MXene-contained paper is a good choice to design ultrathin and flexible electromagnetic interference(EMI)shielding materials.However,the deficiencies in strength and stability of MXene-contained paper impede its practical applications.Herein,a composite paper was proposed to address the problems,in which a filter paper was modified with a three-layer structured surface via a facile layer-by-layer coating procedure.Specifically,the TEMPO-oxidized cellulose nanofibers(TOCN)/cationic starch(CS)/MXene gel layer and TOCN/MXene nacre structure layer ensured the good EMI shielding and mechanical performances of the composite paper,while the uppermost TOCN/CS hydrogel film layer mainly protected MXene.The composite paper achieved an EMI SE of 40.3 dB at a thickness of merely 0.1894 mm(SE/t value of ca.212.8 dB mm^(−1),SSE/t values of ca.13216 dB cm 2 g^(−1))and the total MXene dosage was 20 g m^(−2).Its tensile strength could be up to 11.7 MPa while the original filter paper was 6.4 MPa.Four pieces of this composite papers could be easily packed together to attain an EMI SE of nearly 70 dB.Importantly,the hydrogel film layer efficiently protected the MXene and maintained the EMI shielding performance of the composite paper when immersed in different liquids including water,HCl(1 M)and ethanol,due to the dense and compact structure of hydrogel film layer.This work provides a practical way to develop ultrathin,flexible and durable EMI shielding materials.

The changes in the annual distribution of mountain runoff during the period of 1965–2018 in Hexi Corridor, Northwest China

The annual distribution characteristics of river runoff in arid regions have significant implications for water resource stability and management. Based on the mountain runoff data from 1965 to 2018, this study examines the annual change characteristics of monthly runoff of the Shiyang River Basin, Heihe River Basin, and Shule River Basin in the Hexi Corridor, Northwest China. Many indexes are used and analyzed, including the coefficient of variance, the complete regulation coefficient, the concentration degree and concentration period, the magnitude of change, the skewness coefficient, and the kurtosis coefficient of the annual distribution curves. The results reveal the following:(1) The inhomogeneity of annual runoff distribution in the Taolai River and the rivers to the west of it, except the Shiyou River, show an increasing trend. Conversely, the inhomogeneity of the rivers to the east of the Taolai River generally show a downward trend, but the coefficient of variance value is still very high.(2) In the Shiyang River Basin, the annual distribution of the concentration period is characterized by a relatively discrete pattern. Conversely, the Heihe River Basin exhibits a relatively concentrated pattern, and the distribution pattern of the Shule River Basin is quite different. Notably, all concentration periods in the three basins have shifted backward after the 2000s.(3) The Shiyang River Basin exhibits disordered annual distribution curves of runoff in different years. In contrast, the Heihe River Basin presents a typical 'single-peak' pattern with a prominent right-skewed. The Shule River Basin has regular distribution curves, with a gradually significant'double-peak' pattern from east to west. Overall, there has been a slight change in runoff in the Shiyang River Basin, while the Heihe River Basin and Shule River Basin have experienced significant increases in runoff. Synthetic vascular grafts suitable for small-diameter arteries(<6 mm) are in great need.However,there are still no commercially available small-diameter vascular grafts(SDVGs) in clinical practice due to thrombosis and stenosis after in vivo implantation.When designing SDVGs,many studies emphasized reendothelization but ignored the importance of reconstruction of the smooth muscle layer(SML).To facilitate rapid SML regeneration,a high-resolution 3D printing method was used to create a novel bilayer SDVG with structures and mechanical properties mimicking natural arteries.Bioinspired by the collagen alignment of SML,the inner layer of the grafts had larger pore sizes and high porosity to accelerate the infiltration of cells and their circumferential alignment,which could facilitate SML reconstruction for compliance restoration and spontaneous endothelialization.The outer layer was designed to induce fibroblast recruitment by low porosity and minor pore size and provide SDVG with sufficient mechanical strength.One month after implantation,the arteries regenerated by 3D-printed grafts exhibited better pulsatility than electrospun grafts,with a compliance(8.9%) approaching that of natural arteries(11.36%) and significantly higher than that of electrospun ones(1.9%).The 3D-printed vascular demonstrated a three-layer structure more closely resembling natural arteries while electrospun grafts showed incomplete endothelium and immature SML.Our study shows the importance of SML reconstruction during vascular graft regeneration and provides an effective strategy to reconstruct blood vessels through 3D-printed structures rapidly.

Relapse after CAR-T cell therapy in B-cell malignancies: challenges and future approaches

Chimeric antigen receptor-T(CAR-T)cell therapy,as a novel cellular immunotherapy,has dramatically reshaped the landscape of cancer treatment,especially in hematological malignancies.However,relapse is still one of the most troublesome obstacles to achieving broad clinical application.The intrinsic factors and superior adaptability of tumor cells mark a fundamental aspect of relapse.The unique biological function of CAR-T cells governed by their special CAR construction also affects treatment efficacy.Moreover,complex cross-interactions among CAR-T cells,tumor cells,and the tumor microenvironment(TME)profoundly influence clinical outcomes concerning CAR-T cell function and persistence.Therefore,in this review,based on the most recent discoveries,we focus on the challenges of relapse after CAR-T cell therapy in B-cell malignancies from the perspective of tumor cells,CAR-T cells,and the TME.We also discuss the corresponding basic and clinical approaches that may overcome the problem in the future.We aim to provide a comprehensive understanding for scientists and physicians that will help improve research and clinical practice.

