Vaccinium darrowii is a subtropical wild blueberry species that has been used to breed economically important southern highbush cultivars.The adaptive traits of V.darrowii to subtropical climates can provide valuable information for breeding blueberry and perhaps other plants,especially against the background of global warming.Here,we assembled the V.darrowii genome into 12 pseudochromosomes using Oxford Nanopore long reads complemented with Hi-C scaffolding technologies,and we predicted 41815 genes using RNA-sequencing evidence.Syntenic analysis across three Vaccinium species revealed a highly conserved genome structure,with the highest collinearity between V.darrowii and Vaccinium corymbosum.This conserved genome structure may explain the high fertility observed during crossbreeding of V.darrowii with other blueberry cultivars.Analysis of gene expansion and tandem duplication indicated possible roles for defense-and flowering-associated genes in the adaptation of V.darrowii to the subtropics.Putative SOC1 genes in V.darrowii were identified based on phylogeny and expression analysis.Blueberries are covered in a thick cuticle layer and contain anthocyanins,which confer their powdery blue color.Using RNA sequencing,we delineated the cuticle biosynthesis pathways of Vaccinium species in V.darrowii.This result can serve as a reference for breeding berries whose colors are appealing to customers.The V.darrowii reference genome,together with the unique traits of this species,including its diploid genome,short vegetative phase,and high compatibility in hybridization with other blueberries,make V.darrowii a potential research model for blueberry species.

Abscisic Acid Transport and Homeostasis in the Context of Stomatal Regulation

The discovery of cytosolic ABA receptors is an important breakthrough in stomatal research; signaling via these receptors is involved in determining the basal stomatal conductance and stomatal responsiveness. However, the source of ABA in guard cells is still not fully understood. The level of ABA increases in guard cells by de novo synthesis, recycling from inactive conjugates via β-glucosidases BG1 and BG2 and by import, whereas it decreases by hydroxylation, conjugation, and export. ABA importers include the NRT1/PTR family protein AIT1, ATP-binding cassette protein ABCG40, and possibly ABCG22, whereas the DTX family member DTX50 and ABCG25 function as ABA exporters. Here, we review the proteins involved in ABA transport and homeostasis and their physiological role in stomatal regulation. Recent experiments suggest that functional redundancy probably exists among ABA transporters between vascu- lature and guard cells and ABA recycling proteins, as stomatal functioning remained intact in abcg22, abcg25, abcg40, aitl, and bglbg2 mutants. Only the initial response to reduced air humidity was significantly delayed in abcg22. Considering the reports showing autonomous ABA synthesis in guard cells, we discuss that rapid stomatal responses to atmospheric factors might depend primarily on guard cellsynthesized ABA, whereas in the case of long-term soil water deficit, ABA synthesized in the vasculature might have a significant role.

The cuticle is the outer physical barrier of aerial plant surfaces and an important interaction point between plants and the environment. Many environmental stresses affect cuticle formation, yet the regulatory pathways involved remain undefined. We used a genetics and gene expression analysis in Arabidopsis thaliana to define an abscisic acid （ABA） signaling loop that positively regulates cuticle formation via the core ABA signaling pathway, including the PYR/PYL receptors, PP2C phosphatase, and SNF1-Related Protein Kinase （SnRK） 2.21SnRK2.3/SnRK2.6. Downstream of the SnRK2 kinases, cuticle formation was not regulated by the ABA-responsive element-binding transcription factors but rather by DEWAX, MYB16, MYB94, and MYB96. Additionally, low air humidity increased cuticle formation independent of the core ABA pathway and cell death/reactive oxygen species signaling attenuated expression of cuticle-biosynthesis genes. In Physcornitrella patens, exogenous ABA suppressed expression of cuticle- related genes, whose Arabidopsis orthologs were ABA-induced. Hence, the mechanisms regulating cuticle formation are conserved but sophisticated in land plants. Signaling specifically related to cuticle deficiency was identified to play a major role in the adaptation of ABA signaling pathway mutants to increased humidity and in modulating their immunity to Botrytis cinerea in Arabidopsis. These results define a cuticle-specific downstream branch in the ABA signaling pathway that regulates responses to the external environment.

Challenges facing the USA in matching Germany on advanced manufacturing for green growth

This study explores the relative strength of public policies promoting advanced manufacturing by comparing the government programs and capacities of two world leaders in science and technology:the USA and Germany.Based on insights from’varieties-of-capitalism’literature and using process tracing methodology,this study identifies significant differences in today’s pursuit of advanced manufacturing.The German model of Rhine capitalism has strength due to its long tradition of diversified quality work,built on a long-term commitment to government-industry collaboration,with significant indirect public support provided by vocational training and research institutes.The American model of Manchester capitalism has been challenged by the loss of millions of low-skilled manufacturing jobs due to outsourcing and competition from Asia.The study finds that the“Manufacturing USA”policy initiative of the Obama era,although directly inspired by Germany’s Fraunhofer institutes,is hampered by the contextual and institutional circumstances identified.With biobased carbon fibers offering the potential of a novel,low-cost material,pursued with research and development(R&D)efforts in both countries,it provides a measure for evaluating policy outcomes,and the study confirms Germany’s technological lead.In addition,the study’s process-tracing methodology reveals that significant legacies from German knowledge creation in the past have spilled over to American science and technology,and may continue to do so,lured by government funds.Further studies on advanced manufacturing should test the robustness of these findings.

