The rise in antimicrobial resistance increases researchers’ interest in medicinal plants used for traditional treatment of infectious diseases. The study is based on ten (10) medicinal plants mostly cited in the treatment of diarrhea in West Africa: Khaya senegalensis, Anacardium ouest L., Cassia sieberiana DC., Pterocarpus erinaceus, Diospyros mespiliformis, Ocimum gratissimum, Manihot esculenta, Vernonia amygdalina Delile, Pseudocedrela kotschyi, Daniellia oliveri. 背景:六价铬(Cr(Ⅵ))是人类一级致癌物,对多个器官有损伤毒性效应,肝脏是Cr(Ⅵ)毒性作用的主要靶器官。Cr(Ⅵ)暴露可通过氧化应激反应(Reactive Oxygen Species,ROS)诱导肝脏毒性,但其确切的作用机制仍未明了;本项目以Cr(Ⅵ)暴露的小鼠肝组织为研究对象,通过RNA-seq、Western Blot等多种生物学技术揭示Cr(Ⅵ)的肝脏毒性机制。方法:不同浓度(0,40,80,160和320 mg/***)Cr(Ⅵ)暴露建立急性Cr(Ⅵ)肝损伤模型;RNA-seq检测160 mg/kg Cr(Ⅵ)暴露C57BL/6小鼠肝组织转录组变化,并进行生信分析;H&E观察肝组织结构变化;蛋白杂交印迹(Western Blot)和组织免疫组化检测蛋白表达;荧光定量PCR(RT-q PCR)检测基因表达水平变化。结果:Cr(Ⅵ)暴露后小鼠肝组织结构异常、肝细胞损伤及产生肝脏炎症反应,且呈现剂量依赖效应。RNA-seq转录组学结果显示Cr(Ⅵ)暴露后氧化应激、凋亡和炎症反应通路活性升高;KEGG通路分析发现NF-κB信号通路活性显著上升,与RNA-seq结果一致;免疫组化显示Cr(Ⅵ)暴露导致库普弗细胞和中性粒细胞细胞增多,并发现炎症因子(TNF-α、IL-6、IL-1β)表达升高,以及NF-κB信号通路(p-IKKα/β和p-p65)活化。而用ROS抑制剂N-acetylcysteine(NAC)处理后,可减少Cr(Ⅵ)暴露的枯否细胞和中性粒细胞数量,降低炎症因子(TNF-α、IL-6、IL-1β)的表达,抑制NF-κB信号通路(p-IKKα/β和p-p65)活化,并减少Cr(Ⅵ)诱导的肝组织损伤。结论:本项目首次揭示Cr(Ⅵ)通过NF-κB信号通路介导的炎症反应诱导肝组织损伤,这一研究结果将为明确Cr(Ⅵ)的毒性机制和临床预防治疗奠定良好的基础。

Several thousands of chemical substances are registered every year for different purposes, and sometimes many of them are claimed to play the same role. To establish and compare their toxicities, the determination of the lethal concentrations is usually necessary and should account for natural mortality. However, many of the statistical software packages used for that purpose do not readily integrate control mortality or adjust the best link function to the data during the process. This manuscript proposes an ＂lc＂ function in the R open source that aims at the effective determination of lethal concentrations. Furthermore, it performs the procedure with the appropriate link function. The ＂lc＂ application on the example provided revealed that the complementary log link function is adequate.

In this paper, the author aimed to determine the nutritional value of soursop (Annona muricata L. fruit) consumed in Kinshasa as well as the study of the acute toxicity of its seeds. The fruit of Annona muricata L. was sampled according to the ISO 7002 standard for agricultural and food products. The selected fruits were ripe, without physical damage. The usual analytical methods allowed the determination of the nutritional value of the fruit pulp of Annona muricata L. The water content was measured by the method of loss of mass on drying. The total amount of ash was determined by incineration in the oven at 550°C. The mineral elements were determined by inductively coupled plasma spectrometry (ICP0). The determination of acute toxicity was carried out on 25 female mice of the NMRI SUISSE species according to OECD 425 guidelines. For 100 grams of fresh material from the fruit pulp, we noted a very high water content of 84% ± 6%. We also note a particularly high amount of carbohydrates with a rate of 12.2% ± 2%. Protein and lipid content were relatively low at 1% ± 0.01% and 0.7% ± 0.3% respectively. The dietary fiber content was 0.8% ± 0.2%. An energy value of 49.3 Kcal per 100 grams of pulp was determined. The LD 50 obtained was 3320 mg/kg, indicating slight toxicity of soursop seeds. The results of this study show that the white pulp of the fruit of Annona muricata L. consumed in Kinshasa is rich in carbohydrates when we compare it to others biomolecules. It also contains dietary fiber and mineral salts making soursop an excellent constituent of a weight loss diet with a low energy intake.

