The osteoclastic activity in apical distal region of molar mesial roots affects orthodontic tooth movement and root resorption in rats

The utilization of optimal orthodontic force is crucial to prevent undesirable side effects and ensure efficient tooth movement during orthodontic ***,the sensitivity of existing detection techniques is not sufficient,and the criteria for evaluating optimal force have not been yet ***,by employing 3D finite element analysis methodology,we found that the apical distal region(A-D region)of mesial roots is particularly sensitive to orthodontic force in ***-resistant acidic phosphatase(TRAP)-positive osteoclasts began accumulating in the A-D region under the force of 40 grams(g),leading to alveolar bone resorption and tooth *** the force reached 80 g,TRAP-positive osteoclasts started appearing on the root surface in the A-D ***,micro-computed tomography revealed a significant root resorption at 80 ***,the A-D region was identified as a major contributor to whole root *** was determined that 40 g is the minimum effective force for tooth movement with minimal side effects according to the analysis of tooth movement,inclination,and *** findings suggest that the A-D region with its changes on the root surface is an important consideration and sensitive indicator when evaluating orthodontic forces for a rat ***,our investigations into this region would aid in offering valuable implications for preventing and minimizing root resorption during patients’orthodontic treatment.

Cardiovascular disease is still the leading cause of mortality *** endothelial dysfunction is viewed as the initial step of most cardiovascular *** studies have indicated that periodontal pathogens,especially Porphyromonas gingivalis,are closely correlated with vascular endothelial homeostasis,but the function of *** and the underlying mechanisms are still *** illuminate the effects and elucidate the mechanisms of *** on endothelial structural integrity,we developed *** infection models in vivo and in *** cell proliferation,differentiation and apoptosis were ***,we showed that *** can impair endothelial integrity by inhibiting cell proliferation and inducing endothelial mesenchymal transformation and apoptosis of endothelial cells,which reduce the cell levels and cause the endothelium to lose its ability to repair itself.A mechanistic analysis showed that TLR antagonist or NF-κB signalling inhibitor can largely rescue the damaged integrity of the endothelium caused by ***,suggesting that TLR-NF-κB signalling plays a vital role in vascular endothelial homeostasis destroyed by *** results suggest a potential intervention method for the prevention and treatment of cardiovascular disease.

CD36 is a highly glycosylated integral membrane protein that belongs to the scavenger receptor class B family and regulates the pathological progress of metabolic ***36 was recently found to be widely expressed in various cell types in the nervous system,including endothelial cells,pericytes,astrocytes,and ***36 mediates a number of regulatory processes,such as endothelial dysfunction,oxidative stress,mitochondrial dysfunction,and inflammatory responses,which are involved in many central nervous system diseases,such as stroke,Alzheimer’s disease,Parkinson’s disease,and spinal cord ***36 antagonists can suppress CD36 expression or prevent CD36 binding to its ligand,thereby achieving inhibition of CD36-mediated pathways or ***,we reviewed the mechanisms of action of CD36 antagonists,such as Salvianolic acid B,tanshinone IIA,curcumin,sulfosuccinimidyl oleate,antioxidants,and small-molecule ***,we predicted the structures of binding sites between CD36 and *** sites can provide targets for more efficient and safer CD36 antagonists for the treatment of central nervous system diseases.

目的建立一种基于两对交叉引物PCR技术(PCR with confronting two-pair primers,PCR-CTPP)同时快速检测非缺失型α地中海贫血WS、QS和CS三种突变类型的方法及其在临床中的应用。方法根据α珠蛋白基因中发生WS、QS和CS突变的区域基因序列,分别针对单个位点设计两对引物即通用外引物和特异性引物,经过聚合酶链反应、电泳鉴别基因型,同时检测非缺失型α地中海贫血。将该方法与PCR反向斑点杂交(PCR-RDB)技术比较,验证其检测准确性。结果PCR-CTPP方法可用于准确检测非缺失型α地中海贫血的WS、QS和CS突变类型,结果显示与常规非缺失型α地中海贫血PCR-RDB检测方法一致。结论PCR-CTPP技术检测非缺失型α地中海贫血WS、QS、CS三种突变位点,简单快速、经济省力,可用于非缺失型α地中海贫血的快速筛查和分子流行病学研究,适合在一般实验室推广应用。适用于非缺失型α地中海贫血的人群筛查及婚前或产前筛查,适合各个层次医院应用。

