Immunomodulatory activity of polycaprolactone nanoparticles with calcium phosphate salts against Leishmania infantum infection

Objective:To prepare and characterize polycaprolactone(PCL)nanoparticles loaded with sonicator fragmented(SLA)and freeze-thaw Leishmania antigens(FTLA)and to investigate the in vitro immunogenicity of antigen-encapsulated nanoparticles with calcium phosphate ***:The water/oil/water binary emulsion solvent evaporation method was used to synthesize antigen-loaded PCL *** were characterized by scanning electron microscopy and zeta potential *** cytotoxicity in J774 macrophages in vitro was determined by MTT *** addition,the amount of nitric oxide and the level of cytokines produced by macrophages were determined by griess reaction and ELISA method,*** protective effect of the developed formulations was evaluated by determining the infection index percentage in macrophages infected with Leishmania ***:Compared to the control group,SLA PCL and FTLA PCL nanoparticles with calcium phosphate adjuvant induced a 6-and 7-fold increase in nitric oxide,***,the vaccine formulations promoted the production of IFN-γand *** PCL and FTLA PCL nanoparticles combined with calcium phosphate adjuvant caused an approximately 13-and 11-fold reduction in infection index,respectively,compared to the control ***:The encapsulation of antigens obtained by both sonication and freeze-thawing into PCL nanoparticles and the formulations with calcium phosphate adjuvant show strong in vitro immune stimulating ***,PCL-based antigen delivery systems and calcium phosphate adjuvant are recommended as a potential vaccine candidate against leishmaniasis.

Naringin attenuates oxidative stress,inflammation,apoptosis,and oxidative DNA damage in acrylamide-induced nephrotoxicity in rats

Eocene magmatism(Maden Complex) in the Southeast Anatolian Orogenic Belt: Magma genesis and tectonic implications

The origin and geodynamic setting of the Maden Complex, which is situated in the Bitlis-Zagros Suture Zone in the Southeast Anatolian Orogenic Belt, is still controversial due to lack of systematic geological and geochemical data. Here we present new whole rock major-trace-rare earth element and Sre Nd isotope data from the Middle Eocene volcanic rocks exposed in Maden Complex and discuss their origin in the light of new and old data. The volcanic lithologies are represented mainly by basalt and andesite, and minor dacite that vary from low-k tholeiitic, calc-alkaline, high-k calc-alkaline, and shoshonitic in composition. They exhibit enrichments in large ion lithophile and light rare earth elements, with depletions in high field strength elements. Basaltic rocks have uniform Sr and Nd isotope ratios with high εNd(t) values varying from t5.5 to t6.7, in contrast to, andesitic rocks are characterized by low εNd(t) values ranging from à1.6 to à10. These geochemical and isotopic characteristics indicate that two end-members, a subduction-related mantle source and a continental crust, were involved in the magma genesis. Considering all geological and geochemical data, we suggest that the Eocene Maden magmatism occurred as a post-collisional product by asthenospheric upwelling owing to convective removal of the lithosphere during an extensional collapse of the Southeast Anatolian ranges.

Evaluation of in vitro and in vivo immunostimulatory activities of poly(lactic-co-glycolic acid) nanoparticles loaded with soluble and autoclaved Leishmania infantum antigens: A novel vaccine candidate against visceral leishmaniasis

