BACKGROUND MicroRNAs(miRNAs)regulate gene expression and play a critical role in cancer physiology.However,there is still a limited understanding of the function and regulatory mechanism of miRNAs in gastric cancer(GC).AIM To investigate the role and molecular mechanism of miRNA-145-5p(miR145-5p)in the progression of GC.METHODS Real-time polymerase chain reaction(RT-PCR)was used to detect miRNA expression in human GC tissues and cells.The ability of cancer cells to migrate and invade was assessed using wound-healing and transwell assays,respectively.Cell proliferation was measured using cell counting kit-8 and colony formation assays,and apoptosis was evaluated using flow cytometry.Expression of the epithelial-mesenchymal transition(EMT)-associated protein was determined by Western blot.Targets of miR-145-5p were predicated using bioinformatics analysis and verified using a dual-luciferase reporter system.Serpin family E member 1(SERPINE1)expression in GC tissues and cells was evaluated using RT-PCR and immunohistochemical staining.The correlation between SERPINE1 expression and overall patient survival was determined using Kaplan-meier plot analysis.The association between SERPINE1 and GC progression was also tested.A rescue experiment of SERPINE1 overexpression was conducted to verify the relationship between this protein and miR-145-5p.The mechanism by which miR-145-5p influences GC progression was further explored by assessing tumor formation in nude mice.RESULTS GC tissues and cells had reduced miR-145-5p expression and SERPINE1 was identified as a direct target of this miRNA.Overexpression of miR-145-5p was associated with decreased GC cell proliferation,invasion,migration,and EMT,and these effects were reversed by forcing SERPINE1 expression.Kaplan-meier plot analysis revealed that patients with higher SERPINE1 expression had a shorter survival rate than those with lower SERPINE1 expression.Nude mouse tumorigenesis experiments confirmed that miR-145-5p targets SERPINE1 to regulate extracellular signal-regu

Aims: Sleep disorders are prevalent among stroke survivors and impede stroke recovery, yet they are still insufficiently considered in the management of stroke patients, and the mechanisms by which they occur remain unclear.There is evidence that boosting phasic GABA signaling with zolpidem during the repair phase improves stroke recovery by enhancing neural plasticity;however, as a nonbenzodiazepine hypnotic, the effects of zolpidem on post-stroke sleep disorders remain unclear.Method: Transient ischemic stroke in male rats was induced with 30-minute middle cerebral artery occlusion.Zolpidem or vehicle was intraperitoneally delivered once daily from 2 to 7 days after the stroke, and the electroencephalogram and electromyogram were recorded simultaneously.At 24 hours after ischemia, c-Fos immunostaining was used to assess the effect of transient ischemic stroke and acute zolpidem treatment on neuronal activity.Results: In addition to the effects on reducing brain damage and mitigating behavioral deficits, repeated zolpidem treatment during the subacute phase of stroke quickly ameliorated circadian rhythm disruption, alleviated sleep fragmentation, and increased sleep depth in ischemic rats.Immunohistochemical staining showed that in contrast to robust activation in para-infarct and some remote areas by 24 h after the onset of focal ischemia, the activity of the ipsilateral suprachiasmatic nucleus, the biological rhythm center, was strongly suppressed.A single dose of zolpidem significantly upregulated c-Fos expression in the ipsilateral suprachiasmatic nucleus to levels comparable to the contralateral side.Conclusion: Stroke leads to suprachiasmatic nucleus dysfunction.Zolpidem restores suprachiasmatic nucleus activity and effectively alleviates post-stroke sleep disturbances, indicating its potential to promote stroke recovery.

