Protein arginine methyltransferase-6 regulates heterogeneous nuclear ribonucleoprotein-F expression and is a potential target for the treatment of neuropathic pain

Protein arginine methyltransferase-6 participates in a range of biological functions,particularly RNA processing,transcription,chromatin remodeling,and endosomal ***,it remains unclear whether protein arginine methyl transferase-6 modifies neuropathic pain and,if so,what the mechanisms of this *** this study,protein arginine methyltransferase-6 expression levels and its effect on neuropathic pain were investigated in the spared nerve injury model,chronic constriction injury model and bone cancer pain model,using immunohistochemistry,western blotting,immunoprecipitation,and label-free proteomic *** results showed that protein arginine methyltransferase-6 mostly co-localized withβ-tubulinⅢin the dorsal root ganglion,and that its expression decreased following spared nerve injury,chronic constriction injury and bone cancer *** addition,PRMT6 knockout(Prmt6~(-/-))mice exhibited pain ***,the development of spared nerve injury-induced hypersensitivity to mechanical pain was attenuated by blocking the decrease in protein arginine methyltransferase-6 ***,when protein arginine methyltransferase-6 expression was downregulated in the dorsal root ganglion in mice without spared nerve injury,increased levels of phosphorylated extracellular signal-regulated kinases were observed in the ipsilateral dorsal horn,and the response to mechanical stimuli was ***,protein arginine methyltransferase-6 appeared to contribute to spared nerve injury-induced neuropathic pain by regulating the expression of heterogeneous nuclear ***,protein arginine methyltransfe rase-6-mediated modulation of hete rogeneous nuclear ribonucleoprotein-F expression required amino atids 319 to 388,but not classical H3R2 *** findings indicated that protein arginine methyltransferase-6 is a potential therapeutic target fo r the treatment of peripheral neuro pathic pain.

枫香树叶片变色的物质基础及其地理变异规律

为探究枫香树Liquidambar formosana叶片变色多样的物质基础,2016年,对收集的41个种源592个家系的枫香树种子开展种源试验,同年9月、11月、12月,分别测量枫香树叶片变色前、变色中和变色后3个阶段的叶片组分;2020年11月,测量选取绿、黄绿、黄、黄褐、红黄、粉红、红、深红和紫9个不同色系枫香树叶片的色素组分含量,通过主成分分析、相关分析和回归分析,比较不同颜色叶片的色素含量,分析叶片组分与种源地理和环境的关系,探究叶片变色的物质基础及其地理变异模式。结果表明:(1)枫香树叶片颜色差异主要归因于叶片中总叶绿素和花青素含量的变化,其中总叶绿素含量的变异系数为103.19%,花青素含量的变异系数为74.20%;其是紫色叶片中叶绿素、类胡萝卜素和花青素含量均保持较高水平。(2)主成分分析结果表明,在枫香树叶片变色过程中,种源间叶片颜色的差异主要来自于花青素、总叶绿素含量和pH值,相关分析结果显示,叶片中的花青素、总叶绿素含量与种源地理环境显著相关(P<0.05),但pH值与种源地理环境不相关。(3)回归曲线估计表明,变色后,叶片中的花青素含量与种源地经度呈二次函数模型,其他时间段叶片中的总叶绿素和花青素含量与种源地理环境的关系为一次线性关系,除经度和纬度外,种源地的1月均温也是叶片总叶绿素和花青素含量变化的关键因子。叶片变色前,总叶绿素含量呈东-西变异模式,花青素含量呈南-北变异模式;变色中,总叶绿素含量呈南-北变异模式,花青素含量呈东-西变异模式;变色后,总叶绿素含量呈东南-西北变异模式,花青素含量保持东-西变异模式。叶片色素变异模式的研究在种质资源选择和叶色选优上具有较强的指导价值,可为选育叶色稳定、一致和特异的枫香树品种提供依据。

Lenalidomide regulates the CCL21/CCR7/ERK1/2 axis to inhibit migration and proliferation in diffuse large B-cell lymphoma

Background:The prognostic significance of the chemokine receptor CCR7 in diffuse large B-cell lymphoma(DLBCL)has been reported ***,the detailed mechanisms of CCR7 in DLBCL,particularly regarding its interaction with lenalidomide treatment,are not fully ***:Our study utilized bioinformatics approaches to identify hub genes in SU-DHL-2 cell lines treated with lenalidomide compared to control *** data and clinical information from 122 patients with DLBCL were analyzed to assess the correlation of CCR7 and p-ERK1/2 expression with the prognosis of ***,in vitro and in vivo experiments were conducted to clarify the role of CCR7 in the response of DLBCL to lenalidomide ***:Our bioinformatics analysis pinpointed CCR7 as a hub gene in the context of lenalidomide treatment in ***,31.14%and 36.0%(44/122)of DLBCL cases showed positive expression for CCR7 and ERK1/2 respectively,establishing them as independent prognostic factors for adverse outcomes in DLBCL via multivariate Cox regression ***,our studies demonstrated that the external application of the protein CCL21 promoted proliferation,migration,invasion,and activation of the ERK1/2 pathway in SU-DHL-2 and OCI-LY3 cell lines with high levels of CCR7 *** effect was mitigated by CCR7 silencing through siRNA,application of ERK inhibitors,or lenalidomide *** vivo experiments reinforced the efficacy of lenalidomide,significantly reducing tumor growth rate,tumor mass,serum total LDH levels,and expression of CCR7 and p-ERK1/2 in a SUDHL-2 xenograft model in nude mice(p<0.05).Conclusion:Our study clarifies the potential role of the CCL21/CCR7/ERK1/2 axis in the therapeutic effects of lenalidomide in DLBCL treatment.

