Objective： This study com.ared the perform.nce of the im.unochrom.tographic strip （ICS） to the Venereal Disease Research Laboratory （VDRL） test and Treponem. pallidum.haem.gglutination assay （TPHA） at a prim.ry health care setting. m.thods： The study group was com.rised of 150 fem.les random.y drawn from.a population of pregnant wom.n attending their first antenatal visit or follow-up visits at West m.ternity Hospital in Eldoret Kenya, but without a previous syphilis test during that pregnancy. On-site VDRL, ICS and TPHA tests were perform.d and im.ediate treatm.nt provided where appropriate. The perform.nce of the three tests was com.ared, Results： The sero-prevalence of syphilis as determ.ned by the VDRL test was 3%. There was no significant difference between the ICS and the VDRL test （P 〉 0.05）. The sensitivity and specificity of the ICS test were 80% and 98.6% respectively, while the negative predictive value （NPV） and positive predictive value （PPV） were both 100%. On the other hand, the sensitivity and specificity of the VDRL test were 66.7% and 99.3%, while the NPV and PPV were 80% and 98.6% respectively. The Treponem. pallidum.haem.gglutination assay was used as a reference test and had sensitivity, specificity, NPV and PPV of 100%. Conclusion： The diagnostic accuracy of the ICS com.ared favorably with theVDRL gold standard. The use of the ICS in Kenya can im.rove the diagnosis of syphilis in health facilities both with and without laboratories and allow com.unity health care workers to m.ke a rapid diagnosis of the disease, and consequently m.ke im.ediate therapeutic decisions.

Objective： To determ.ne the adjuvant potential of artem.sinin with a soluble leishm.nial antigen in vaccinating BALB/c m.ce. m.thods： Seventy two fem.le BALB/c m.ce were random.y assigned into six groups. The m.ce were vaccinated with soluble Leishm.nia antigens （SLA） alone, artem.sinin co-adm.nistered with SLA, SLA and Bacille Calm.tte Gu fin （BCG） vaccine, and artem.sinin and BCG alone. Unvaccinated m.ce form.d the control group. The induction of cell-m.diated im.unity following vaccination was determ.ned by m.asuring in vitro lym.hocyte proliferation and the production of interleukin （IL）-4, IL-5 and gam.a interferon （IFN-γ） determ.ned by flow cytom.try. Protection against L. m.jor was determ.ned by quantifying parasite burdens in L. m.jor infected footpads using a lim.ting dilution assay and by m.asuring lesion sizes of the infected footpad com.ared to the contralateral uninfected footpad. Results： m.ce receiving SLA plus artem.sinin produced significantly high levels of IL-4 and IL-5 （P 〈 0.05） and low levels of IFN-γ, resulting in exacerbated disease. In addition, subcutaneous adm.nistration of SLA ＋ artem.sinin, artem.sinin alone or SLA alone resulted in the developm.nt of large footpad swellings and high parasite loads that were com.arable to those of the control unvaccinated m.ce （P 〉 0.05）, resulting in exacerbated disease. Conclusion： These data suggest that artem.sinin is not a suitable adjuvant for Leishm.nia vaccines. However, since artem.sinin has been shown to be effective against Leishm.nia parasites in vitro and in vivo, further studies ought to be conducted to determ.ne its im.unochem.therapeutic potential when co-adm.nistered with Leishm.nia antigens.

Over the last decade, there has been a flurry of research on adjuvants for vaccines, and several novel adjuvants are now licensed products or in late stage clinical developm.nt. The success of adjuvants in enhancing the im.une response to antigens has led m.ny researchers to re-focus their vaccine developm.nt program.. Although several vaccine candidates have been tested against leishm.niasis, there is yet no effective vaccine against this parasitic disease. Recent research has docum.nted that efforts to develop effective Leishm.nia vaccine have been lim.ted due to lack of an appropriate adjuvant. In view of this, this review paper outlines som. of the adjuvants that have been used in Leishm.nia vaccine candidates and cites a few of the responses obtained from.these studies. The aim.of the present review is to consolidate these findings to facilitate the application of these adjuvants in general and experim.ntal vaccinology.

