Background: Changes in the day-to-day variability in the glucose-lowering effect of insulin [fluctuations of blood glucose levels (BG) seen during the same time period] that occur when insulin glargine (glargine) is replaced with insulin degludec (degludec) have not been sufficiently evaluated. Subjects: Five diabetics with unstable BG undergoing basal-bolus treatment using insulin glargine as basal insulin. Methods: Basal insulin was changed from glargine to same-dose degludec. The subjects’ HbA1c, glycoalbumin, and 1.5-anhydro-D-glucitol were measured before and after the switchover. Fasting blood glucose concentration (FBG) and predinner blood glucose concentration (PDBG) were measured continuously for 28 days immediately before the switchover, and 28 days immediately thereafter, to compare and evaluate 1) the changes in their mean values and standard deviations (SDs) before and after the switchover, and 2) the frequency of appearance of BG of over 180 mg/dL (BG ≥ 180) and under 70 mg/dL (BG Results: The levels of HbA1c, glycoalbumin, FBG’s mean value, SDs, BG ≥ 180 and BG Conclusion: The possibility was shown that degludec, to a greater extent than glargine, suppressed daily fluctuations of FBG and PDBG, suppressed the occurrence frequency of hyperglycemia and hypoglycemia, and exerted more steady hypoglycemic actions.

Aims: We evaluated whether urinary excretion of type IV collagen (U-COL) may predict an increase in the urinary albumin-to-creatinine ratio (ACR) and what factors regulate U-COL in 145 normoalbuminuric patients with type 2 diabetes. Methods: We measured HbA1c, systolic blood pressure (SBP), urinary 8-hydroxydeoxyguanosine (8-OHdG) and monocyte chemoattractant protein (MCP)-1 at start of this study (Baseline), ACR and U-COL in addition to these measurements at one year later (Evaluation-1), and ACR and SBP after two years of the Evaluation-1 (Evaluation-2). The relationships were investigated between the increase of ACR and the U-COL. The effect of angiotensin receptor blockers (ARB) treatment on the correlations between U-COL and ACR at Evaluation-2 on one hand, and between U-COL and percent change of ACR on the other, was also analyzed. Furthermore, we investigated whether the increase in 8-OHdG and in MCP-1 in a year prior to the Evaluation-1 were risk factors of the rise in U-COL levels. Results: Both U-COL and SBP at Evaluation-1, but not ARB treatment, were independent risk factors for an increased ACR after 2 years. ARB treatment significantly suppressed the increase in ACR after 2 years in patients with higher U-COL excretion. The percentage changes in 8-OHdG (%8-OHdG) and MCP-1 (%MCP-1) in one year prior to Evaluation-1 measurements are independent risk factors for U-COL. HbA1c and SBP values one year prior to Evaluation-1 are independent risk factors not only for %8-OHdG but also, for baseline U-COL. The %8-OHdG is an independent risk factor for %MCP-1. Conclusions: U-COL may predict an increase in the ACR. The U-COL seems to be increased with oxidative stress and inflammation induced by past hyperglycemia.

The bone extracellular matrix(ECM) contains minerals deposited on highly crosslinked collagen fibrils and hundreds of noncollagenous proteins. Some of these proteins are key to the regulation of bone formation and regeneration via signaling pathways,and play important regulatory and structural roles. However, the complete list of bone extracellular matrix proteins, their roles, and the extent of individual and cross-species variations have not been fully captured in both humans and model organisms. Here, we introduce the most comprehensive resource of bone extracellular matrix(ECM) proteins that can be used in research fields such as bone regeneration, osteoporosis, and mechanobiology. The Phylobone database(available at https://***) includes 255proteins potentially expressed in the bone extracellular matrix(ECM) of humans and 30 species of vertebrates. A bioinformatics pipeline was used to identify the evolutionary relationships of bone ECM proteins. The analysis facilitated the identification of potential model organisms to study the molecular mechanisms of bone regeneration. A network analysis showed high connectivity of bone ECM proteins. A total of 214 functional protein domains were identified, including collagen and the domains involved in bone formation and resorption. Information from public drug repositories was used to identify potential repurposing of existing drugs. The Phylobone database provides a platform to study bone regeneration and osteoporosis in light of(biological) evolution,and will substantially contribute to the identification of molecular mechanisms and drug targets.

