Objective： To investigate the effect on erythroid differentiation and proliferation of K562 cells by IER3IP1-knockdown with RNA interference targeting at IER3IP1 gene. Methods： The shRNA eukaryotic expression vectors targeting at IER3IP1 gene were designed and constructed. Inhibitory effect was detected by semiquantitative RT-PCR. The impacts on K562 cells by RNAi were studied by MTT assay, benzidine staining, light microscope and electron microscopy observation, cell cycles analysis, colony formation assay and RT-PCR. The expressions of erythroid differentiation correlated genes Gfi-lB, GPA and 7-globin were studied after being exposed to 0.2μmol/L imatinib for two days. Results： The shRNA eukaryotic expression vectors were successfully constructed. The expression of IER3IP1 gene was significantly inhibited with an inhibition efficiency of 76% （P〈0.01）. Compared with the control groups, bcr/abl mRNA level was increased in K562/shRNA-IER3IP1 group （P〈0.01）. The proliferation ability was enhanced （P〈0.01） and the proportion of cells at G0/G1 phase decreased but S phase increased （P〈0.05） in K562/shRNA-IER3IP1 group. Under electron microscopy, the amount of euchromatin increased but heterochromatin decreased. There were structural abnomalities in endocytoplasmic reticulum and clusters of vesicular. The percentage of benzidine staining positive cells and mRNA expression levels of Gfi-1B, GPA and γ-globin were all decreased after being exposed to 0.2 μmol/L STI571 for two days in K562/shRNA-IER3IP1 group （P〈0.01）. Conclusion： IER3IP1-knockdown can hinder the erythroid differentiation and elevate the proliferation level of K562 cells. IER3IP1 may play a role in erythroid differentiation and proliferation of K562 cells.

BACKGROUND Gastrointestinal(GI)discomforts are common in the elderly population;however,whether such discomforts are associated with dietary intake has not been *** To evaluate GI discomforts in Chinese urban elders and the associated *** The gastrointestinal symptom rating scale(GSRS)was used to identify GI discomforts in 688 elders from eight cities of *** semi-quantitative food frequency questionnaire and one time of 24 h dietary recall were used to access the food intake,identify dietary pattern,and calculate the nutrients *** About 83%of studied elders experienced at least one of GI discomforts in the past 6 mo;dyspepsia was the most commonly reported(66.7%),followed by reflux(44.8%),abdominal pain(35.9%),constipation(35.8%),and diarrhea(34.7%).Female gender,lower education level,and lower family income were associated with a higher GSRS *** who were diagnosed with a GI-related disease had a higher score of constipation,but a lower score of *** diseases were positively associated with certain GI *** dietary patterns were identified by the method of principal component analysis,which were characterized as high intake of salt and tea;more frequent intake of tubers,fruits,aquatic products,and soybeans;and high intake of cereal,vegetables,and meat,***,no associations between dietary patterns and GSRS score were *** elders with a higher GSRS score had significantly lower intake of bean *** elders whose GSRS score was≥21 and 18-20 decreased their bean production intake by 7.2(0.3,14.3)g/d and 14.3(1.2,27.3)g/d,respectively,compared with those who had a GSRS score≤*** were no differences in other food categories,calories,or nutrients intake among elders with different GSRS *** GI discomforts are common in Chinese urban *** discomforts might be associated with the choice of food.

Predictive validity of BODE index for anxious and depressive symptoms in patients with chronic obstructive pulmonary disease

Background Anxiety and depression are two of the commonest and most modifiable comorbidities of chronic obstructive pulmonary disease （COPD） and have an independent effect on health and prognosis. FEV1% has been shown to be a poor predictor of anxiety and depression. The body mass index, degree of airflow obstruction, dyspnea, and exercise capacity （BODE） index is a multidimensional assessment system which may predict health outcome in COPD patients. The purpose of this study was to investigate the predictive validity of the BODE index for anxious and depressive symptoms in COPD patients. Methods This was a multicenter prospective cross-sectional study in 256 patients with stable COPD. Anxious and depressive symptoms were assessed using the Hospital Anxiety and Depression Scale （HADS）. The relationships between anxiety, depression and potential predictors （including the BODE index） were analyzed by a binary Logistic regression model. Results Subjects who were anxious and depressive waJked a shorter sJx-mJnute waJkJng distance （6MWD~, had more dyspnea, a higher BODE index, and lower health-related quality of life （P 〈0.01）. Anxiety and depression score was significantly correlated with BODE index, respectively （r=0.335, P 〈0.001 ; r=0.306, P 〈0.001）. The prevalence of anxiety and depression increased with BODE stage increasing （P 〈0.05）. On the basis of binary Logistic regression, the BODE index was a good and independent predictor of anxiety and depression because it comprised dyspnea and 6MWD, which were shown to be the main determinants. Conclusions The predictive validity of the BODE index for anxiety and depression was demonstrated. We propose that the BODE index should be included in assessment of COPD severity.

Scorpion venom heat-resistant peptide(SVHRP)is a component purified from Buthus martensii Karsch scorpion venom. Our previous studies have shown that SVHRP is neuroprotective in models of Alzheimer’s disease and Parkinson’s disease. The present study aimed to explore the potential neuroprotective effects of SVHRP on cerebral ischemia/reperfusion(I/R) injury, using a mouse model of middle cerebral artery occlusion/reperfusion(MCAO/R) and a cellular model of oxygen-glucose deprivation/reoxygenation(OGD/R). Our results showed that SVHRP treatment decreased the neurological deficit scores, edema formation, infarct volume and neuronal loss in the MCAO/R mice, and protected primary neurons against OGD/R insult. SVHRP pretreatment suppressed the alterations in protein levels of N-methyl-D-aspartate receptors(NMDARs) and phosphorylated p38 MAPK as well as some proinflammatory factors in both the animal and cellular models. These results suggest that SVHRP has neuroprotective effects against cerebral I/R injury, which might be associated with inhibition of the NMDA-MAPKmediated excitotoxicity.
