Objective Radix salviae miltiorrhizae（SM） and Lignum dalbergiae odoriferae（DO） are both traditional Chinese medicinal herbs for the treatment of ischemic heart disease and other cardiovascular diseases,but few of literature is about the systematic ***-Dawley rats were pretreated with SM,aqueous extract of DO（DOA） and volatile oil of DO（DOO）,either as monotherapy or in combination for 7 days,then subjected to 30 min of ischemia followed by 180 min of *** pharmacodynamic evaluation and metabonomics based on gas chromatography/time-of-flight mass spectrometry（GC/TOF-MS） were used to identify the therapeutic effect of these traditional Chinese *** results showed that SM,DOA and DOO ameliorated cardiac function respectively,and this effect was further strengthened by their *** all combinations,SM+DOO showed predominant potential（P＜0.01） to improve electrocardiogram and heart rate,reduce heart weight index and myocardial infarct size,decrease the levels of CK-MB and ***,metabonomics-based findings such as the principal component analysis（PCA） and partial least squares discriminant analysis（PLS-DA） score plot of metabolic state in rat serum correlated well to the preceding results,confirming that SM+DOO exerted synergistic therapeutic efficacies to exhibit better effect on MI/R injury rats compared with other pretreatment ***,the maximum effective point of SM+DOO was 30 min and the minimum effective point was 180 *** addition,SM+DOO group markedly（P＜0.01） reduced the number of TUNEL-positive cells,the levels of TNF-α,IL-6 and MDA,while increased the activity of SOD both in serum and *** results indicated that SM+DOO have combined effects that are highly effective than single pretreatment against MI/R injury *** effect might be achieved partly through anti-apoptosis,anti-oxidant and *** SM+DOO might be an effective and promising medicine for both prophylaxis

Traditional Chinese medicine uses a systemic treatment approach, targeting multiple etiological factors simultaneously. Danhong injection (DHI), a very popular Chinese medicine injection composed of water-soluble extracts of Radix Salvia miltiorrhiza (Danshen) and Flos Carthamus tinctorius L. (Honghua), is reported to be effective for many cardiovascular conditions. The primary active ingredients of DHI, and their systemic and interrelated mechanism have not been evaluated in an established myocardial ischemia/reperfusion (MI/R) injury model. We identified the main active constituents in DHI, including hydroxysafflor yellow A (A), salvianolic acid B (B), and Danshensu (C), by HPLC fingerprint analysis and assessed their effect on I/R injury rats and cardiomyocytes. These 3 compounds and DHI all decreased the levels of IL-1, TNF-α, and MDA, and increased those of IL-10 and SOD activity in vivo and in vitro, and had anti-apoptotic effects, as shown by flow cytometric analysis and TUNEL assay. Moreover, these compounds increased phosphorylation of Akt and ERK1/2 in cardiomyocytes. Interestingly, we found A, B, and C, exerted prominent effect compared with two other ingredients on inhibiting inflammation, reducing oxidative stress, alleviating apoptosis through decreasing NF-κB level and increasing Nrf2 expression and lower caspase-3 activity, respectively. Compounds A, B, and C are the main active ingredients of DHI. Compound A exerted a more prominent anti-inflammatory effect than B and C, by decreasing NF-κB levels;compound B had more powerful anti-oxidative capacity than A and C, by increasing Nrf2 expression;and compound C had stronger anti-apoptotic ability than A and B, by lowering caspase-3 activity. Our results elucidate the mechanisms by which DHI protects against MI/R induced injury.

Background: Patients with type 2 diabetes have increased cardiovascular disease risk compared with those without diabetes. Hyperglycemia can induce reactive oxygen species （ROS） generation, which contributes to development of diabetic cardiomyopathy. Our previous study has demonstrated that total saponins of Aralia taibaiensis （sAT）, a frequently-used antidiabetic in traditional Chinese medicine （TCM）, can scavenge free radicals in vivo and have good antioxidant ability on lipid peroxidation of rat liver microsomes. To investigate whether sAT could protect the heart, while it was in the treatment of diabetes, we designed this study to elucidate cardioprotective effects of sAT. Methods and results: Oxidative stress was induced in H9c2 cells by high glucose （33 mM） and glucose oxidase （15mU） and the protective effects of sAT were evaluated. Treatment of H9c2 cells with G/GO resulted in an increase in cell death, intracellular ROS level and cell oxidative injury, which were markedly reduced by sAT treatment. Further study revealed that sAT induced the nuclear translocation of Nrf2, enhanced the expression of Nrf2-related antioxidant proteins heme oxygenase （HO-1） and NAD （P） H quinone oxidoreductase-1 （NQO-1）. Inhibition of Nrf2 by siRNA reduced sAT-induced upregulation of these antioxidant genes. Western blotting and luciferase assay revealed sAT activated Nrf2 via the PI3K/Akt pathway. Moreover, Nrf2 siRNA markedly reduced the cytoprotective effects of sAT. Conclusions: sAT exerts cytoprotective effects against oxidative stress induced by hyperglycemia. This cardioprotection is mediated, at least in part, through PI3K/Akt/Nrf2 pathways, which further activates Nrf2-regulated antioxidant enzymes. sAT might be a promising candidate for the treatment of diabetic cardiomyopathy.
