Engineering microscale systems for fully autonomous intracellular neural interfaces

Conventional electrodes and associated positioning systems for intracellular recording from single neurons in vitro and in vivo are large and bulky,which has largely limited their scalability.Further,acquiring successful intracellular recordings is very tedious,requiring a high degree of skill not readily achieved in a typical laboratory.We report here a robotic,MEMs-based intracellular recording system to overcome the above limitations associated with form factor,scalability,and highly skilled and tedious manual operations required for intracellular recordings.This system combines three distinct technologies:(1)novel microscale,glass–polysilicon penetrating electrode for intracellular recording;(2)electrothermal microactuators for precise microscale movement of each electrode;and(3)closed-loop control algorithm for autonomous positioning of electrode inside single neurons.Here we demonstrate the novel,fully integrated system of glass–polysilicon microelectrode,microscale actuators,and controller for autonomous intracellular recordings from single neurons in the abdominal ganglion of Aplysia californica(n=5 cells).Consistent resting potentials(s(>60 mV)were recorded after each successful penetration attempt with the controller and microactuated glass–polysilicon microelectrodes.The success rate of penetration and quality of intracellular recordings achieved using electrothermal microactuators were comparable to that of conventional positioning systems.Preliminary data from in vivo experiments in anesthetized rats show successful intracellular recordings.The MEMs-based system offers significant advantages:(1)reduction in overall size for potential use in behaving animals,(2)scalable approach to potentially realize multi-channel recordings,and(3)a viable method to fully automate measurement of intracellular recordings.This system will be evaluated in vivo in future rodent studies.

Loss to specialized Cardiology Follow-Up in Adults Living with Congenital Heart Disease

Background:Much has been written about the loss to follow-up in the transition between pediatric and adult Congenital Heart Disease(CHD)care centers.Much less is understood about the loss to follow-up(LTF)after a successful transition.This is critical too,as patients lost to specialised care are more likely to experience mor-bidity and premature mortality.Aims:To understand the prevalence and reasons for loss to follow-up(LTF)at a large Australian Adult Congenital Heart Disease(ACHD)centre.Methods:Patients with moderate or highly complex CHD and gaps in care of>3 years(deﬁned as LTF)were identiﬁed from a comprehensive ACHD data-base.structured telephone interviews examined current care and barriers to clinic attendance.Results:Overall,407(22%)of ACHD patients(n=1842)were LTF.The mean age at LTF was 31(sD 11.5)years and 54%were male;311(76%)were uncontactable.Compared to adults seen regularly,lost patients were younger,with a greater socio-economic disadvantage,and had less complex CHD(ps(14%).The top 3 responses for care absences were“feeling well”(61%),losing track of time(36%),and not needing fol-low-up care(25%).Conclusions:A large proportion of the ACHD population becomes lost to specialised cardiac care,even after a successful transition.This Australian study reports younger age,moderate complexity defects,and socio-economic disadvantage as predictive of loss to follow-up.This study highlights the need for novel approaches to patient-centered service delivery even beyond the age of transition and resources to maintain patient engagement within the ACHD service.