Invariant natural killer T(iNKT)cells are part of the family of unconventional T cells,and they recognize glycolipid antigens presented on the MHC class I like molecule CD1^([1]).In humans there are five CD1 molecules named CD1a to CD1e,whereas mice use only CD1d for lipid presentation.Functionally,iNKT cells can elicit both cytotoxicity similarly to conventional CD8 T cells and secrete cytokines that influence the adaptive immune response including providing direct T cell help to B cells^([2]).In the context of tumors,they can use both release of granzyme B and use Fas-mediated mechanisms for killing of tumor cells.As iNKT cells use a restricted TCR repertoire and CD1 molecules are non-heterogenous the risk of graft vs host disease is limited and thus these T cells have been an attractive option for off the shelf immunotherapy to treat cancer^([3]).

Background:Adenosine triphosphatase inhibitory factor 1(IF1)is a key protein involved in energy metabolism.IF1 has been linked to various agerelated diseases,although its relationship with physical activity(PA)remains unclear.Additionally,the apolipoprotein A-I(apoA-I),a PA-modulated lipoprotein,could play a role in this relationship because it shares a binding site with IF1 on the cell-surface ATP synthase.We examined here the associations between chronic PA and plasma IF1 concentrations among older adults,and we investigated whether apoA-I mediated these associations.Methods:In the present work,1096 healthy adults(63.8%females)aged 70 years and over who were involved in the Multidomain Alzheimer Prevention Trial study were included.IF1 plasma concentrations(square root of ng/mL)were measured at the 1-year visit of the Multidomain Alzheimer Prevention Trial,while PA levels(square root of metabolic equivalent task min/week)were assessed using questionnaires administered each year from baseline to the 3-year visit.Multiple linear regressions were performed to investigate the associations between the first-year mean PA levels and IF1 concentrations.Mediation analyses were conducted to examine whether apoA-I mediated these associations.Mixedeffect linear regressions were carried out to investigate whether the 1-year visit IF1 concentrations predicted subsequent changes in PA.Results:Multiple linear regressions indicated that first-year mean PA levels were positively associated with IF1 concentrations(B=0.021;SE=0.010;p=0.043).Mediation analyses revealed that about 37.7%of this relationship was mediated by apoA-I(B_(ab)=0.008;SE=0.004;p=0.023).Longitudinal investigations demonstrated that higher concentrations of IF1 at the 1-year visit predicted a faster decline in PA levels over the subsequent 2 years(time×IF1:B=0.148;SE=0.066;p=0.025).Conclusion:This study demonstrates that regular PA is associated with plasma IF1 concentrations,and it suggests that apoA-I partly mediates this association.Additionally,this study f

现代医学认为,脑主导机体的一切生命过程和精神活动,且与周围环境的平衡和调节存在关系。而中医认为“心者,君主之官,神明出焉”,心有主宰身体和神志的功能。褪黑素(MT)是脑室内松果体分泌的一种激素,其在人体的效应有类似中医理论中的“心”“神”作用。如何理解并且融通中医和现代医学两个不同的理论体系,是目前在临床和科研中的重要问题。“心脑一体观”是结合不同理论体系的对同一个生理现象的探讨。本研究基于“心脑一体观”探讨MT失调所致疾病的相关研究,阐释针灸通过调理“心”“神”相关的穴位组合,不仅可改善失眠、抑郁、肥胖、血管性痴呆、胃溃疡等疾病症状,同时具有调节MT水平的作用,为临床针灸治疗MT相关疾病的腧穴配伍提供理论依据。

Effects of Fresh Cupressus sempervirens Leaves Infusion on Growth Performance, Intestinal Microbiota and Haemato-Biochemical Parameters in Broilers

The accumulation of growth-promoting antibiotic residues in animal products and the resistance developed by bacteria in poultry farms has led to a search for natural compounds derived from plants. This study was designed to promote the production performance of broiler chickens using fresh Cupressus sempervirens leaves infusion. Fresh Cupressus sempervirens leaves were harvested, washed, chopped and ground to a paste using a blender and fermented for three days in a closed container at a rate of 500 g/L of water. The solution obtained was filtered and added at the rate of 2, 4, 6, 8 and 10 ml/L of drinking water. The chickens fed on the graded level of the solution were compared to a control ration without an additive and positive control group supplemented with 1 g antibiotic/kg feed. At the finisher phase and throughout the study period, water intake increased significantly (P 0.05) affected by the inclusion of Cupressus sempervirens infusion. Haematological parameters significantly (P < 0.05) increase independently of the rate of incorporation of the infusion into the drinking water, with the exception of RBCs, MCHT and PCT. Serum content in total protein, globulins, LDL cholesterol and triglycerides were significantly (P < 0.05) high with 8 and 10 ml Cupressus sempervirens infusion/litre drinking water as compared to all other treatments. AST, ALT, urea, creatine, albumin, total cholesterol and HDL-cholesterol were not significantly affected.
