Surfen and oxalyl surfen decrease tau hyperphosphorylation and mitigate neuron deficits in vivo in a zebrafish model of tauopathy

Background:Tauopathies comprise a family of neurodegenerative disorders including Alzheimer’s disease for which there is an urgent and unmet need for disease-modifying *** are characterized by pathological tau hyperphosphorylation,which has been shown to correlate tightly with disease progression and memory loss in patients suffering from Alzheimer’s *** recently demonstrated an essential requirement for 3-O-sulfated heparan sulfate in pathological tau hyperphosphorylation in zebrafish,a prominent model organism for human drug ***,we investigated whether in vivo treatment with surfen or its derivatives oxalyl surfen and hemisurfen,small molecules with heparan sulfate antagonist properties,could mitigate tau hyperphosphorylation and neuronal deficits in a zebrafish model of ***:In vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos for 2 days with surfen or oxalyl surfen significantly reduced the accumulation of the pThr181 tau phospho-epitope measured by ELISA by 30%and 51%,*** blot analysis also showed a significant decrease of pThr181 and pSer396/pSer404 in embryos treated with surfen or oxalyl *** analysis further confirmed that treatment with surfen or oxalyl surfen significantly decreased the AT8 tau epitope in spinal *** addition,in vivo treatment of Tg[HuC::hTau^(P301L);DsRed]embryos with surfen or oxalyl surfen significantly rescued spinal motoneuron axon-branching defects and,as a likely consequence,the impaired stereotypical touch-evoked escape ***,treatment with hemisurfen,a surfen derivative devoid of heparan sulfate antagonist activity,does not affect tau hyperphosphorylation,nor neuronal or behavioural deficits in Tg[HuC::hTau^(P301L);DsRed]***:Our findings demonstrate for the first time that surfen,a well-tolerated molecule in clinical settings,and its derivative,oxalyl surfen,could mitigate or delay neuronal defects

Evaluation of Environmental Requirements for Sustainable Ready-Mix Concrete Production in Abu Dhabi Emirate

Sustainability is the main concern for current and future construction projects in the world. In any project, attaining sustainability requires the integrity of four main elements. These elements cover economical, societal, cultural, and environmental aspects to its successful implementation and integration in an anthropogenic framework. Concrete is one of the main building materials used in current construction activities around the world;therefore, many issues pertaining to concrete long term sustainability arise. This makes issues related to environmental evaluation and assessment for the ready-mix concrete production process of vital importance. The release of particulate matter (PM) including PM10 and PM2.5 could be considered the main air pollutant that is emitted from this industrial sector;where PM can be released from different stages of ready-mix concrete production process. Another concern arising from the ready-mix concrete operations which may affect the soil and ground water quality is the spills or leaks that may occur from the storage containers used for diesel, motor oil, and chemical admixture. In 2010, Environmental Agency-Abu Dhabi (EAD) collected data pertaining to the operations of 73 ready-mix concrete facilities in the Emirate of Abu Dhabi. The data showed that the average production rate of concrete is around 250,000 m3/year, which indicates that the concrete industry is a very active industry in the Emirate. Hence, EAD has established a set of plans and requirements to minimize the environmental impacts of concrete production on the environment and public health. The environmental impact of the concrete manufacturing can be reduced by conserving use of raw material ingredients, conserving energy consumption, and following Best Management Practices (BMPs) related to readymix concrete production. Phobia Treatment with Face-Your-Fear VR App

Technology plays an important role in people’s daily life, for they depend on it to perform many tasks. Among the prominent technologies are smartphones and Virtual Reality (VR). Developing mobile apps and integrating VR technology allow users to experience an artificial environment as if it is real such as the simulation of phobias. This research paper proposes a mobile application named Face-Your-Fear (FyF) integrating VR to help any user to live a cybernetic experience, overcome the illness, know about phobia symptoms, and get extra information on two types of phobia: aquaphobia and achluophobia. Its architecture, features, development, interfaces and limitations are presented, including the main results of the qualitative survey to users.

Neonatal Outcome of Induced Prematurity for Severe Preeclampsia in Four Great Kinshasa Maternities

Severe preeclampsia (SPE) is associated with fetal complications including intrauterine growth retardation (IUGR), prematurity and in utero fetal death. Its treatment remains child birth that often is planned before term. However, this attitude can lead to fetal complications related to prematurity. Several studies on preeclampsia have already been studied in the DRC and several aspects have already been realized, but to date, the neonatal outcome has not yet been addressed. Methods: This is cross-sectional study performed in four public hospitals in Kinshasa (Democratic Republic of Congo). We included 400 cases of induced prematurity (IP) for SPE;the analysis compared pregnant women who gave birth before 34 weeks of amenorrhea (WA) and those after 34 WA. The comparison of the proportions was made by the Chi-square test and the calculation of Means by the Student’s test. The significance level was set at P Objective: To determine the frequency of induced prematurity for severe preeclampsia (SPE), to identify the indications and to evaluate neonatal outcome. Results: The IP frequency for SPE was 46.2%. The retro placental hematoma was the most indication in pregnancies before 34 WA 24.9%, while high blood pressure 54.5% in the after 34 WA group. Modeling and Optimization of Relocation of Some Heavy Metals and Micro-nutrients in the Argan Trees

