GUN4 Is Required for Posttranslational control of Plant Tetrapyrrole Biosynthesis

In aerobic photosynthetic organisms, GUN4 binds the chlorophyll intermediates protoporphyrin and Mg protoporphyrin, stimulates Mg chelatase activity, and is implicated in plastidic retrograde signaling. GUN4 expression is most abundant in young and greening tissues and parallels the activity of 5-aminolevulinic acid （ALA） ALA and Mg porphyrin biosynthesis during photoperiodic growth. We explored function and mode of action of GUN4 using GUN4- deficient and overexpressing plants. GUN4 overexpression leads to a general activation of the enzymes of chlorophyll biosynthesis. During photoperiodic growth GUN4 deficiency prevents ALA synthesis and chlorophyll accumulation. All these metabolic changes do not correlate with altered gene expression or changes of protein abundance in tetrapyrrole biosynthesis. While ALA feeding fails to compensate GUN4 deficiency during light-dark growth, this approach results in chlorophyll accumulation under continuous dim light. A new model defines the involvement of GUN4 in posttranslational regulation of ALA and Mg porphyrin synthesis, to sustain chlorophyll synthesis, namely under varying environmental conditions.

Plastids communicate their developmental and physiological status to the nucleus via retrograde signaling,allowing nuclear gene expression to be adjusted *** during plastid biogenesis and responses of mature chloroplasts to environmental changes are designated“biogenic”and“operational”controls,respectively.A prominent example of the investigation of biogenic signaling is the screen for gun(genomes uncoupled)*** the first five gun mutants were identified 30 years ago,the functions of GUN proteins in retrograde signaling remain controversial,and that of GUN1 is hotly ***,we provide background information and critically discuss recently proposed concepts that address GUN-related signaling and some novel gun ***,considering heme as a candidate in retrograde signaling,we revisit the spatial organization of heme biosynthesis and export from *** this review focuses on GUN pathways,we also highlight recent progress in the identification and elucidation of chloroplast-derived signals that regulate the acclimation response in green algae and ***,stress-induced accumulation of unfolded/misassembled chloroplast proteins evokes a chloroplast-specific unfolded protein response,which leads to changes in the expression levels of nucleus-encoded chaperones and proteases to restore plastid protein *** also address the importance of chloroplast-derived signals for activation of flavonoid biosynthesis leading to production of anthocyanins during stress acclimation through sucrose non-fermenting 1-related protein kinase ***,a framework for identification and quantification of intercompartmental signaling cascades at the proteomic and metabolomic levels is provided,and we discuss future directions of dissection of organelle-nucleus communication.

In a recently published study in The New England Journal of Medicine,Liu and colleagues were able to demonstrate that-apart from the antileukemic effects previously reported for hypomethylating agents(HMAs)-treatment with these drugs can also lead to demethylation and subsequent upregulation of oncogenes resulting in shorter survival in patients with myelo-dysplastic syndrome(MDS).

An engineering and economic analysis of energy loss through single paned windows in a large college building is *** goal of this work was to determine an economically viable solution for Marietta college to save on energy *** on the model presented,the economics of double paned window installation are *** alternative solution employing the addition of storm windows to the existing single paned windows has yielded very exciting *** double paned windows are generally the first thought when it comes to energy efficient windows,this work shows that storm window addition yields a much lower investment and quicker payback period for this specific *** engineering analysis and experimental technique presented can be used to investigate the cost benefit of single paned window upgrades for other buildings.

A fleshy fruit is commonly assumed to resemble a thin-walled pressure vessel containing a homogenous carbohydrate *** sweet cherry(Prunus avium L.)as a model system,we investigate how local differences in cell water potential affect H2O and D2O(heavy water)*** partitioning of H2O and D2O was mapped nondestructively using magnetic resonance imaging(MRI).The change in size of mesocarp cells due to water movement was monitored by optical coherence tomography(OcT,non-destructive).Osmotic potential was mapped using microosmometry(destructive).Virtual sections through the fruit revealed that the H2O distribution followed a net pattern in the outer mesocarp and a radial pattern in the inner *** patterns align with the disposition of the vascular bundles.D2O uptake through the skin paralleled the acropetal gradient in cell osmotic potential gradient(from less negative to more negative).cells in the vicinity of a vascular bundle were of more negative osmotic potential than cells more distant from a vascular *** revealed net H2O uptake was the result of some cells loosing volume and other cells increasing volume.H2O and D2O partitioning following uptake is non-uniform and related to the spatial heterogeneity in the osmotic potential of mesocarp cells.

Surgical Treatment of craniopharyngiomas in Adults:comparison between Primary Surgery and Surgery for Recurrence

Objective Few studies have investigated the differences in outcomes between primary and repeat surgery for a craniopharyngioma in *** a result,a treatment concept for adult patients with a craniopharyngioma has not yet been *** present study aimed to retrospectively analyze adult patients with craniopharyngioma to compare surgical outcomes between primary surgery and surgery for *** The demographic and clinical data of 68 adult patients with craniopharyngioma who had primary surgery(n=50)or surgery for recurrence(n=18)were retrospectively *** addition,the patients were followed up for an average of 38.6 months(range:1–133 months).Results The cohorts of patients undergoing primary surgery or repeat surgery did not differ preoperatively in terms of demographic data,or radiological tumor ***,patients with recurrent craniopharyngioma had significantly more pituitary hormone deficits and hypothalamo-pituitary disorders before surgery compared with patients with newly diagnosed *** success rate of complete resection in primary surgery was 53.2%.Even after repeat surgery,a satisfactory rate of complete resection of 35.7%was *** morbidity was increased neither in patients with repeat surgery compared with those with primary surgery(postoperative bleeding P=0.560;meningitis P=1.000;cSF leak P=0.666;visual disturbance P=0.717)nor in patients with complete resection compared with those with partial *** found no difference in recurrence-free survival between initial surgery and repeat surgery(P=0.733).The recurrence rate was significantly lower after complete resection(6.9%)than after partial resection(47.8%;Pconclusion Attempting complete resection is justified for not only those with newly diagnosed craniopharyngioma but also for those with recurrent ***,the surgeon must settle for less than total resection if postoperative morbidity is anticipated.

The so-called rain-cracking of sweet cherry fruit severely threatens commercial *** observation tells us that cuticular microcracking(invisible)always precedes skin macrocracking(visible).The objective here was to investigate how a macrocrack *** detached sweet cherry fruit in deionized water induces *** fruit in D2O and concurrent magnetic resonance imaging demonstrates that water penetration occurs only(principally)through the microcracks,with nondetectable amounts penetrating the intact *** coherence tomography of detached,whole fruit incubated in deionized water,allowed generation of virtual cross-sections through the zone of a developing *** mesocarp cell volume increased before macrocracks developed but increased at a markedly higher rate *** change in mesocarp cell volume occurred in a control zone distant from the *** water incubation continued,the cell volume in the crack zone decreased,indicating leaking/bursting of individual mesocarp *** incubation continued still longer,the crack propagated between cells both to form a long,deep *** mesocarp cell turgor did not differ significantly before and after incubation between fruit with or without macrocracks;nor between cells within the crack zone and those in a control zone distant from the *** cumulative frequency distribution of the logtransformed turgor pressure of a population of outer mesocarp cells reveals all cell turgor data followed a normal *** results demonstrate that microcracks develop into macrocracks following the volume increase of a few outer mesocarp cells and is soon accompanied by cell bursting.