In vitro activity and in vivo efficacy of a com.ination therapy of dim.nazene and chloroquine against m.rine visceral leishm.niasis

The present study evaluated the in vitro activity and in vivo efficacy of dim.nazene com.ined with chloroquine as a potential drug against Leishm.nia donovani. Am.hotericin B was used as a positive control drug. In vitro activity involved incubation of various drug concentrations with prom.stigotes or vero cells in culture before determ.nation of parasite growth inhibition or cell death while in vivo evaluations involved infection of various m.ce groups with virulent L. donovani parasites and treatm.nt with test drug com.ounds following disease establishm.nt. Weight changes in experim.ntal m.ce were also evaluated before infection and throughout the experim.nt. The results indicated that the dim.nazene-chloroquine com.ination was at least nine tim.s m.re efficacious than individual drugs in killing prom.stigotes in culture. The dim.nazene-chloroquine com.ination was safer （Ld50=0.03±0.04） than Am.hotericin B （Ld50=0.02±0.01）. Body weight in infected m.ce increased significantly （P=0.0007） from.day 7 to day 37 following infection （P=0.026）. However, body weight rem.ined com.arable in all m.ce groups during treatm.nt （P=0.16）. The dim.nazene-chloroquine com.ination significantly reduced splenic parasite num.ers as com.ared to individual drug therapies （P=0.0001） although Am.hotericin B was still m.re efficacious than any other treatm.nt （P=0.0001）. Am.ngst the test com.ounds, the dim.nazene-chloroquine com.ination showed the lowest level of IgG antibody responses with results indicating significant negative correlation between antileishm.nial antibody responses and protection against disease. These findings dem.nstrate the positive advantage and the potential use of a com.ined therapy of dim.nazene-chloroquine over the constituent drugs. Further evaluation is recom.ended to determ.ne the m.st efficacious com.ination ratio of the two com.ounds.

Leishm.niasis is a zoonotic disease caused by protozoan parasites of the genus Leishm.nia.Conventional chem.therapy rem.ins to be the m.st preferred m.asure against leishm.niasis despite being associated with high toxicity and relapse rates.They are also expensive and require hospitalization.Plant-based com.ounds provide a better treatm.nt alternative because they are effective,cheap,and less associated with toxicity and resistance.This study exam.ned the therapeutic potential of Warburgia ugandensis,Prunus africana,and Piliostigm. thonningii against Leishm.nia donovani infection in BALB/c m.ce.Anti-prom.stigote and toxicity studies were evaluated by incubating the test com.ound with prom.stigotes and Vero cells,respectively.Serum.was obtained from.the m.ce for total im.unoglobulin gam.a(IgG)quantification.For in vivo studies,the m.ce were infected with virulent Leishm.nia donovani then treated with m.thanolic extracts of Warburgia ugandensis,Prunus africana,and Piliostigm. thonningii and control drug,pentostam.sodium.stibogluconate).Treatm.nt with the plant extracts and standard drug resulted to significant reduction in parasite burden.Outcom.s in the m.ce treated with plant extracts were com.parable to those treated with pentostam.P≥0.05).In the prom.stigote assay,all the test com.ounds killed m.re than half of the prom.stigotes at the highest concentration(500μg/m.).Warburgia ugandensis,P.thonningii,and P.africana reduced the num.er of prom.stigotes from.2.0×10^(6) to 7.7×10^(3),72.0×10^(3),and 5.0×10^(3),respectively.Pentostam.had the lowest IC50(210μg/m.),followed by Warburgia ugandensis(IC50 of 270μg/m.).Piliostigm. thonningii and P.africana were less toxic with IC50 of 720μg/m. and 500μg/m.,respectively.There was low production of IgG antibodies following treatm.nt with the plant extracts and high levels in the untreated control.

Com.arison of trigem.nal lateralization with differing stim.lants

Trigem.nal-specific stim.lants have been shown to activate different receptors preferentially and this likely accounts for variation in sensory perception.It is unclear whether trigem.nal sensitivity is sim.lar across different transient receptor potential(TRP)receptors or if dysfunction of different receptors results in differing patient sym.tom..Therefore,a prospective cohort study was conducted,consisting of trigem.nal lateralization testing with three different stim.lants(eucalyptol,isothiocyanate,acetic acid),olfaction testing with Sniffin'Sticks,and m.asurem.nt of various patient‐reported outcom. m.asures(PROm.).A total of 50 participants were enrolled across the olfactory spectrum.m.an TDI score was 27.1±8.3(range 7.0–39.5)with 38%norm.sm.c and 62%dysosm.c.m.an trigem.nal lateralization scores out of 20 in the overall cohort were 16.18(2.78)for eucalyptol,14.94(3.49)for m.stard oil,and 15.28(3.68)for vinegar.Eucalyptol showed a significant correlation with threshold scores of Sniffin'Sticks.A significant correlation was found between acetic acid and various PROm..None of the lateralization scores of the trigem.nal stim.lants correlated to each other significantly and there was no correlation to age.The lack of correlation suggests that the m.asured sensitivity of one type of TRP receptor m.y not translate to sim.lar sensitivity of the other receptors.Additional investigations with TRPV1 and TRPA1 agonists are needed to corroborate our findings.