Objective: To prepare and characterize poly lactic-co-glycolic acid(PLgA) nanoparticles loaded with soluble leishmanial antigen or autoclaved leishmanial antigen and explore in vitro and in vivo immunogenicity of antigen encapsulated nanoparticles. Methods: Water/oil/water double emulsion technique was employed to synthesize PLgA nanoparticles, and scanning electron microscopy, Fourier transform infrared spectroscopy and Zeta-potential measurements were used to identify the characteristics of nanoparticles. Cytotoxicity of synthetized nanoparticles on J774 macrophage were investigated by MTT assays. To determine the in vitro immunostimulatory efficacies of nanoparticles, griess reaction and ELISA was used to measure the amounts of NO and cytokines. During the in vivo analysis, Balb/c mice were immunized with vaccine formulations, and protective properties of nanoparticles were measured by Leishman Donovan unit in the liver following the infection. Cytokine levels in spleens of mice were determined by ELISA. Results: MTT assay showed that neither soluble leishmanial antigen nor autoclaved leishmanial antigen encapsulated nanoparticles showed cytotoxicity against J774 macrophage cells. Contrary to free antigens, both autoclaved leishmanial antigen-nanoparticle and soluble leishmanial antigen-nanoparticle formulations led to a 10 and 16-fold increase in NO amounts by macrophages, respectively. Leishman Donovan unit calculations revealed that soluble leishmanial antigen-nanoparticles and autoclaved leishmanial antigen-nanoparticles yielded 52% and 64% protection against visceral leishmaniasis in mouse models. Besides, in vitro and in vivo tests demonstrated that by increasing IFN-γ and IL-12 levels and inhibiting IL-4 and IL-10 secretions, autoclaved leishmanial antigen-nanoparticles and soluble leishmanial antigennanoparticles triggered Th1 immune response. Effect of Breathing on Exposed Lung Volumes and Doses in Patients with Breast Carcinoma Receiving Radiotherapy

Introduction: This study evaluates the changes in the lung volume (LV) exposed radiation during the breath cycle and whether these volume differences have an effect on both lung and target doses in breast carcinoma patients. Material and Methods: Ten patients with left breast carcinoma underwent breast conservative surgery or mastectomy receiving radiotherapy (RT) (breast or chest wall and regional lymph nodes) were included. For this study, planning computerized tomography (CT) images were obtained during deep inspiration (DI) and end of expiration (EE), besides free breathing (FB) to simulate breath cycles. Three-dimensional conformal or intensity-modulated RT planning was done to obtain dose-volume information using CT series taken FB, DI and EE. The treatment plan was done with FB images and exported to the DI and EE scans and re-calculated. Volume changes and calculated dose differences according to breath cycles were compared. Results: There were significant differences in the whole LV, ipsilateral LV and contralateral LV between FB-DI and EE-DI while no significant difference was seen between FB and EE. V20 was lower during DI than FB and EE but the difference was not significant. There was no significant variation in whole breast dose although significant dose variations were observed in mean MI, supraclaviculary and level III axillary lymph node doses between breath cycles. Conclusion: Breath cycle had no significant effect on whole breast dose although significantly changed regional lymph node doses in patients with breast carcinoma receiving whole breast and regional lymph nodes radio-therapy. V20 dose was lower during DI than FB and EE, but the difference was not significant.

A new approach for development of vaccine against visceral leishmaniasis: Lipophosphoglycan and polyacrylic acid conjugates

Objective: To determine the antileishmanial vaccine effectiveness of lipophosphoglycan(LPg) and polyacrylic acids(PAA) conjugates on in vivo mice ***: LPg molecule was isolated and purified from large-scale Leishmania donovani parasite culture. Protection efficacies of LPg alone, in combination with Freund's adjuvant, in a physical mixture and in conjugate(consisting of various LPg concentrations) with PAA, were comparatively determined by various techniques, such as cultivation with the micro-culture method, assessment of in vitro infection rates of peritoneal macrophages, determination of parasite load in liver with Leishman-Donovan Units, and detection of cytokine ***: Obtained results demonstrated that the highest vaccine-mediated immune protection was provided by LPg-PAA conjugate due to all parameters investigated. According to the Leishman-Donovan Units results, the sharpest decline in parasite load was seen with a ratio of 81.17% when 35 mg LPg containing conjugate was applied. This value was 44.93% for the control group immunized only with LPg. Moreover, decreases in parasite load were 53.37%, 55.2% and 65.8% for the groups immunized with 10 mg LPg containing LPg-PAA conjugate, a physical mixture of the LPg–PAA, and a mixture of LPg + Freund's adjuvant, respectively. Furthermore, cytokine results supported that Th1 mediated protection occurred when mice were immunized with LPg-PAA ***: It has been demonstrated in this study that conjugate of LPg and PAA has an antileishmanial vaccine effect against visceral leishmaniasis. In this respect, the present study may lead to new vaccine approaches based on high immunogenic LPg molecule and adjuvant polymers in fighting against Leishmania infection.

