Noise-induced hearing loss is the primary non-genetic factor contributing to auditory ***,there are currently no effective pharmacological interventions for patients with noise-induced hearing ***,we present evidence suggesting that the lysine-specific demethylase 1 inhibitor–tranylcypromine is an otoprotective agent that could be used to treat noise-induced hearing loss,and elucidate its underlying regulatory *** established a mouse model of permanent threshold shift hearing loss by exposing the mice to white broadband noise at a sound pressure level of 120 d B for 4 *** found that tranylcypromine treatment led to the upregulation of Sestrin2(SESN2)and activation of the autophagy markers light chain 3B and lysosome-associated membrane glycoprotein 1 in the cochleae of mice treated with *** noise exposure group treated with tranylcypromine showed significantly lower average auditory brainstem response hearing thresholds at click,4,8,and 16 k Hz frequencies compared with the noise exposure group treated with *** findings indicate that tranylcypromine treatment resulted in increased SESN2,light chain 3B,and lysosome-associated membrane glycoprotein 1 expression after noise exposure,leading to a reduction in levels of 4-hydroxynonenal and cleaved caspase-3,thereby reducing noise-induced hair cell ***,immunoblot analysis demonstrated that treatment with tranylcypromine upregulated SESN2 expression via the autophagy *** treatment also reduced the production of NOD-like receptor family pyrin domaincontaining 3(NLRP3)*** conclusion,our results showed that tranylcypromine treatment ameliorated cochlear inflammation by promoting the expression of SESN2,which induced autophagy,thereby restricting NLRP3-related inflammasome signaling,alleviating cochlear hair cell loss,and protecting hearing *** findings suggest that inhibiting lysine-specific demethylase 1 is a

This work addresses the distributed fixed-time trajectory formation collaborative control for heterogeneous multiagent systems comprising unmanned surface vessels(USVs) and unmanned aerial vehicles(UAVs), with inherent dynamic uncertainties. By transforming the model coordinates, the heterogeneous USV-UAV systems can be converted into a fully actuated second-order multi-agent framework. Subsequently, a fixed-time dynamic observer is developed to estimate the uncertain dynamics of each agent. Integrating the backstepping technique with the virtual leader, a fixed-time distributed formation control based on uncertain dynamic estimation protocol is designed for the heterogeneous USV-UAV systems. The convergence of the formation errors is rigorously demonstrated by constructing a suitable Lyapunov function within a fixed time frame. Finally, the efficiency of the developed fixed-time formation control algorithm is empirically validated through two numerical simulations in both constant and time-varying formation scenarios.

目的应用多普勒超声心动图评估法洛四联症(TOF)胎儿的肺动脉干(MPA)及左、右分支远段的血流阻力特征。方法前瞻性收集2022年8月至2023年1月中南大学湘雅二医院42例TOF胎儿(TOF组)和84例胎龄匹配的正常胎儿(对照组)。按肺动脉瓣环Z score将TOF分为轻度(Z score≥-2)、中度(-40.05),重度TOF组DLPA-PI明显低于轻度TOF组(P<0.05)。结论中晚孕期TOF胎儿的肺动脉干阻力明显大于正常胎儿。TOF胎儿肺动脉分支远段的阻力降低,尤其是左肺动脉远段。需要进一步的大型随访研究证明TOF患者的血管阻力改变与肺动脉发育之间的关系。

