Proper regulation of synapse formation and elimination is critical for establishing mature neuronal circuits and maintaining brain function.Synaptic abnormalities,such as defects in the density and morphology of postsynaptic dendritic spines,underlie the pathology of various neuropsychiatric disorders.Protocadherin 17(PCDH17)is associated with major mood disorders,including bipolar disorder and depression.However,the molecular mechanisms by which PCDH17 regulates spine number,morphology,and behavior remain elusive.In this study,we found that PCDH17 functions at postsynaptic sites,restricting the number and size of dendritic spines in excitatory neurons.Selective overexpression of PCDH17 in the ventral hippocampal CA1 results in spine loss and anxiety-and depression-like behaviors in mice.Mechanistically,PCDH17 interacts with actin-relevant proteins and regulates actin filament(F-actin)organization.Specifically,PCDH17 binds to ROCK2,increasing its expression and subsequently enhancing the activity of downstream targets such as LIMK1 and the phosphorylation of cofilin serine-3(Ser3).Inhibition of ROCK2 activity with belumosudil(KD025)ameliorates the defective F-actin organization and spine structure induced by PCDH17 overexpression,suggesting that ROCK2 mediates the effects of PCDH17 on F-actin content and spine development.Hence,these findings reveal a novel mechanism by which PCDH17 regulates synapse development and behavior,providing pathological insights into the neurobiological basis of mood disorders.

Investigation of Solidification and Segregation Characteristics of Cast Ni-Base Superalloy K417G

Differential scanning calorimetry （DSC） analysis, isothermal solidification experiment and Thermo-Calc simulation were employed to investigate solidification characteristics of K417G Ni-base superalloy. Elec- tron probe microanalysis （EPMA） was employed to analyze the segregation characteristics. Liquidus, solidus and the formation temperatures of main phases were measured. In the process of solidification, the volume fraction of liquid dropped dramatically in the initial stage, while the dropping rate became very low in the final stage due to severe segregation of positive segregation elements into the residual liquid. The solidification began with the formation of primary γ. Then with solidification proceeding, Ti and Mo were enriched in the liquid interdendrite, which resulted in the precipitation of MC carbides in the interdendrite. A1 accumulated into liquid at the initial stage, but gathered to solid later due to the precipitation of γ/γ＇ eutectic at the intermediate stage of solidification. However, Co tended to segregate toward the solid phase. In the case of K417G alloy, combining DSC analysis and isothermal solidification experiment is a good way to investigate the solidification characteristics. Thermo-Calc simulation can serve as reference to investigate K417G alloy.

A wide spectrum of metabolites(mainly,the three major nutrients and their derivatives)can be sensed by specific sensors,then trigger a series of signal transduction pathways and affect the expression levels of genes in epigenetics,which is called metabolite sensing.Life body regulates metabolism,immunity,and inflammation by metabolite sensing,coordinating the pathophysiology of the host to achieve balance with the external environment.Metabolic reprogramming in cancers cause different phenotypic characteristics of cancer cell from normal cell,including cell proliferation,migration,invasion,angiogenesis,etc.Metabolic disorders in cancer cells further create a microenvironment including many kinds of oncometabolites that are conducive to the growth of cancer,thus forming a vicious circle.At the same time,exogenous metabolites can also affect the biological behavior of tumors.Here,we discuss the metabolite sensing mechanisms of the three major nutrients and their derivatives,as well as their abnormalities in the development of various cancers,and discuss the potential therapeutic targets based on metabolite-sensing signaling pathways to prevent the progression of cancer.

为密织防范网络,清理挖矿木马病毒,有效治理校园网虚拟货币挖矿行为,提出了一种校园网恶意挖矿行为的检测与阻断模型。该模型采用基于签名的深度包检测技术,结合动态威胁情报,建立了挖矿协议的状态机模型,对报文进行深度包分析,以识别挖矿协议,在校园网出口实现挖矿流量的检测、识别与阻断。实践证明,该模型能够实时检测出虚拟货币相关流量,动态拦截受害矿机与矿池的通信流量,并实时定位受感染主机,有效地遏制校园网的恶意挖矿行为。

Application potential of carbon nanotubes in water treatment:A review

Water treatment is the key to coping with the conflict between people＇s increasing demand for water and the world-wide water shortage. Owing to their unique and tunable structural, physical, and chemical properties, carbon nanotubes （CNTs） have exhibited great potentials in water treatment. This review makes an attempt to provide an overview of potential solutions to various environmental challenges by using CNTs as adsorbents, catalysts or catalyst support, membranes, and electrodes. The merits of incorporating CNT to conventional water-treatment material are emphasized, and the remaining challenges are discussed.

Powdery mildew of wheat is a destructive disease seriously threatening yield and quality worldwide.Comprehensive dissection of new resistance-related loci/genes is necessary to control this disease.LS5082 is a Chinese wheat breeding line with resistance to powdery mildew.Genetic analysis,using the populations of LS5082 and three susceptible parents(Shannong 29,Shimai 22 and Huixianhong),indicated that a single dominant gene,tentatively designated PmLS5082,conferred seedling resistance to different Blumeria graminis f.***.tritici(Bgt)isolates.Bulked segregant RNA-Seq was carried out to map PmLS5082 and to profile differentially expressed genes associated with PmLS5082.PmLS5082 was mapped to a 0.7 cM genetic interval on chromosome arm 2BL,which was aligned to a 0.7 Mb physical interval of 710.3–711.0 Mb.PmLS5082 differs from the known powdery mildew(Pm)resistance genes on chromosome arm 2BL based on their origin,chromosome positions and/or resistance spectrum,suggesting PmLS5082 is most likely a new Pm gene/allele.Through clusters of orthologous groups and kyoto encyclopedia of genes and genomes analyses,differentially expressed genes(DEGs)associated with PmLS5082 were profiled.Six DEGs in the PmLS5082 interval were confirmed to be associated with PmLS5082 via qPCR analysis,using an additional set of wheat samples and time-course analysis postinoculation with Bgt isolate E09.Ten closely linked markers,including two kompetitive allele-specific PCR markers,were confirmed to be suitable for marker-assisted selection of PmLS5082 in different genetic backgrounds,thus can be used to detect PmLS5082 and pyramid it with other genes in breeding programs.

