Elevated axonal membrane permeability and its correlation with motor deficits in an animal model of multiple sclerosis

Background:It is increasingly clear that in addition to myelin disruption,axonal degeneration may also represent a key pathology in multiple sclerosis(MS).Hence,elucidating the mechanisms of axonal degeneration may not only enhance our understanding of the overall MS pathology,but also elucidate additional therapeutic *** objective of this study is assess the degree of axonal membrane disruption and its significance in motor deficits in EAE ***:Experimental Autoimmune Encephalomyelitis was induced in mice by subcutaneous injection of myelin oligodendrocyte glycoprotein/complete Freud’s adjuvant emulsion,followed by two intraperitoneal injections of pertussis *** assessment was performed using a 5-point *** Peroxidase Exclusion test was used to quantify the disruption of axonal *** glycol was prepared as a 30%(w/v)solution in phosphate buffered saline and injected ***:We have found evidence of axonal membrane disruption in EAE mice when symptoms peak and to a lesser degree,in the pre-symptomatic stage of EAE ***,polyethylene glycol(PEG),a known membrane fusogen,significantly reduces axonal membrane disruption in EAE *** PEG-mediated membrane repair was accompanied by significant amelioration of behavioral deficits,including a delay in the emergence of motor deficits,a delay of the emergence of peak symptom,and a reduction in the severity of peak ***:The current study is the first indication that axonal membrane disruption may be an important part of the pathology in EAE mice and may underlies behavioral *** study also presents the initial observation that PEG may be a therapeutic agent that can repair axolemma,arrest axonal degeneration and reduce motor deficits in EAE mice.

Synchronous colorectal cancer: Clinical, pathological and molecular implications

Synchronous colorectal carcinoma refers to more than one primary colorectal carcinoma detected in a single patient at initial presentation. A literature review has shown that the prevalence of the disease is approximately 3.5% of all colorectal carcinomas. This disease has a male to female ratio of 1.8:1. The mean age at presentation of patients with synchronous colorectal cancer is in the early half of the seventh decade. Patients with inflammatory bowel diseases (ulcerative colitis and Crohn’s disease), hereditary non-polyposis colorectal cancer, familial adenomatous polyposis and serrated polyps/hyperplastic polyposis are known to have a higher risk of synchronous colorectal carcinoma. These predisposing factors account for slightly more than 10% of synchronous colorectal carcinomas. Synchronous colorectal carcinoma is more common in the right colon when compared to solitary colorectal cancer. On pathological examination, some synchronous colorectal carcinomas are mucinous adenocarcinomas. They are usually associated with adenomas and metachronous colorectal carcinomas. Most of the patients with synchronous colorectal cancer have two carcinomas but up to six have been reported in one patient. Patients with synchronous colorectal carcinoma have a higher proportion of microsatellite instability cancer than patients with a solitary colorectal carcinoma. Also, limited data have revealed that in many synchronous colorectal carcinomas, carcinomas in the same patient have different patterns of microsatellite instability status, p53 mutation and K-ras mutation. Overall, the prognosis of patients with synchronous colorectal carcinoma is not significantly different from that in patients with solitary colorectal carcinoma, although a marginally better prognosis has been reported in patients with synchronous colorectal carcinoma in some series. The effects of glycyrrhizic acid and glabridin in the regulation of CXCL5 inflammation gene on acceleration of wound healing

Objective: To evaluate the anti-inflammatory property of both glycyrrhizic acid(GA)and glabridin in reducing inflammation to accelerate wound regeneration on 3T3-L1 and NIH-3T3 fibroblast cell ***: Cell proliferation and viability assay(MTT assay), scratch wound healing assays,and quantitative real-time PCR were conducted to investigate the effects on cell proliferation,cell migration, and expression of CXC chemokine ligand 5 inflammation gene ***: Results showed that at a low concentration of 1 × 10^(-8)mol/L, glabridin down regulated cell proliferation in NIH-3T3 significantly, suggesting its involvement in ERK1/2 signaling pathway. GA and glabridin significantly accelerated cell migration through wound healing in both 3T3-L1 and NIH-3T3 and significantly down regulated the expression of CXC chemokine ligand 5 in 3T3-L1 at concentration 1 × 10^(-8)mol/L,indicating the possible involvement of nuclear factor-k B and cyclooxygenase 2 transcriptions ***: Both GA and glabridin can serve as potential treatment for chronic inflammatory disease, and glabridin as an oncogenic inhibitor due to its anti-proliferative property.