This work constitutes a contribution to the study of the physical-chemical performance of some heavy metals and micro-nutrients in the Argan trees. The samples are harvested from small Argan trees, three samples （soil, wood, leaves） per tree. The content of various heavy metals and micro-nutrient in aerial parts （leaves, wood and soil） were analyzed in the Argan tree by the method of ICP-ES. The results of analysis of data for the chemical variables were measured for several trees at different measurement times, do not affect the dispersion of the data. The use of chemometrics has allowed us to interpret the results obtained especially those related to the characterization of heavy metals and micro-nutrient after calibration by variable regression of these results in order to get an overall estimate of the optimal content, and to follow changes in the methods of treatment. The results of the characteristics of Argan trees have been treated using the statistical method of Principal Component Analysis （MFA）, The MFA was used to analyze the three groups simultaneously. These groups are fairly similar.

Determinants of Outcomes and Prognosis Score in Obstetric Vesico-Vaginal Fistula Repair

An estimated 2 million women living in countries with limited resources currently have vesico-vaginal, recto-vaginal, or mixed types of fistulae because of the necrosis that occurs with obstructed labor. We evaluated factors readily assessed by the examining practitioner in a consecutive case series of surgical repairs of obstetrical fistulae, for the ability of those factors to stand as a prognostic guide, in a clinical score. Objectives: To identify the predictors of surgical repair outcomes and establish a Score combining these different determinants to facilitate the care of obstetric fistula. Methods: We conducted a multicentric prospective study between 2011 and 2014 in Democratic Republic of the Congo (DRC). Outcomes: We measured 3 months post-surgery in a series of 483 patients with obstetrical fistula repaired by the same surgeon included closure and failure appreciated by dye test. Multivariable generalized estimating equation models were used to generate adjusted odd ratios (OR) and 95% confidence intervals (CIs). The scores ranging from 3 to 14 were established from the outcomes determinants identified. Results: In total, 483 women were enrolled, and 390 cases were at their first surgery and were included in the obstetric fistula (OF) prognosis score and classification. Their mean age was 35 years at the time of the surgery and 25 years at the onset of OF. In 28.6%, the fistula patient was primigravida. Mean duration between onset of the fistula and surgical treatment was 8 years. In 24%, the fistula patients lived separated from their partners. Overall closure rate of the fistulas was 85.7%. Severe vaginal fibrosis (p 0.01), big fistula size (0.01), small distance from fistula to external urethral meatus (0.01) and prior surgery (p 0.48) predicted failed fistula closure. Objective: Pelvic organ prolapse is an emerging public health problem affecting adult women of all ages with a negative impact on social, physical well-being, and psychological. Its presents several challenges in countries with low resources. This literature review aims to examine POP in its epidemiological aspects, risk factors, and staging by taking up the challenges associated with low-resource settings and identifying some avenues for future research. Methods: We searched the PubMed, Google Scholar, and Scopus databases. The other studies were identified by checking the secondary references in the original citation. We have collected studies on adult women published in English for the last 30 years. In total, 71 articles were read. We excluded studies from all newspaper articles, Studies presenting co-morbidities (fistulas, cervical cancer, pregnancy), those evaluating treatment, letters, comments, case reports, practice guidelines, news, historical articles, legal cases, published erratum, and congresses. Results: 16 studies examining the epidemiology have been identified with 11 in countries defined by the World Bank as limited or intermediate resources. 18 on risk factors whose 10 in countries with limited or intermediate resources, 10 on staging and 27 on physiopathology. Conclusion: POP affects the young more in low-resource settings. Its prevalence remains underestimated for several reasons. Several risk factors found are the same as those of women in countries with a high standard of living. However, there are some specific risk factors for these resource-limited settings.

Severe Wolfram Syndrome Caused by a Novel Frameshift Mutation in <i>WFS1</i>Gene: Effect on the WFS1/CaM Interaction and Phenotype-Genotype Correlation

Mutations in the WFS1 gene have been reported in Wolfram syndrome (WFS), a rare and autosomal recessive disorder defined by early onset of diabetes mellitus and progressive optic and hearing impairment. Only few data are available concerning the association between clinical and molecular aspects of the WFS. We present a consanguineous family with a patient presenting an early onset of WFS and severe manifestations. Sequencing of WFS1 gene was performed for all the family members to search for responsible mutation and bioinformatics tools were conducted to predict its effect on structure and function of the protein. We have detected a novel frameshift mutation in the proband at homozygous state and at the heterozygous state in the parents who have no WFS manifestations. In silico analysis predicted the pathogenicity of the mutation and could lead to a complete loss of its function. Thus, 3D modeling showed that the mutation abolishes the interaction of the CaM binding region to the N-terminal of WFS1 and then impairs the WFS1-CaM complex formation. Genotype-phenotype correlation study shows that the novel mutation predisposes to early onset of diabetes and severe symptoms observed in the proband. We also report the effect of the frameshift mutation on the CaM-WFS1 impaired binding, and we discuss its possible consequence in pancreatic β-cells dysfunction and its role in the early onset of diabetes.
