Micro and Nanofabrication methods to control cell-substrate interactions and cell behavior: A review from the tissue engineering perspective

Cell-substrate interactions play a crucial role in the design of better biomaterials and integration ofimplants with the tissues. Adhesion is the binding process of the cells to the substrate through interactionsbetween the surface molecules of the cell membrane and the substrate. There are severalfactors that affect cell adhesion including substrate surface chemistry, topography, and stiffness. Thesefactors physically and chemically guide and influence the adhesion strength, spreading, shape and fate ofthe cell. Recently, technological advances enabled us to precisely engineer the geometry and chemistry ofsubstrate surfaces enabling the control of the interaction cells with the substrate. Some of the mostcommonly used surface engineering methods for eliciting the desired cellular responses on biomaterialsare photolithography, electron beam lithography, microcontact printing, and microfluidics. Thesemethods allow production of nano- and micron level substrate features that can control cell adhesion,migration, differentiation, shape of the cells and the nuclei as well as measurement of the forces involvedin such activities. This review aims to summarize the current techniques and associate these techniqueswith cellular responses in order to emphasize the effect of chemistry, dimensions, density and design ofsurface patterns on cell-substrate interactions. We conclude with future projections in the field of cellsubstrateinteractions in the hope of providing an outlook for the future studies.

The tumor microenvironment consists of diverse,complex etiological *** matrix component of pancreatic ductal adenocarcinoma(PDAC)plays an important role not only in physical properties such as tissue rigidity but also in cancer progression and therapeutic *** significant efforts have been made to model desmoplastic PDAC,existing models could not fully recapitulate the etiology to mimic and understand the progression of ***,two major components in desmoplastic pancreatic matrices,hyaluronic acid-and gelatin-based hydrogels,are engineered to provide matrices for tumor spheroids composed of PDAC and cancer-associated fibroblasts(CAF).Shape analysis profiles reveals that incorporating CAF contributes to a more compact tissue *** expression levels of markers associated with proliferation,epithelial to mesenchymal transition,mechanotransduction,and progression are observed for cancer-CAF spheroids cultured in hyper desmoplastic matrix-mimicking hydrogels,while the trend can be observed when those are cultured in desmoplastic matrix-mimicking hydrogels with the presence of transforming growth factor-β1(TGF-β1).The proposed multicellular pancreatic tumor model,in combination with proper mechanical properties and TGF-β1 supplement,makes strides in developing advanced pancreatic models for resembling and monitoring the progression of pancreatic tumors,which could be potentially applicable for realizing personalized medicine and drug testing applications.

Comparison of the simulated gamma-ray attenuation coefficients with the real measurements

The gamma-ray linear and the mass attenuation coefficients of Pb, Al, Cu, and plexiglass materials were calculated from both experimental and theoretical(simulation) methods. For the experimental results, a spectrometer, which was consisted of a Na I(Tl) inorganic scintillation detector, was used. The theoretical attenuation values were calculated by means of the FLUKA Monte Carlo(MC) and XCOM programs. Obtained attenuation coefficients from the experiment and the theoretical methods were compared with each other and literature values.

A comprehensive study for mass attenuation coefficients of different parts of the human body through Monte Carlo methods

The gamma-ray mass attenuation coefficients of blood,bone,lung,eye lens,adipose,tissue,muscle,brain and skin were calculated at different energies(60,80,150,400,500,600,1000,1250,1500,and 2000 keV) by various theoretical methods such as FLUKA,GEANT4 Monte Carlo(MC) methods and XCOM program in this *** coefficients were also compared with the National Institute of Standards and Technology(NIST) *** results were highly in accordance with each other and NIST *** results showed that FLUKA was quite convenient in comparison to GEANT4 in the calculation of the mass attenuation coefficients of the used human body samples for low-energy photons(60,80,and 150 keV) when compared with the NIST values.

New treatment strategies for hepatitis c infection

Hepatitis C infection can lead to cirrhosis and hepatocellular carcinoma and it is an important cause of mortality and morbidity. Achieving a sustained virological response has been the major aim for decades. Interferon treatment was the primarily developed therapy against the infection. Addition of the guanosine analog ribavirin to stop viral RNA synthesis increased the response rates as well as the adverse effects of the treatment. The increasing demands for alternative regimens led to the development of direct-acting antivirals(DAAs). The approval of sofosbuvir and simeprevir signaled a new era of antiviral treatment for hepatitis C infection. Although the majority of studies have been performed with DAAs in combination with interferon and resulted in a decrease in treatment duration and increase in response rates, the response rates achieved with interferon-free regimens provided hope for patients ineligible for therapy with interferon. Most DAA studies are in phase Ⅱ leading to phase Ⅲ. In the near future more DAAs are expected to be approved. The main disadvantage of the therapy remains the cost of the drugs. Here, we focus on new treatment strategies for hepatitis C infection as well as agents targeting hepatitis C virus replication that are in clinical development.

Comparison of pre-treatment and post-treatment use of selenium in retinal ischemia reperfusion injury

AIM: To investigate the effects of selenium in rat retinal ischemia reperfusion(IR) model and compare pretreatment and post-treatment ***: Selenium pre-treatment group(n =8) was treated with intraperitoneal(i.p.) selenium 0.5 mg/kg for7 d and terminated 24 h after the IR injury. Selenium posttreatment group( n = 8) was treated with i. p. selenium0.5 mg/kg for 7d after the IR injury with termination at the end of the 7d period. Sham group(n =8) received *** injections identical to the selenium volume for 7d with termination 24 h after the IR injury. Control group(n =8) received no intervention. Main outcome measures were retina superoxide dismutase(SOD), glutathione(GSH),total antioxidant status(TAS), malondialdehyde(MDA),DNA fragmentation levels, and immunohistological apoptosis ***: Compared to the Sham group, selenium pre-treatment had a statistical difference in all parameters except SOD. Post-treatment selenium also resulted in statistical differences in all parameters except the MDA levels. When comparing selenium groups, the pre-treatment selenium group had a statistically higher success in reduction of markers of cell damage such as MDA and DNA fragmentation. In contrast, the post-selenium treatment group had resulted in statisticallyhigher levels of GSH. Histologically both selenium groups succeeded to limit retinal thickening and apoptosis. Pre-treatment use was statistically more successful in decreasing apoptosis in ganglion cell layer compared to post-treatment ***: Selenium was successful in retinal protection in IR injuries. Pre-treatment efficacy was superior in terms of prevention of tissue damage and apoptosis.
