Incidental focal colorectal ^(18)F-fluorodeoxyglucose uptake on positron emission tomography/computed tomography

AIM: To assess the clinical significance of incidental focal colorectal 18 F-fluorodeoxyglucose ( 18 F-FDG) uptake on 18 F-FDG-positron emission tomography/computed tomography (PET/CT). METHODS: The records of all the cases which had undergone colonoscopy after PET/CT within a two weeks interval were reviewed. Adenomas were considered advanced when they were villous, ≥ 10 mm in size, or had high-grade dysplasia. Colorectal cancers and advanced adenomas are collectively referred to as advanced colorectal neoplasms. Receiver-operating characteristic curve analysis was used to determine thesignificant predictive maximum standardized uptake value (SUVmax) cutoff value for advanced colorectal neoplasms and cancer. RESULTS: Ninety-five colorectal lesions matched the site of incidental focal colorectal 18 F-FDG uptake on PET/CT and 146 did not. Colonoscopy showed advanced colorectal neoplasms corresponding to the site of 18 F-FDG uptake in 49 of the 95 (51.5%) lesions with incidental uptake. Of the lesions without incidental uptake, only 6 of 146 (4.1%) had advanced colorectal neoplasms on colonoscopy, indicating a statistically significant difference between the two groups (P < 0.001). The sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of incidental focal 18 F-FDG uptake in identifying advanced colorectal neoplasms were 89.1%, 75.3%, 51.6%, 95.9%, and 78.4%, respectively. In detecting only CRC, these values were 89.2%, 69.6%, 34.7%, 97.3%, and 72.6%, respectively. The significant SUVmax cutoff value for advanced colorectal neoplasms (area under the curve 0.755, P < 0.001) was 4.35, with a sensitivity, specificity, PPV, NPV, and accuracy of 75.5%, 65.2%, 69.8%, 71.4% and 70.5%, respectively. For CRC, 5.05 was the significant SUVmax cutoff value (area under the curve 0.817, P < 0.001), with a sensitivity, specificity, PPV, NPV, and accuracy of 84.8%, 71.0%, 80.9%, 89.8%, and 75.8%, respectively. Formulation of a film-coated dutasteride tablet bioequivalent to a soft gelatin capsule(Avodart~?): Effect of γ-cyclodextrin and solubilizers

The aim of this study was to optimize a tablet formulation of dutasteride that is bioequivalent to a commercially available soft gelatin capsule(Avodart~?). The effect of cyclodextrin on enhancing the aqueous solubility of dutasteride was investigated, after which the formulation was further optimized with solubilizing polymer and surfactant. Among the cyclodextrins tested, the highest solubility was observed when dutasteride was complexed with γ-cyclodextrin. Moreover, the addition of polyvinylpyrrolidone and Gelucire/TPGS further enhanced the solubility of dutasteride. Differential scanning calorimetry(DSC) and powder X-ray diffraction(pX RD) studies demonstrated that dutasteride existed in the amorphous form in the complex. Optimized dutasteride complexes were selected after a pharmacokinetic study in rats, and film-coated tablets were prepared by the direct compression method. In vitro dissolution profiles for the tablets of dutasteride complexes were similar to those of the reference. Moreover, pharmacokinetic parameters including the C max and AUC values after oral administration in beagle dogs were not significantly different from those of the reference with a relative bioavailability of 92.4%. These results suggest the feasibility of developing a tablet formulation of dutasteride using cyclodextrin complex in addition to a solubilizing polymer and surfactant.

Polyurethane-based Actuators with Various Polyols

This study dealt with the electrostrictive responses of polyurethane （PU） actuators with different microphase separation structure, which was a promising candidate for a material used in polymer actuators. In order to construct PUs with different higher-order structure, PUs with various types of polyol were synthesized： poly（neopentyl glycol adipate）（PNAD）, poly（tetramethylene glycol）（PTMG）, and poly（dimethyl siloxnae）（PDMS）. Synthesized PU was characterized by Fourier transform-infrared （FT-IR） spectroscopy and gel per- meation chromatography （GPC）. Thermal analysis and mechanical properties of PU films were carried out with differential scanning calorimetry （DSC） and UTM （universal testing machine）, respectively. And PU actuator was formed in a type of monomorph, which was made by carbon black electrodes on the both surfaces of PU film by spin coating method. Actuation behavior was mainly influenced on microphase separation struc- ture and mechanical property of PU. In result, PU actuator with PNAD, polyester urethane, had the largest field-induced displacement.

Home Range Size and Overlap of the Small Nocturnal Schlegel’s Japanese Gecko (Gekko japonicus), Introduced into a City park in Korea

Studying the home range of an organism is important in understanding its ecology. Due to being cryptic, few studies have been conducted on the home range studies of small, nocturnal geckos. We conducted radio-tracking surveys for 23 individuals to estimate the home range size and home range overlap of the Schlegel’s Japanese gecko(Gekko japonicus) previously introduced into a suburban city park. Individuals were commonly found in artificial structures(buildings and accessory structures) and on nearby natural trees. Daily moved distance was positively correlated with home range size. minimum convex polygon(MCP) home range was 97.8 m^2 for females and 99.5 m^2 for males, on average. Gekko japonicus moved farther daily distances and used wider MCP and Kernel 95 home ranges in breeding season compared to non-breeding season, while the size of Kernel 50 home range did not differ between seasons. Both daily moved distance and home range size were not significantly different between sexes. In the breeding season, MCP and Kernel 50 home ranges of each gecko overlapped with 32.4% and 13.8% of remaining geckos, respectively. Our results not only show that 1) G. japonicus uses both artificial structures and adjacent natural trees as microhabitat, but also suggest that 2) G. japonicus is non-territorial, but has a core habitat that is shared with few other individuals, and 3) the reproductive system of G. japonicus is polygamous.

Green coffee bean extract improves obesity by decreasing body fat in high-fat diet-induced obese mice

Post-translational modifications （PTMs） of transcription factors play a crucial role in regulating metabolic homeostasis. These modifications include phosphory- lation, methylation, acetylation, ubiqulUnation, SUMOy- lation, and O-GIcNAcylation. Recent studies have shed light on the importance of lysine acetylation at nonhis- tone proteins including transcription factors. Acetyla- tion of transcription factors affects subcellular distribution, DNA affinity, stability, transcriptional activity, and current investigations are aiming to further expand our understanding of the role of lysine acetyla- tion of transcription factors. In this review, we summarize recent studies that provide new insights into the role of protein lysine-acetylation in the transcriptional regulation of metabolic homeostasis.