RNA sequencing of exosomes secreted by fibroblast and Schwann cells elucidates mechanisms underlying peripheral nerve regeneration

Exosomes exhibit complex biological functions and mediate a variety of biological processes,such as promoting axonal regeneration and functional recove ry after *** non-coding RNAs(IncRNAs)have been reported to play a crucial role in axonal *** r,the role of the IncRNA-microRNAmessenger RNA(mRNA)-competitive endogenous RNA(ceRNA)network in exosome-mediated axonal regeneration remains *** this study,we performed RNA transcriptome sequencing analysis to assess mRNA expression patterns in exosomes produced by cultured fibroblasts(FC-EXOs)and Schwann cells(SCEXOs).Diffe rential gene expression analysis,Gene Ontology analysis,Kyoto Encyclopedia of Genes and Genomes analysis,and protein-protein intera ction network analysis were used to explo re the functions and related pathways of RNAs isolated from FC-EXOs and *** found that the ribosome-related central gene Rps5 was enriched in FC-EXOs and SC-EXOs,which suggests that it may promote axonal *** addition,using the miRWalk and Starbase prediction databases,we constructed a regulatory network of ceRNAs targeting Rps5,including 27 microRNAs and five *** ceRNA regulatory network,which included Ftx and Miat,revealed that exsosome-derived Rps5 inhibits scar formation and promotes axonal regeneration and functional recovery after nerve *** findings suggest that exosomes derived from fibro blast and Schwann cells could be used to treat injuries of peripheral nervous system.

Lead-acid batteries have been contributing to energy storage since they were invented,and today they still occupy an important position in the battery ***,due to the special process and operating conditions of lead-acid batteries,their failure is also a major problem that plagues *** the failure mechanism of lead-acid batteries is of great significance for improving lead-acid battery manufacturing technology and promoting the development of the energy *** failure of lead-acid batteries is mainly concentrated on softening of the positive plate,sulphation of the negative plate,corrosion of the grid,acid stratification and *** investigation of these failure modes,especially softening of the positive electrode plate—the most complicated one,has been the focus of the *** in-situ electrochemical analysis methods of lead-acid batteries,including in-situ XRD and in-situ AFM spectating techniques,will do good to the research of battery failure mechanisms.

Turbulence Statistics of Thermo-Buoyancy Supercritical Fuel Flow in a Regenerative Cooling Channel

Active regenerative cooling with supercritical hydrocarbon fuel is considered as the most promising thermal protection *** existence of buoyancy force would lead to strongly anisotropic flow and thermal transport *** is closely related to the cooling performance of the *** elucidate the mechanisms of turbulent transport,the large eddy simulation(LES) was performed to assess turbulence statistics within different turbulence *** results indicated that the buoyancy and inertial force together dominated the change of turbulent ***,the spatial thermal buoyancy effect significantly suppressed the vertical velocity *** is due to the laminar motion caused by the buoyancy force,thereby weakening the thermal *** the statistics of velocity fluctuation,it was found that the buoyancy force and inertial force greatly weaken the vertical and streamwise velocity fluctuation,*** the statistics of thermal transport,the results pointed out that the near-wall heat transport characteristics need to be paid more *** thickness of the temperature mixing boundary layer led to the attenuation of vertical heat flux,which inhibited vertical temperature diffusion and predisposed to extreme conditions of heat transfer *** results can enhance the academic understanding of the heat transfer mechanism of supercritical fluids,and give guidance for further applications of thermal protection.
