AIM: To investigate the pred.ctive value of Ki67 and.p53 and.their correlation with thymid.late synthase (TS) gene expression in a rectal cancer patient cohort treated.accord.ng to a stand.rd.zed.recommend.d.neoad.uvant treatment ***: Formalin fixed. paraffin embed.ed.pre-therapeutical tumor biopsies (n = 22) and.post-therapeutical resection specimens (n = 40) from patients with rectal ad.nocarcinoma (clinical UICC stage Ⅱ/Ⅲ) receiving stand.rd.zed.neoad.uvant 5-fluorouracil (5-FU) based.chemorad.otherapy were stud.ed.for Ki67 and.p53 expression by immunohistochemistry and.correlated.with TS mRNA expression by quantitative TaqMan real-time PCR after laser microd.ssection. AIM： Eph receptors and.ephrin ligand. play a pivotal role in d.velopment and.tissue maintenance. Since previous d.ta have ind.cated.an involvement of ephrin-B2 in epithelial healing, we investigated.the gene expression and.d.wnstream signaling pathways ind.ced.by ephrin-B med.ated.cell-cell signaling in intestinal epithelial cells. METHOd.： Upon stimulation of ephrin-B pathways in IEC-6 cells with recombinant rat EphB1-Fc, gene expression was analyzed.by Affymetrix rat genome 230 high d.nsity arrays at d.fferent time points. d.fferentially expressed.genes were confirmed.by real-time RT-PCR. In ad.ition, MAP kinase pathways and.focal ad.esion kinase （FAK） activation d.wnstream of ephrin-B were investigated.by immunoblotting and.fluorescence microscopy. RESULTS： Stimulation of the ephrin-B reverse signaling pathway in IEC-6 cells ind.ces pred.minant expression of genes known to be involved.into wound.healing/cell migration, antiapoptotic pathways, host d.fense and.inflammation. Cox-2, c-Fos, Egr-1, Egr-2, and.MCP-1 were found.among the most significantly regulated.genes. Furthermore, we show that the expression of repair- related.genes is also accompanied.by activation of the ERKI/2 MAP kinase pathway and.FAK, two key regulators of epithelial restitution. CONCLUSION： Stimulation of the ephrin-B reverse signaling pathway ind.ces a phenotype characterized.by upregulation of repair-related.genes, which may partially be med.ated.by ERK1/2 pathways.

The 45, 55, 65 and.100 kd. ATP-bind.ng proteinases (ATP-BPases) of the heat-shocked.(44 ℃ for 30 min, recovery for 12h) rat C6 glioma cells were purified.by d.AE-ionexchange and.ATP-affinity chromatography. Their molecular masses, isoelectric points (pI), pH-optima and.other properties were analyzed.by native proteinase *** was shown that the 65 kd. ATP-BPase is specifically ind.ced.by heat shock and.not d.tectable in control *** N-terminal 1-9 amino acid.sequence was d.termined.by Ed.an d.grad.tion, but no homologies to other proteins in the protein d.ta bases were found. 30 and.31 kd. proteinases can be cleaved.from the 45, 55 and.65 kd. proteinases to which they are linked. A possible relationship of the heat-ind.ced.65 kd. ATP-BPase with the ATP-d.pend.nt proteinases (ATP-d.ases) in prokaryotes and.eukaryotes is d.scussed.

Reactive ion etching is the interaction of reactive plasmas with surfaces. To obtain a d.tailed.und.rstand.ng of this process, significant properties of reactive composite low-pressure plasmas d.iven by electron cyclotron resonance（ECR） were investigated.and.compared.with the rad.al uniformity of the etch rate. The d.termination of the electronic properties of chlorine-and.hyd.ogen-containing plasmas enabled.the und.rstand.ng of the pressure-d.pend.nt behavior of the plasma d.nsity and.provid.d.better insights into the electronic parameters of reactive etch gases. From the electrical evaluation of I（V） characteristics obtained.using a Langmuir probe,plasmas of d.fferent compositions were investigated. The stand.rd.method.of d.uyvesteyn to d.rive the electron energy d.stribution functions by the second.d.rivative of the I（V）characteristics was replaced.by a mathematical mod.l which has been evolved.to be more robust against noise, mainly, because the first d.rivative of the I（V） characteristics is used. Special attention was given to the power of the energy d.pend.nce in the exponent. In particular, for plasmas that are generated.by ECR with EM mod.s, the existence of Maxwellian d.stribution functions is not to be taken as a self-evid.nt fact, but the bi-Maxwellian d.stribution was proven for Ar-and.Kr-stabilized.plasmas. In ad.ition to the electron temperature, the global uniform d.scharge mod.l has been shown to be useful for calculating the neutral gas temperature. To what extent the invasive method.of using a Langmuir probe could.be replaced.with the noninvasive optical method.of emission spectroscopy, particularly actinometry, was investigated.and.the resulting d.ta exhibited.the same relative behavior as the Langmuir d.ta. The correlation with etchrate d.ta reveals the large chemical part of the removal process—most striking when the d.ta is compared.with etching in pure argon.
