Active compounds in RenShenJian decoction ameliorate insulin resistance in vitro

Background:RenShenJian decoction,a combination of Pueraria lobata(Willd.)Ohwi and Panax ginseng ***,has been used in China since the Song Dynasty(960-1279 C.E.)to relieve symptoms of diabetes ***,the key compounds in RenShenJian that ameliorate insulin resistance remain *** study identified the anti-diabetic compounds in RenShenJian by rescuing the decreased function of adenosine 5’-monophosphate-activated protein kinase(AMPK),sirtuin 3(SIRT3),or glucose transporter isoform 4(GLUT4).Methods:After streptozotocin-induced diabetic mice were treated with RenShenJian,fasting blood glucose levels and protein expression of SIRT3,p-AMPK,and AMPK were *** from RenShenJian in plasma were monitored using multiple responses by liquid chromatography-mass ***,two insulin-resistant cell models were incubated with compounds identified in RenShenJian in the *** uptake was determined using the fluorescent analog 2-(N-(7-nitrobenz-2-oxa-1,3-dia-xol-4-yl)amino)-*** expression levels of p-AMPK,AMPK,SIRT3,and GLUT4 were detected by western ***:RenShenJian decreased FBG levels and upregulated SIRT3 expression and AMPK phosphorylation in diabetic *** RenShenJian extracts were identified in the blood,11 of which increased the ratios of 2-(N-(7-nitrobenz-2-oxa-1,3-dia-xol 4-yl)amino)-2-deoxyglucose uptake in two insulin-resistant cell *** extracts increased AMPK phosphorylation,nine increased SIRT3 expression,and six elevated GLUT4 expression in palmitate-induced HepG2 *** extracts-puerarin,puerarin 6″-O-xyloside,genistein,ginsenoside Rb1,and ginsenoside Rd-simultaneously activated AMPK,SIRT3 and ***:A series of compounds in RenShenJian that target AMPK,SIRT3,and/or GLUT4 was confirmed and indicate the chemical material basis of amelioration of insulin resistance by RenShenJian.

Pulp health and calcific healing of a complicated crown–root fracture with additional root fracture in a maxillary incisor: A case report

BACKGROUND Complicated crown–root fracture (CRF) involves severe injury to the crown, root,and pulp, and may be accompanied by multiple root fractures. The loss of a toothhas lifelong consequences for children and teenagers, but the maintenance of pulphealth and the calcific healing of multiple root fractures are rarely reported in *** SUMMARY This case reports healing of a permanent tooth with complicated crown–root andadditional root fractures, in which pulp health was maintained. A 10-year-old girlfell and fractured the root of her maxillary left central incisor at the cervical *** the coronal fragment was repositioned, the tooth was splinted until thetooth was no longer mobile, 2 years later. Eight years after treatment, the toothhas remained asymptomatic with vital pulp and localized gingival ***-beam computed tomography revealed not only calcified healing of the CRFbut also spontaneous healing in an additional undiagnosed root fracture. Thefracture line on the enamel could not be healed by hard tissue and formed agroove in the cervical crown. It was speculated that the groove was related to thelocalized gingival *** This case provides a clinical perspective of the treatment of a tooth with acomplicated CRF and an additional root fracture.

FINITE ELEMENT ANALYSIS OF CARDIAC MYOCYTE DEBONDING AND REORIENTATION DURING CYCLIC SUBSTRATE STRETCH EXPERIMENTS

The substrate stretch experiment, which is carried out on several kinds of adherent cells, is usually used to catch the physiological variation and morphological response to cyclic substrate deformation. In this paper, stretch loading was exerted on cardiac myocytes cultured on silica substrates using a custom-made substrate stretch device. The effect of stretch on the alignment orientation of cardiac myocytes was studied through morphocytological statistics. Under cyclic stretch stimulus, the long axes of cardiac myocytes oriented perpendicularly to the stretch direction for continuous stretch acting. However, the mechanism underlying these behaviors is not well understood from such in vitro tests. Finite element （FE） model was developed in the analysis to investigate these behaviors. Xu-Needleman formulation was used to define the interaction behavior for contact surfaces between cell and substrate. The role of cell viscoelasticity nature is studied in adherent cell debonding with the substrate and aligning perpendicular to the stretch direction during long time cyclic stretch stimulation. There were four different strain magnitudes considered in the simulation to find out the cell debonding affected by the cyclic strains. The potential role of cyclic strain frequency in regulating cell debonding and alignment was also studied using FE analysis.