Background:The mobilization and redistribution of severe acute respiratory syndrome coronavirus 2(sARs-CoV-2)specific T-cells and neutralizing antibodies(nAbs)during exercise is purported to increase immune surveillance and protect against severe coronavirus disease 2019(COVID-19).We sought to determine if COVID-19 vaccination would elicit exercise-responsive sARs-CoV-2 T-cells and transiently alter nAb titers.Methods:Eighteen healthy participants completed a 20-min bout of graded cycling exercise before and/or after receiving a COVID-19 vaccine.All major leukocyte subtypes were enumerated before,during,and after exercise by flow cytometry,and immune responses to sARs-CoV-2 were determined using whole blood peptide stimulation assays,T-cell receptor(TCR)-βsequencing,and sARs-CoV-2 nAb serology.Results:COVID-19 vaccination had no effect on the mobilization or egress of major leukocyte subsets in response to intensity-controlled graded exercise.However,non-infected participants had a significantly reduced mobilization of CD4+and CD8+naive T-cells,as well as CD4+central memory T-cells,after vaccination(synthetic immunity group);this was not seen after vaccination in those with prior sARs-CoV-2 infection(hybrid immunity group).Acute exercise after vaccination robustly mobilized sARs-CoV-2 specific T-cells to blood in an intensity-dependent manner.Both groups mobilized T-cells that reacted to spike protein;however,only the hybrid immunity group mobilized T-cells that reacted to membrane and nucleocapsid antigens.nAbs increased significantly during exercise only in the hybrid immunity group.Conclusion:These data indicate that acute exercise mobilizes sARs-CoV-2 specific T-cells that recognize spike protein and increases the redistribution of nAbs in individuals with hybrid immunity.

Cu-Based Materials for Enhanced C_(2+) Product selectivity in Photo-/Electro-Catalytic CO_(2) Reduction: Challenges and Prospects

Carbon dioxide conversion into valuable products using photocatalysis and electrocatalysis is an effective approach to mitigate global environmental issues and the energy shortages. Among the materials utilized for catalytic reduction of CO_(2), Cu-based materials are highly advantageous owing to their widespread availability, cost-effectiveness, and environmental sustainability. Furthermore, Cu-based materials demonstrate interesting abilities in the adsorption and activation of carbon dioxide, allowing the formation of C_(2+) compounds through C–C coupling process. Herein, the basic principles of photocatalytic CO_(2) reduction reactions(PCO_(2)RR) and electrocatalytic CO_(2) reduction reaction(ECO_(2)RR) and the pathways for the generation C_(2+) products are introduced. This review categorizes Cu-based materials into different groups including Cu metal, Cu oxides, Cu alloys, and Cu sACs, Cu heterojunctions based on their catalytic applications. The relationship between the Cu surfaces and their efficiency in both PCO_(2)RR and ECO_(2)RR is emphasized. Through a review of recent studies on PCO_(2)RR and ECO_(2)RR using Cu-based catalysts, the focus is on understanding the underlying reasons for the enhanced selectivity toward C_(2+) products. Finally, the opportunities and challenges associated with Cu-based materials in the CO_(2) catalytic reduction applications are presented, along with research directions that can guide for the design of highly active and selective Cu-based materials for CO_(2) reduction processes in the future.

Objective Hypertriglyceridemic waist(HW),hypertriglyceridemic waist-to-height ratio(HWHtR),and waist-to-hip ratio(WHR)have been shown to be indicators of cardiometabolic risk factors.However,it is not clear which indicator is more suitable for children and adolescents.We aimed to investigate the relationship between HW,HWHtR,WHR,and cardiovascular risk factors clustering to determine the best screening tools for cardiometabolic risk in children and adolescents.Methods This was a national cross-sectional study.Anthropometric and biochemical variables were assessed in approximately 70,000 participants aged 6–18 years from seven provinces in China.Demographics,physical activity,dietary intake,and family history of chronic diseases were obtained through questionnaires.ANOVA,χ2 and logistic regression analysis was conducted.Results A significant sex difference was observed for HWHtR and WHR,but not for HW phenotype.The risk of cardiometabolic health risk factor clustering with HW phenotype or the HWHtR phenotype was significantly higher than that with the non-HW or non-HWHtR phenotypes among children and adolescents(HW:OR=12.22,95%CI:9.54-15.67;HWHtR:OR=9.70,95%CI:6.93-13.58).Compared with the HW and HWHtR phenotypes,the association between risk of cardiometabolic health risk factors(CHRF)clustering and high WHR was much weaker and not significant(WHR:OR=1.14,95%CI:0.97-1.34).Conclusion Compared with HWHtR and WHR,the HW phenotype is a more convenient indicator with higher applicability to screen children and adolescents for cardiovascular risk factors.

