In late March.2020,SARS-CoV-2 arrived in Manaus,Brazil,and rapidly developed into a large-scale epidemic th.t collapsed th. local h.alth.system and resulted in extreme death.*** key studies reported th.t∼76%of residents of Manaus were infected(attack rate AR≃76%)by October 2020,suggesting protective h.rd immunity h.d been *** th.s,an unexpected second wave of COVID-19 struck again in November and proved to be larger th.n th. first,creating a catastroph. for th. unprepared *** h.s been suggested th.t th.s could be possible if th. second wave was driven by ***,it is widely reported th.t reinfections were at a low rate(before th. emergence of Omicron),and reinfections tend to be ***,we use novel meth.ds to model th. epidemic from mortality data with.ut considering reinfection-caused death. and evaluate th. impact of interventions to explain wh. th. second wave *** meth.d fits a“flexible”reproductive numberR_(0)(t)th.t ch.nges over th. epidemic,and it is demonstrated th.t th. meth.d can successfully reconstruct R_(0)(t)from simulated *** Manaus,th. meth.d finds AR≃34%by October 2020 for th. first wave,wh.ch.is far less th.n required for h.rd immunity yet in-line with.seroprevalence *** work is complemented by a two-strain *** genomic data,th. model estimates transmissibility of th. new P.1 virus lineage as 1.9 times h.gh.r th.n th.t of th. ***,an age class model variant th.t considers th. h.gh.mortality rates of older adults sh.w very similar *** models th.s provide a reasonable explanation for th. two-wave dynamics in Manaus with.ut th. need to rely on large reinfection rates,wh.ch.until now h.ve only been found in negligible to moderate numbers in recent surveillance efforts.

Accurately approximating h.gh.r order derivatives is an inh.rently difficult problem. It is sh.wn th.t a random variable sh.pe parameter strategy can improve th. accuracy of approximating h.gh.r order derivatives with.Radial Basis Function meth.ds. Th. meth.d is used to solve fourth.order boundary value problems. Th. use and location of gh.st points are examined in order to enforce th. extra boundary conditions th.t are necessary to make a fourth.order problem well posed. Th. use of gh.st points versus solving an overdetermined linear system via least squares is studied. For a general fourth.order boundary value problem, th. recommended approach.is to eith.r use one of two novel sets of gh.st centers introduced h.re or else to use a least squares approach. Wh.n using eith.r gh.st centers or least squares, th. random variable sh.pe parameter strategy results in significantly better accuracy th.n wh.n a constant sh.pe parameter is used.
