Comparison of methods for deriving phenotypes from incomplete observation data with an application to age at puberty in dairy cattle

Background Many phenotypes in animal breeding are derived from incomplete measures,especially if they are challenging or expensive to measure *** include time-dependent traits such as reproductive status,or *** measures for these traits result in phenotypes that are subject to left-,interval-and rightcensoring,where phenotypes are only known to fall below an upper bound,between a lower and upper bound,or above a lower bound *** we compare three methods for deriving phenotypes from incomplete data using age at first elevation(>1 ng/mL)in blood plasma progesterone(AGEP4),which generally coincides with onset of puberty,as an example *** We produced AGEP4 phenotypes from three blood samples collected at about 30-day intervals from approximately 5,000 Holstein–Friesian or Holstein–Friesian×Jersey cross-bred dairy heifers managed in 54 seasonal-calving,pasture-based herds in New *** used these actual data to simulate 7 different visit scenarios,increasing the extent of censoring by disregarding data from one or two of the three *** methods for deriving phenotypes from these data were explored:1)ordinal categorical variables which were analysed using categorical threshold analysis;2)continuous variables,with a penalty of 31 d assigned to right-censored phenotypes;and 3)continuous variables,sampled from within a lower and upper bound using a data augmentation *** Credibility intervals for heritability estimations overlapped across all methods and visit scenarios,but estimated heritabilities tended to be higher when left censoring was *** sires with at least 5 daughters,the correlations between estimated breeding values(EBVs)from our three-visit scenario and each reduced data scenario varied by method,ranging from 0.65 to *** estimated breed effects also varied by method,but breed differences were smaller as phenotype censoring *** Our results indicate that using

Following the recent report by Dasenbrock-Gammon et al.[Nature 615,244–250(2023)]of near-ambient superconductivity in nitrogendoped lutetium trihydride(LuH_(3-δ)N_(ε)),significant debate has emerged surrounding the composition and interpretation of the observed sharp resistance ***,we meticulously revisit these claims through comprehensive characterization and *** definitively identify the reported material as lutetium dihydride(LuH_(2)),resolving the ambiguity surrounding its *** similar conditions(270–295 K and 1–2 GPa),we replicate the reported sharp decrease in electrical resistance with a 30%success rate,aligning with the observations by Dasenbrock-Gammon et ***,our extensive investigations reveal this phenomenon to be a novel pressure-induced metal-to-metal transition intrinsic to LuH_(2),distinct from ***,nitrogen doping exerts minimal impact on this *** work not only elucidates the fundamental properties of LuH_(2)andLuH_(3),but also critically challenges the notion of superconductivity in these lutetium hydride *** findings pave the way for future research on lutetium hydride systems,while emphasizing the crucial importance of rigorous verification in claims of ambient-temperature superconductivity.

The introduction of alleles into commercial crop breeding pipelines is both time consuming and *** technologies that are disrupting traditional breeding processes are doubled haploid(DH)breeding and genome editing(GE).Recently,these techniques were combined into a GE trait delivery system called HI-Edit(Haploid Inducer-Edit).In HI-Edit,the pollen of a haploid inducer line is reprogrammed to deliver GE traits to any variety,obviating recurrent *** HI-Edit to operate at scale,an efficient transformable HI line is needed,but most maize varieties are recalcitrant to transformation,and haploid inducers are especially difficult to transform given their aberrant reproductive *** marker assisted selection and a three-tiered testing scheme,we report the development of new Iodent and Stiff Stalk maize germplasm that are transformable,have high haploid induction rates,and exhibit a robust,genetically-dominant anthocyanin native trait that may be used for rapid haploid *** show that transformation of these elite‘‘HI-Edit”lines is enhanced using the BABYBOOM and WUSCHEL morphogenetic ***,we evaluate the HI-Edit performance of one of the lines against both Stiff Stalk and non-Stiff Stalk *** strategy and results of this study should facilitate the development of commercially scalable HI-Edit systems in diverse crops.

Synthesis of Fréchet-type dendritic homotriptycenes

A rapid and efficient synthesis of novel dendrimer homotriptycenes is presented. The dendronized 9,10-dihydroanthracen-9-ols 4, having high electron densities in the benzene rings of the Fréchet-type dendrons, exhibited in the presence of acid a quantitative transannular ring closure to the corresponding dendritic homotriptycenes. The electron-donating Fréchet-type dendrons enabled the intramolecular FC alkylation by a regioselective 1,7-elimination of H2O.

Current treatment of ulcerative colitis

Ulcerative colitis (UC) is a chronic disease featuring re- current inflammation of the colonic mucosa. The goal of medical treatment is to rapidly induce a steroid-free remission while at the same time preventing complica- tions of the disease itself and its treatment. The choice of treatment depends on severity, localization and the course of the disease. For proctitis, topical therapy with 5-aminosalicylic acid (5-ASA) compounds is used. More extensive or severe disease should be treated with oral and local 5-ASA compounds and corticosteroids to induce remission. Patients who do not respond to this treatment require hospitalization. Intravenous steroids or, when refractory, calcineurin inhibitors (cyclosporine, tacrolimus), tumor necrosis factor-α antibodies (infliximab) or immunomodulators (azathioprine, 6-mercaptopurine) are then called for. Indications for emergency surgery include refractory toxic megacolon, perforation, and continuous severe colorectal bleeding. Close collaboration between gastroenterologist and surgeon is mandatory in order not to delay surgical therapy when needed. This article is intended to give a general, practice-orientated overview of the key issues in ulcerative colitis treatment. Recommendations are based on published consensus guidelines derived from national and international guidelines on the treatment of ulcerative colitis.

