A novel triple immunoenzyme staining enables simultaneous identification of all muscle fiber types on a single skeletal muscle cryosection from normal, denervated or reinnervated rats

Triple immunofluorescence staining has recently been developed to simultaneously identify all muscle fibers on a single cryosection which is helpful for clinical and basic research, but it has disadvantages such as fast photobleaching and unclear outlines of muscle fibers. Triple immunoenzyme staining（TIE） is likely to avoid these disadvantages. In this study, we aimed to establish a sensitive and specific TIE technique to identify fiber types in normal, denervated, and reinnervated rat muscles, and to develop a systematic sampling method for muscle fiber quantification. Tibialis anterior and soleus from normal, denervated, and reinnervated Lewis rat hind limbs were used. Five consecutive cryosections were cut from each muscle, including one for TIE and four for single immunoenzyme staining（SIE）. The TIE was performed using the polymerized reporter enzyme staining system for the first two antigens（A4.74 for My HC-IIA, BA-F8 for My HC-I） and alkaline phosphatase staining system for the third antigen（BF-F3 for My HC-IIB）, followed by corresponding detective systems and respective chromogens. The type of muscle fibers was quantified by systematic sampling at 12.5%, 25%, 33% and 50% of all muscle fibers, and was compared with that acquired from counting all the fibers（100%）. All muscle fiber phenotypes, including pure and hybrid, could be simultaneously identified on a single TIE cryosection with clear outlines. The fiber types on TIE slides matched well with their respective counterpart on the consecutive SIE slides with a 95% match rate. Systematic sampling of 12.5% fibers could represent the true fiber type distribution of the entire muscle section. Our results suggest that novel TIE can effectively visualize fiber types in normal, denervated or reinnervated rat muscles.

DNA autoionization is a fundamental process wherein ultraviolet （UV）- photoexcited nucleobases dissipate energy by charge transfer to the environment without undergoing chemical damage. Here, single-wall carbon nanotubes （SWNT） are explored as a photoluminescent reporter for the study of the mechanism and rates of DNA autoionization. Two-color photoluminescence spectroscopy allows separate photoexcitation of the DNA and the SWNTs in the UV and visible range, respectively. A strong SWNT photoluminescence quenching is observed when the UV pump is resonant with the DNA absorption, consistent with charge transfer from the excited states of the DNA to the SWNT. Semiempirical calculations of the DNA-SWNT electronic structure, combined with a Green＇s function theory for charge transfer, show a 20 fs autoionization rate, dominated by hole transfer. Rate-equation analysis of the spectroscopy data confirms that the quenching rate is limited by thermalization of the free charge carriers transferred to the nanotube reservoir. This approach has great potential for monitoring DNA excitation, autoionization, and chemical damage, both in vivo and in vitro.

We characterize the agreement and disagreement of four publically available burned products(Fire CCI,Copernicus Burnt Area,MODIS MCD45A1,and MODIS MCD64A1)at a finer spatial and temporal scale than previous assessments using a grid of three-dimensional cells defined both in space and in *** analysis,conducted using seven years of data(2005–2011),shows that estimates of burned area vary greatly between products in terms of total area burned,the location of burning,and the timing of the *** use regional and monthly units for analysis to provide insight into the variation between products that can be lost when considering products yearly and/or *** with independent,contemporaneous MODIS active fire observations provides one indication of which products most reasonably capture the burning *** results have implications for the use of global burned area products in fire ecology,management and emissions applications.