Melt extrusion-based additive manufacturing(ME-AM)is a promising technique to fabricate porous scaffolds for tissue engi-neering applications.However,most synthetic semicrystalline polymers do not possess the intrinsic biological activity required to control cell fate.Grafting of biomolecules on polymeric surfaces of AM scaffolds enhances the bioactivity of a construct;however,there are limited strategies available to control the surface density.Here,we report a strategy to tune the surface density of bioactive groups by blending a low molecular weight poly(ε-caprolactone)5k(PCL5k)containing orthogonally reactive azide groups with an unfunctionalized high molecular weight PCL75k at different ratios.stable porous three-dimensional(3D)scaf-folds were then fabricated using a high weight percentage(75 wt.%)of the low molecular weight PCL 5k.As a proof-of-concept test,we prepared films of three different mass ratios of low and high molecular weight polymers with a thermopress and reacted with an alkynated fluorescent model compound on the surface,yielding a density of 201-561 pmol/cm^(2).subsequently,a bone morphogenetic protein 2(BMP-2)-derived peptide was grafted onto the films comprising different blend compositions,and the effect of peptide surface density on the osteogenic differentiation of human mesenchymal stromal cells(hMsCs)was assessed.After two weeks of culturing in a basic medium,cells expressed higher levels of BMP receptor II(BMPRII)on films with the conjugated peptide.In addition,we found that alkaline phosphatase activity was only significantly enhanced on films contain-ing the highest peptide density(i.e.,561 pmol/cm^(2)),indicating the importance of the surface density.Taken together,these results emphasize that the density of surface peptides on cell differentiation must be considered at the cell-material interface.Moreover,we have presented a viable strategy for ME-AM community that desires to tune the bulk and surface functionality via blending of(modified)polymers.Furthermore,the use of alkyne-a

We examined the transverse momentum(pT)spectra of various identified particles,encompassing both light-flavored and strange hadrons(π++π−,K++K−,p+p¯,ϕ,K0s,Λ+Λ¯,Ξ−+Ξ¯+,andΩ−+Ω¯+),across different multiplicity classes in proton-proton collisions(p-p)at a center-of-mass energy of s√=7 TeV.Utilizing the Tsallis and Hagedorn models,parameters relevant to the bulk properties of nuclear matter were extracted.Both models exhibit good agreement with experimental data.In our analyses,we observed a consistent decrease in the effective temperature(T)for the Tsallis model and the kinetic or thermal freeze-out temperature(T0)for the Hagedorn model,as we transitioned from higher multiplicity(class-I)to lower multiplicity(class-X).This trend is attributed to the diminished energy transfer in higher multiplicity classes.Additionally,we observed that the transverse flow velocity(βT)experiences a decline from class-I to class-X.The normalization constant,which represents the multiplicity of produced particles,was observed to decrease as we moved toward higher multiplicity classes.While the effective and kinetic freeze-out temperatures,as well as the transverse flow velocity,show a mild dependency on multiplicity for lighter particles,this dependency becomes more pronounced for heavier particles.The multiplicity parameter for heavier particles was observed to be smaller than that of lighter particles,indicating a greater abundance of lighter hadrons compared to heavier ones.Various particle species were observed to undergo decoupling from the fireball at distinct temperatures:lighter particles exhibit lower temperatures,while heavier ones show higher temperatures,thereby supporting the concept of multiple freeze-out scenarios.Moreover,we identified a positive correlation between the kinetic freeze-out temperature and transverse flow velocity,a scenario where particles experience stronger collective motion at a higher freeze-out temperature.The reason for this positive correlation is that,as the multiplicity increases,more energy is

Relationship between abdominal adiposity, cardiovascular fitness, and biomarkers of cardiovascular risk in British adolescents

