Wheat-rye T1BL·1RS translocation lines are widely used,especially in China,but their processing quality is generally poor.An interfering expression vector targeting theω-secalin genes was constructed with the 1Bx7 seed-specific promoter.Biolistic-mediated genetic transformation of the wheat cultivar KN199 carrying the T1BL·1RS translocation generated 10 transgenic lines.Two representative transgenic lines,8-2 and 13-7,were selected for analysis.Compared with the control,the two transformants showed an up to 4.5-fold decrease in totalω-secalins and various levels of decrease inω-gliadins,γ-gliadins,and low-molecular-weight glutenins.A decrease in high molecular weight(HMW)glutenin 1Bx7 was detected only in 8-2,owing possibly to promoter methylation.Increased levels ofα-gliadins were observed in both transformants,but increased levels of HMW glutenins were observed only in 13-7.Line 13-7 showed increases in gluten index,Zeleny sedimentation value,stabilization time,and maximum resistance.Its bread volume was 849.6 mL,an 11.9%increase over that of the control.Line 8-2 showed decreases in these parameters,but its total cake-making quality score was 88,an 17.3%increase over that of the control.The study demonstrates that the same RNAi construct may produce different effects on wheat processing quality and highlights the influence of the vector promoter in RNA interference.

目的:探讨人类免疫缺陷病毒(HIV)/艾滋病(AIDS)患者的快感缺失的相关因素。方法:选取某传染病医院HIV/AIDS患者237例,采用自编一般资料调查表、时间性愉快体验量表(TEPS)、自我接纳量表(SAQ)、歧视知觉量表(SIS)、领悟社会支持量表(PSSS)进行调查。结果:患者TEPS得分为(73.4±16.1)。逐步线性回归分析显示PSSS总分、受教育程度、个人月收入与TEPS总分正向关联(β=0.41、5.17、4.63),年龄与TEPS总分负向关联(β=-0.30)。结论:HIV/AIDS患者的快感缺失情况需引起重视,患者的快感缺失与个人月收入、受教育程度、年龄、领悟社会支持相关。

[Objectives]This study was conducted to improve the genetic transformation rate of maize. [Methods]The seeds of maize ＂ Zheng 58＂ as the experimental material were germinated and treated under freezing temperature of-18 ℃ for 30 and 35 min. The EGFP gene was transformed into the growth points of these seed buds by Agrobacterium tumefaciens EHA105. The transformation effect was determined by fluorescence protein detection on transformed buds. [Results]After a certain period of treatment at-18 ℃ following germination and transformation of maize bud growth points with A. tumefaciens,although the survival rate of maize buds was reduced,the percentage of transformed cells significantly increased. [Conclusions]Low temperature treatment can improve the transformation efficiency of A. tumefaciens to the growth point of maize bud.

[Objectives]The effect of acetosyringone seed soaking on the transformation of maize seed buds was analyzed,so as to improve the genetic transformation efficiency of maize and to provide technical support for transgenic breeding of maize.[Methods]The seeds of the"Zheng 58"maize inbred line were used as experimental materials.When the seeds were germinated,AS was added to the water at concentrations of 70,140,210,and 280μmol/L,respectively,and the seeds germinated without the addition of AS served as the CK.The Agrobacterium-mediated method was used to transform bud growth points of maize seeds,and green fluorescent protein detection was performed on the young shoots transformed with EGFP(enhanced green fluorescent protein)gene.The effect of soaking seeds with acetosyringone solution on the transformation of maize bud growth points by Agrobacterium was studied according to the detection results.[Results]Soaking seeds in acetosyringone solutions for germination had the effect of inhibiting the germination of maize seeds and inhibiting sprout elongation,and the higher the concentration of acetosyringone,the stronger the inhibition.When the concentration of acetosyringone solution was 280μmol/L,the germination rate of seeds was only 36.2%of the CK,while soaking seeds with 70-140μmol/L acetosyringone solution for germination could not only ensure a higher germination rate of maize seeds,but also significantly increased the transformation efficiency of maize bud growth points.When the seeds were soaked with 70μmol/L acetosyringone solution for germination,the positive rate of transformed maize buds was the highest,reaching 32.1%.[Conclusions]When maize bud growth points were used as the receptor of Agrobacterium transformation,soaking seeds with 70-140μmol/L acetosyringone for germination basically did not affect the germination of seeds,and was beneficial to the activation of Agrobacterium,thereby promoting the transformation.

[Objectives] This study was conducted to improve the efficiency of genetic transformation using growing points of corn sprouts. [Methods] A mixed liquid of plant hormones 6-BA and KT at a suitable concentration and ratio was added dropwise to the growing points of corn sprouts, followed by culture under appropriate conditions. [Results] Corn sprouts could be induced to form multiple plants, thereby improving the transformation efficiency of growing points in corn sprouts. [Conclusions] This study provides a breeding method using growing points of corn sprouts as transformation receptors for direct plantlet formation without tissue culture by the Agrobacterium transgenic technology.

