Enteric glia mediate neuronal outgrowth through release of neurotrop.ic factors

p.evious studies have shown that transp.anted enteric glia enhance axonal regeneration, reduce tissue damage, and p.omote functional recovery following sp.nal cord injury. However, the mechanisms by which enteric glia mediate these beneficial effects are unknown. Neurotrop.ic factors can p.omote neuronal differentiation, survival and neurite extension. We hyp.thesized that enteric glia may exert their p.otective effects against sp.nal cord injury p.rtially through the secretion of neurotrop.ic factors. In the p.esent study, we demonstrated that p.imary enteric glia cells release nerve growth factor, brain-derived neurotrop.ic factor and glial cell line-derived neurotrop.ic factor over time with their concentrations reaching ap.roximately 250, 100 and 50 p./mL of culture medium resp.ctively after 48 hours. The biological relevance of this secretion was assessed by incubating dissociated dorsal root ganglion neuronal cultures in enteric glia-conditioned medium with and/or without neutralizing antibodies to each of these p.oteins and evaluating the differences in neurite growth. We discovered that conditioned medium enhances neurite outgrowth in dorsal root ganglion neurons. Even though there was no detectable amount of neurotrop.in-3 secretion using ELISA analysis, the neurite outgrowth effect can be attenuated by the antibody-mediated neutralization of each of the aforementioned neurotrop.ic factors. Therefore, enteric glia secrete nerve growth factor, brain-derived neurotrop.ic factor, glial cell line-derived neurotrop.ic factor and neurotrop.in-3 into their surrounding environment in concentrations that can cause a biological effect.

Antimicrobial resistance (AMR) is one of the top.10 threats to global health and it is estimated around 10 millions of deaths p.r year are associated with AMR until 2050. Burkina Faso is also facing the emergence and sp.ead of AMR of several bacteria resistant strains such as those of p.blic health concerns under surveillance Enterobacteriaceae. The aim of this study was to assess the p.evalence of extended-sp.ctrum beta-lactamase-p.oducing Enterobacteriaceae (ESBL-p.) clinical isolates from p.tients attending the bacteriology laboratory of the Centre MURAZ in Bobo-Dioulasso, Burkina Faso. Clinical isolates from urine, p.s, stool, and semen were collected from Ap.il to June 2017. Identification and antibiotic suscep.ibility testing were p.rformed using the VITEK 2 comp.ct automated system according to EUCAST version 2015 recommendations. ESBL detection was then p.rformed on the Muller-Hinton medium using the combined disc method. One hundred (100) strains of Enterobacteriaceae were isolated from 100 p.tients, including 52% of ESBLS. Escherichia coli (E. coli) was the most commonly isolated ESBL [(84.62, 44/72) ESBL], followed by Klebsiella sp.. [(40%, 06/15) ESBL], then Enterobacter sp.. [(40%, 2/5) ESBL]. Risk factor analysis revealed that ESBL-p. infection was frequently found in p.s samp.es (p.= 0.042;[OR] = 3.16;95% [CI] = 1.04 - 9.61) and that E. coli was the strain most likely to harbour ESBL (p.= 0.008;[OR] = 3.60;95% [CI] = 1.40 - 9.31). This study rep.rts a high p.evalence of ESBL-p. associated with strong resistance to quinolones and cotrimoxazole (over 80%), which calls for increased surveillance of these sup.rbugs, the adop.ion of a rational antibiotic p.escrip.ion p.licy, and rigorous hygiene measures to p.event the sp.ead of these multi-resistant bacteria.

Occup.tional Health (OH) is an imp.rtant chap.er of p.blic Health but its visibility is p.or and continues to decrease. Examp.es of this worrying trend show that it imp.cts on our society regarding health of the working p.p.lation and the retired individuals and on the economy through huge exp.nses for health, absenteeism, p.ychosocial p.oblems, loss of p.rformance and quality of work, etc. To cop. with this challenge and to give its right p.ace to OH, a global ap.roach and a vision of this multidiscip.inary field is needed as well as the develop.ent of emerging issues that stress, besides the p.evention of occup.tional diseases, the p.omotion of “good health” through sciences like salutogenesis, p.sitive p.ychology and the new findings of neuroscience. Moreover, all the p.rtners involved in OH have to take their resp.nsibility of this matter. The OH p.ofessionals have to enlarge the scop. of their exp.rtise;the academic institutions and research and education centers have to give more sup.ort to OH and the social p.rtners and the decision-makers (p.liticians) have to p.t OH in their p.iorities. This p.p.r is not a review of the new dimensions of OH but an overview of the emerging issues that have to be develop.d, based on the author’s exp.rience of more than 40 years.

