The study of modified RNA known as epitranscriptomics has become increasingly relevant in our understanding of disease-modifying mechanisms. Methylation of N6 adenosine(m6A) and C5 cytosine(m5C) bases occur on mRNAs, tRNA, mt-tRNA, and rRNA species as well as non-coding RNAs. With emerging knowledge of RNA binding proteins that act as writer, reader, and eraser effector proteins, comes a new understanding of physiological processes controlled by these systems. Such processes when spatiotemporally disrupted within cellular nanodomains in highly specialized tissues such as the brain, give rise to different forms of disease. In this review, we discuss accumulating evidence that changes in the m6A and m5C methylation systems contribute to neurocognitive disorders. Early studies first identified mutations within FMR1 to cause intellectual disability Fragile X syndromes several years before FMR1 was identified as an m6A RNA reader protein. Subsequently, familial mutations within the m6A writer gene METTL5, m5C writer genes NSUN2, NSUN3, NSUN5, and NSUN6, as well as THOC2 and THOC6 that form a protein complex with the m5C reader protein ALYREF, were recognized to cause intellectual development disorders. Similarly, differences in expression of the m5C writer and reader effector proteins, NSUN6, NSUN7, and ALYREF in brain tissue are indicated in individuals with Alzheimer’s disease, individuals with a high neuropathological load or have suffered traumatic brain injury. Likewise, an abundance of m6A reader and anti-reader proteins are reported to change across brain regions in Lewy bodies diseases, Alzheimer’s disease, and individuals with high cognitive reserve. m6A-modified RNAs are also reported significantly more abundant in dementia with Lewy bodies brain tissue but significantly reduced in Parkinson’s disease tissue, whilst modified RNAs are misplaced within diseased cells, particularly where synapses are located. Investigation of the Acceptability of Cultured Meat in University Students

Background: Over the past 20 years, cultured meat has drawn a lot of public attention as a potential solution to issues with animal husbandry, including inadequate use of natural sources, improper animal welfare practices, and possible risks to public health and safety. The novel method of producing meat through culture reduces the need for animals to produce muscle fiber, thereby obviating the necessity for animal slaughter. Apart from its ethical advantages, cultured meat presents a possible way to fulfill the expanding need for food among growing populations. The purpose of this research was to find out whether Turkish students would be willing to pay for and accept cultured meat. Technique: Method: 371 university students who willingly consented to fill out a questionnaire and provide demographic data make up the research sample. Questions from previous studies on the acceptability of cultured meat were compiled to create the survey. The research’s data collection took place in March and April of 2022. The research was completed in June 2022 after the data had been processed and analyzed. Results: The results showed that the majority of participants were female and had omnivorous eating habits. Based on the results of the Bonferroni correction test, students with a higher intention to purchase and consume cultured meat were those who received economics and business education. Students with two years of university education had a higher overall survey score than those with four years of education (p < 0.05). Furthermore, it is discovered that there is a negative correlation between the participants’ ages and their Factor 2 (using cultured meat as an alternative to industrial meat) and Factor 3 (consuming and purchasing it) section points (r = -109, p = 0.036) (r = -0.121, p = 0.019). In conclusion, university students generally have a negative outlook on health-related issues, such as eating cultured meat as an alternative.

We read with great interest the article by Cheung et al.(1)recently published in Hepatobiliary Surgery and Nutrition,presenting a narrative review of various ablation systems for hepatocellular carcinoma(HCC)in cirrhotic patients.We would like to provide several comments.We would first like to offer our congratulations to the authors for their review,which extensively discusses radiofrequency(RFA),high-intensity focused ultrasound(HIFU),and microwave ablation(MWA)ablation techniques.This review is a succinct summary to better understand the utility of various ablation techniques used in HCC treatment(1).