One of the greatest challenges in the agroecosystem is to improve cropland intensification while preserving agroecosystem services.While many studies have investigated the effect of cropland intensification on agroecosystem service,the interactive coupling and coordination among these factors remain largely unexplored.In view of this,this study performed a case study of the Loess Plateau in Shaanxi Province,China and constructed comprehensive evaluation models to quantify the cropland intensification and agroecosystem service in this area.Balance analysis and the coupling coordination degree model were used to evaluate the interactive relationship between cropland intensification and agroecosystem service,and statistical analysis and spatial autocorrelation were used to analyze the spatial characteristics and potential mechanism of the coupling coordination.Results show that both the cropland intensification and agroecosystem service in the study area were relatively low yet gradually increased from 2000 to 2020.Agroecosystem service lag was identified as the dominant unbalanced development type.Improving the supply capacity of agroecosystem services plays a key role in the balanced development of cropland in the Loess Plateau.The coupling coordination degree between cropland intensification and agroecosystem service ranges from basic coordination to serious incoordination.Therefore,cropland intensification practices in the area should be optimized to enhance this coordination degree.An upward trend was also observed in the coupling coordination degree from2000 to 2020.The withdrawal of marginal cropland in the Grain for Green program is one of the most important reasons for this trend,especially for the northern region.Around 83.6%of the high-high clusters are concentrated in the southern region of the Loess Plateau,whereas 70.5%of the low-low clusters are distributed in the northern region.These clustering characteristics are mainly attributed to the environmental suitability of these areas for agriculture and

CEOs’Diverse Career Experiences and the Effectiveness of Compensation Contracts:Evidence from Compensation Stickiness

CEO compensation stickiness represents an important indicator to measure the effectiveness of compensation contracts.This study uses CEO career experience data and compensation stickiness data from Shanghai and Shenzhen A-share listed companies from 2015 to 2020 to investigate the compensation contracts’effectiveness of CEOs with diverse career experiences.The findings are as follows:1)Compensation stickiness is more pronounced for CEOs with diverse career experiences.According to the mechanism test,these CEOs with diverse career experiences can obtain compensation incentives by reducing corporate uncertainty perception and improving total factor productivity.This approach leads to increased compensation stickiness and the effectiveness of compensation contracts.CEOs with diverse career experiences may receive excess compensation by raising agency costs,which intensifies compensation stickiness and weakens the effectiveness of compensation contracts.2)Compensation stickiness of CEOs with diverse career experiences is more significant in companies with lower investor protection,which brings about less effective compensation contracts.In contrast,in companies with higher diversification,the compensation stickiness of CEOs with diverse career experiences is more significant,which delivers more effective compensation contracts.The conclusions deepen the research of CEO compensation contracts and provide a helpful reference for CEO compensation management practices.

Effect of initial ammonium concentration on a one-stage partial nitrification/anammox biofilm system:Nitrogen removal performance and the microbial community

One-stage partial nitrification coupled with anammox(PN/A)technology effectively reduces the energy consumption of a biological nitrogen removal system.Inhibiting nitrite-oxidizing bacteria(NOB)is essential for this technology to maintain efficient nitrogen removal performance.Initial ammonium concentration(IAC)affects the degree of inhibited NOB.In this study,the effect of the IAC on a PN/A biofilm was investigated in a moving bed biofilm reactor.The results showed that nitrogen removal efficiency decreased from 82.49%±1.90%to 64.57%±3.96%after the IAC was reduced from 60 to 20 mg N/L,while the nitrate production ratio increased from 13.87%±0.90%to 26.50%±3.76%.NOB activity increased to1,133.86 mg N/m^(2)/day after the IAC decreased,approximately 4-fold,indicating that the IAC plays an important inhibitory role in NOB.The rate-limiting step in the mature biofilm of the PN/A system is the nitritation process and is not shifted by the IAC.The analysis of the microbial community structure in the biofilm indicates that the IAC was the dominant factor in changes in community structure.Ca.Brocadia and Ca.Jettenia were the main anammox bacteria,and Nitrosomonas and Nitrospira were the main AOB and NOB,respectively.IAC did not affect the difference in growth between Ca.Brocadia and Ca.Jettenia.Thus,modulating the IAC promoted the PN/A process with efficient nitrogen removal performance at medium to low ammonium concentrations.

Geometric error is the main factor affecting the machining accuracy of hybrid machine tools.Kinematic calibration is an effective way to improve the geometric accuracy of hybrid machine tools.The necessity to measure both position and orientation at each pose,as well as the instability of identification in case of incomplete measurements,severely affects the application of traditional calibration methods.In this study,a kinematic calibration method with high measurement efficiency and robust identification is proposed to improve the kinematic accuracy of a five-axis hybrid machine tool.First,the configuration is introduced,and an error model is derived.Further,by investigating the mechanism error characteristics,a measurement scheme that only requires tool centre point position error measurement and one alignment operation is proposed.Subsequently,by analysing the effects of unmeasured degrees of freedom(DOFs)on other DOFs,an improved nonlinear least squares method based on virtual measurement values is proposed to achieve stable parameter identification in case of incomplete measurement,without introducing additional parameters.Finally,the proposed calibration method is verified through simulations and experiments.The proposed method can efficiently accomplish the kinematic calibration of the hybrid machine tool.The accuracy of the hybrid machine tool is significantly improved after calibration,satisfying actual aerospace machining requirements.

Cancer is a leading cause of death worldwide,and a series of strategies has been reported for tumor-specific therapy.Currently,chemodynamic therapy(CDT)has become a research hotspot for antitumor treatment due to its advantages of high specificity,endogenous stimulation,and high biosafety.However,the therapeutic effects of CDT are normally limited in the complex tumor microenvironment(TME),such as insufficient acidity,tumor hypoxia,low hydrogen peroxide(H2O2),and high glutathione(GSH).Consequently,different kinds of multifunctional nanomaterials have been designed to manipulate TME conditions,which provided more opportunities to improve the efficiency of CDT.This review focuses on nanomaterial-based strategies for enhancing CDT through manipulating TME.Upon CDT enhancements,this review would provide a reference for the future development of efficient CDT nanomaterials.