Identification and Physical Mapping of New PCR-Based Markers Specific for the Long Arm of Rye(Secale cereale L.) Chromosome 6

To effectively use elite genes on the long arm of rye chromosome 6（the 6RL arm） in wheat breeding programs,precise and fast identification of 6RL chromatin in wheat backgrounds is ***-based 6RL-specific markers can facilitate the detection of elite genes on 6RL in wheat ***,only a limited number of 6RL-specific markers have been *** the present study.300 new PCR-based 6RL-specific markers were identified using specific length amplified fragment sequencing（SLAF-seq） technology,and were further physically mapped to four regions on the 6RL arm using 6R and 6RL deletion ***,127 of the 300 markers were physically localized to a region from the site between 2.3 and 2.5 to the telomere,the same region where the powdery mildew resistance gene was *** addition,95 of the 300 markers exhibit polymorphisms,which can be used to investigate the diversity of rye 6RL *** markers developed in this study can be used to identify given segments of 6RL in wheat backgrounds and accelerate the utilization of elite genes on 6RL in wheat breeding.

Cu/Cr co-stabilization mechanisms in a simulated Al_(2)O_(3)-Fe_(2)O_(3)-Cr_(2)O_(3)-CuO waste system

Chromium slag usually contains various heavy metals,making its safe treatment *** sintering has been applied to resolve this issue and emerged as an effective method for metal immobilization by incorporating heavy metals into stable crystal ***,there is limited knowledge about the reaction pathways adopted by multiple heavy metals and the co-stabilization functions of the crystal *** study the Cu/Cr co-stabilization mechanisms during thermal treatment,a simulated system was prepared using a mixture with a molar ratio of Al_(2)O_(3):Fe_(2)O_(3):Cr_(2)O_(3):CuO=1:1:1:*** samples were sintered at temperatures 600–1300℃ followed by intensive analysis of phase constitutions and microstructure development.A spinel phase(CuFe_(x)Al_(y)Cr_(2–x–y)O_(4))started to generate at 700℃ and the incorporation of Cu/Cr into the spinel largely complete at 900℃,although the spinel peak intensity continued increasing slightly at temperatures above 900℃.Fe_(2)O_(3)/Cr_(2)O_(3) was more easily incorporated into the spinel at lower temperatures,while more Al_(2)O_(3) was gradually incorporated into the spinel at higher ***,sintered sample microstructures became more condensed and smoother with increased sintering ***/Cr leachability substantially decreased after Cu/Cr incorporation into the spinel phase at elevated *** 600℃,the leached ratios for Cu and Cr were 6.28%and 0.65%,*** sintering temperature was increased to 1300℃,the leached ratios for all metal components in the system were below 0.2%.This study proposes a sustainable method for managing Cu/Cr co-exist slag at reasonable temperatures.

Sensitive detection of dopamine and quinone drugs based on the quenching of the fluorescence of carbon dots

In this work,we demonstrated that the quinone structure can quench the fluorescence of the carbon dots(CDs).The sensitive determination of dopamine(DA) was studied primarily based on this *** would be transformed into DA quinone under alkaline conditions,which resulted in fluorescence quenching of the CDs.A good linear range from 5 nmol/L to 0.4 mmol/L was obtained and the detection limit was 1 nmol/***,the quenching effect of quinone structure on the fluorescence of CDs was confirmed by Fourier transform infrared spectra,time-correlated single-photon counting and X-ray photoelectron ***,CDs were firstly applied to detect the quinone drugs quantitatively which contained typical quinone structure based on the quenching *** than this,the sensing platform was demonstrated to provide credible selectivity and satisfactory stability in human serum solution with good liner ***,our practical application and mechanism have showed great potential for diagnostic purposes.

Machine learning methods have proven to be powerful in various research *** this paper,we show that research on radiation effects could benefit from such methods and present a machine learning-based scientific discovery *** total ionizing dose(TID)effects usually cause gain degradation of bipolar junction transistors(BJTs),leading to functional failures of bipolar integrated ***,many experiments of TID effects on BJTs have been conducted at different laboratories worldwide,producing a large amount of experimental data which provides a wealth of ***,it is difficult to utilize these data *** this study,we proposed a new artificial neural network(ANN)approach to analyze the experimental data of TID effects on BJTs An ANN model was built and trained using data collected from different *** results indicate that the proposed ANN model has advantages in capturing nonlinear correlations and predicting the *** trained ANN model suggests that the TID hardness of a BJT tends to increase with base current I.A possible cause for this finding was analyzed and confirmed through irradiation experiments.