Genetic mutation of Tas2r104/Tas2r105/Tas2r114 cluster leads to a loss of taste perception to denatonium benzoate and cucurbitacin B

Background:Bitter taste receptors(Tas2rs)are generally considered to sense various bitter compounds to escape the intake of toxic substances.Bitter taste receptors have been found to widely express in extraoral tissues and have important physiological functions outside the gustatory system in vivo.Methods:To investigate the physiological functions of the bitter taste receptor cluster Tas2r106/Tas2r104/Tas2r105/Tas2r114 in lingual and extraoral tissues,multiple Tas2rs mutant mice and Gnat3 were produced using CRISPR/Cas9 gene-editing technique.A mixture containing Cas9 and sgRNA mRNAs for Tas2rs and Gnat3 gene was microinjected into the cytoplasm of the zygotes.Then,T7EN1 assays and sequencing were used to screen genetic mutation at the target sites in founder mice.Quantitative real-time polymerase chain reaction(qRT-PCR)and immunostaining were used to study the expression level of taste signaling cascade and bitter taste receptor in taste buds.Perception to taste substance was also studied using twobottle preference tests.Results:We successfully produced several Tas2rs and Gnat3 mutant mice using the CRISPR/Cas9 technique.Immunostaining results showed that the expression of GNAT3 and PLCB2 was not altered in Tas2rs mutant mice.But qRT-PCR results revealed the changed expression profile of m Tas2rs gene in taste buds of these mutant mice.With two-bottle preference tests,these mutant mice eliminate responses to cycloheximide due to genetic mutation of Tas2r105.In addition,these mutant mice showed a loss of taste perception to quinine dihydrochloride,denatonium benzoate,and cucurbitacin B(CuB).Gnat3-mediated taste receptor and its signal pathway contribute to CuB perception.Conclusions:These findings implied that these mutant mice would be a valuable means to understand the biological functions of TAS2Rs in extraoral tissues and investigate bitter compound-induced responses mediated by these TAS2Rs in many extraoral tissues.

Recent research efforts in the field of electromagnetic interference shielding(EMI)materials have focused on biomass as a green and sustainable resource.More specifically,wood and cellulose nano fiber(CNF)have many advantages,some of which include lightweight,porosity,widespread availability,low cost,and easy processing.These favorable properties have led researchers to consider these types of biomass as an EMI shielding material with great potential.At present,while many excellent published works in EMI shielding materials have investigated wood and CNF,this research area is still new,compared with non-biomass EMI shielding materials.More specifically,there is still a lack of in-depth research and summary on the preparation process,pore structure regulation,component optimization,and other factors affecting the EMI shielding of wood and CNF based EMI shielding materials.Thus,this review paper presents a comprehensive summary of recent research on wood and CNF based EMI shielding materials in recent three years in terms of the preparation methods,material structure design,component synergy,and EMI mechanism,and a forward future perspective for existing problems,challenges,and development trend.The ultimate goal is to provide a comprehensive and informative reference for the further development and exploration of biomass EMI shielding materials.

Construction and evaluation of the functional polygenic risk score for gastric cancer in a prospective cohort of the European population

Background:A polygenic risk score(PRS)derived from 112 single-nucleotide polymorphisms(SNPs)for gastric cancer has been reported in Chinese populations(PRS-112).However,its performance in other populations is unknown.A functional PRS(fPRS)using functional SNPs(fSNPs)may improve the generalizability of the PRS across populations with distinct ethnicities.Methods:We performed functional annotations on SNPs in strong linkage disequilibrium(LD)with the 112 previously reported SNPs to identify fSNPs that affect protein-coding or transcriptional regulation.Subsequently,we constructed an fPRS based on the fSNPs by using the LDpred2-infinitesimal model and then analyzed the performance of the PRS-112 and fPRS in the risk prediction of gastric cancer in 457,521 European participants of the UK Biobank cohort.Finally,the performance of the fPRS in combination with lifestyle factors were evaluated in predicting the risk of gastric cancer.Results:During 4,582,045 person-years of follow-up with a total of 623 incident gastric cancer cases,we found no significant association between the PRS-112 and gastric cancer risk in the European population(hazard ratio[HR]=1.00[95%confidence interval(CI)0.93–1.09],P=0.846).We identified 125 fSNPs,including seven deleterious protein-coding SNPs and 118 regulatory non-coding SNPs,and used them to construct the fPRS-125.Our result showed that the fPRS-125 was significantly associated with gastric cancer risk(HR=1.11[95%CI,1.03–1.20],P=0.009).Compared to participants with a low fPRS-125(bottom quintile),those with a high fPRS-125(top quintile)had a higher risk of incident gastric cancer(HR=1.43[95%CI,1.12–1.84],P=0.005).Moreover,we observed that participants with both an unfavorable lifestyle and a high genetic risk had the highest risk of incident gastric cancer(HR=4.99[95%CI,1.55–16.10],P=0.007)compared to those with both a favorable lifestyle and a low genetic risk.Conclusion:These results indicate that the fPRS-125 derived from fSNPs may act as an indicator to measure the genetic risk of g

sEMG-Based Lower Limb Motion Prediction Using CNN-LSTM with Improved PCA Optimization Algorithm

