historical museum database information and new survey work on Mount Hilong-hilong, we provide species accounts and describe microhabitat preferences of the anurans （frogs and toads） present in the area. Twenty-seven species representing seven anuran families were studied in detail at elevations between 700 to 1300 meters above sea level; 16 of these species are Mindanao faunal region endemics. Qualitative overlap in microhabitat use was observed, suggesting that, for the species recorded, intact forest may ensure species persistence to some levels of anthropogenic disturbance. A more extensive herpetofaunal survey is needed to fully estimate the herpetofaunal diversity of Mount Hilong-hilong. because amphibians represent ifne-scale indicators of environmental quality and microendemism, we recommend appropriate ifne-scaled regional strategies geared towards the conservation of amphibians in the Caraga area, northeast Mindanao Island.

Unsteady generalized Couette flow in composite microchannel

A numerical study is reported on the fully developed unsteady laminar fluid flow in microchannel parallel-plates partially filled with a uniform porous medium and partially filled with a clear fluid. The flow is induced by the movement of one of the plates and the pressure gradient. The brinkman-extended Darcy model is utilized to model the flow in the porous region, while the Stokes equation is used in the clear fluid region. A theoretical analysis is also presented for the fully developed steady flow to find closed- form expressions for the interracial velocity and the velocity and skin frictions at the bounding plates. Numerical computations shows excellent agreement between the closedform solutions for fully developed steady flow and the numerical solution to unsteady flow at large values of time.

Phase transformation and micro structural change of an as-cast ZA27 alloy were investigated during compressive creep by X-ray diffraction technique, SEM and TEM. Compressive creep induced decomposition of α metastable η' phase and a four-phase transformation, α + ε → T' + η and T' → θ, occurred during the compressive creep testing. The occurrence of negative creep in the alloy resulted from the volume expansion caused by the four-phase transformation. A micro structural change was also observed from a lamellar structure into a spheroidized structure in higher creep strain of tested specimens. It provided evidence of compressive creep induced phase transformation which occurred in ageing process.

NUMERICAl SIMUlATION OF TEMPERATURE FIElDS IN ElECTROSlAG REMElTING Sl.b INGOTS

The method based on transient heat transfer model is adopted to simulate electro-slag remelting process. The calculated results of the model show that the process is in the quasi-steady state, and the shape of pool remains unchanged when the height of ingot is approximately 2.5-3 times the thickness of sl.b ingot. The change in the shape of pool is found to be strongly dependent on the pattern of melting rate, and hence, the power input; the depth of the molten pool increases with the increase in melting speed. It is concluded that a transient heat transfer model has to be used to obtain reliable input information for the entire, operatina time.

MicroRNA-let-7a promotes E2F-mediated cell proliferation and NFKb activation in vitro

Epigenetic factors, including altered microRNA （miRNA） expression, may contribute to aberrant immune cell function in systemic lupus erythematosus （SlE）. MiRNA-let-7a （let-7a） has been shown to directly alter cell cycle progression and proinflammatory cytokine production. Due to the crucial role of let-7a in cell division and inflammation, we investigated let-7a-mediated proliferation and NFKb translocation in J774A. 1 macrophages and MES 13 mesangial cells in vitro. In immune-stimulated cells transfected with let-7a, cell proliferation was significantly increased over time. There was a significant increase in the number of immune-stimulated cells in S and G2 phases. Immune-stimulated cells overexpressing let-7a had increased nuclear translocation of NFKb. bioinformatical analysis revealed that the E2F family, critical regulators of the G1-S transition, has potential.binding sites for let-7a in their mRNA transcripts. let-7a overexpression significantly increased the expression of the cell cycle activator E2F2 and increased retinoblastoma protein （Rb） phosphorylation in immune-stimulated cells. The cell cycle inhibitor E2F5 was significantly decreased in let-7a-transfected cells that were immune-stimulated. bioinformatical analysis revealed E2F2 and NFKb are transcription factors predicted to regulate the let-7a promoter. We analyzed transcriptional regulation of let-7a by real-time RT-PCR using chromatin immunoprecipitation with E2F2 and NFKb antibodies. There was an increase in E2F2 and NFKb binding in DNA enriched for the let-7a promoter in immune-stimulated cells. Silencing E2F2 or NFKb significantly decreased let-7a expression and Il-6 production in immune-stimulated cells. Taken together, our results suggest that overexpression of let-7a may contribute to hyperplasia and the proinflammatory response in SlE.

The effect of Argan plant extract （APE） on the corrosion of the steel in hydrochloric acid medium was studied using gravimetric, electrochemical polarization and impedance spectroscopy （EIS） measurements. Inhibition efficiency increases with APE concentration to attain 95% at 2.5 g/l. We note good agreement between gravimetric and electrochemical methods （potentiodynamic polarization and EIS）. Effect of temperature is also made in the 298-328 K range. Polarization measurements show also that APE act as a mixed inhibitor. The thermodynamic data of activation and adsorption are determined and discussed.

Thermal nanoimprinting is a fast and versatile method for transferring the anti-reflective properties of subwavelength nanostructures onto the surface of highly reflective substrates, such as chalcogenide glass optical fiber end faces. In this paper, the technique is explored experimentally on a range of different types of commercial and custom-drawn optical fibers to evaluate the influence of geometric design, core/cladding material, and thermo-mechanical properties. Up to32.4% increased transmission and 88.3% total transmission are demonstrated in the 2–4.3 μm band using a mid-infrared(IR) supercontinuum laser.