目的观察脓毒症急性呼吸窘迫综合征(ARDS)合并急性胃肠功能损伤(AGI)不同分级患者的临床特征及预后,进一步探讨患者不良预后的相关危险因素。方法收集2023年3月至10月天津市第一中心医院重症监护病房(ICU)收治的脓毒症ARDS患者的临床资料。根据2012年欧洲重症医学协会AGI定义及分级标准,将患者分为AGI 0~Ⅳ级组。观察患者的临床特征及28 d临床结局;采用单因素和多因素Logistic回归法分析脓毒症ARDS合并AGI患者预后的相关危险因素;绘制受试者工作特征曲线(ROC曲线)和校准曲线,评估各危险因素对脓毒症ARDS合并AGI患者预后的预测价值。结果共纳入92例脓毒症ARDS患者,其中AGI 0级组7例、AGIⅠ级组20例、AGIⅡ级组38例、AGIⅢ级组23例、AGIⅣ级组4例,AGI发生率为92.39%。随AGI分级增加,ARDS分级越高,急性生理学与慢性健康状况评分Ⅱ(APACHEⅡ)、序贯器官衰竭评分(SOFA)、腹内压(IAP)、白细胞计数(WBC)、中性粒细胞计数(NEU)、淋巴细胞计数(LYM)、淋巴细胞比例(LYM%)、28 d病死率均呈明显升高趋势,氧合指数(PaO2/FiO2)呈明显降低趋势(均P<0.05)。Pearson相关分析显示,APACHEⅡ评分、SOFA评分和ARDS分级与患者AGI分级呈显著正相关(Pearson相关性指数分别为0.386、0.473、0.372,均P<0.001),PaO2/FiO2与患者AGI分级呈显著负相关(Pearson相关性指数为-0.425,P<0.001)。脓毒症ARDS合并AGI患者中,28 d存活68例,28 d死亡17例。存活组与死亡组APACHEⅡ评分、SOFA评分、ARDS分级、AGI分级、PaO2/FiO2、IAP、AGI 7 d最差值、ICU住院时间及总住院时间比较差异均有统计学意义(均P<0.05)。单因素Logistic回归分析显示,SOFA评分〔优势比(OR)=1.350,95%可信区间(95%CI)为1.071~1.702,P=0.011〕、PaO2/FiO2(OR=0.964,95%CI为0.933~0.996,P=0.027)和AGI 7 d最差值(OR=2.103,95%CI为1.194~3.702,P=0.010)是脓毒症ARDS合并AGI患者28 d死亡的危险因素。多因素Logistic回归分析显示,SOFA评分(OR=1.384,95%CI为1.153~1.661,P<0.001)、PaO2/FiO2(OR=0.983,95%CI为0.968~0.999,P=0.035)和AGI 7 d最差值(OR=1.992,95%CI为1.141~3.478,P=0.015)是脓毒症ARDS合并AGI患者28 d死亡的独立危险因素。ROC曲线分析显示,SOFA评分、PaO2/FiO2和AGI 7 d最差值对脓毒症ARDS合并AGI患者28 d预后均有一定预测价值,ROC曲线下面积(AUC)分别为0.824(95%CI为0.697~0.950)、0.760(95%CI为0.642~0.877)和0.721(95%CI为0.586~0.857),均P<0.01;当上述指标最佳截断值分别为5.50分、163.45 mmHg(1 mmHg≈0.133 kPa)、2.50级时,敏感度分别为94.1%、94.1%、31.9%,特异度分别为80.9%、67.6%、88.2%。结论脓毒症ARDS患者AGI的发生率约为90%,AGI分级越高患者预后越差,SOFA评分、PaO2/FiO2和AGI 7 d最差值对脓毒症ARDS合并AGI患者的预后均有一定的预测价值,其中SOFA评分和AGI 7 d最差值越大、PaO2/FiO2越小,患者病死率越高。

Sphingolipid inhibitor response gene GhMYB86 controls fiber elongation by regulating microtubule arrangement

Although the cell membrane and cytoskeleton play essential roles in cellular morphogenesis, the interaction between the membrane and cytoskeleton is poorly understood. Cotton fibers are extremely elongated single cells, which makes them an ideal model for studying cell development. Here, we used the sphingolipid biosynthesis inhibitor, fumonisin B1(FB1), and found that it effectively suppressed the myeloblastosis(MYB) transcription factor Gh MYB86,thereby negatively affecting fiber elongation. A direct target of Gh MYB86 is Gh TUB7, which encodes the tubulin protein, the major component of the microtubule cytoskeleton. Interestingly, both the overexpression of Gh MYB86 and Gh TUB7 caused an ectopic microtubule arrangement at the fiber tips,and then leading to shortened fibers. Moreover, we found that Gh MBE2 interacted with Gh MYB86 and that FB1 and reactive oxygen species induced its transport into the nucleus, thereby enhancing the promotion of Gh TUB7 by Gh MYB86. Overall, we established a Gh MBE2-Gh MYB86-Gh TUB7 regulation module for fiber elongation and revealed that membrane sphingolipids affect fiber elongation by altering microtubule arrangement.

