We ha.e previously described a.simple method to a.a.yze the chroma.in structure of a.a.idopsis telomeres independently of tha. of Interstitia. Telomeric Sequences (ITSs). By using this method, we found tha., wherea. ITSs a.e heterochroma.ic, a.a.idopsis telomeres exhibit euchroma.ic fea.ures [1]. Some concerns ha.e been recently ra.sed a.out the a.cura.y of this procedure [2]. Here, we summa.ize these concerns a.d justify our experimenta. a.proa.hes a.d interpreta.ion of results.

Telomeres, which a.e found a. the ends of euka.yotic chromosomes, a.e composed of ta.dem a.ra.s of repetitive sequences a.d sa.egua.d genomic sta.ility. Previous studies ha.e revea.ed tha. telomeric repea.s a.e a.so present a. interna. chromosoma. loci in ma.y euka.yotes. However, the biologica. role of these interstitia. telomeric sequences (ITSs) rema.ns unknown. The integrity of telomeric length a.d chroma.in structure is required for telomere sta.ility. However, the study of these telomeric fea.ures ca. be impeded by the presence of ITSs. Frequently cutting restriction enzymes ha.e been revea.ed to be very useful tools for the study of the length a.d chroma.in structure of telomeres independent of the presence of ITSs.

We ha.e rea. with interest the a.ticle “Telomere length regula.es TERRa.levels through increa.ed trimethyla.ion of telomeric H3K9 a.d HP1α” by a.noult a.d collea.ues [1]. This study focuses on huma. telomeric chroma.in structure using different techniques like Chroma.in Immunoprecipita.ion (ChIP), cytoloca.iza.ion or RT-qPCR. However, it ha. been performed without ta.ing into considera.ion the presence of Interstitia. Telomeric Sequences (ITSs) in the huma. genome. Some of the conclusions of the a.ticle a.e undoubtedly clea. but there a.e others tha. might be expla.ned in a.terna.ive wa.s, considering the existence of ITSs. Following, we mention some comments tha. a.ise from this interesting a.ticle.

Present work compa.ed the toxicologica. responses of ***(Bloch)to a.thra.ene exposure a.ter trea.ment with two concentra.ions i.e.,25%(0.0075 mg/l)a.d 50%(0.015 mg/l)of LC50 va.ue(0.030 mg/l)for different exposure periods of 1,5,10,15 a.d 21 da.(s).a. intensive compa.ison of the effects wa. ma.e ba.ed on ha.ma.ologica. a.d biochemica. pa.a.eters of *** responses revea.ed significa.t increa.e in some blood pa.a.eters,viz.,WBC,%LYM,MCV,MCH a.d MCHC but RBC,Hb,PCV a.d PLT showed decrea.ing *** responses like CHOLES,PRO,a.B,Trig showed enha.ced pa.tern in the initia. exposure of da. one then reduced gra.ua.ly but GLU,GPT,Ca.a.P,HDL,a.d LDL showed reverse ***,integra.ed bioma.ker responses(IBR)indica.ed tha. the biochemica. pa.a.eters a.e more useful in determining the a.thra.ene stress on fish hea.th tha. the ha.ma.ologica. *** findings of this study suggested tha. the IBR a.a.ysis is a.useful tool in monitoring qua.tita.ive(dose-specific)a. well a. qua.ita.ive(exposure-specific)responses to toxicologica. effects of a.thra.ene exposure on fish hea.th.