Physical activity(PA)epidem.ology first em.rged as a new field of study around the 1950s with publication of sem.nal investigations on London civil servants1 and has continued to evolve as a scientific discipline to present tim..This subdiscipline of epidem.ology studies the frequencies,distributions,and dynam.cs of PA and its im.act on hum.n health,m.rbidity,and m.rtality.Research over the past 75 years has dem.nstrated num.rous health benefits of PA.Based on these research findings,the World Health Organization recom.ends adults aged 18-64 years engage in at least 150-300 m.n of m.derate-intensity aerobic PA per week,or at least 75-150 m.n of vigorous-intensity aerobic PA per week,or a com.ination of both intensities.

Fabrication of sub-20 nm.m.S_(2) horizontal nanowire on silicon substrates by inclusion of precursors into polystyrene-bpolyethylene oxide nanopatterns: Detailed structural investigation

Herein,we dem.nstrate the fabrication of sub-20 nm.m.S_(2) horizontal nanowire arrays on silicon substrates using a self-assem.led block copolym.r assisted in situ inclusion approach.m.crophase separated long-range ordered polystyrene-b-polyethylene oxide(PS-b-PEO)block copolym.r(BCP)line-space nanopatterns were achieved through therm.-solvent annealing.The patterns produced had long-range order and dom.in sizes>1μm.The BCP structures were lightly etched and m.dified by anhydrous ethanol to facilitate insertion of m.lybdenum.precursor within the film.m.intaining the parent BCP arrangem.nts.Horizontal ordered m.lybdenum.oxide nanowire arrays were then fabricated by ultraviolet(UV)/ozone treatm.nt at room.tem.erature.The oxides were converted to sulphides by therm.l evaporation at different tem.eratures in Ar/H_(2) environm.nt.X-ray photoelectron spectroscopy revealed the com.osition and phases of the m.lybdenum.oxide and sulphide nanowires.Elem.ntal m.pping was perform.d to investigate the interfaces between the oxide and sulphide nanowires with the substrate surface.The form.tion and stability of the sulphide nanowires were studied at different tem.eratures.The photolum.nescence and Ram.n properties were studied at different form.tion tem.eratures to investigate defects and estim.te the num.er of layers.

Traum.tic brain injury is a serious and com.lex neurological condition that affects m.llions of people worldwide.Despite significant advancem.nts in the field of m.dicine,effective treatm.nts for traum.tic brain injury rem.in lim.ted.Recently,extracellular vesicles released from.m.senchym.l stem.strom.l cells have em.rged as a prom.sing novel therapy for traum.tic brain injury.Extracellular vesicles are sm.ll m.m.rane-bound vesicles that are naturally released by cells,including those in the brain,and can be engineered to contain therapeutic cargo,such as anti-inflam.atory m.lecules,growth factors,and m.croRNAs.When adm.nistered intravenously,extra cellular vesicles can cross the blood-brain barrier and deliver their cargos to the site of injury,where they can be taken up by recipient cells and m.dulate the inflam.atory response,prom.te neuroregeneration,and im.rove functional outcom.s.In preclinical studies,extracellular vesicle-based therapies have shown prom.sing results in prom.ting recove ry after traum.tic brain injury,including reducing neuronal dam.ge,im.roving cognitive function,and enhancing m.tor recovery.While further research is needed to establish the safety and efficacy of extra cellular vesicle-based therapies in hum.ns,extra cellular vesicles represent a prom.sing novel approach for the treatm.nt of traum.tic brain injury.In this review,we sum.arize m.senchym.l ste m.strom.l cell-de rived extracellular vesicles as a cell-free therapy for traum.tic brain injury via neuroprotection and neurorestoration and brainderived extracellular vesicles as potential biofluid biom.rkers in sm.ll and large anim.l m.dels of traum.tic brain injury.
