Human microorganisms,including bacteria,fungi,and viruses,play key roles in several physiological and pathological processes.Some studies discovered that tumour tissues once considered sterile actually host a variety of microorganisms,which have been confirmed to be closely related to oncogenesis.The concept of intratumoural microbiota was subsequently proposed.Microbiota could colonise tumour tissues through mucosal destruction,adjacent tissue migration,and hematogenic invasion and affect the biological behaviour of tumours as an important part of the tumour microenvironment.Mechanistic studies have demonstrated that intratumoural microbiota potentially promote the initiation and progression of tumours by inducing genomic instability and mutations,affecting epigenetic modifications,promoting inflammation response,avoiding immune destruction,regulating metabolism,and activating invasion and metastasis.Since more comprehensive and profound insights about intratumoral microbiota are continuously emerging,new methods for the early diagnosis and prognostic assessment of cancer patients have been under examination.In addition,interventions based on intratumoural microbiota show great potential to open a new chapter in antitumour therapy,especially immunotherapy,although there are some inevitable challenges.Here,we aim to provide an extensive review of the concept,development history,potential sources,heterogeneity,and carcinogenic mechanisms of intratumoural microorganisms,explore the potential role of microorganisms in tumour prognosis,and discuss current antitumour treatment regimens that target intratumoural microorganisms and the research prospects and limitations in this field.

甘薯作为我国第四大粮食作物,不仅在解决人们温饱方面发挥了重要作用,而且作为一种营养丰富的食材也广受青睐。甘薯的经济价值主要取决于其产量和营养品质,营养品质的评定主要包括淀粉、还原糖、蛋白质、膳食纤维、矿质元素等方面。为了探究适合在邢台地区种植的甘薯品种,选取河北省邢台市柏乡县甘薯产业园试验田规范种植的11个甘薯品种,测定其产量、水分、干物质、蛋白质、粗脂肪、还原糖、淀粉和粗灰分8个指标。实验结果表明:徐薯22、大开花和秦五这三个品种的干物质含量和淀粉含量较好,口感粉面;济薯26、商19、烟薯25、西瓜红、紫薯的蛋白质含量较好,营养品质较高;秦五、烟薯25、秦八的还原糖含量较高,口感香甜;徐薯32、徐薯22、玛沙利的粗脂肪含量较高。

Although it is usually latent on citrus, apple, and pear, apple stem grooving virus(ASGV) poses a great risk to many sensitive cultivars. Since severe leaf yellow mottle mosaic(LYMM) symptoms have been observed on Huangjinmiyou(HJY) pummelos(Citrus grandis cv. Huangjinmiyou), a commercial variety that is widely cultivated in South China, high throughput sequencing(HTS) was used to find potential pathogens and only three divergent ASGV variants were identified. The three ASGV variants shared 81.03–82.34% genome-wide pairwise identities with each other, and were separately closest to other ASGV variants from different hosts and/or geographical regions, as indicated by viral phylogenies. However, these new variants may have developed from viral interstrain interactions, based on the results of recombination analysis. A large-scale survey using reverse transcription-PCR(RT-PCR) protocols designed for the three ASGV variants revealed a high incidence(92.7–100%) of ASGV in symptomatic HJY trees from 11 major citrusproducing regions in China. None of ASGV were detected in asymptomatic trees. Temperature treatments applied to the symptomatic HJY plants showed that ASGV is sensitive to high temperatures(30–35°C), at which not only the plants recovered, but also the viruses were not detected by RT-PCR, while at low temperatures(20–24°C), both the symptoms and viruses remained detectable. These data show that ASGV is associated with the LYMM disease prevalent on HJY in China, and this is the significant basis especially of taking appropriate measures timely to manage the disease.

This study presents experimental evidence of the dependence of non-radiative recombination processes on the electron-phonon coupling of perovskite in perovskite solar cells(PSCs).Via A-site cation engineering,a weaker electron-phonon coupling in perovskite has been achieved by introducing the structurally soft cyclohexane methylamine(CMA^(+))cation,which could serve as a damper to alleviate the mechanical stress caused by lattice oscillations,compared to the rigid phenethyl methylamine(PEA^(+))analog.It demonstrates a significantly lower non-radiative recombination rate,even though the two types of bulky cations have similar chemical passivation effects on perovskite,which might be explained by the suppressed carrier capture process and improved lattice geometry relaxation.The resulting PSCs achieve an exceptional power conversion efficiency(PCE)of 25.5%with a record-high opencircuit voltage(V_(OC))of 1.20 V for narrow bandgap perovskite(FAPbI_(3)).The established correlations between electron-phonon coupling and non-radiative decay provide design and screening criteria for more effective passivators for highly efficient PSCs approaching the Shockley-Queisser limit.

