The rheological behavior of the semi-solid Ti B2 reinforced Al-Mg alloy comp.site slurry was investigated,which is required for the mould filling simulations during the semi-solid *** B2 reinforced in-situ Al-Mg alloy comp.site samp.es were remelted and subsequently brought to the semi-solid temp.rature regime within the heating chamber of a searle typ. *** order to understand the rheological behavior of comp.sites,three different typ.s of exp.riment were carried out,namely,steady state test,continuous cooling test and isothermal *** from that,the thixotrop.c nature of the slurry was confirmed from the obtained hysteresis loop. during the *** results indicate that when isothermally held,the comp.site slurry exhibits p.eudo-p.asticity and shows shear-thinning behavior up.to the shear rate of 1300 s^-1,and at higher shear rates（1300 s^-1）,it shows a shear thickening tendency,which is p.obably due to the agglomeration of non-deformable nano-Ti B2 *** p.eudo-p.astic behavior of the slurry was also estimated emp.oying intermittent step.changes of shear rate（shear jump.test）.

New monazite U\|p. geochronological data from the Everest region suggest that 20～25Ma elap.ed between the initial India—Asia collision and kyanite\|sillimanite grade metamorp.ism. Our results indicate a two\|p.ase metamorp.ic history, with p.ak Barrovian metamorp.ism at (32 2±0 4)Ma and a later high\|temp.rature, low\|p.essure event (620℃, 400Mp.) at (22 7±0 2)Ma.. Emp.acement and crystallization of the Everest granite subsequently occurred at 20 5～21 3Ma. The monazite crystallization ages that differ by 10Ma are recorded in two structurally adjacent rocks of different lithology, which have the same p.st collisional p.T history.. Scanning electron microscop. reveals that the younger monazite is elaborately shap.d and grew in close association with ap.tite at grain boundaries and trip.e junctions, suggesting that growth was stimulated by a change in the fluid regime. The older monazite is euhedral, is not associated with ap.tite, and is commonly armoured within silicate minerals. During the low\|p.essure metamorp.ic event, the armouring p.otected the older monazites, and a lack of excess ap.tite in this samp.e p.evented new growth. Textural relationship. suggest that ap.tite is one of the necessary monazite\|p.oducing reactants, and sp.ts within monazite that are rich in Ca, Fe, Al and Si suggest that allanite acted as a p.eexisting rare earth element host. We p.op.se a simp.ified reaction for monazite crystallization based on this evidence.

p.ogranulin(p.RN)has recently emerged as a key p.ayer in a subset of frontotemp.ral dementias(FTD).Numerous mutations in the p.ogranulin gene have been identified in p.tients with familial or sp.radic frontotemp.ral lobar degeneration(FTLD).In order to understand the molecular mechanisms by which p.RN deficiency leads to FTLD,we examined activity of p.RN in mouse cortical and hip.ocamp.l neurons and in human neuroblastoma SH-SY5Y *** of mouse neurons with p.RN p.otein resulted in an increase in neurite outgrowth,sup.orting the role of p.RN as a neurotrop.ic *** treatment stimulated p.osp.orylation of glycogen synthase kinase-3 beta(GSK-3β)in cultured *** of p.RN in SH-SY5Y cells imp.ired retinoic acid induced differentiation and reduced the level of p.osp.orylated GSK-3β.p.RN knockdown cells were also more sensitized to staurosp.rineinduced *** results reveal an imp.rtant role of p.RN in neurite outgrowth and involvement of GSK-3βin mediating p.RN *** of GSK-3βactivation as a downstream event for p.RN signaling p.ovides a mechanistic exp.anation for p.RN activity in the nervous *** work also suggest that loss of axonal growth stimulation during neural injury rep.ir or deficits in axonal rep.ir may contribute to neuronal damage or axonal loss in FTLD associated with p.RN ***,our study suggests that modulating GSK-3βor similar signaling events may p.ovide therap.utic benefits for FTLD cases associated with p.RN mutations.

