Development of a cell adhesion-based prognostic model for multiple myeloma:Insights into chemotherapy response and potential reversal of adhesion effects

Multiple myeloma(MM)is a hematologic malignancy notorious for its high relapse rate and development of drug resistance,in which cell adhesion-mediated drug resistance plays a critical role.This study integrated four RNA sequencing datasets(CoMMpass,GSE136337,GSE9782,and GSE2658)and focused on analyzing 1706 adhesionrelated genes.Rigorous univariate Cox regression analysis identified 18 key prognosis-related genes,including KIF14,TROAP,FLNA,MSN,LGALS1,PECAM1,and ALCAM,which demonstrated the strongest associations with poor overall survival(OS)in MM patients.To comprehensively evaluate the impact of cell adhesion on MM prognosis,an adhesion-related risk score(ARRS)model was constructed using Lasso Cox regression analysis.The ARRS model emerged as an independent prognostic factor for predicting OS.Furthermore,our findings revealed that a heightened cell adhesion effect correlated with tumor resistance to DNA-damaging drugs,protein kinase inhibitors,and drugs targeting the PI3K/Akt/mTOR signaling pathway.Nevertheless,we identified promising drug candidates,such as tirofiban,pirenzepine,erlotinib,and bosutinib,which exhibit potential in reversing this resistance.In vitro,experiments employing NCIH929,RPMI8226,and AMO1 cell lines confirmed that MM cell lines with high ARRS exhibited poor sensitivity to the aforementioned candidate drugs.By employing siRNA-mediated knockdown of the key ARRS model gene KIF14,we observed suppressed proliferation of NCIH929 cells,along with decreased adhesion to BMSCs and fibronectin.This study presents compelling evidence establishing cell adhesion as a significant prognostic factor in MM.Additionally,potential molecular mechanisms underlying adhesion-related resistance are proposed,along with viable strategies to overcome such resistance.These findings provide a solid scientific foundation for facilitating clinically stratified treatment of MM.

Detecting genes that affect specific traits(such as human diseases and crop yields)is important for treating complex diseases and improving crop quality.A genome-wide association study(GWAS)provides new insights and directions for understanding complex traits by identifying important single nucleotide polymorphisms.Many GWAS summary statistics data related to various complex traits have been gathered recently.Studies have shown that GWAS risk loci and expression quantitative trait loci(e QTLs)often have a lot of overlaps,which makes gene expression gradually become an important intermediary to reveal the regulatory role of GWAS.In this review,we review three types of gene-trait association detection methods of integrating GWAS summary statistics and e QTLs data,namely colocalization methods,transcriptome-wide association study-oriented approaches,and Mendelian randomization-related methods.At the theoretical level,we discussed the differences,relationships,advantages,and disadvantages of various algorithms in the three kinds of gene-trait association detection methods.To further discuss the performance of various methods,we summarize the significant gene sets that influence highdensity lipoprotein,low-density lipoprotein,total cholesterol,and triglyceride reported in 16 studies.We discuss the performance of various algorithms using the datasets of the four lipid traits.The advantages and limitations of various algorithms are analyzed based on experimental results,and we suggest directions for follow-up studies on detecting gene-trait associations.

Multiple grain-size fraction analysis of heavy minerals and the provenance identification of sediments from the abandoned Huanghe River,eastern China

The quantitative analysis of sediment sources in a sink is an important scientific topic and challenge in provenance research.The characteristics of heavy minerals,combined with the geochemical constituents of detrital grains,provide a reliable provenance-tracing approach.We developed a mineral identification method to analyze the multiple grain-size fraction of sediments,from which the elemental geochemistry of hornblende was used to compare the characteristics of sediments from the Huaihe River and Huanghe(Yellow)River in eastern China.Elements that were statistically identified as being able to discriminate sediment provenance were employed to perform a quantitative analysis of the sources of sediments of the abandoned Huanghe River.Results reveal that the Huaihe River is characterized by a high amphibole content of>60%and that the Huanghe and abandoned Huanghe rivers have greater abundances of limonite and carbonate minerals compared with those of the Huaihe River.The contents of trace elements and rare earth elements in hornblende show that the sediments of the abandoned Huanghe River are similar to those of the Huanghe River but different from those of the Huaihe River.Furthermore,chemical mass balance was used to calculate the relative contributions of different provenances of sediment from the abandoned Huanghe River,and nine trace elements of hornblende were identified as discriminators of provenance.Approximately 2%of the hornblende in the abandoned Huanghe River is derived from the Huaihe River and 98%from the Huanghe River.Considering the proportion of hornblende in the total sediment,it is inferred that the contribution of Huaihe River sediment to the abandoned Huanghe River is approximately 0.5%.This study shows that mineral analysis using multiple grain-size fractions(within the wide range of 1Φto 6Φ)with assessment in elemental geochemistry of hornblende can characterize the provenance of fluvial material in coastal zones.

Design of artificial ligaments possessing both osteogenic activity and antibacterial effect that promotes ligament-bone healing and prevents bacterial infection in bone tunnels for anterior cruciate ligament(ACL)reconstruction remains a significant challenge.In this study,black tantalic oxide(BTO)submicro-particles with oxygen vacancies and structure defects were fabricated by using traditional white tan-talic oxide(WTO)through magnesium thermal reduction(MTR)method,and BTO was coated on polyetheretherketone(PEEK)fibers(PKF),which were woven into fabrics(PBT)as artificial ligaments.PBT with BTO coating exhibited excellent photothermal performance,which possessed not only antibac-terial effects in vitro but also anti-infective ability in vivo.PBT with optimized surface properties(e.g.,submicro-topography and hydrophilicity)not only significantly facilitated rat bone mesenchymal stem cells(BMSC)responses(e.g.,proliferation and osteogenic differentiation)in vitro but also stimulated new bone formation for ligament-bone healing in vivo.The presence of oxygen vacancies and structure de-fects in BTO did not change the surface properties and osteogenic activity of BPT while displaying an outstanding photothermal antibacterial effect.In summary,BPT with osteogenic activity and photother-mal antibacterial effect promoted bone regeneration and prevented bacterial infection,thereby promoting ligament-bone healing.Therefore,PBT would have tremendous potential as a novel artificial ligament for ACL reconstruction.