AIM:To define the anatomic and functional outcomes of pars plana vitrectomy(PPV)with internal limiting membrane(ILM)peeling,inverted ILM flap and free ILM patch graft technique for the treatment of myopic macular hole(MH)without retinal detachment.METHODS:Sixty-four eyes of 64 patients who underwent PPV for myopic MH were included.Group 1 consists of patients underwent ILM peeling(n=26),and Groups 2 and 3 consists of patient underwent free ILM patch graft(n=20)and inverted ILM flap procedure(n=18)respectively.Outcomes following surgery were MH closure and best corrected visual acuity(BCVA)in logMAR at 6mo.RESULTS:Closure of MH was obtained in 20 eyes(76.9%)of the Group 1,in 16 eyes(80%)of the Group 2 and in 16 eyes(88.9%)of the Group 3.The mean preoperative and postoperative BCVA was 1.60±0.53 logMAR and 1.27±0.58 logMAR,respectively(P<0.05).There was no significant difference in the postoperative BCVA and anatomical closure rates in the three groups.Although the anatomical closure rate did not differ significantly in the groups,closure of MH tended to be better in the inverted ILM flap technique group at 6mo.CONCLUSION:Different surgical techniques may provide favorable visual and anatomical results for myopic MH surgery.ILM flap techniques offer higher closure rates compared to ILM peeling technique.However,in terms of visual outcomes,the study reveals no difference in three surgical techniques.

Patients with inflammatory bowel disease(IBD)require lifelong and personalized care by a multidisciplinary healthcare team.However,the traditional medical model is not ideal for patients who require continuous close monitoring and whose symptoms may dramatically worsen between regularly scheduled visits.Additionally,close dietary follow-up and monitoring of IBD in a traditional setting are challenging because of the disease complexity,high pressure on outpatient clinics with a small number of IBD specialist dietitians,and rising incidence.Given the significant burden of IBD,there is a need to develop effective dietary management strategies.The coronavirus disease 2019 pandemic caused an unprecedented shift from in-person care to delivering health care via technological remote devices.Traditional nutrition therapy and consultation can be provided by telenutrition through remote electronic communication applications that could greatly benefit patient care.Telenutrition might be useful,safe,and cost-effective compared with standard care.It is likely that virtual care for chronic diseases including IBD will continue in some form into the future.This review article summarizes the evidence about telenutrition applications in the management of IBD patients,and we gave an overview of the acceptance and impact of these interventions on health outcomes.

Geochemistry of the Aptian bituminous limestones in Gümuü?hane area,Eastern Black Sea region:new insight into paleogeography and paleoclimate conditions

Aptian is characterized by widespread deposition of organic-rich sediment.The Aptian bitumen limestone horizon,which is thin decimetre-thick sequences,locally crops out in the Kircaova area,Eastern Black Sea Region(Eastern Pontides).They are well correlated with Aptian bitumen limestone in the other Tethys Reams.They are proposed as episodes of increased organic matter.However,background factors controlling organic matter enrichment are poorly known.In this study,we present new inorganic geochemistry,including trace elements,rare earth elements(REE),redox-sensitive elements(RSE),stable-isotopes(δ~(18)O andδ~(13)C),and total organic carbon(TOC).We integrated new geochemical data with existing stratigraphy,paleontology,and organic chemistry data to provide new insight into the depositional environment and paleoclimate conditions during Aptian.The lacustrine bitumen limestone(LBL)samples have variedδ~(13)C(ave.-1.45‰)andδ~(18)O(ave.-4.50‰).They possess distinct REE patterns,with an average of REE(ave.14.45 ppm)and Y/Ho(ave.35)ratios.In addition,they have variable Nd/YbN(0.28-0.81;ave.0.56)and Ce/Ce*(0.68-0.97;ave.0.86),and relatively high Eu^(*)/Eu(1.23-1.53;ave.1.35).They display seawater signatures with reduced oxygen conditions.The enrichment in RSE(Mo,Cu,Ni,and Zn)and the low Mo/TOC(0.70-3.69;ave.2.41)support a certain degree of water restriction.The high Sr/Ba,Sr/Cu,Ga/Rb,and K/Al records of the LBL facies suggest hot house climatic conditions.The sedimentary environment was probably an isolated basin that is transformed from the marine basin.In addition to depositional conditions,the regional parameters such as the climate,increased run-off period,nutrient levels,alkalinity level,and dominant carbonate producers favored the enrichment in organic matter of LBL facies.Thus,extreme greenhouse palaeoclimate conditions have an important role in organic matter enrichment in the isolated basin.Our results are conformable with the published data from marine,semi-restricted basin,and lacustrine settings in the differen