Applications of deep learning for detecting ophthalmic diseases with ultrawide-field fundus

The synaptic gene Shank3 is a high confidence candidate gene for autism spectrum disorder(ASD). We previously reported a new line of Shank3 mutant rat which results in a complete loss of Shank3 by deleting exons 11-21(Δe11-21) globally. The Shank3Δe11-21 rats display obvious ASD-like behaviors and abnormal pain and thermal sensation, which mimic the symptoms of patients with Shank3 mutations. Since ASD strikes early in life, moreover it is often co-presenting with sleep problems, which can predict the severity of ASD core symptoms and may worsen the symptoms of the disease, here we further investigated the effect of shank3 deficiency on sleep architecture in juvenile rats, to better understand the biological characteristics of the Shank3Δe11-21 rats. Spontaneously locomotor activity and EEG/EMGs of the rats were recorded at postnatal day 23 and 30-31, respectively. We found that although male juvenile Shank3-/-rats displayed hypoactivity, they spent less time in NREM sleep during the dark period. The sleep profile of female juvenile Shank3-/-rats were similar to those of males, while they slept less during both the dark and light period, and the amount of REM sleep in dark phase was also significantly decreased. Further in-depth analysis of the duration of sleep-wake episodes revealed that the male juvenile Shank3-/-rats exhibited sleep fragmentation, as manifested by an increase in short wake bouts. Instead, the increase of wakefulness in the female juvenile Shank3-/-rats was mainly due to their longer duration of wakefulness episodes. EEG power spectra analysis showed that both male and female juvenile Shank3-/-rats exhibited reduced sleep intensity. Overall, our study shows that Shank3 deficiency causes sleep disturbances in rats at an early age. Together with the results of our previous behavior studies, this new rat model well presented the behavioral phenotypes associated with ASD comorbidities, which will facilitate to further promote the research for proper understanding of Shank3 related neurodevelopment

AIM:To summarize the application of deep learning in detecting ophthalmic disease with ultrawide-field fundus images and analyze the advantages,limitations,and possible solutions common to all tasks.METHODS:We searched three academic databases,including PubMed,Web of Science,and Ovid,with the date of August 2022.We matched and screened according to the target keywords and publication year and retrieved a total of 4358 research papers according to the keywords,of which 23 studies were retrieved on applying deep learning in diagnosing ophthalmic disease with ultrawide-field images.RESULTS:Deep learning in ultrawide-field images can detect various ophthalmic diseases and achieve great performance,including diabetic retinopathy,glaucoma,age-related macular degeneration,retinal vein occlusions,retinal detachment,and other peripheral retinal diseases.Compared to fundus images,the ultrawide-field fundus scanning laser ophthalmoscopy enables the capture of the ocular fundus up to 200°in a single exposure,which can observe more areas of the retina.CONCLUSION:The combination of ultrawide-field fundus images and artificial intelligence will achieve great performance in diagnosing multiple ophthalmic diseases in the future.

BACKGROUND Hypoparathyroidism with basal ganglia calcification is clinically rare.Here,we report a case of Fahr’s syndrome due to hypoparathyroidism and review the literature in terms of etiology,clinical manifestation,diagnosis,and treatment.CASE SUMMARY A 62-year-old man experienced repeated twitching of both hands in recent 10 years.On July 28,2017,the patient was admitted to our hospital due to slow response and speech difficulties.On medical examinations,he had a positive Chvostek sign,while no Albright’s hereditary osteodystrophy signs or history of neck surgery or radiation,and his family members had no similar medical history.Laboratory examinations revealed hypocalcemia,hyperphosphatemia,and low parathyroid hormone(PTH)levels.Computed tomography revealed basal ganglia calcification.Based on these investigations,a diagnosis of Fahr’s syndrome due to hypoparathyroidism was suggested.After receiving intravenous calcium gluconate to relieve symptoms,the patient continued to take oral calcium carbonate and calcitriol for treatment.CONCLUSION The possibility of hypoparathyroidism should be considered in patients with chronic hypocalcemia,recurrent tetany,and even neuropsychiatric symptoms.Hypoparathyroidism is a common cause of basal ganglia calcification.Therefore,it is recommended that blood calcium,phosphorus,and PTH levels should be measured in all individuals with basal ganglia calcification to exclude hypoparathyroidism.

