Dysfunction of synaptic endocytic trafficking in Parkinson's disease

Parkinson's disease is characterized by the selective degeneration of dopamine neurons in the nigrostriatal pathway and dopamine deficiency in the *** precise reasons behind the specific degeneration of these dopamine neurons remain largely *** investigations have identified over 20 causative PARK genes and 90 genomic risk loci associated with both familial and sporadic Parkinson's ***,several of these genes are linked to the synaptic vesicle recycling process,particularly the clathrinmediated endocytosis *** suggests that impaired synaptic vesicle recycling might represent an early feature of Parkinson's disease,followed by axonal degeneration and the eventual loss of dopamine cell bodies in the midbrain via a"dying back"***,several new animal and cellular models with Parkinson's disease-linked mutations affecting the endocytic pathway have been created and extensively *** models faithfully recapitulate certain Parkinson's disease-like features at the animal,circuit,and cellular levels,and exhibit defects in synaptic membrane trafficking,further supporting the findings from human genetics and clinical *** this review,we will first summarize the cellular and molecular findings from the models of two Parkinson's disease-linked clathrin uncoating proteins:auxilin(DNAJC6/PARK19)and synaptojanin 1(SYNJ1/PARK20).The mouse models carrying these two PARK gene mutations phenocopy each other with specific dopamine terminal pathology and display a potent synergistic ***,we will delve into the involvement of several clathrin-mediated endocytosis-related proteins(GAK,endophilin A1,SAC2/INPP5 F,synaptotagmin-11),identified as Parkinson's disease risk factors through genome-wide association studies,in Parkinson's disease *** will also explore the direct or indirect roles of some common Parkinson's disease-linked proteins(alpha-synuclein(PARK1/4),Parkin(PARK2),and LRRK2(PARK

Modification in Cu-Zn Alloy Properties by 2 MeV Ni<sup>+</sup>Ions Irradiation

We investigate the effects of 2 MeV Ni+ ion beam irradiation with various fluence ranging from 15 × 1011 to 60 × 1014 ions/cm2 on the surface, structural and mechanical properties of Cu-Zn alloy. The modification in target properties after irradiation is confirmed by using various characterization techniques viz. SEM, XRD, UTM and Vickers micro-hardness tester. The SEM results illustrate the formation of nano sized craters with different diameters. Their average diameter decreases from 190 nm to 90 nm by increasing ion fluence. The XRD analysis of irradiated targets reveals that Ni+ ion irradiation enhances the growth of (111) phase and its peak position varies due to ion induced tensile stresses in target matrix. Tensile and Vickers micro-hardness tests verify the mechanical properties of Cu-Zn alloy reduce monotonically upon irradiation. Various mechanisms such as generation, recombination, augmentation and annihilation of ion induced defects are responsible for this reduction. Understanding the relationships between various modified properties of irradiated target is essential for growing new advanced material by irradiation.

Wellbore Cleaning Degree and Hydraulic Extension in Shale Oil Horizontal Wells

The efficient development and exploitation of shale oil depends on long-distance horizontal wells. As the degreeof cleaning of the wellbore plays a key role in these processes, in this study, this problem is investigated experimentallyby focusing on the dimensionless cuttings bed height. A method is proposed to calculate the horizontalwellhydraulic extension taking into account the influence of the wellbore cleaning degree on the wellborepressure distribution and assess the effect of a variety of factors such as the bottom hole pressure, the circulatingpressure drop, the drilling pump performance and the formation properties. The analysis shows that the hydraulicextension of horizontal wells decreases with an increase in the cuttings bed height, and the higher the displacementof drilling fluid, the faster the hydraulic extension declines. The annular pressure drop of the horizontalsection increases with the increase of the cuttings bed height, resulting in a higher bottom-hole pressure. Severalarguments are provided to guide the safe drilling of shale oil horizontal wells and overcome the limits of currenttechnological approaches.

Numerical simulation of the dynamic migration mechanism and prediction of saturation of tight sandstone oil

Quantitative characterization of tight sandstone oil migration and accumulation is an emerging research frontier in the field of oil and gas *** this study,a conceptual model containing multiple basic geological elements is developed,and a nonlinear seepage numerical model for tight sandstone oil migration and accumulation is *** effects of the slip effect,overpressure driving force,buoyancy,and capillary force on the migration and accumulation of tight oil are *** results showed that(1)the differences in oil migration and accumulation between tight and conventional reservoirs are reflected in the growth mode of oil saturation,distribution characteristics of oil and water,and extent of the effect of the formation dip angle;(2)the slip effect has a significant impact when the average pore throat radius is less than 150 nm and the overpressure driving force and capillary force are the main mechanical mechanisms controlling oil migration and accumulation in tight sandstone,while the coupling effect of buoyancy,capillary force,and overpressure driving force controls the upper and lower limits of oil ***,a dimensional and dimensionless identification chart for rapidly predicting the oil saturation of tight sandstone is proposed and verified using the measured *** study provides a basis for analyzing the migration and accumulation mechanisms of tight sandstone oil and a new approach for predicting oil ***,we developed digital and visual analysis methods for the migration results,enriching the expression of the dynamics of hydrocarbon accumulation.