BACKGROUND Adolescents with autism spectrum disorder(ASD)may encounter many difficulties with their menstrual cycles.Potential challenges that adolescents with ASD may face include understanding physical changes,coping with symptoms,emotional sensitivity,communication,personal care,and hygiene.AIM To evaluate the effect of menstrual hygiene skills training given to adolescents with ASD on their menstrual hygiene skills.METHODS The study was conducted with 15 adolescents diagnosed with ASD by the single group pre-test and post-test model in three special education centers in Türkiye.Data were collected with the Adolescent and Parent Information Form and the Adolescent-Specific Menstrual Hygiene Skill Registration Form.RESULTS While the mean age of adolescents was 16.06±0.88 years,the mean age of individuals responsible for adolescent care was 43.66±5.56 years.While 60.0%of the adolescents noticed the onset of bleeding before training,this rate was 93.3%after training.The Adolescent-Specific Menstrual Hygiene Skill Registration Form showed a statistically significant increase in the application steps after the training.The difference between the menstrual hygiene skill scores of adolescents CONCLUSION The menstrual hygiene skills training given to adolescents with ASD was beneficial in increasing their menstrual hygiene skills.These individuals must take responsibility during menstruation and independently manage their continuous care activities.

Outcome of patients with severe COVID-19 pneumonia treated with high-dose corticosteroid pulse therapy:A retrospective study

Objective:To assess the effectiveness of high-dose corticosteroid pulse therapy and evaluate possible factors associated with 28-day mortality in hospitalised patients with severe COVID-19 pneumonia.Methods:We conducted a single-centre retrospective cohort study on hospitalised patients with clinical,epidemiological,and/or radiologically confirmed and suspected COVID-19 at Bitlis Tatvan State Hospital in Turkey between December 1,2020 and June 1,2021.All data of the study participants were recorded,and all patients received intravenous high-dose corticosteroid pulse therapy.The Ordinal Scale for Clinical Improvement(OSCI),Charlson Comorbidity Index and Total Severity Score were calculated.Univariate and multivariate Cox regression models were performed to evaluate the clinical and laboratory parameters that may affect the 28-day mortality.Results:A total of 126 patients were included in the analysis.The 28-day mortality rate of the patients was 22.2%.Laboratory and clinical improvement were observed in 77.8%(98/126)of patients after high-dose corticosteroid pulse therapy.There was a statistically significant difference between the survivors and non-survivors in terms of age,platelet count,neutrophil/lymphocyte ratio,and OSCI,Charlson Comorbidity Index,and Total Severity Score(P<0.001).Multivariate Cox regression analysis revealed that age[HR 1.047(95%CI 1.01-1.08)],use of prophylactic anticoagulation[HR 0.838(95%CI 0.79-0.89)],and bacterial co-infection[HR 3.966(95%CI 1.40-11.21)]were significant determinants of mortality.Early C-reactive protein(CRP)response,decreased oxygen requirement,and improving respiratory rate/OSCI scores after administration of high-dose corticosteroid pulse therapy could contribute to clinical improvement.Conclusions:CRP response,needed oxygen and OSCI scores can be used as prognostic factors to select patients who will benefit from high-dose corticosteroid pulse therapy.

Ozone(O) gas is the triatomic state of oxygen and it is used as a disinfection agent due to its strong oxidizing effect, since its discovery in the mid-nineteenth century. Ozone therapy is also an alternative therapeutic approach for some diseases like circulatory disorders, AIDS, asthma, cardiovascular diseases, and certain types of cancer by increasing the oxygen levels in the blood by external addition of ozone to the body. In this study, the therapeutic potential of ozone therapy was examined by inhibiting the growth of breast cancer cells in a dose-dependent procedure. Ozone concentrations varying from 5 to 20 μg/ml were applied to the MDA-MB-231, human breast adenocarcinoma and HUVEC, human umbilical vein endothelium, cell lines, and MDA cells demonstrated an increased rate of death while its migration potential decreases. RT-PCR analysis showed mRNA expression levels of pro-apoptotic genes demonstrated higher folds in MDA cells after 10 μg/ml treatment. In the same context,Annexin V/PI and cell cycle analysis also concluded that ozone therapy causes apoptotic cell death on breast tumor cells. The use of ozone therapy for cancer treatment requires further and extensive research. However, this research has shown that ozone therapy is a promising source for cancer treatment in a way by inhibiting the proliferation of breast tumor cells.