Nowadays,optimization techniques are required in various engineering domains to find optimal solutions for complex problems.As a result,there is a growing tendency among scientists to enhance existing nature-inspired algorithms using various evolutionary strategies and to develop new nature-inspired optimization methods that can properly explore the feature space.The recently designed nature-inspired meta-heuristic,named the Golden Jackal Optimization(GJO),was inspired by the collaborative hunting actions of the golden jackal in nature to solve various challenging problems.However,like other approaches,the GJO has the limitations of poor exploitation ability,the ease of getting stuck in a local optimal region,and an improper balancing of exploration and exploitation.To overcome these limitations,this paper proposes an improved GJO algorithm based on multi-strategy mixing(LGJO).First,using a chaotic mapping strategy to initialize the population instead of using random parameters,this algorithm can generate initial solutions with good diversity in the search space.Second,a dynamic inertia weight based on cosine variation is proposed to make the search process more realistic and effectively balance the algorithm's global and local search capabilities.Finally,a position update strategy based on Gaussian mutation was introduced,fully utilizing the guidance role of the optimal individual to improve population diversity,effectively exploring unknown regions,and avoiding the algorithm falling into local optima.To evaluate the proposed algorithm,23 mathematical benchmark functions,CEC-2019 and CEC2021 tests are employed.The results are compared to high-quality,well-known optimization methods.The results of the proposed method are compared from different points of view,including the quality of the results,convergence behavior,and robustness.The superiority and high-quality performance of the proposed method are demonstrated by comparing the results.Furthermore,to demonstrate its applicability,it is employed to solve four c

In this paper,we propose a novel Local Macroscopic Conservative(LoMaC)low rank tensor method with discontinuous Galerkin(DG)discretization for the physical and phase spaces for simulating the Vlasov-Poisson(VP)system.The LoMaC property refers to the exact local conservation of macroscopic mass,momentum,and energy at the discrete level.The recently developed LoMaC low rank tensor algorithm(arXiv:2207.00518)simultaneously evolves the macroscopic conservation laws of mass,momentum,and energy using the kinetic flux vector splitting;then the LoMaC property is realized by projecting the low rank kinetic solution onto a subspace that shares the same macroscopic observables.This paper is a generalization of our previous work,but with DG discretization to take advantage of its compactness and flexibility in handling boundary conditions and its superior accuracy in the long term.The algorithm is developed in a similar fashion as that for a finite difference scheme,by observing that the DG method can be viewed equivalently in a nodal fashion.With the nodal DG method,assuming a tensorized computational grid,one will be able to(i)derive differentiation matrices for different nodal points based on a DG upwind discretization of transport terms,and(ii)define a weighted inner product space based on the nodal DG grid points.The algorithm can be extended to the high dimensional problems by hierarchical Tucker(HT)decomposition of solution tensors and a corresponding conservative projection algorithm.In a similar spirit,the algorithm can be extended to DG methods on nodal points of an unstructured mesh,or to other types of discretization,e.g.,the spectral method in velocity direction.Extensive numerical results are performed to showcase the efficacy of the method.

Dendrite growth is a complex liquid-solid phase transition process involving multiple physical factors.A phase-field lattice-Boltzmann method was developed to simulate the two-and three-dimension dendrite growth of Al-Cu alloy.The effect of fully coupled thermal-solute-convection interaction on the dendrite growth was investigated by incorporating a parallel-adaptive mesh refinement algorithm into the numerical model.By accurately reproducing the latent heat release,solute diffusion and convective transport behaviors at the liquidsolid interface,the interaction mechanism among thermal-solute-convection transport as well as their coupling effects on the dendrite growth dynamics were discussed.The simulation results show that the release of latent heat slows down the dendrite growth rate,and both natural and forced convection disrupt the symmetrical growth of dendrites.Their combination makes the growth of dendrites more complex,capturing important physical aspects such as recalescence,dendrite tip splitting,dendrite tilting,dendrite remelting,and solute plume in the simulation case.Based on the robustness and powerful ability of the numerical model,the formation mechanisms of these physical aspects were revealed.

The effects of hydrogen charging time and pressure on the hydrogen embrittlement(HE)susceptibility of X52 pipeline steel material are studied by slow strain rate tensile tests.The fracture morphologies of the specimens are observed by scanning electron microscopy.The HE susceptibility of the X52 pipeline steel material increases with an increase in both hydrogen charging time and hydrogen pressure.At a charging time of 96 h,the HE susceptibility index reaches 45.86%,approximately 3.6 times that at a charging time of 0 h.Similarly,a charging pressure of 4 MPa results in a HE susceptibility index of 31.61%,approximately 2.5 times higher than that at a charging pressure of 0.3 MPa.