Background: Maternal death is a major public health problem worldwide, particularly in sub-Saharan Africa. Objective: This study sought to investigate dysfunctions in the management of patients whose outcome was classified as “maternal death” in the Gynaecology-Obstetrics section of the Departmental University Teaching Hospital of Borgou Alibori (CHUD-BA) from 2017 to 2021. Method: This was a retrospective cross-sectional study with descriptive and analytical purposes. The study population consisted of pregnant women, parturients and puerperas admitted into the CHUD-BA maternity ward from 2017 to 2021. Result: A total of 2011 patients were included in this study. The in-hospital maternal mortality ratio was 1526 per 100,000 live births. The dysfunctions identified were the amount of time spent in the referring center (more than 48 hours) (p = 0.001), delay of more than 2 hours between referral and admission into the referral center (p < 0.001), means of transport (motorcycle or public transport) (p compliance to the protocol for emergency obstetric and neonatal care (SONU) (p < 0.001) and delay of more than 2 hours in the etiological management of pregnant women (p Conclusion: Particular attention should be paid to the management of pregnant women in our healthcare system if we are looking forward to reducing maternal mortality.

Background: Gastric ulcer is one of the most common gastrointestinal diseases with a worldwide prevalence of about 40% in the developed countries and 80% in Africa. Acid inhibitors, anticholinergics, histamine H2-antagonists and antibiotics are commonly used to treat gastric ulcer. However, the accumulating evidence for resistance to antibiotics and the side effects of antibiotics and acid inhibitors, anticholinergics and histamine H2-antagonists. Therefore, there is an urgent need for approaches to treat and prevent gastric ulcer. One alternative strategy is the use of probiotic lactic acid bacteria. Aim of the study: This study aimed to isolate and characterised probiotic lactic acid bacteria from palm wine and fermented milk, and to evaluate their antiulcerogenic potentials on ethanol-induced gastric ulcer in mice. Methods: Probiotic lactic acid bacteria were isolated from the fermented milk and palm wine using pour plate technique on MRS agar and identified using the 16S r RNA gene sequencing. For functional properties and selection, acid and bile salt tolerance were evaluated based on viable colony count on MRS agar. Two probiotic lactic acid bacteria were selected for in vivo studies. Fifty-four healthy young adult Balb/c mice were randomly divided into 9 groups of 6 mice each. Gastric ulcer was induced in mice using one oral dose of absolute ethanol (10 mL/kg body weight). The probiotic lactic acid bacteria (F1 and F2) at different doses (MF3 = 9 × 108 CFU/mL, MF6 = 1.8 × 109 CFU/mL and MF9 = 2.7 × 109 CFU/mL) and omeprazole (20 mg/kg) (a reference drug) were orally administrated daily for 14 days before ulcer induction. These mice were sacrificed 1hour after induction and the stomach contents were collected for volume and pH determination. The untreated effluents generated by hospital activities contribute to the dissemination of pathogenic germs and multi-resistant bacteria, thus presenting a great potential danger for health and the environment. The objective of this study was to evaluate the microbiological and physico-chemical quality of the effluents of the Yaoundé University Hospital Centre and their impact on the environment. It was a prospective and analytical study on three sites where hospital effluents from the intensive care unit (Station A), the upstream of the wastewater treatment plant (Station B), and the gynaecology, surgery and hospitalisation departments (Station C) were sampled. Samples were collected in sterile glass bottles for bacteriological analyses and polyethylene bottles for physico-chemical analyses. The bacteriological parameters measured showed that the density of the bacterial species sought was very high at Station B with a predominance of the species Escherichia coli (57.36%). At Station A, total coliforms were very abundant (50.12%) and at Station C, the genus Pseudomonas was predominant (14.69%). Bacteria of the genus Streptococcus were represented by three species, namely: Streptococcus agalactiae, Streptococcus faecalis, and Streptococcus pneumoniae. The Pseudomonas genus was also represented by 3 species, namely Pseudomonas aeruginosa, Pseudomonas maltophilia and Pseudomonas putida. The physico-chemical parameters showed that apart from temperature and conductivity, which were in compliance with the standards, the other had values higher than these standards. This study shows that untreated hospital effluent contains most of the bacteria involved in community, nosocomial infections and would be a potential source of risk to the surrounding population.

Background: Preeclampsia (PE) is one of the forms of hypertensive diseases that occur during pregnancy. Early-onset preeclampsia (EOP), which occurred before 34 weeks, proved to be the deadliest. Indeed, it is characterized by a poor maternal and fetal prognosis. EOP has a disparate incidence in the world varying between 0.9% and 31%. Several risks factors are associated with the occurrence of EOP, which is responsible of several adverse obstetrical outcomes. Complications can affect up to 85% of pregnant women with EOP, especially when EOP appears very early, before 28 or even 25 weeks’ gestation. Objectives: To determine frequency of EOP at the University Clinics of Kinshasa, to describe sociodemographic and clinical characteristics of pregnant women with EOP and to identify its risks factors and its association adverse obstetrical outcomes. Methods: The study will be a cross-sectional analytical study in University Clinics of Kinshasa from January 2016 to December 2022. The minimal size will be 119. Our study population will consist of pregnant women who consult for antenatal best care and are neonates in our Clinic. Result will be presented as percentage proportion. Comparison and proportion means between groups will be made using Student’s test and Pearson’s chi-square test, respectively. Our test will be statistically significant for a p-value ≤ than less 0.05. Data will be collected and analysed anonymously and confidentiality. Conclusion: We believe that our study should enable us to identify profile of gestational carriers at risk of EOP in our environment, as well as prognosis associated with this entity, with a view to arousing particular interest in EOP.