Background:Puberty is a critical time in the development of overweight and obesity.The aim of this study was to examine relationships between measures of adiposity,cardiovascular fitness,and biomarkers of cardiovascular disease risk in adolescents.Methods:In a cross-sectional study design,129 girls and 95 boys aged 12.9-14.4 years at various stages of puberty were included,along with their mothers(n=217)and fathers(n=207).Anthropometric assessments of adiposity were made,along with cardiovascular physical fitness,using the 20-m shuttle run test,and biomarkers associated with cardiovascular risk,including glucose,insulin,triglyceride,fibrinogen,and C-reactive protein(CRP)concentrations.Results:Waist-to-height ratio values were similar in boys and girls and correlated positively with diastolic blood pressure,insulin,triglyceride,fibrinogen,and CRP concentrations,and inversely with cardiovascular fitness scores.skinfold thickness measurements were higher in girls.High-molecular-weight adiponectin concentrations were lower in boys than girls,particularly in late puberty,and CRP levels were higher.Cardiovascular fitness,maternal body mass index(BMI),and paternal BMI contributed independently to the variance in waist measurements in girls and boys.Gender,triceps skinfold thickness,and weight-to-height ratio,but not parental BMI,contributed independently to the variance in cardiovascular fitness.Conclusion:There is a relationship between measures of adolescent adiposity and parental weight that involves factors other than cardiovascular fitness.Adolescent boys have relatively more abdominal fat than girls and a tendency to have a proinflammatory profile of biomarkers.These observations suggest that family and social environmental interventions are best undertaken earlier in childhood,particularly among boys.

Nicotiana tabacum and Nicotiana benthamiana are widely used models in plant biology research.However,genomic studies of these species have lagged.Here we report the chromosome-level reference genome assemblies for N.benthamiana and N.tabacum with an estimated 99.5%and 99.8%completeness,respec-tively.sensitive transcription start and termination site sequencing methods were developed and used for accurate gene annotation in N.tabacum.Comparative analyses revealed evidence for the parental origins and chromosome structural changes,leading to hybrid genome formation of each species.Interestingly,theantiviral silencinggenesRDR1,RDR6,DCL2,DCL3,andAGO2were lost from one or both subgenomes in N.benthamiana,while both homeologs were kept in N.tabacum.Furthermore,the N.benthamiana genome encodes fewer immune receptors and signaling components than that of N.tabacum.These find-ings uncover possible reasons underlying the hypersusceptible nature of N.benthamiana.We developed the user-friendly Nicomics(http:/***/Nicomics/)web server to facilitate better use of Nicotiana genomic resources as well as gene structure and expression analyses.

Introduction:Older adults are prone to high levels of depression due to their deteriorating physical functions and shrinking social networks after retirement.Volunteering as an important social activity is essential for alleviating depression by building social network.This paper aims to examine the effect of volunteering on depression among older adults by using China Longitudinal Aging social survey(CLAss 2018)data.Methods:This study uses descriptive analysis and chi-square tests to show differences in demographic factors of older adults’volun-teerism participation,followed by bivariate correlation analysis to examine the correlation between the vital vari-ables.Afterward,stratified linear regression analysis is used to research the significant level and impact between volunteering and degree of expertise,frequency,and variety of participation.Results:8,459 older adults are included in study.The research reveals that older adults who are younger,live in urban areas,are married,or have a higher degree of education tend to have fewer depressive symptoms.Meanwhile,participation in volun-teering(OR=0.90,95%CI:0.8,1.1,ps well as that demands specialized skills(OR=0.51,95%CI:0.30,0.2,ps(OR=0.21,95%CI:0.12,0.29,psitive inﬂuence on depression levels.Discussion/Conclusion:Older adults who participate in voluntary services have lower depression symptoms and should be encouraged to use their professional skills and increase participation frequency and variety in this process.This article suggests that governments should help older adults participate in voluntary services by time bank which will further strengthen social ties,rebuild social networks and alleviate depression symptoms of older adults.
