A combined computational pipeline to detect circular RNAs in human cancer cells under hypoxic stress

Hypoxia is associated with several diseases,including *** that are deprived of adequate oxygen supply trigger tran-scriptional and post-transcriptional responses,which control cellular pathways such as angiogenesis,proliferation,and metabolic *** RNAs(circRNAs)are a novel class of mainly non-coding RNAs,which have been implicated in multiple cancers and attract increasing attention as potential ***,we characterize the circRNA signatures of three different cancer cell lines from cervical(HeLa),breast(MCF-7),and lung(A549)cancer under *** order to reliably detect circRNAs,we integrate available tools with custom approaches for quantification and statistical analysis?Using this consolidated computational pipeline,we identify?12000 circRNAs in the three cancer *** molecular characteristics point to an involvement of complementary RNA sequences as well as trans-acting factors in circRNA biogenesis,such as the RNA-binding protein HNRNPC-Notably,we detect a number of circRNAs that are more abundant than their linear *** addition,64 circRNAs significantly change in abundance upon hypoxia,in most cases in a cell type-specific *** summary,we present a comparative circRNA profiling in human cancer cell lines,which promises novel insights into the biogenesis and function of circRNAs under hypoxic stress.

Comparison of a new stent and Wallstent for transjugular intrahepatic portosystemic shunt in a porcine model

AIM To evaluate a new balloon-expandablestainless steel stent (Cordis stent ) in atransjugular intrahepatic portosystemic shunt(TIPS) porcine model and compared *** TIPS was performed in 26 normaldomestic pigs weighing 20 kg-30 kg using aCordis stent or Wallstent (13 pigs in each stent).All pigs were sacrificed at the 14th day afterTIPS. The stent deployment delivery system,stent patency, and stent recoil after placementwere evaluated. Proliferative response inrepresentative histological sections from thecenter, hepatic and portal regions of the twostent designs were *** The shunt was widely patent in 4pigs in the Cordis stent group (4/12, prematuredead in 1 pig), and in 5 pigs in the Wallstentgroup (5/13). All remaining stents of bothdesigns were occluded or stenotic. The barren stalk2 Gene Is Required for Axillary Meristem Development in Maize

The diversity of plant architecture is determined by axillary meristems (AMs). AMs are produced from small groups of stem cells in the axils of leaf primordia and generate vegetative branches and reproductive inflorescences . Previous studies identified genes critical for AM development that function in auxin biosynthesis, transport, and signaling. barren stalkl (ba1), a basic helix-loop-helix transcription factor, acts downstream of auxin to control AM formation. Here, we report the cloning and characterization of barren stalk2 (ba2), a mutant that fails to produce ears and has fewer branches and spikelets in the tassel, indicating that ba2 functions in reproductive AM development. Furthermore, the ba2 mutation suppresses tiller growth in the teosinte branchedl mutant, indicating that ba2 also plays an essential role in vegetative AM development. The ba2 gene encodes a protein that co-localizes and heterodimerizes with BA1 in the nucleus . Characterization of the genetic interaction between ba2 and ba1 demonstrates that ba1 shows a gene dosage effect in ba2 mutants, providing further evidence that BA1 and BA2 act together in the same pathway. Characterization of the molecular and genetic interaction between ba2 and additional genes required for the regulation of ba1 further supports this finding. The ba1 and ba2 genes are orthologs of rice genes, LAX PANICLE1 (LAX1) and LAX2, respectively, hence providing insights into pathways controlling AMs development in grasses.
