Nicotinamide adenine dinucleotide treatment confers resistance to neonatal ischemia and hypoxia:effects on neurobehavioral phenotypes

Neonatal hypoxic-ischemic brain injury is the main cause of hypoxic-ischemic encephalopathy and cerebral palsy.Currently,there are few effective clinical treatments for neonatal hypoxic-ischemic brain injury.Here,we investigated the neuroprotective and molecular mechanisms of exogenous nicotinamide adenine dinucleotide,which can protect against hypoxic injury in adulthood,in a mouse model of neonatal hypoxic-ischemic brain injury.In this study,nicotinamide adenine dinucleotide(5 mg/kg)was intraperitoneally administered 30 minutes befo re surgery and every 24 hours thereafter.The results showed that nicotinamide adenine dinucleotide treatment improved body weight,brain structure,adenosine triphosphate levels,oxidative damage,neurobehavioral test outcomes,and seizure threshold in experimental mice.Tandem mass tag proteomics revealed that numerous proteins were altered after nicotinamide adenine dinucleotide treatment in hypoxic-ischemic brain injury mice.Parallel reaction monitoring and western blotting confirmed changes in the expression levels of proteins including serine(or cysteine)peptidase inhibitor,clade A,member 3N,fibronectin 1,5'-nucleotidase,cytosolic IA,microtubule associated protein 2,and complexin 2.Proteomics analyses showed that nicotinamide adenine dinucleotide ameliorated hypoxic-ischemic injury through inflammation-related signaling pathways(e.g.,nuclear factor-kappa B,mitogen-activated protein kinase,and phosphatidylinositol 3 kinase/protein kinase B).These findings suggest that nicotinamide adenine dinucleotide treatment can improve neurobehavioral phenotypes in hypoxic-ischemic brain injury mice through inflammation-related pathways.

Haploinsufficiency of Lipin3 leads to hypertriglyceridemia and obesity by disrupting the expression and nucleocytoplasmic localization of Lipin1

Lipin proteins including Lipin 1–3 act as transcriptional co-activators and phosphatidic acid phosphohydrolase enzymes,which play crucial roles in lipid metabolism.However,little is known about the function of Lipin3 in triglyceride(TG)metabolism.Here,we identified a novel mutation(NM_001301860:p.1835A>T/p.D612V)of Lipin3 in a large family with hypertriglyceridemia(HTG)and obesity through whole-exome sequencing and Sanger sequencing.Functional studies revealed that the novel variant altered the half-life and stability of the Lipin3 protein.Hence,we generated Lipin3 heterozygous knockout(Lipin3-heKO)mice and cultured primary hepatocytes to explore the pathophysiological roles of Lipin3 in TG metabolism.We found that Lipin3-heKO mice exhibited obvious obesity,HTG,and non-alcoholic fatty liver disorder.Mechanistic study demonstrated that the haploinsufficiency of Lipin3 in primary hepatocytes may induce the overexpression and abnormal distribution of Lipin1 in cytosol and nucleoplasm.The increased expression of Lipin1 in cytosol may contribute to TG anabolism,and the decreased Lipin1 in nucleoplasm can reduce PGC1α,further leading to mitochondrial dysfunction and reduced TG catabolism.Our study suggested that Lipin3 was a novel disease-causing gene inducing obesity and HTG.We also established a relationship between Lipin3 and mitochondrial dysfunction.

A Skin-Inspired Self-Adaptive System for Temperature Control During Dynamic Wound Healing

The thermoregulating function of skin that is capable of maintaining body temperature within a thermostatic state is critical.However,patients suffering from skin damage are struggling with the surrounding scene and situational awareness.Here,we report an interactive self-regulation electronic system by mimicking the human thermos-reception system.The skin-inspired self-adaptive system is composed of two highly sensitive thermistors(thermal-response composite materials),and a low-power temperature control unit(Laserinduced graphene array).The biomimetic skin can realize self-adjusting in the range of 35–42℃,which is around physiological temperature.This thermoregulation system also contributed to skin barrier formation and wound healing.Across wound models,the treatment group healed~10%more rapidly compared with the control group,and showed reduced inflammation,thus enhancing skin tissue regeneration.The skin-inspired self-adaptive system holds substantial promise for nextgeneration robotic and medical devices.

Nanoplatform-based cellular reactive oxygen species regulation for enhanced oncotherapy and tumor resistance alleviation

The cancer cells realize their proliferation and metastasis activities based on the special redox adaptation to increased reactive oxygen species(ROS)level,which inversely makes them sensitive to external interference with their redox state.In view of this,in recent decades,researchers have made great efforts to construct a series of novel nanoplatform-based ROS-mediated cancer therapies through increasing ROS generation and inhibiting the ROS elimination.Besides,the multidrug resistance and thermoresistance of tumor are closely related to tumor redox state.Recently,numerous works have shown that ROS regulation in cancer cells can intervene in the expression,function and stability of related proteins to achieve reversal of tumor resistance.In this review,the recent researches about ROS-regulating nanoagents on cancer therapy and tumor resistance alleviation have been well summarized.Finally,the challenges and research directions of ROS-regulating nanoagents for future clinical translation are also discussed.