背景本研究分别采用全血阻抗法与传统透光聚集度（LTA）测定血小板聚集率，以探讨抗血小板治疗药物是否会降低心脏手术或体外循环（CPB）后的血小板聚集功能。方法选择70例拟行择期冠状动脉旁路移植术的患者，分别于麻醉诱导前、诱导中和肝素化后15分钟及3小时采取动脉血，应用Multiplate（M）测定随时间变化的电阻值作为聚集单位，同时使用以胶原（COL）、二磷酸腺苷（ADP）或花生四烯酸（AA）为激活剂的LTA（％聚集率）法进行检测。A组（n=48）为对照组，手术前停用抗血小板药物至少7天，B组（n=11）和C组（n=11）患者分别服用阿司匹林100mg／d或阿司匹林100mg／d与氯吡格雷75mg／d（双重抗血小板治疗）至手术前一天。结果未行抗血小板治疗的患者，给予鱼精蛋白15分钟及3小时后，应用3种激活剂及两种聚集方法均可观察到血小板聚集率明显降低。单纯使用阿司匹林的患者，LTA—COL、LTA—ADP及M—ADP随时间发生显著改变；接受双重抗血小板治疗的患者，两种方法的ADP检测均提示给予鱼精蛋白15分钟后血小板聚集显著减弱。通过计算受试者工作特征曲线（receiver-operatingcharacteristiccurves，ROC曲线）下面积来区分抗血小板药物，LTA-COL可以在CPB后15分钟和3小时区分出对照组与阿司匹林组或双重抗血小板治疗组，M—ADP则可在鱼精蛋白拮抗后3小时区分对照组和双重抗血小板治疗组。结论对于未经抗血小板治疗的患者，全血电阻抗法及使用了常用激活剂的传统LTA法均可检测出CPB所致的血小板聚集功能改变。而对于接受了抗血小板治疗的患者，ADP激活的抗血小板检测方法能更好地检测出CPB所致的血小板聚集功能减退。

背景肝脏低灌注被认为是围手术期肝脏损害的一个重要病理生理因素。尽管硬膜外麻醉（EDA）应用广泛，但目前还没有关于阻滞平面仅局限在胸段的EDA对肝脏血流影响的相关数据。方法选择20例行全身麻醉的患者，通过经食道超声心动图（TEE）评估EDA前后肝右静脉和肝中静脉的肝血流指数。在T7～9间行硬膜外穿刺置管，推注１％的甲哌卡因，剂量中位数10ml（变化范围为8—16ml）。若EDA后，患者平均动脉压下降低于60mmHg，则持续输注去甲肾上腺素（NE）（***组）。在研究过程中，其他患者不用任何儿茶酚胺类药物（EDA组）。另外将10例未接受EDA的患者作为对照组。结果本研究中，有5例患者必须连续输注NE以避免平均动脉压降至60intoHg以下。因此，***组有5例患者，而EDA组有15例患者。在EDA组，肝脏两条静脉的血流指数下降中位数是24％（P〈0．01）。在EDA—NE组，所有5例患者的肝血流指数均下降，其中，肝右静脉下降的中位数为39（１１～45）％，而肝中静脉下降的中位数为32（7—49）％。对照纽患者肝血流指数保持恒定。与对照组相比，EDA组和EDA-NE组肝血流指数的减少差异具有显著性（Ｐ〈0．05）。相较于肝血流，心输出量不受EDA的影响。结论我们得出结论，胸段硬膜外阻滞与肝血流下降相关。胸段EDA复合连续静脉输注NE可能会导致肝血流进一步降低。

Human semen quality and the secondary sex ratio

The aim of this study was to evaluate the association between semen quality and the secondary sex ratio （SSR）, defined as the ratio of male to female live births. Our study cohort comprised 227 male partners who were enrolled prior to conception in Michigan and Texas between 2005 and 2009, and prospectively followed through delivery of a singleton birth. The male partners provided a baseline and a follow-up semen sample a month apart. Semen analysis was conducted to assess 27 parameters including five general characteristics, six sperm head measures, 14 morphology measures, and two sperm chromatin stability assay measures. Modified Poisson regression models with a robust error variance were used to estimate the relative risk （RR） and 95% confidence interval （95% CI） of a male birth for each semen parameter, after adjusting for potential confounders. Of the 27 semen parameters, only the percentage of bicephalic sperm was significantly associated with the SSR （2nd VS Ist quartile, RR, 0.65, 95% CI, 0.45-0.95, P= 0.03; 4TM vs 1St quartile, RR, 0.61, 95% CI, 0.38-1.00, P 〈 0.05 before rounding to two decimal places）, suggestive of a higher percentage of bicephalic sperm being associated with an excess of female births. Given the exploratory design of the present study, this preconception cohort study suggests no clear signal that human semen quality is associated with offspring sex determination.