Mesenchymal stem cells(MSCs)are ubiquitously-existing multip.tent p.ogenitors that can self-renew and differentiate into multip.e lineages including osteocytes,chondrocytes,adip.cytes,tenocytes and *** rep.esent one of the most commonly-used adult p.ogenitors and serve as excellent p.ogenitor cell models for investigating lineagesp.cific differentiation regulated by various cellular signaling p.thways,such as bone morp.ogenetic p.oteins(BMp.).As members of TGFb sup.rfamily,BMp. p.ay diverse and imp.rtant roles in develop.ent and adult *** least 14 BMp. have been identified in *** BMp. exert distinct but overlap.ing biological *** a comp.ehensive analysis of 14 BMp. in MSCs,we demonstrated that BMp. is one of the most p.tent BMp. in inducing osteogenic differentiation of ***,a global mechanistic view of BMp.signaling in regulating the p.oliferation and differentiation of MSCs remains to be fully ***,we conducted a comp.ehensive transcrip.omic p.ofiling in the MSCs stimulated by 14 typ.s of *** clustering analysis classifies 14 BMp. into three subclusters:an osteo/chondrogenic/adip.genic cluster,a tenogenic cluster,and BMp. *** also demonstrate that six BMp.(e.g.,BMp.,BMp.,BMp.,BMp.,BMp.,and BMp.)can induce ISmads effectively,while BMp.,BMp.,BMp.,BMp.,and BMp.1 up.regulate Smad-indep.ndent MAp.kinase ***,we show that many BMp. can up.egulate the exp.ession of the signal mediators of Wnt,Notch and p.3K/AKT/mTOR *** the rep.rted transcrip.omic changes need to be further validated,our exp.ession p.ofiling rep.esents the first-of-its-kind to interrogate a comp.ehensive transcrip.omic landscap. regulated by the 14 typ.s of BMp. in MSCs.

As multip.tent p.ogenitor cells,mesenchymal stem cells(MSCs)can renew themselves and give rise to multip.e lineages including osteoblastic,chondrogenic and adip.genic ***’s p.eviously shown that BMp. is the most p.tent BMp.and induces osteogenic and adip.genic differentiation of ***,the molecular mechanism through which BMp. regulates MSC differentiation remains p.orly *** evidence indicates that noncoding RNAs,esp.cially microRNAs,may p.ay imp.rtant roles in regulating MSC differentiation and bone *** highly conserved RNA binding p.oteins,Argonaute(AGO)p.oteins are essential comp.nents of the multi-p.otein RNA-induced silencing comp.exes(RISCs),which are critical for small RNA ***,we investigate p.ssible roles of AGO p.oteins in BMp.-induced lineage-sp.cific differentiation of *** first found that BMp. up.egulated the exp.ession of Ago1,Ago2 and Ago3 in *** engineering multip.ex siRNA vectors that exp.ess multip.e siRNAs targeting individual Ago genes or all four Ago genes,we found that silencing individual Ago exp.ession led to a decrease in BMp.-induced early osteogenic marker alkaline p.osp.atase(ALp.activity in ***,we demonstrated that simultaneously silencing all four Ago genes significantly diminished BMp.-induced osteogenic and adip.genic differentiation of MSCs and matrix mineralization,and ectop.c bone ***,our findings strongly indicate that AGO p.oteins and associated small RNA biogenesis p.thway p.ay an essential role in mediating BMp.-induced osteogenic differentiation of MSCs.

Bioactivity-guided p.ytochemical investigation of p.docarp.s neriifolius ***.(p.docarp.ceae)has led to the isolation of one new(2)and three known(1,3,and 4)B-typ. p.dolactones,along with three totarane-typ. diterp.nes(5-7).Their structures were determined by interp.etation of High Resolution ElectroSp.ay Ionization Mass Sp.ctrometry(HRESIMS)and 1D and 2D NMR data,and comp.rison with the values rep.rted in the *** structure of comp.und 1,p.eviously identifed as 3-deoxy-2α-hydroxynagilactone E(8),was revised as its 2β-ep.mer,which has been rep.rted recently as a new *** of the isolates were evaluated for their antip.oliferative activity against a p.nel of four human cancer cell lines,namely,ovarian(OVCAR3),breast(MDA-MB-231),colon(HT-29),and melanoma(MDA-MB-435),and comp.unds 1 and 3 were found to be cytotoxic with IC_(50) values in the low micromolar range for most of the cell lines *** major comp.und,inumakilactone A(3),was further tested in vivo using the HT-29,MDA-MB-435,and OVCAR3 cells in a murine hollow fber model,for the frst time.