Alternative splicing plays important roles in gene regulation and contributes to protein complexity. Previous studies suggest that alternative splicing exists in members of the villin/gelsolin/fragmin superfamily. In this study, a ser- ine/argine-rich （SR） protein cDNA with 28kDa protein （LISR28） was isolated from a lily （Lilium Iongiflorum） expression library. Protein domain analysis showed that LISR28 had similar structures to Arabidopsis SR45 （AtSR45）, and LISR28 could complement the phenotype of loss of AtSR45 function. Therefore, overexpression of LISR28 and AtSR45 mutant （atsr45-1） were used in the following experiments. Overexpression of LISR28 in Arabidopsis completely inhibited pollen germina- tion. In contrast, the pollen germination of atsr45-1 was earlier than that of wild-type. In addition, pollen of atsr45-1 contained less F-actin at the corresponding hydration stage during pollen germination compared to that of wild-type. Alternative splicing analysis showed that Arabidopsis villinl （AtVLN1） transcript encoding the full-length protein was increased, and that encoding the truncated protein was decreased in atst45-1. Moreover, the mRNA expression level of other actin-binding proteins （ABPs） abundant in Arabidopsis pollen was also changed in atsr45-1. In conclusion, we hypothesize that LISR28 alters F-actin dynamics probably through its alternative splicing activities to affect directly or indirectly the alternative splicing of AtVLN1 and the expression of different ABPs, which then affects the pollen germination.

Improvement in Mechanical Properties and Biocompatibility ofBiosynthetic Bacterial Cellulose/Lotus Root Starch Composites

Introduction:Previously, we have documented that isolated autophagosomes from tumor cells after blocking protein degradation to enrich tumor specific antigens, used as therapeutic vaccines and cross-presented by dendritic cells, can efficiently cross-prime nave T cells and mediate tumor regression.However, the effect of tumor-derived autophagosomes, here we refer as to DRibbles, on B cells has not been studied so far.Purpose:In this study, we examined whether tumor-derived DRibbles could induce B-cell activation and proliferation and production of tumor-specific antibodies in vivo.In addition, we investigated whether B cells could uptake and cross-present DRibbles antigens and serves as efficient antigen presenting cells for T cell activation.Methods:DRibbles were firstly isolated from Hep1-6 cells, and then intravenously injected into C57/BL6 mice.Hep1-6 tumor cell specific antibodies in serum were analyzed by flow cytometry and fluorescence microscope.B cells were purified using a combination of anti-CD43 coupled magnetic beads, and then coincubated with CFSE-labeled DRibbles in vitro.B cell proliferation was assessed by measuring the dilution of CFSE via flow cytometry.B cell activation was analyzed by measuring the activation markers on B cell surface, and supernatants were harvested for detection of cytokines and antibodies via ELISA.For DRibblesantigens presentation by B cells assay, C57/BL6 mice were intro-node vaccinated with DRibbles, CD4+T and CD8+ T cells were purified from lymph node of the vaccinated mice, and then co-incubated with purified B cells plus DRibbles for 72 hours respectively.IFN-γ or IL-4 in the supernatants was tested by ELISA.Results:At present study, we found that DRibbles generated from a murine hepatoma cell line Hep1-6, induced B cell activation after intravenous injection into mice.B cell populations were significantly expanded and the production of Hep1-6 tumor-specific antibodies was successfully induced.Moreover, in vitro studies showed that DRibbles directly induce B cell pr

Bacterial cellulose/lotus root starch(BC/LRS) composites were prepared by cultivating Acetobacter xylinum in nutrient media containing gelatinized lotus root starch. Low concentrations of gelatinized LRS had increased BC production with the maximum value at 6.67 g/L when 5 g/L of LRS was added in the culture media and the composites had thicker and denser fibrils compared with those of BC with low concentrations of LRS(2.5 and 5 g/L). When the concentration of LRS was increased above 7.5 g/L, the morphology of the BC/LRS composites contained more fibril layers that were linked with LRS. The results from X-ray diffraction(XRD) demonstrated that there was no significant difference in structure between BC and BC/LRS composites except a slight increase in crystallinity for BC/LRS composites as the concentration of LRS was lifted up. The tensile tests were performed to display BC/LRS composites prepared with LRS concentration at 2.5 and 5 g/L in media had the tensile strength of 54 and 60 MPa, respectively, which indicated an improvement in mechanical property compared to the unmodified BC(45 MPa). Live/dead assay with chondrocytes seeded on BC/LRS composite revealed higher cell viability ranging from 85% to 90% than BC. Furthermore, cell morphology with typical spindle shape was observed on the surfaces of BC/LRS composite by confocal microscope. Through the overall results, it shows that this study has provided a guidance to prepare BC/LRS composites with better cell biocompatibility and higher mechanical strength than those of BC for the potential use in cartilage tissue engineering.