A Survey on Mixed Spin P-Fields

The mixed spin P-fields （MSP for short） theory sets up a geometric platform to relate Gromov-Witten invariants of the quintic three-fold and Fan-Jarvis-Ruan-Witten invariants of the quintic polynomial in five variables. It starts with Wittens vision and the P-fields treatment of GW invariants and FJRW invaxiants. Then it briefly discusses the master space technique and its application to the set-up of the MSP moduli. Some key results in MSP theory are explained and some examples are provided.

Effectiveness of modified pedestrian crossing signs in an urban area

The District of Columbia currently uses the standard pedestrian warning signs and diagonal arrow plaques at a substantial number of uncontrolled crosswalks within the ***, the widespread use of these measures appears to be ineffective in curbing the incidence of pedestrian involved crashes or pedestrian-vehicle conflicts. To compensate for the perceived lack of effectiveness of the standard pedestrian warning sign the District Department of Transportation developed a new side-of-street crossing sign to improve driver compliance based on pedestrian right-of-way *** study was aimed at determining the effectiveness(defined as the proportion of drivers approaching a crosswalk who stop or yield the right of way to a pedestrian in the crosswalk) of the experimental side-of-street pedestrian crossing sign compared to the standard sign, with and without rectangular rapid flashing beacons. Effectiveness of the side-of-street pedestrian sign and standard sign were observed at a total of 32 locations in the District over a one-year period using the “control” and “experimental” comparison approach. Video data for each location was obtained from March 2018 through February 2019 during typical weekdays for the morning and afternoon peak periods. The results of the study showed that the experimental signs with RRFBs provided higher driver compliance rates(yielding to pedestrians) compared to the standard signs for both the morning and afternoon peak periods. However, the differences in compliance rates for the experimental and standard signs were not statistically significant at a 95% confidence *** evaluation of the signs is recommended using the “before” and “after” approach in addition to an assessment of crash statistics at the selected locations.

Within the consistent daily rhythm of human life,intervertebral discs endure a variety of complex loads beyond the influences of gravity and muscle forces,leading to significant morphological changes(in terms of volume,area,and height)as well as biomechanical alterations,including an increase in disc stiffness and a decrease in intradiscal ***,the discs demonstrate an ability to regain their original morphological and biomechanical characteristics after a period of nocturnal *** preservation of normal disc function is critically dependent on this recovery phase,which serves to forestall premature disc *** phenomenon of disc recovery has been extensively documented through numerous in vivo studies employing advanced clinical techniques such as Magnetic Resonance Imaging(MRI),stadiometry,and intradiscal pressure ***,the findings from in vitro studies present a more complex picture,with reports varying between full recovery and only partial recuperation of the disc ***,research focusing on degenerated discs in vitro has shed light on the quantifiable impact of degeneration on the disc ability to *** dynamics within the disc are considered a primary factor in recovery,yet the disc intricate multiscale structure and its viscoelastic properties also play key *** elements interact in complex ways to influence the recovery mechanism,particularly in relation to the overall health of the *** objective of this review is to collate,analyze,and critically evaluate the existing body of in vivo and in vitro research on this topic,providing a comprehensive understanding of disc recovery *** understanding offers a blueprint for future advancements in medical treatments and bionic engineering solutions designed to mimic,support,and enhance the natural recovery processes of intervertebral discs.

Human settlement and agricultural development are closely linked to local geomorphological and climatic ***,the variation in agricultural systems in different environmental and prehistoric contexts remains *** report new archaeobotanical and radiocarbon dates from 34 Neolithic and Bronze Age sites in the Hutuo River Valley(HTRV)in north-central China and compare them with updated archaeobotanical studies in the Sushui River Valley(SSRV)and Henan Province(HNP),to explore the similarities and differences of agricultural patterns under different geomorphologic and climatic environments in north-central *** results reveal that humans consistently cultivated foxtail and broomcorn millet in the HTRV from the Miaodigou(6000–5700 cal.a BP)to the Shang-Zhou(3600–2256 cal.a BP)period,despite the introduction of wheat and barley into the area around 4000 a *** conditions and hilly landforms in the HTRV,SSRV and some parts of the HNP led people to develop foxtail and broomcorn millet dry land farming practices between 7000 and 3000 a ***,in other areas of the HNP,the climate conditions and alluvial plains enabled people to develop a mixed agriculture of millets,soybeans,and rice from 7000 to 4000 a BP,with the addition of wheat between 4000 and 3000 a ***’different agricultural technologies and interactions with foreign cultures may have also influenced the formation of different agricultural patterns in the three regions between 7000 and 3000 a *** growth during 7000–3000 a BP can explain the overall propensity of the higher-yield foxtail millet rather than broomcorn millet as the main cultivated *** the HTRV,however,higher proportions of broomcorn millet(as compared to the SSRV and HNP),may reflect the greater drought and heat tolerance of broomcorn millet,representing an adaptive agricultural strategy in the river valley.