Symplastic communication in the root cap directs auxin distribution to modulate root development

Root cap not only protects root meristem,but also detects and transduces the signals of environmental changes to affect root development.The symplastic communication is an important way for plants to transduce signals to coordinate the development and physiology in response to the changing enviroments.However,it is unclear how the symplastic communication between root cap cells affects root growth.Here we exploit an inducible system to specifically block the symplastic communication in the root cap.Transient blockage of plasmodesmata(PD)in differentiated collumella cells severely impairs the root development in Arabidopsis,in particular in the stem cell niche and the proximal meristem.The neighboring stem cell niche is the region that is most sensitive to the disrupted symplastic communication and responds rapidly via the alteration of auxin distribution.In the later stage,the cell division in proximal meristem is inhibited,presumably due to the reduced auxin level in the root cap.Our results reveal the essential role of the differentiated collumella cells in the root cap mediated signaling system that directs root development.

The declines in laying performance during the late productionperiod have adverse effects on the length of theproduction cycle. Improving the nutrition of laying hens is a crucialmeasure to reverse this declination. Thisstudy investigatedthe effect of seleniumyeast (SY)on egg production, ileal gene expressionandmicrobiota, aswell as elucidating their associations in aged laying hens. A total of 375 Jinghong laying hens at 76weeks oldwere randomly assigned into 5 dietary treatments, which included a selenium-deficient basal diet based oncorn-soybean meal, and dietary supplementation of SY at 0.15, 0.30 and 0.45 mg/kg, and sodium selenite at0.45mg/kg.The results showed that SYamelioratedthe depressionin aged layingperformance inthe 0.30mg/kg group (P < 0.01). Selenium yeast significantly increased ileum selenium concentration (P < 0.05), and SYgroups had higher seleniumdeposition efficiency than the sodiumselenite group. Functional enrichment andShort Time-series Expression Miner (STEM) analysis indicated that SY activated metabolic progress (e.g.,glycerolipidmetabolism, glycerophospholipidmetabolism, andfattyacidmetabolism),immune responseandoxidative stress response. Four hub genes including thioredoxin reductase 1 (TXNRD1), dihydrolipoamidedehydrogenase (DLD), integrin linked kinase (ILK) and leucine zipper tumor suppressor 2 (LZTS2) wereinvolved in intestinal metabolismwhich was closely associated with selenium deposition/status. Moreover,the relative abundance of Veillonella, Turicibacter and Lactobacillus was significantly increased, but the relativeabundance of Stenotrophomonas was significantly decreased by SY supplementation. Multi-omics dataintegration and Canonical correspondence analysis (CCA) showed that both the ileumselenium content andthe laying rate were highly correlated with pathways and bacteria enriched in metabolism and immuneresponse. Detecting gravitational wave with an interferometric seismometer array on lunar nearside

Our Moon is by far the easiest natural, extraterrestrial celestial object for us to reach, and many space agencies, in particular the China National Space Administration, have publicized plans for manned missions there. Such high-profile projects must yield commensurate scientific outputs to justify their cost to the taxpayers. An important criterion for the selection of candidate experiments is the extent to which the Moon plays an integral and irreplaceable role, as opposed to offering only marginal advantages over Earth-orbit, space-based alternatives. In this study, we propose an experiment that excels in this regard, which considers the Moon as part of the experimental apparatus. Specifically, the Moon acts as a Weber bar-style resonant mass that responds to deci-Hertz gravitational waves(GWs). GW-excited lunar vibrations can be picked up by an array of(prefabricated on Earth and easy to transport to the Moon) small laser interferometric seismometers yielding GW detection. Such a setup would plug a gap in the GW frequency range already covered by the existing or underdevelopment detectors, and it can inform us on important astrophysics topics, such as the progenitors of Type Ia supernovae(SNe Ia) or intermediate mass black holes. We provide the instrument design as well as its deployment strategy, and we also summarize relevant science cases according to the estimated viable sensitivity.

Seasonal Variations of Observed Raindrop Size Distribution in East China

Seasonal variations of rainfall microphysics in East China are investigated using data from the observations of a twodimensional video disdrometer and a vertically pointing micro rain radar. The precipitation and rain drop size distribution(DSD) characteristics are revealed for different rain types and seasons. Summer rainfall is dominated by convective rain,while during the other seasons the contribution of stratiform rain to rainfall amount is equal to or even larger than that of convective rain. The mean mass-weighted diameter versus the generalized intercept parameter pairs of convective rain are plotted roughly around the "maritime" cluster, indicating a maritime nature of convective precipitation throughout the year in East China. The localized rainfall estimators, i.e., rainfall kinetic energy–rain rate, shape–slope, and radar reflectivity–rain rate relations are further derived. DSD variability is believed to be a major source of diversity of the aforementioned derived estimators. These newly derived relations would certainly improve the accuracy of rainfall kinetic energy estimation, DSD retrieval, and quantitative precipitation estimation in this specific region.