Nitrated and parent PAHs in the surface water of Lake Taihu,China:Occurrence,distribution,source,and human health risk assessment

Nitrated polycyclic aromatic hydrocarbons(NPAHs)have toxic potentials that are higher than those of their corresponding parent polycyclic aromatic hydrocarbons(PAHs)and thus have received increasing attention in recent years.In this study,the occurrence,distribution,source,and human health risk assessment of 15 NPAHs and 16 PAHs were investigated in the surface water from 20 sampling sites of Lake Taihu during the dry,normal,and flood seasons of 2018.The PAH concentrations ranged from 255 to 7298 ng/L and the NPAH concentrations ranged from not-detected(ND)to 212 ng/L.Among the target analytes,2-nitrofluorene(2-n Flu)was the predominant NPAH,with a detection frequency ranging from 85%to 90%and a maximum concentration of 56.2 ng/L.The three-ringed and four-ringed NPAHs and PAHs comprised the majority of the detected compounds.In terms of seasonal variation,the highest levels of the NPAHs and PAHs were in the dry season and flood season,respectively.Diagnostic ratio analysis indicated that the prime source of NPAHs was direct combustion,whereas in the case of PAHs the contribution was predominantly from a mixed pattern including pollution from unburned petroleum and petroleum combustion.The human health risk of NPAHs and PAHs was evaluated using a lifetime carcinogenic risk assessment model.The carcinogenic risk level of the targets ranged from 2.09×10^(-7)to 5.75×10^(-5)and some surface water samples posed a potential health risk.

能量管理是确保无源节点在不稳定供能环境下稳健运行的必要条件,但也带来了额外的计算负担,致使能量效率下降。为解决这一问题,研究了无源节点的能量理论,并针对性地提出无须本地计算辅助的“边端一体化”能量管理方案。首先,将高开销的细粒度能量规划从节点(端)解耦至网关(边),以降低节点上能量管理的复杂度。同时,创新性地提出了无采样感知和免计算决策电路的设计,解决了节点上无计算单元辅助下能量探测和决策执行所面临的两大关键技术挑战。至此,节点无须额外分配计算资源用于能量管理,从而在原理上避免了传统方式因将能量管理作为“附加任务”执行所导致的高开销问题。此外,在网关侧建立了远程开销预算模型,为多样化任务的供能需求实现了远程动态匹配。最后,实现了设计原型,并通过原型测试验证了设计的可行性。结果表明,与四种参考方式对比,该设计可降低92%的能量管理开销,并能对多变的供能环境作出灵活响应。

以低成本、易变形的Ti-42Al-5Mn合金(原子分数,%)为研究对象,利用DSC、EPMA、EBSD及Pandat热力学计算软件系统研究了W含量(0.5%~1.0%)对合金相变行为和组织的影响。结果表明,随着W含量由0.5%提高至1.0%,合金β单相区开始温度(T_(β))和γ相溶解温度(T_(γ-solv))几乎未发生变化,而共析转变温度(T_(eut))稍有增加。W添加会在一定程度上影响合金固态相变路线,随着W含量增加至0.5%,合金近服役温度下的平衡相由α_(2)+γ+Laves逐渐演变为β_(o)+α_(2)+γ+Laves。W添加会对合金片层组织特征产生显著影响。在(γ+α+β)三相区处理时,随着W含量的提高,合金缓冷后组织中片层含量显著降低,当W含量为0.8%和1.0%时,合金组织以γ晶粒和βo相为主,几乎消除了α_(2)/γ层片组织。在(α+β)两相区处理时,合金缓冷后均为近片层组织,且随着W含量由0.5%提高至1.0%,片层组织晶团得到明显细化。

设计并制备了一种新型低成本、易热变形的Ti-43Al-3.5Mn-0.5W(at%)合金锻锭,并对其组织、室/高温拉伸力学性能、抗高温氧化性、热变形能力进行了系统分析。结果表明,与Ti-42Al-5Mn相比,研制的合金强度、抗高温氧化性、热变形能力更佳,且该合金α2和βo相中具有更低的Mn含量,降低了合金近服役温度下富锰Laves相的析出倾向。合金的固态相变路线为:β→β+α→β+α+γ→β+βo+α+α2+γ→βo+α2+γ,其中γ相完全溶解的温度(T_(γ-solv))约为1250℃,β单相区温度(Tβ或Tα)约为1360℃。锻态合金显微组织为α2/γ片层和片层界面处大量βo和γ混合相,高温强度降低明显。通过两步热处理,锻态合金的高温强度和稳定性均有一定提升,这主要归结为片层组织含量的提高和片层晶团尺寸的细化。1260℃/0.5 h/AC+800℃/3 h/FC热处理合金800℃的屈服强度、抗拉强度和延伸率分别为320 MPa、555 MPa、16%。