Constructing on-demand single/multi-color transitioning fabrics with photocatalysis/photothermal-armed deficient semiconductors

On-demand color switching systems that utilize synchronized semiconductor-catalyzed reduction and photothermal-accelerated oxidation in liquid/solid are highly appealing.Herein,on-demand single/multi-color switching fabrics have been constructed by using defective SnO_(2):Sb-based color switching systems.SnO_(2):Sb nanocrystals with the suitable doping concentration accord lattices with abundant free electrons,conferring high photocatalytic and photothermal performances.A well-crafted set of dual light-responsive semiconductor-catalyzed systems with rapid color change can be attained via the homogenous mixture of SnO_(2):Sb with suitable redox dyes to produce single-color(RGB(red,green,blue))and multi-color transitioning(purple and green)systems.The illumination of these systems by 450 nm light triggers rapid photocatalytic discoloration,while irradiation by 980 nm light confers the photothermal effect that accelerates recoloration in air.Besides,the inks can be extended to rewritable fabrics by embedding the nanocrystals and redox dyes into hydroxyethyl cellulose(as the polymer matrix)and then coating on hydrophobic cotton fabrics to produce photo-switchable fabrics with excellent single/multi-color response.By exploiting the dual light interactions with the semiconductor-mediated systems,various images/letters can be remotely printed and erased on the rewritable fabrics which show promise for potential applications as information storage media and visual sensors.Importantly,the present rewritable fabric shows good stability and reversibility.The present work provides insights into the development of novel color-switching materials.

Characteristics of transfer zones under the influence of preexisting faults and regional stress transformation:Wenchang A subsag,Zhujiang River Mouth Basin,northern South China Sea

A transfer zone in rift basins preserves important information on regional tectonic evolution and plays significant roles in hydrocarbon accumulation.Based on the systematic analysis of 3D seismic data and hydrocarbon accumulation conditions,the geometry,kinematics,and reservoir control of a large synthetic overlapping transfer zone in the south of the Wenchang A subsag in the Zhujiang(Pearl)River Mouth basin were investigated.Results indicate that the development and evolution of the transfer zone was controlled by the interaction between pre-existing faults and regional stress transformation.The intense rifting of the main faults of the transfer zone controlled the development of source rocks and faultcontrolled slope break paleogeomorphology.The strike-slip overprint since the Oligocene is conducive to the formation of a large-scale fault-anticline trap,and the secondary faults in the transfer zone contribute to the hydrocarbon transportation.The conjugate intersection area of the NE-and NW-trending faults offers more opportunity for hydrocarbon migration and accumulation.

为考察父母体罚与儿童日常执行功能、执行功能任务水平之间的关系,揭示长期父母体罚可能存在的累积效应。本研究采用连续3年追踪研究设计,以亲子冲突解决策略量表、执行功能行为评定量表和威斯康星卡片分类测验为工具对710名三、四年级小学生及其父母双亲进行测查。结果发现:(1)父母体罚初始水平越高、随时间下降越慢,儿童日常执行功能问题越多,父母体罚对儿童日常执行功能的影响均存在累积效应;(2)父母体罚初始水平越高,儿童执行功能任务水平越差,但父母体罚的发展变化不能预测儿童执行功能任务水平,父母体罚对儿童执行功能任务水平的影响不存在累积效应。研究结果表明长期父母体罚对儿童日常认知功能的消极影响存在累积效应。

Antibody-mediated rejection(ABMR)is a major cause of dysfunction and loss of transplanted kidney.The current treatments for ABMR involve nonspecific inhibition and clearance of T/B cells or plasma cells.However,the prognosis of patients following current treatment is poor.T follicular helper cells(Tfh)play an important role in allograft-specific antibodies secreting plasma cell(PC)development.Tfh cells are therefore considered to be important therapeutic targets for the treatment of antibody hypersecretion disorders,such as transplant rejection and autoimmune diseases.Tacrolimus(Tac),the primary immunosuppressant,prevents rejection by reducing T cell activation.However,its administration should be closely monitored to avoid serious side effects.In this study,we investigated whether Tac delivery to helper T(CD4^(+))cells using functionalized mesoporous nanoparticles can block Tfh cell differentiation after alloantigen exposure.Results showed that Tac delivery ameliorated humoral rejection injury in rodent kidney graft by suppressing Tfh cell development,PC,and donor-specific antibody(DSA)generation without causing severe side effects compared with delivery through the drug administration pathway.This study provides a promising therapeutic strategy for preventing humoral rejection in solid organ transplantation.The specific and controllable drug delivery avoids multiple disorder risks and side effects observed in currently used clinical approaches.

“通体性”是一种融合了新物质主义和物质女性主义相关理论成果的后人类主义理论。它认为,所有生命本质上都是物质的具身,同属包括人类在内的“超人类”整体,并发生持续不断的、物质性的、动态的及内在交互式的相互融通和塑造。“通体性”是西方学者在反对身心二元对立道路上的又一个理论成果,但是其理论缺陷也显而易见:首先,从物质性具身的“通”过渡到人类个体的道德和责任,这其中的逻辑转换比较生硬;其次,淡化了因为人类社会内部政治经济的不公平所带来的环境非正义等问题。我国儒家传统的“仁民爱物”哲学思想可为消解上述缺陷提供参照。它从情感的角度出发,从“亲亲”到“仁民”到“爱物”,推己及人,将“仁”延展到世间万物,其伦理逻辑更清晰,有助于弥补西方通体性理论的缺陷。

核心素养落地呼唤“新课型”,其理论阐述和实施模型本身就是学习科学与教学设计研究的有益探索,代表着利用教育科学技术改进课堂教学实践、提高效能的合理尝试。将新课型区分为“5+1”课型模式,探讨各种课型的含义以及彼此之间的关系,发现新手、熟手和高手在解决问题中的差异,将事实、概念、规则、原理、策略和动力六种知识与了解、理解和应用三种水平组成一个矩阵,作为新课型教育目标分类框架。鉴于提高教学质量同教师上好概念课、规则课和策略课直接相关,进一步重点探讨概念教学策略,包括采用扶放有度原则、抓住重要概念、做好三种表征和实现三个阶段,落实“讲解”和“示例”基本方法等,以阐明新课型中概念教学的有效路径。
