Clones identification of Sequoia sempervirens (D. Don) Endl. in Chile by using PCR-RAPDs technique

A protocol of polymerase chain reaction-random amplified polymorphic DNAs (PCR-RAPDs) was established to analyse the gene diversity and genotype identification for clones of Sequoia sempervirens (D. Don) Endl. in Chile. Ten (out of 34) clones from introduction trial located in Voipir-Villarrica, Chile, were studied. The PCR-RAPDs technique and a modified hexadecyltrimethylammonium bromide (CTAB) protocol were used for genomic DNA extraction. The PCR tests were carried out employing 10-mer random primers. The amplification products were detected by electrophoresis in agarose gels. Forty nine polymorphic bands were obtained with the selected primers (BG04, BF07, BF12, BF13, and BF14) and were ordered according to their molecular size. The genetic similarity between samples was calculated by the Jaccard index and a dendrogram was con- structed using a cluster analysis of unweighted pair group method using arithmetic averages (UPGMA). Of the primers tested, 5 (out of 60) RAPD primers were selected for their reproducibility and high polymorphism. A total of 49 polymorphic RAPD bands were detected out of 252 bands. The genetic similarity analysis demonstrates an extensive genetic variability between the tested clones and the dendrogram depicts the genetic relationships among the clones, suggesting a geographic relationship. The results indicate that the RAPD markers permitted the identification of the assayed clones, although they are derived from the same geo- graphic origin.

Optimal response to drought is critical for plant survival and will affect biodiversity and crop performance during climate change. Mitotically heritable epigenetic or dynamic chromatin state changes have been implicated in the plant response to the drought stress hormone abscisic acid （ABA）. The Arabidopsis SWI/SNF chromatin-remodeling ATPase BRAHMA （BRM） modulates response to ABA by preventing pre- mature activation of stress response pathways during germination. We show that core ABA signaling pathway components physically interact with BRM and post-translationally modify BRM by phosphoryla- tion/dephosphorylation. Genetic evidence suggests that BRM acts downstream of SnRK2.2/2.3 kinases, and biochemical studies identified phosphorylation sites in the C-terminal region of BRM at SnRK2 target sites that are evolutionarUy conserved. Finally, the phosphomimetic BRMs17s^D S1762D mutant displays ABA hypersensitivity. Prior studies showed that BRM resides at target loci in the ABA pathway in the presence and absence of the stimulus, but is only active in the absence of ABA. Our data suggest that SnRK2- dependent phosphorylation of BRM leads to its inhibition, and PP2CA-mediated dephosphorylation of BRM restores the ability of BRM to repress ABA response. These findings point to the presence of a rapid phosphorylation-based switch to control BRM activity; this property could be potentially harnessed to improve drought tolerance in plants.

In the present study,the green synthesis of potassium nanoparticles(K-NPs)was assessed using aqueous extract of Sideroxylon *** potassium nanoparticles were analyzed by UV-visible spectroscopic techniques,X-ray spectrometers of energy dispersive(SEM-EDS)and dynamic light *** results showed high values at 3.5 keV confirming the formation of potassium nanoparticles and the SEM analysis showed an agglomerated particles size between 360 to 200 nm with a spherical *** K-NPs showed an effective antibacterial activity against the test organisms mainly with Bacillus cereus,Enterobacter aerogenes,Fusarium solani and Botrytis *** further studies about nanotoxicity of K-NPs are needed to confirm their potential in the control of the pathogen microorganisms under field conditions.

Comparison of clinical outcomes between unrelated single umbilical cord blood and"ex-vivo"T-cell depleted haploidentical transplantation in children with hematological malignancies

Background Over the last two decades,umbilical cord blood(UCB)and haploidentical transplantation(HaploHSCT)have emerged as alternative sources of hematopoietic stem cell for allogeneic *** are few retrospective studies and no prospective studies comparing both types of alternative transplantation in pediatric *** We analyzed the data of 134 children with hematological malignancies who received a hematopoietic stem cell transplantation from a single umbilical cord blood(UCB)(n=42)or an"tex-vivo"T-cell depleted transplant from a haploi-dentical-related donor(HaploHSCT)(n=92)between 1996 and *** recovery was faster after HaploHSCT than the UCB transplant group(median times to neutrophil and platelet recovery:13 vs.16 days,10 vs.57 days,respectively)(P<0.001).The HaploHSCT group had a significantly early immune reconstitution based on NK and CD8+T cells compared with the UCB ***,after the first year *** had a lower number of CD4+ T and B lymphocytes compared with the UCB transplant *** cumulative incidence of TRM was 29±8%in the HaploHSCT group versus 40±5%in the UCB *** incidence was 21±7%in the HaploHSCT group and 19±8%in the UCB *** of DFS was 58±8%in the HaploHSCT group versus 40±9%in the UCB group(P=0.051).Conclusions TCD haploidentical transplant is associated with advantages in terms of engraftment and early immune reconstitution *** haploidentical transplant was associated with lower incidence of infectious and non-infectious complications,especially in the early phases of the transplant compared with UCB transplant ***,there are no advantages in transplant outcomes compared with UCB transplant.