The Coordination Compound of ga(III) with Stillbazo in the Presence of Cetyl Pyridinium Chloride as a Surface Active Compound

The coordination compound of gallium (III) with stillbazo-stillbene-4,4'-bis [(1-azo)3,4-dihydroxybenzene]-2,2'-disulfonic acid has been investigated by spectrophotometric method in the presence of cetyl pyridinium chloride (CPC) as surface active compound. It has been determined that the mixed ligand complexes were formed with the ratio of 1:2:4 in CPC-mediated phase. The molar coefficient and the stability constant of the complex compound βк = 6.8 × 109, εк = 66,834 have been studied by various spectrophotometric methods. The effect of pH, time, temperature, concentration of the reagents and CPC on formation of the unmixed and mixed ligand coordination compounds has been also studied. It has been determined that the Beer’s law is respected within 0.008 - 10 μg of ga(III) in 25 mL solution. The technique of the photometric determination of gallium in natural compositive objects and industrial objects has been developed.

The guleman ophiolite,one of the most important ophiolitic massifs of the Southeast Anatolian Ophiolitic Belt,consists of a core of serpentinized mantle rocks overlain by an ultramafic sequence,layered and isotropic gabbro,and sheeted *** ophiolite structurally overlies the Lower Miocene Lice Formation and is overlain by young sandstones and shales of the Upper Maashtrichtian-Lower Eocene Hazar Complex and Middle Eocene Maden *** guleman ophiolite tectonically overlain by Precambrian to Upper Triassic Bitlis metamorphic *** mantle peridotites compose mainly of fresh and in place serpentinized harzburgite tectonite with local bands and lenses of dunites with large-sized chromitite *** guleman peridotites commonly show porphyroclastic texture,high-temperature fabrics such as kink-bands in *** to microprobe analyses,the harzburgite and dunite have low Ca O and Al2O3 abundance similar to Mariana forearc,and their average Cr-(=Cr/(Cr+Al)atomic)ratio of Cr-spinelsis surprisingly high(>0.63)besides Fo content of olivine is between 90.9 to 92.3 in *** to Mg#(Mg/(Mg+Fe2+))versus Cr#in spinel diagram,the degree of partial melting is higher than 35%and spinel values plot in the forearc peridotites *** gulemanharzburgites have low Ca O,Al2O3 and Ti O2 contents in orthopyroxene and clinopyroxene lammelles,resembling those of depleted harzburgites from modern forearcs and different from moderately depleted abyssal ***,we propose that the guleman peridotites form in a forearc setting during the subduction initiation that developed as a result of northward subduction of the southern branch of the Neo-Tethys in response to the convergence between Arabian and Anatolian plates.

AIM:To investigate the performance of the microcapillary culture method(MCM) in Helicobacter pylori(***) isolation and ***:Microcapillary culture(MC),classical culture(CC),rapid urease(CLO) test,and histopathologic examination(HE) were performed with biopsy *** biopsy samples were loaded into capillary tubes and incubated for 48 h at 37 ℃ without providing a microaerophilic ***,three or four loops of the homogenized sample were inoculated in a ready-to-use selective medium(Becton Dickinson,Helicobacter Agar,Modified) specific for the isolation of *** and incubated at 37 ℃ in a microaerophilic atmosphere provided by Campy gen(Becton Dickinson,gas Pack).Bacteria reproducing in microcapillary tubes were evaluated in an inverted microscope and also were evaluated after performing a CC with the *** obtained by CC,CLO test,and HE were compared with those of *** diagnostic performances of the methods used in this study were evaluated for specificity,sensitivity,positive predictive value(PPV),negative predictive value(NPV),and ***:*** was found positive by CLO test +HE and/or CC culture in 26 patient antrum and corpus biopsy *** 25(25/26) patient biopsy samples,*** was isolated by MCM,whereas in only 14(14/26) patient biopsy samples,*** was isolatedby *** test and HE were found positive in 17(17/26) patient biopsy *** the results of the isolation of *** by MCM,CC,CLO test,and HE,the sensitivity of the MCM was found as 96%,the specificity as 80%,the PPV as 83%,the NPV as 95%,and the 95%CI as 0.76(χ2 =31.51,P gh diagnostic sensitivity compared with CC,HE,and CLO tests.