Rapid and scalable preparation of highly quaternized polymer networks to achieve efficient simultaneous adsorption of iodide and pertechnetate

The efficient separation of fission products,such as TcO4-and I-,holds strategic significance for the management of radioactive wastes and environmental *** this study,we propose an ultrafast strategy for scalable preparation of a highly quaternized organic network to facilitate efficient and synchronous separation of iodide and *** network can be prepared within a few minutes at room temperature,using polyethylenimine and 1,2,4,5-tetrakis（bromomethyl）benzene as building *** chlorine replacement,the network,abundantly decorated with quaternary ammonium groups （Cl@QPN）,exhibits an ultrahigh positive charge density of 7.6 mmol/*** enables the rapid and efficient enrichment of target anions through strong electrostatic Coulomb *** a result,Cl@QPN exhibits significantly higher adsorption rate constants of 0.830 g/（mg min） for ReO4-（a nonradioactive surrogate of TcO4-） and 0.677 g/（mg min） for I-compared to other ***,it possesses high adsorption capacities,reaching 1,681 mg/g for ReO4-and 917.4 mg/g for I-.Cl@QPN also demonstrates good selectivity towards target ions and shows efficient adsorption for99TcO4-.Additionally,Cl@QPN exhibits high dynamic processing capacities,handling up to 3,100 and 7,400 kg of simulated streams per kilogram of material for I-and ReO4-,respectively.

Photoinduced radical sulfur dioxide insertion with asymmetric cyclization of alkenes: accessing β-chiral sulfones bearing S-stereocentric cyclic sulfinamides

The generation of various β-chiral sulfones bearing chiral cyclic sulfinamides through photoredox-catalyzed sulfur dioxide insertion with asymmetric radical cyclization is described. This method features a broad substrate scope and excellent stereospecificity. Computational studies reveal that the dispersion interaction between sulfinyl group and CH2SO2Ph group plays a crucial role in the stereoselectivity control.

Establishing a new testing protocol of the electrochemical stability window for electric double-layer capacitors

The electrochemical stability window(ESW) is crucial for determining the energy density of electric double-layer capacitors(EDLCs). However, current ESW testing protocols of EDLCs are strongly affected by artificial factors, thereby leading to unreliable result. To solve this problem, we first identify the key shortcomings of the traditional protocols by analyzing their test principles. Subsequently, taking an aqueous carbon-based EDLC as an example, we design a side reaction descriptor to represent the response difference in the charging capacitance to side reactions between the cyclic voltammetry and galvanostatic chargedischarge methods. With the aid of this descriptor, we propose a new ESW testing protocol, which is not only able to completely avoid the adverse influence of subjective factors but also fully consider the operating condition limitations. The protocol can be extended from the single electrode to the full device. This work proposes a more rigorous definition of ESW and provides a reliable protocol for evaluating the ESW of EDLCs.

目的了解海珠区居民高血压的患病现状及其影响因素,为高血压的防治提供依据。方法采用多阶段整群随机抽样方法,对年满45岁以上的海珠区常住居民进行横断面调查,共调查1667人,调查内容包括:一般情况、高血压危险因素及体格检查,对所取得的年龄、性别、文化程度、吸烟、饮酒、家族史、体质指数、腰臀比等资料用SPSS17.0软件进行统计分析。结果 45岁以上人群高血压粗患病率为36.4%。多因素Logistic回归分析显示,年龄、文化程度、高血压家族史、BMI、血脂水平和糖尿病是高血压的主要影响因素。结论海珠区防治高血压应当采取全人群预防与高危人群干预相结合的策略,加强对中老年、文化程度低以及超重肥胖人群的健康教育,提倡合理膳食、加强体育锻炼、控制超重和肥胖,对防治高血压和控制心脑血管疾病具有重要意义。

采用X射线粉末衍射和扫描电子显微镜研究了保温时间对废催化裂化平衡剂合成莫来石的影响。结果表明,二次莫来石化反应发生在1 450℃,保温3 h后,莫来石质量分数达94%,晶体发育良好;当保温时间超过4 h,由于反应物中Si4+被Al3+取代,导致空位增多,过渡金属进入结构的空位,使莫来石熔点降低,表现为保温时间越长,液相越多,莫来石晶体粘结于液相之中,使机械性能降低。由此可见,保温时间对二次莫来石生成反应影响较大。