Pillararene-based fluorescent sensors for the tracking of organic compounds

Fluorescent chemosensors based on pillararene complexes represent a new, promising branch in sensor technology. Because of CH-π interactions, aliphatic chains are well suited for the columnar cavities of pillararenes and bulky or sheet-like(sub)structures can be arranged on the portals. Thus, pillararenes form versatile receptors and an alteration of the fluorescence behavior upon complexation ensures the function of these chemosensors as the reporter. Although this field of research exists only since a few years, remarkable chemosensors were developed for substances as diverse as medical drugs,biochemicals, herbicides and explosives.

Towards high strength cast Mg-RE based alloys:Phase diagrams and strengthening mechanisms

Mg-rare earth(RE)based systems provide several important commercial alloys and many alloy development opportunities for high strength applications,especially in aerospace and defense *** phase diagrams,microstructure,and strengthening mechanisms of these multicomponent systems are very complex and often not well understood in *** have calculated phase diagrams of important binary,ternary,and multicomponent RE-containing alloy systems,using CALPHAD(CALculation of PHAse Diagrams).Based on these phase diagrams,this paper offers a critical overview on phase equilibria and strengthening mechanisms in these alloy systems,including precipitation,long period stacking order(LPSO),and other intermetallic *** review also summarized several promising Mg-RE based cast alloys in comparison with commercial WE54 and WE43 alloys;and explored new strategies for future alloy development for high strength *** is pointed out that the combination of precipitation and LPSO phases can lead to superior strength and ductility in Mg-RE based cast *** precipitates and LPSO phases can form a complex three-dimensional network that effectively impedes dislocation motion on the basal and non-basal *** LPSO phases can also prevent the coarsening of precipitates when they interact,thus providing good thermal stability at elevated *** research is needed to determine how the combination of these two types of phases can be used in alloy design and industrial scale applications.

Crystal Growth of[Ca3Al(OH)6·12H2O]2·(SO4)3·2H2O (Ettringite) Studied Under Microgravity Conditions

On parabolic flights,the growth of ettringite,[Ca3Al(OH)6·12H2O]2·(SO4)3·2H2O,a major reaction product of cement with water which forms instantaneously,was crystallized under microgravity conditions and *** the experiments,Ca(OH)2/Al2(SO4)3 solutions were combined and reacted for 10?s,followed by immediate filtration of the suspension and subsequent quenching with *** the ettringite crystals,the size,aspect ratios,quantity and morphology were determined and the results were compared with those from identical experiments performed under terrestric *** microgravity,generally smaller crystals (l-2.9 μm) precipitated in larger amount than under normal gravity (l-3.5 μm).The aspect ratios of the crystals grown under terrestric or microgravity condition were comparable at about *** is assumed that the reason for the smaller ettringite crystals is the absence of convection leading to more initial nuclei,but slower crystal growth which is diffusion ***,no preference relative to the ion transport to the different faces of the crystals *** results contribute to the understanding of the mineralization of inorganic salts under microgravity conditions for which hitherto only a handful of examples were reported.

Excitonic Eigenstates of Disordered Semiconductor Quantum Wires:Adaptive Wavelet Computation of Eigenvalues for the Electron-Hole Schrodinger Equation

A novel adaptive approach to compute the eigenenergies and eigenfunctions of the two-particle(electron-hole)Schrodinger equation including Coulomb attraction is *** an example,we analyze the energetically lowest exciton state of a thin one-dimensional semiconductor quantum wire in the presence of disorder which arises from the non-smooth interface between the wire and surrounding *** eigenvalues of the corresponding Schrodinger equation,i.e.,the onedimensional exciton Wannier equation with disorder,correspond to the energies of excitons in the quantum *** wavefunctions,in turn,provide information on the optical properties of the *** reformulate the problem of two interacting particles that both can move in one dimension as a stationary eigenvalue problem with two spacial dimensions in an appropriate weak form whose bilinear form is arranged to be symmetric,continuous,and *** disorder of the wire is modelled by adding a potential in the Hamiltonian which is generated by normally distributed random *** numerical solution of this problem is based on adaptive *** scheme allows for a convergence proof of the resulting scheme together with complexity *** examples demonstrate the behavior of the smallest eigenvalue,the ground state energies of the exciton,together with the eigenstates depending on the strength and spatial correlation of disorder.

AIM： To study the association of three common ABCB11 and ABCC2 polymorphisms （ABCB11： 1331T〉C→V444A; ABCC2： 3563T〉A → V1188E and 4544G 〉A → C1515Y） with intrahepatic cholestasis of pregnancy （ICP） and contraceptive-induced cholestasis （CIC）. METHODS： ABCB11 and ABCC2 genotyping data were available from four CIC patients and from 42 and 33 ICP patients, respectively. Allele-frequencies of the studied polymorphisms were compared with those in healthy pregnant controls and Caucasian individuals. Furthermore, serum bile acid levels were correlated with the presence or absence of the 1331 C allele. RESULTS： The ABCB11 1331T〉C polymorphism was significantly more frequent in cholestatic patients than in pregnant controls： C allele 76.2% （CI, 58.0-94.4） vs 51.3% （CI 35.8-66.7）, respectively （P = 0.0007）; and CC allele 57.1% （CI 36.0-78.3） vs 20% （CI 7.6-32.4）, respectively （P = 0.0065）. All four CIC patients were homozygous carriers of the C allele. In contrast, none of the studied ABCC2 polymorphism was overrepresented in ICP or CIC patients. Higher serum bile acid levels were found in carriers of the 1331CC genotype compared to carriers of the TT genotype. CONCLUSION： Our data support a role for the ABCB11 1331T〉C polymorphism as a susceptibility factor for the development of estrogen-induced cholestasis, whereas no such association was found for ABCC2. Serum bile acid and 7-glutamyl transferase levels might help to distinguish ABCB4- and ABCB11-related forms of ICP and CIC.