An adaptive physics-informed deep learning method for pore pressure prediction using seismic data

Accurate prediction of formation pore pressure is essential to predict fluid flow and manage hydrocarbon production in petroleum *** deep learning technique has been receiving more interest due to the great potential to deal with pore pressure ***,most of the traditional deep learning models are less efficient to address generalization *** fill this technical gap,in this work,we developed a new adaptive physics-informed deep learning model with high generalization capability to predict pore pressure values directly from seismic ***,the new model,named CGP-NN,consists of a novel parametric features extraction approach(1DCPP),a stacked multilayer gated recurrent model(multilayer GRU),and an adaptive physics-informed loss *** machine training,the developed model can automatically select the optimal physical model to constrain the results for each pore pressure *** CGP-NN model has the best generalization when the physicsrelated metricλ=0.5.A hybrid approach combining Eaton and Bowers methods is also proposed to build machine-learnable labels for solving the problem of few *** validate the developed model and methodology,a case study on a complex reservoir in Tarim Basin was further performed to demonstrate the high accuracy on the pore pressure prediction of new wells along with the strong generalization *** adaptive physics-informed deep learning approach presented here has potential application in the prediction of pore pressures coupled with multiple genesis mechanisms using seismic data.

Model checking is an automated formal verification method to verify whether epistemic multi-agent systems adhere to property *** there is an extensive literature on qualitative properties such as safety and liveness,there is still a lack of quantitative and uncertain property verifications for these *** uncertain environments,agents must make judicious decisions based on subjective *** verify epistemic and measurable properties in multi-agent systems,this paper extends fuzzy computation tree logic by introducing epistemic modalities and proposing a new Fuzzy Computation Tree Logic of Knowledge(FCTLK).We represent fuzzy multi-agent systems as distributed knowledge bases with fuzzy epistemic interpreted *** addition,we provide a transformation algorithm from fuzzy epistemic interpreted systems to fuzzy Kripke structures,as well as transformation rules from FCTLK formulas to Fuzzy Computation Tree Logic(FCTL)***,we transform the FCTLK model checking problem into the FCTL model *** enables the verification of FCTLK formulas by using the fuzzy model checking algorithm of FCTL without additional computational ***,we present correctness proofs and complexity analyses of the proposed ***,we further illustrate the practical application of our approach through an example of a train control system.

A continuous and long-term in-situ stress measuring method based on fiber optic. Part I: Theory of inverse differential strain analysis

A method for in-situ stress measurement via fiber optics was proposed. The method utilizes the relationship between rock mass elastic parameters and in-situ stress. The approach offers the advantage of long-term stress measurements with high spatial resolution and frequency, significantly enhancing the ability to measure in-situ stress. The sensing casing, spirally wrapped with fiber optic, is cemented into the formation to establish a formation sensing nerve. Injecting fluid into the casing generates strain disturbance, establishing the relationship between rock mass properties and treatment ***, an optimization algorithm is established to invert the elastic parameters of formation via fiber optic strains. In the first part of this paper series, we established the theoretical basis for the inverse differential strain analysis method for in-situ stress measurement, which was subsequently verified using an analytical model. This paper is the fundamental basis for the inverse differential strain analysis method.

Comparison between sepsis-induced coagulopathy and sepsis-associated coagulopathy criteria in identifying sepsis-associated disseminated intravascular coagulation

BACKGROUND:Disseminated intravascular coagulation(DIC)is associated with increased mortality in sepsis *** this study,we aimed to assess the clinical ability of sepsis-induced coagulopathy(SIC)and sepsis-associated coagulopathy(SAC)criteria in identifying overt-DIC and preDIC status in sepsis ***:Data from 419 sepsis patients were retrospectively collected from July 2018 to December *** performances of the SIC and SAC were assessed to identify overt-DIC on days 1,3,7,or *** SIC status or SIC score on day 1,the SAC status or SAC score on day 1,and the sum of the SIC or SAC scores on days 1 and 3 were compared in terms of their ability to identify *** SIC or SAC status on day 1 was evaluated as a pre-DIC indicator for anticoagulant ***:On day 1,the incidences of coagulopathy according to overt-DIC,SIC and SAC criteria were 11.7%,22.0%and 31.5%,*** specificity of SIC for identifying overt-DIC was significantly higher than that of the SAC criteria from day 1 to day 14(P3 had a significantly higher sensitivity(72.00%)and area under the curve(AUC)(0.69)in identifying pre-DIC than did the SIC or SAC status(sensitivity:SIC status 44.00%,SAC status 52.00%;AUC:SIC status 0.62,SAC status 0.61).The sum of the SIC scores on days 1 and 3 had a higher AUC value for identifying the pre-DIC state than that of SAC(0.79 vs.0.69,P<0.001).Favorable effects of anticoagulant therapy were observed in SIC(adjusted hazard ratio[HR]=0.216,95%confidence interval[95%CI]:0.060–0.783,P=0.018)and SAC(adjusted HR=0.146,95%CI:0.041–0.513,P=0.003).CONCLUSION:The SIC and SAC seem to be valuable for predicting *** sum of SIC scores on days 1 and 3 has the potential to help identify pre-DIC.