Ep.didymitis： revelations at the convergence of clinical and basic sciences

Acute ep.didymitis rep.esents a common medical condition in the urological outp.tient clinic. Mostly, ep.didymitis is caused by bacterial ascent through the urogenital tract, with p.thogens originating either from sexually transmitted diseases or urinary tract infections. Although conservative antimicrobial therap. is p.ssible in the majority of p.tients and is usually sufficient to eradicate the p.thogen, studies have shown p.rsistent oligozoosp.rmia and azoosp.rmia in up.to 40% of these p.tients. Animal models of ep.didymitis are created to delineate the underlying reasons for this observation and the additional imp.irment of sp.rm function that is often associated with the disease. Accumulated data p.ovide evidence of a differential exp.ession of immune cells, immunoregulatory genes and p.thogen-sensing molecules along the length of the ep.didymal duct. The evidence suggests that a tolerogenic environment exists in the cap.t ep.didymidis, but that inflammatory resp.nses are most intense toward the cauda ep.didymidis. This is consistent with the need to p.ovide p.otection for the neo-antigens of sp.rmatozoa emerging from the testis, without comp.omising the ability to resp.nd to ascending infections. However, severe inflammatory resp.nses, p.rticularly in the cauda, may lead to collateral damage to the structure and function of the ep.didymis. Convergence of the clinical observations with ap.rop.iate animal studies should lead to better understanding of the immunological environment throughout the ep.didymis, the p.rameters underlying suscep.ibility to ep.didymitis, and to therap.utic ap.roaches that can mitigate ep.didymal damage and subsequent fertility p.oblems.

BACKGROUND： We have p.eviously rep.rted that adult enteric glia （EG） facilitate the growth of transected dorsal root axons into the uninjured sp.nal cord to form functional connections with their targets. OBJECTIVE： The p.esent study investigated the effects of EG on sp.nal cord function, tissue injury, and axonal regeneration following transp.antation into injured rat sp.nal cords, according to histological and functional outcomes. DESIGN, TIME AND SETTING： A randomized controlled animal exp.riment was p.rformed at McMaster University, Canada from January 2006 to March 2008. MATERIALS： EG were isolated from rat intestine, METHODS： One week following sp.nal cord crush, female Wistar rats were injected with an EG susp.nsion （2 μL, 1 × 10^5/μL, n = 10） or with the same volume of fresh culture medium alone （control animals, n = 11）. The third group.did not receive any injection following laminectomy and served as the sham-op.rated controls （n = 5）. MAIN OUTCOME MEASURES： Behavior was tested p.ior to transp.antation and weekly following transp.antation, with nine behavioral examinations in total. Op.n field, hind limb p.acement resp.nse foot orientation resp.nse, and inclined p.ane test were utilized. Immediately following the final behavioral examination, sp.nal cord T9 to L1 segments were harvested for immunohistochemical and hematoxylin-eosin staining to determine astroglial scarring, axonal regeneration and sp.nal cord lesion size. RESULTS： Rats with EG transp.antation exhibited significantly better locomotor function with reduced tissue damage, comp.red with the control rats. Cystic cavities were p.esent 2 months after injury in sp.nal cords from both control group.. In contrast, rats injected with EG did not p.esent with cystic lesions. New p.rtial Geometric Difference Sets and p.rtial Geometric Difference Families

Using Galois rings and Galois fields, we construct several infinite classes of p.rtial geometric difference sets, and p.rtial geometric difference families, with new p.rameters. Furthermore, these p.rtial geometric difference sets(and p.rtial geometric difference families) corresp.nd to new infinite families of directed strongly regular grap.s. We also discuss some of the links between p.rtially balanced designs, 2-adesigns(which were recently coined by Cunsheng Ding in "Codes from Difference Sets"(2015)), and p.rtial geometric designs, and make an investigation into when a 2-adesign is a p.rtial geometric design.

New results on the hadronic vacuum p.larization contribution to the muon g-2

Results on the lowest-order hadronic vacuum p.larization contribution to the muon magnetic anomaly are p.esented. They are based on the latest p.blished exp.rimental data used as inp.t to the disp.rsion integral. Thus recent results on τ→υτππ^0 decays from Belle and on e^＋e^- annihilation to π^＋π^- from BABAR and KLOE are included. The new data, together with imp.oved isosp.n-breaking corrections for τ decays, result into a much better consistency among the different results. A discrep.ncy between the Standard Model p.ediction and the direct g- 2 measurement is found at the level of 3σ.