In recent years,sEMG(surface electromyography)signals have been increasingly used to operate wearable devices.The development of mechanical lower limbs or exoskeletons controlled by the nervous system requires greater accuracy in recognizing lower limb activity.There is less research on devices to assist the human body in uphill movements.However,developing controllers that can accurately predict and control human upward movements in real-time requires the employment of appropriate signal pre-processing methods and prediction algorithms.For this purpose,this paper investigates the effects of various sEMG pre-processing methods and algorithms on the prediction results.This investigation involved ten subjects(five males and five females)with normal knee joints.The relevant data of the subjects were collected on a constructed ramp.To obtain feature values that reflect the gait characteristics,an improved PCA algorithm based on the kernel method is proposed for dimensionality reduction to remove redundant information.Then,a new model(CNN+LSTM)was proposed to predict the knee joint angle.Multiple approaches were utilized to validate the superiority of the improved PCA method and CNN-LSTM model.The feasibility of the method was verified by analyzing the gait prediction results of different subjects.Overall,the prediction time of the method was 25 ms,and the prediction error was 1.34±0.25 deg.By comparing with traditional machine learning methods such as BP(backpropagation)neural network,RF(random forest),and SVR(support vector machine),the improved PCA algorithm processed data performed the best in terms of convergence time and prediction accuracy in CNN-LSTM model.The experimental results demonstrate that the proposed method(improved PCA+CNN-LSTM)effectively recognizes lower limb activity from sEMG signals.For the same data input,the EMG signal processed using the improved PCA method performed better in terms of prediction results.This is the first step toward myoelectric control of aided exoskeleton robots using disc

Exploring an advanced and efficient electromagnetic(EM) wave absorbing material by improving dielectric loss capacity and adjusting impendence matching is crucial yet challenging. Herein, the bacterial cellulose(BC) derived carbon aerogel(CA) with a robust nanofibrous network was used as a conductive loss scaffold to dissipate EM waves effectively, and the Zn O microparticles with excellent dielectric properties and low electrical conductivity were decorated on the scaffold to adjust dielectric parameters and impedance matching adequately. By using different zinc precursors, the tunable size and morphologies of Zn O crystals were obtained due to the growth rate of different crystallographic, including flowerlike, nanorod like, and cauliflower-like morphologies, which is beneficial to strong multiple reflections,intensive interfacial polarization, better impendence matching, as well as excellent maintenance of the hierarchical structure. Owing to the appropriate impendence matching and the considerable EM wave dissipation, the CA@ZnO composites achieve a superior EM absorbing performance with a broad effective absorbing bandwidth(whole X band) and a minimum reflection coefficient(-53.3 d B). This work paves a new way for developing lightweight and highly efficient EM absorbing materials comprising the carbon scaffold and semiconductor microparticles.

New insights into the suppression of inflammation and lipid accumulation by JAZF1

Atherosclerosis is one of the leading causes of disease and death worldwide.The identification of new therapeutic targets and agents is critical.JAZF1 is expressed in many tissues and is found at particularly high levels in adipose tissue(AT).JAZF1 suppresses inflammation(including IL-1β,IL-4,IL-6,IL-8,IL-10,TNFα,IFN-γ,IAR-20,COL3A1,laminin,and MCP-1)by reducing NF-κB pathway activation and AT immune cell infiltration.JAZF1 reduces lipid accumulation by regulating the liver X receptor response element(LXRE)of the SREBP-1c promoter,the cAMP-response element(CRE)of HMGCR,and the TR4 axis.LXRE and CRE sites are present in many cytokine and lipid metabolism gene promoters,which suggests that JAZF1 regulates these genes through these sites.NF-κB is the center of the JAZF1-mediated inhibition of the inflammatory response.JAZF1 suppresses NF-κB expression by suppressing TAK1 expression.Interestingly,TAK1 inhibition also decreases lipid accumulation.A dual-targeting strategy of NF-κB and TAK1 could inhibit both inflammation and lipid accumulation.Dual-target compounds(including prodrugs)1–5 exhibit nanomolar inhibition by targeting NF-κB and TAK1,EGFR,or COX-2.However,the NF-κB suppressing activity of these compounds is relatively low(IC50>300 nM).Compounds 6–14 suppress NF-κB expression with IC50 values ranging from 1.8 nM to 38.6 nM.HS-276 is a highly selective,orally bioavailable TAK1 inhibitor.Combined structural modifications of compounds using a prodrug strategy may enhance NF-κB inhibition.This review focused on the role and mechanism of JAZF1 in inflammation and lipid accumulation for the identification of new anti-atherosclerotic targets.