Pax6 and its Drosophila homolog Eyeless(Ey) play essential roles during eye ***/Pax6 contains two distinct DNA binding domains,a Paired domain(PD) and a Homeodomain(HD).While Ey/Pax6 PD is required for the expression of key regulators of retinal development,relatively little is known about the HD-dependent Ey *** this study,we used the UAS/GAL4 system to determine the functions of different Ey domains on cell growth and on retinal *** showed that Ey can promote cell growth,which requires the HD but not the *** contrast,the ability of Ey to activate Ato expression and induce ectopic eye formation requires the PD but not the ***,deletion of the HD enhanced Ey-dependent ectopic eye induction while overexpression of the HD only Ey forms antagonizes ectopic eye *** studies revealed a novel function of Ey HD on cell growth and a novel antagonistic effect of Ey HD on Ey PD-dependent eye *** further show the third helix of the Ey HD can directly interact with the RED subdomain in Ey PD and that deletion of the HD increased the binding of Ey PD to its *** results suggest that the direct interaction between the HD and the PD potentially mediates their antagonistic *** different Ey splicing forms are expressed in overlapping regions during normal development,we speculate that the expression ratios of the different Ey splice forms potentially contribute to the regulation of growth and differentiation of these tissues.

We report a case of hemangiopericytoma of the soft palate of 60-year-old patient, who noticed a mass of the soft palate and experienced difficulty in speaking. We found a pediculate, hard, elastic mass measuring 38 mm （cross-sectional diameter）. Computed tomography （CT） scans and dynamic magnetic resonance imaging （MRI） confirmed irregularly shaped mass and revealed a heterogeneous internal composition, consistent with vascular tumors. We excised the tumor under general anesthesia. Histopathological diagnosis was based on positive immunoreactivity of CD99 and vimentin and weak, positive staining of CD34. Three and half years following tumor excision, there is no recurrence or metastasis.

We perform an extensive review of the numerous studies and methods used to determine the total mass of the Milky *** group the various studies into seven broad classes according to their modeling *** classes include:i)estimating Galactic escape velocity using high velocity objects;ii)measuring the rotation curve through terminal and circular velocities;iii)modeling halo stars,globular clusters and satellite galaxies with the spherical Jeans equation and iv)with phase-space distribution functions;v)simulating and modeling the dynamics of stellar streams and their progenitors;vi)modeling the motion of the Milky Way,M31 and other distant satellites under the framework of Local Group timing argument;and vii)measurements made by linking the brightest Galactic satellites to their counterparts in *** each class of methods,we introduce their theoretical and observational background,the method itself,the sample of available tracer objects,model assumptions,uncertainties,limits and the corresponding measurements that have been achieved in the *** the measured total masses within the radial range probed by tracer objects and the extrapolated virial masses are discussed and *** also discuss the role of modern numerical simulations in terms of helping to validate model assumptions,understanding systematic uncertainties and calibrating the *** measurements in the last two decades show a factor of two scatters,recent measurements using Gaia DR2 data are approaching a higher *** end with a detailed discussion of future developments in the field,especially as the size and quality of the observational data will increase tremendously with current and future *** such cases,the systematic uncertainties will be dominant and thus will necessitate a much more rigorous testing and characterization of the various mass determination methods.

Objective:To evaluate the neuroprotective effects of the organic components of scallop shells(scallop shell extract) on memory impairment and locomotor activity induced by scopolamine or 5-methyl-10,11-dihydro-5H-dibenzo(a,d) cyclohepten-5,10-imine(MK801).Methods:Effect of the scallop shell extract on memory impairment and locomotor activity was investigated using the Y-maze test,the Morris water maze test,and the open field ***:Scallop shell extract significantly reduced scopolamine-induced short-term memory impairment and partially reduced scopolamine-induced spatial memory impairment in the Morris water maze *** shell extract suppressed scopolamine-induced elevation of acetylcholine esterase activity in the cerebral *** with scallop shell extract reversed the increase in locomotor activity induced by *** shell extract also suppressed the increase in locomotor activity induced by ***:Our results provide initial evidence that scallop shell extract reduces scopolamine-induced memory impairment and suppresses MK-801-induced hyperlocomotion.