Ulcerative colitis is a global health problem,affecting millions of individuals *** an inflammatory condition localised in the large intestine,rectal delivery of immunosuppressive therapies such as tacrolimus is a promising strategy to maximise drug concentration at the site of action whilst minimising systemic side ***,for the first time,self-supporting 3D-printed tacrolimus suppositories were prepared without the aid of moulds using a pharmaceutical semi-solid extrusion(SSE)3D *** suppositories were printed vertically in three different sizes using combinations of two lipid pharmaceutical excipients(Gelucire 44/14 or Gelucire 48/16)and coconut *** both suppository formulations had the appropriate viscosity characteristics for printing,the Gel 44 formulation required less energy and force for extrusion compared to the Gel 48 *** Gel 44 disintegrated more rapidly but released tacrolimus more slowly than the Gel 48 *** the tacrolimus release profiles were significantly different,both suppository systems released more than 80% drug within 120 *** and XRD analysis was inconclusive in determining the solid-state properties of the drug in the *** summary,this article reports on the fabrication of 3D printed selfsupporting suppositories to deliver personalised doses of a narrow therapeutic index drug,with potential benefits for patients with ulcerative colitis.

A Comparative Study of Non-Volatile Compounds Present in 3R4F Cigarettes and iQOS Heatsticks Utilizing GC-MS

It is now widely suggested that people who are dependent on nicotine should switch from ordinary tobacco smoking to alternative products, which at least reduce the overall harm from smoking. A number of alternatives are now popular, including electronic cigarettes and heatsticks. In this work comparative analysis of the smoke/aerosol emission from 3R4F standard cigarettes and iQOS heatsticks was undertaken. For this, gas chromatography-mass spectrometry (GC-MS) analysis was applied, to measure the non-volatile compounds of smoke/aerosol emission from individual samples, with the specific aim to determine their content of nicotine and selected other main components. All measurement data were collected under the Health Canada Intense (HCI) puffing regime. The most relevant findings of the present investigation can be summarized as follows. First, the number of measured aerosol components in the iQOS samples, with respect to those of 3R4F samples, was significantly lower (notably 37 versus 12 components). Second, the analysis of the iQOS and 3R4F GC-MS chromatographic fingerprints indicated a non-nicotine global component reduction (number and areas excluding nicotine) of larger than 80% for the iQOS samples in comparison to 3R4F samples. Third, the nicotine content of the iQOS aerosol was less than half that contained in the 3R4F smoke. The results from the present investigation indicate that—except for nicotine—smokers are exposed to a largely reduced number and amount of non-volatile, non-nicotine components in the iQOS heatstick aerosol, in comparison to those in the 3R4F cigarette smoke.

Gelatin based adhesives have been used in the last decades in different biomedical applications due to the excellent biocompatibility,easy processability,transparency,non-toxicity,and reasonable mechanical properties to mimic the extracellular matrix(ECM).Gelatin adhesives can be easily tuned to gain different viscoelastic and mechanical properties that facilitate its ocular *** herein grafted glycidyl methacrylate on the gelatin backbone with a simple chemical modification of the precursor,utilizing epoxide ring-opening reactions and visible *** chemical modification allows the obtaining of an elastic protein-based hydrogel(GELGYM)with excellent biomimetic properties,approaching those of the native *** can be modulated to be stretched up to 4 times its initial length and withstand high tensile stresses up to 1.95 MPa with compressive strains as high as 80%compared to Gelatin-methacryloyl(GeIMA),the most studied derivative of gelatin used as a *** is also highly biocompatible and supports cellular adhesion,proliferation,and migration in both 2 and 3-dimensional *** characteristics along with its super adhesion to biological tissues such as cornea,aorta,heart,muscle,kidney,liver,and spleen suggest widespread applications of this hydrogel in many biomedical areas such as transplantation,tissue adhesive,wound dressing,bioprinting,and drug and cell delivery.

Dear Editor Abscisic acid （ABA） is the main phytohormone involved in adaptive crop responses to drought. ABA signaling relies on the family of pyrabactin resistance 1/PYRl-like/regulatory components of ABA receptors （PYR/PYL/RCAR）, which upon ABA binding form high-affinity ternary complexes with clade A protein phosphatases type 2C （PP2Cs） and inhibit them （Cutler et al., 2010）. This results in the activation of an SnRK2-dependent phosphorylation cascade affecting a high number of targets in the plant cells, regulating stomatal aper- ture and the expression of stress-responsive genes （Cutler et al., 2010）.

Electroencephalographic Correlates of Sexual Arousal Induced by Sexually-Explicit Reading in Human Females

This study explored the cortical functionality in young women while reading a sexually-explicit text. Electroencephalographic activity (EEG) was recorded in heterosexual women while reading either a sexually-explicit text or one with neutral content. Absolute power (AP) and the degree of EEG synchronization among the prefrontal, temporal and parietal cortices were analyzed for the different EEG bands. To evaluate sexual arousal, valence and general activation, Likert-type and Manikin scales were applied. Subjects scored the sexually-explicit text as more pleasurable, and reported greater general activation and sexual arousal than while reading the neutral one. During reading of the sexually-explicit text only, they showed a higher AP in the beta and gamma bands in temporal areas in both hemispheres, and in the theta band in parietal ones, with a lower degree of EEG synchronization between prefronto-temporal areas in the fast bands. Results show that both hemispheres were activated in these sexually-aroused women. The low bilateral synchronization between prefrontal and temporal regions indicates the independent functionality of these cortices, which could be a requirement for inducing and maintaining sexual arousal during reading of sexually-explicit texts in women. These data should broaden our knowledge of the cortical mechanisms that underlie sexual arousal in women.
