Screening of hepatocyte proteins binding to complete S protein of hepatitis B virus by yeast-two hybrid system

AIM: To investigate the biological function of complete S protein and to look for proteins interacting with complete S protein in hepatocytes. METHODS: We constructed bait plasmid expressing complete S protein of HBV by cloning the gene of complete S protein into pGBKT7, then the recombinant plasmid DNA was transformed into yeast AH109 (a type). The transformed yeast AH109 was mated with yeast Y187 (α type) containing liver cDNA library plasmid in 2×YPDA medium. Diploid yeast was plated on synthetic dropout nutrient medium (SD/-Trp-Leu-His-Ade) containing X-α-gal for selection and screening. After extracting and sequencing of plasmids from positive (blue) colonies, we underwent sequence analysis by bioinformatics. RESULTS: Nineteen colonies were selected and sequenced. Among them, five colonies were Homo sapiens solute carrier family 25, member 23 (SLC25A23), one was Homo sapiens ***, one was human serum albumin (ALB) gene, one was Homo sapiens metallothionein 2A, two were Homo sapiens betaine-homocysteine methyltransferase, three were Homo sapiens Na+ and H+coupled amino acid transport system N, one was Homo sapiens CD81 antigen (target of anti-proliferative antibody 1) (CD81), three were Homo sapiens diazepam binding inhibitor, two colonies were new genes with unknown function. CONCLUSION: The yeast-two hybrid system is an effective method for identifying hepatocyte proteins interacting with complete S protein of HBV. The complete S protein may bind to different proteins i.e., its multiple functions in vivo.

Incubation strategies of the Black-necked Crane(Grus nigricollis) in relation to ambient temperature and time of day

Background: The behavior of cranes reflects many of their survival strategies, but little has been known of the incubation strategies of cranes, in which both parents share incubation duties, in response to cold temperatures in alpine environments. The lack of information may restrict the effective conservation of the threatened Black-necked Crane(Grus nigricollis), a biparental bird nesting in high elevation ***: We directly observed and used infrared video cameras from 2014 to 2015 to study the incubation behavior and quantitatively measured the frequency and details of egg turning behavior in the Black-necked Crane at the Yanchiwan National Nature Reserve in western Gansu Province, ***: At lower ambient temperatures in the morning, crane parents spent more time on the nest with less recess frequency and prolonged on-bout duration, while at higher temperatures around noon, the parents had more frequent recesses from incubation and shorter periods between nest exchanges. They adjusted the amount of time incubating by varying the recess frequency and the length of on-bout duration. Mean nest attendance and egg turning frequency of females were significantly higher than those of the males. The nest attendance and on-bout duration of females showed a significantly negative relationship with those of males. The two parents responded differently to the change of temperature. Females spent more time on the nest at lower morning temperatures, while males increased their time on the nest at higher temperatures after noon. Higher incubation recess frequency and egg turning frequency were observed at noon, probably because parents spent more time foraging, taking advantage of the lower egg cooling ***: Both Black-necked Crane parents in the alpine environment adjusted their behavior in response to the thermal requirements of eggs and the weather conditions experienced. Engineering phonon transport in low-dimensional materials has great significance not only for fundamental research,but also for thermal management applications of electric ***,due to the difficulties of micro and nano processing and characterization techniques,the work on tuning phonon transport at nanoscale are *** this work,by introducing Ar+plasma,we probed the phonon transport in two-dimensional(2D)layered semiconductor PdSe_(2)under different defect *** using thermal bridge method,the thermal conductivity was measured to decrease by 50%after a certain Ar+irradiation,which implied a possible phase ***,Raman characterizations were performed to show that the Raman sensitive peaks of PdSe_(2)was red-shifted and finally became disappeared with the increase of defect concentration.“Defect engineering”proves be a practical strategy in tuning the phonon thermal transport in low-dimensional materials,thus providing guidance for potential application in designing thermoelectric devices with various emerging materials.

This paper investigates the consensus control of multi-agent systems(MASs) with constrained input using the dynamic event-triggered mechanism(ETM).Consider the MASs with small-scale networks where a centralized dynamic ETM with global information of the MASs is first ***,a distributed dynamic ETM which only uses local information is developed for the MASs with large-scale *** is shown that the semi-global consensus of the MASs can be achieved by the designed bounded control protocol where the Zeno phenomenon is eliminated by a designable minimum inter-event *** addition,it is easier to find a trade-off between the convergence rate and the minimum inter-event time by an adjustable ***,the results are extended to regional consensus of the MASs with the bounded control *** simulations show the effectiveness of the proposed approach.

