Dielectric spectroscopy coupled with artificial neural network for classification and quantification of sesame oil adulteration

Adulteration using cheap vegetable oils into expensive oils such as sesame oil is a considerable challenge in the edible oil market. To discriminate pure and adulterated sesame oilwith sunflower and canola oils (commonly used as an adulterant to the high-price oils),dielectric spectroscopy was applied in the range of 40 kHz–20 MHz. The principal component analysis (PCA) plots were able to distinguish the pure sesame oil, while it was impossible to separate the adulterated oils based on the kind of adulteration. The correlationbased feature selection (CFS) method was used to select the more relevant dielectric datawithin the spectrum and to reduce the dimensionality of the input vector belongs to theartificial neural network (ANN). The ANN classifier with topology of 19-5-4 structureshowed a perfect accuracy of 100% in detecting the authentic and the adulterated sesameoil. The regression ANN with the topology of 15-5-1, 21-8-1 and 10-11-1 were the mostrobust models in quantifying the amount of adulteration in sesame oil generated by sun-flower oil, canola oil and sunflower + canola oils, with R2Test of 1, 1 and 0.999 9, *** proposed technique is a powerful and simple method to detect and quantify adulteration of sesame oil. The novelty of this research is capability of used system for authentication of adulterated sesame oil using low frequency. Furthermore, the developed systemhas a good capability for other types of sesame oil adulterations as well as to detect adulteration in other expensive edible oils.

Simplified microsatellite instability detection protocol provides equivalent sensitivity to robust detection strategies in Lynch syndrome patients

Objective:Germline mutations in mismatch repair(MMR)genes cause Lynch syndrome(LS).LS is an inherited disease,and an important consequence of MMR deficiency is microsatellite instability(MSI)*** phenotype influences the efficacy of5 fluorouracil(5-FU)***,cost effective,and easy to perform laboratory tests are required to include MSI detection in routine laboratory *** of CAT25 as monomorphic short tandem repeat sequence enables CAT25 to be an efficient screening tool among hereditary nonpolyposis colorectal cancer(HNPCC)patients compared with other methods used ***:Based on Amsterdam II criteria,31 patients in 31 families were shortlisted from a total number of 1,659 colorectal cancer *** status was examined in these patients using CAT25 and a commercially available Promega MSI five-markerbased detection system as well as immunohistochemical(IHC)staining of four important MMR *** were scored as high microsatellite instable(MSI-H),low(MSI-L),or stable(MSS).MSI status determined by CAT25 single mononucleotide marker was compared with that of five mononucleotide markers,Promega commercial kit,and IHC ***:MMR protein deficiency was observed on 7/31 probands using IHC methodology and 6/31 categorized as MSI-H using commercial kit or CAT25 single *** sensitivity and specificity of the CAT25 single marker were the same as those detected by five-marker Promega commercial kit in our ***:Based on our results,the performance of the CAT25 single mononucleotide marker for MSI status determination in our HNPCC patients is the same as that of the five-marker-based commercial kit.

Direct remyelinating effect of bone marrow mesenchymal stem cells in the rat model of spinal cord injury

Background:Several studies have tried to evaluate the effects of stem cell therapy for spinal cord injury in animal models and human *** marrow mesenchymal stem cells are one of the safe and available candidate cells with established beneficial therapeutic effects for bone and cartilage ***,there are some doubts regarding their therapeutic efficacy for nervous system *** marrow mesenchymal stem cells have been used in some clinical trials for spinal cord injury but their precise mechanism of action and also their safety has not been *** experimental reports do not provide sufficient and convincing evidence to persuade therapists to perform further clinical *** the present study we have used detailed staining methods to clearly show the entire structure of the injured spinal cord and the effects of bone marrow mesenchymal stem cells treatment on this ***:Intrathecal injection of neutral red stained bone marrow mesenchymal stem cells were done in a rat model of spinal cord injury and the fate of the transplanted cells,immunologic responses in the host tissue,and tumor generation were assessed by using brightfield and fluorescence ***-motor functions were also evaluated in the ***:Injected cells could migrate to the site of the injury and differentiate into oligodendrocytes and ***-histological assays showed no cells expressing CD2,CD3,CD15 and CD45 in host tissues,and sensory-motor assessments revealed that there were some partial ***:Present study provides strong evidence that mesenchymal stem cell therapy is safe and could be helpful for the remyelination of injured nerve fibers in the acute phase of injury.