Alterations in the gut microbiota and the faecal metabolomes are potentially associated with gestational diabetes mellitus through inflammatory response

Gestational diabetes mellitus (GDM) is a disease of glucose intolerance that first occurs during pregnancy. Accumulating evidence underlined a link between gut microbiota dysbiosis and GDM， and microbial metabolites represent a unique way to explore microbiota–host interactions. However， the associations between changes in the gut microbiota and microbial metabolites and immune homeostasis in the GDM pathogenesis remain largely unclear. In this prospective study， the characteristics of gut microbiota in both first trimester (T1) and second trimester (T2 ) were investigated in 46 GDM patients and 44 matched controls. We comprehensively profiled the microbial metabolites using non-targeted metabolomics and quantitatively targeted metabolomics， measurements of inflammatory cytokines and biomarkers of intestinal barrier function， and combined with correlation analysis in T2. Gut microbiota dybiosis was observed in GDM patients in both T1 and T2， and was characterised by the enrichment of multiple potentially harmful bacteria， such as UBA1819 and Erysipelatoclostridium. Besides， alterations in the microbiota were accompanied by a disturbance in tryptophan metabolism， mainly manifested as a shift towards the production of more kynurenine and less indole derivatives. Most importantly， correlation network analysis indicated that overgrowth of potential pathogens and tryptophan metabolism disorder were associated with inflammatory imbalance and disrupted epithelial barrier in GDM patients. Theses findings provide a greater understanding of the pathogenesis and new targets for microecological interventions by mediating tryptophan metabolism in GDM.

Industrial process data exhibits multi-mode and nonstationary characteristics in the spatial and temporal domain, causing unsatisfactory performance of conventional multivariate statistical fault detection methods. Therefore, we propose a novel round-robin multivariate state estimation fault detection method based on cross-domain collaboration learning, which aims to fully excavate the collaborative information for accurate fault detection. Moreover, to overcome the underdetermined problem of conventional multivariate state estimation fault detection methods, the round-robin multivariate state estimation model in collaborative feature space is constructed for fault detection. Simulation on the Tennessee Eastman process has validated the effectiveness of the proposed method.

本文提出了一种具有简单结构且能实现优异性能的石墨烯基器件.该器件能实现三重动态可调太赫兹表面等离子体诱导透明效应以及慢光效果.该器件的功能结构单元由双层石墨烯和硅-空气光栅组成;在周期性硅-空气光栅的辅助激励下,下层石墨烯中的电子可以被入射太赫兹波/能量完全激发,可被视为明模式,并且容易被检测.上层石墨烯中的电子对入射太赫兹波/能量反应十分迟钝,甚至没有反应,被视为暗模式.当明暗模式的共振频率相同或接近时,两者就会发生干涉,从而形成等离子体诱导透明现象.此外,该器件中的双层石墨烯均是完整的石墨烯条而非间断结构,通过外部电压可以十分方便地调节石墨烯的载流子,进而改变石墨烯的费米能级,从而在不改变器件结构参数的情况下实现动态调谐的功能.本文还研究了该器件的慢光性能,器件的群延迟和群折射率分别能够达到0.304 ps和607.6,有望为慢光、传感、光存储等潜在应用提供理论基础.

为高效、准确、自动化地实现空箱结构的优化设计,以空箱式挡土墙为研究对象,基于Python语言和ABAQUS软件进行参数化建模,实现了结构建模及有限元计算分析全套流程的自动化。结合某水闸工程中的一座空箱式挡土墙结构,以多个强度和稳定性指标的最优化和混凝土体积的最小化为目标,以规范要求的指标阈值、几何尺寸和体积限制为约束条件,采用非支配排序遗传算法(NSGA-Ⅱ算法)进行空箱式挡土墙多目标优化设计。为进一步提高计算效率,基于人工神经网络算法建立了代理模型,实现了与实际有限元模型的高度近似。结果表明,依据所提出的优化设计方法进行空箱式挡土墙多目标优化设计,可以在有效控制强度和稳定性的前提下尽可能地缩减混凝土用量,具有显著的经济效益。

BBX9 forms feedback loops with PIFs and BBX21 to promote photomorphogenic development

Light is one of the most essential environmental factors that tightly and precisely control various physiological and developmental processes in plants. B-box CONTAINING PROTEINs(BBXs)play central roles in the regulation of lightdependent development. In this study, we report that BBX9 is a positive regulator of light ***9 interacts with the red light photoreceptor PHYTOCHROME B(phy B) and transcription factors PHYTOCHROME-INTERACTING FACTORs(PIFs). phy B promotes the stabilization of BBX9 in light, while BBX9 inhibits the transcriptional activation activity of PIFs. In turn, PIFs directly bind to the promoter of BBX9 to repress its *** the other hand, BBX9 associates with the positive regulator of light signaling, BBX21, and enhances its biochemical activity. BBX21 associates with the promoter regions of BBX9 and transcriptionally up-regulates its expression. Collectively, this study unveiled that BBX9 forms a negative feedback loop with PIFs and a positive one with BBX21 to ensure that plants adapt to fluctuating light conditions.