Due to the small models and compact structures,small harvesters have also caused problems such as poor threshing separation performance and large loss *** order to solve unstable cleaning effects of small harvesters when they facing different working conditions,this study selected rice plants in hilly areas for the *** breaking force of different parts of mature rice was tested,which revealed the fracture mechanism of each part under the combined *** threshing method was used to simulate artificial plate bin and design three kinds of non-circular pulley variable speed transmission threshing *** the help of transient inertia force,threshing force was *** paper tested the harvesting performance of the variable speed threshing device with the help of the harvest performance *** show when facing the small rice planting area,the T/2 variable speed threshing device has better cleaning performance,and also the harvest loss rate of T/4 variable speed threshing device is the *** with the constant speed threshing device,the impurity content rate of the variable speed threshing device is increased by 0.64%to 8.76%;the loss rate is reduced by 0.45%to 1.79%,which provides a basis for the optimization design of small combine harvester in hilly areas.

Evapotranspiration(ET)is a crucial variable in the terrestrial water,carbon,and energy *** present,a large number of multi source ET products *** to sparse observations,however,great challenges exist in the evaluation and integration of ET products in remote and complex areas such as the Tibetan Plateau(TP).In this paper,the applicability of the multiple collocation(MC)method over the TP is evaluated for the first time,and the uncertainty of multisource ET products(based on reanalysis,remote sensing,and land surface models)is further analyzed,which provides a theoretical basis for ET data *** results show that 1)ET uncertainties quantified via the MC method are lower in RS-based ET products(5.95 vs.7.06 mm month^(-1))than in LSM ET products(10.22 vs.17.97 mm month^(-1))and reanalysis ET estimates(7.27 vs.12.26 mm month-1).2)A multisource evapotranspiration(MET)dataset is generated at a monthly temporal scale with a spatial resolution of 0.25°across the TP during *** has better performance than any individual product.3)Based on the fusion product,the total ET amount over the TP and its patterns of spatiotemporal variability are clearly *** annual total ET over the entire TP is approximately 380.60 ***,an increasing trend of 1.59±0.85 mm yr^(-1)over the TP is shown during *** study provides a basis for future studies on water and energy cycles and water resource management over the TP and surrounding regions.

In this study,a new image-based method for the extraction and characterization of pore-throat network for unconventional hydrocarbon storage and exploitation is proposed.“Pore-throat solidity”,which is analogous to particle solidity,and a new method for automatic identification of pores and throats in tight sandstone oil reservoirs are ***,the“pore-throat combination”and“pure pore”are defined and distinguished by drawing the cumulative probability curve of the pore-throat solidity and by selecting an appropriate cutoff *** the discrete grid set is recognized as a pore-throat combination,Legendre ellipse fitting and minimum Feret diameter are *** the pore and throat grid sets are identified as pure pores,the pore diameter can be directly *** the new method,the analytical results for the physical parameters and pore radius agree well with most prior *** results comparing the maximum ball and the new model could also prove the accuracy of the latter's in micro and nano *** new model provides a more practical theoretical basis and a new calculation method for the rapid and accurate evaluation of the complex processes of oil migration.

In this work,a variety of Cu_(x)Ni_(2-x)Fe(CN)_(6)(x=0,0.4,0.8,1.2,1.6,2)cathodes for ammonium ion batteries are prepared and their electrochemical performances are *** the introduction of copper in nickel hexacyanoferrate,the electrochemical performance varies without changing the structure of nickel *** increase of Cu content in nickel hexacyanoferrate leads to the enhancement of reaction potential and *** results suggest that the substitution of Cu for Ni has a positive effect on improving the cycling stability and rate capacity of nickel hexacyanoferrate when x in Cu_(x)Ni_(2-x)Fe(CN)_(6)is less than ***,Cu_(0.4)Ni_(1.6)Fe(CN)_(6)exhibits the best cycling per-formance(capacity retention of 97.54%at 0.3 C)and the highest rate capacity(41.4 mAh g^(-1)at 10 C)in Cu_(x)Ni_(2-x)Fe(CN)_(6).Additionally,the X-ray diffraction(XRD)and X-ray photoelectron spectroscopy(XPS)tests also reveal that the structural evolution of Cu_(0.4)Ni_(1.6)Fe(CN)_(6)is highly reversible upon NH_(4)^(+)***,this work proposes a candidate material for ammoniumion batteries and offers a novel avenue for adjusting the operating potential of the material.
