Long-term prognosis of focal segmental glomerulosclerosis treated with therapeutic low-density lipoprotein-apheresis in patients with severe kidney dysfunction and proteinuria

Background:The prognosis of focal segmental glomerulosclerosis patients with nephrotic syndrome is estimated to be 10%-20%in 5 years and 30%-50%in 10 years,leading to end-stage kidney *** response rate with steroid therapy is 40%-60%.Therapeutic low-density lipoprotein-apheresis(LDL-A)may be effective in patients with steroid *** regarding the long-term prognosis of patients with focal segmental glomerulosclerosis receiving this therapy is ***:We investigated the effectiveness of treatment in 50 patients with primary focal segmental glomerulosclerosis diagnosed between 1961 and 2017 at Kanazawa University Hospital and related *** patients were observed at least 12 months after biopsy or until end-stage kidney disease occurrence or ***:LDL-A was performed in four patients who presented with steroidresistant nephrotic syndrome(two patients had concurrent acute renal failure for which hemodialysis was performed).In comparison with 17 patients who did not receive LDL-A after 1989,the LDL-A group had higher urinary protein excretion(13.7 vs.5.2 g/day,P=0.053)and serum creatinine(4.11 vs.1.65 mg/dL)levels at onset,and a numerically higher remission rate(75.0%vs.58.7%)compared with the nonlipoprotein-apheresis ***:Therapeutic LDL-A can be performed for critical cases and may improve the remission rate.

Dynamic methylation driven by neuronal activity in hippocampal neurons impacts complex behavior

Epigenetic processes are well-known to play critical roles in learning and memory. Among epigenetic processes, accumulating data suggests that DNA methylation in particular is a critical determinant of learning and memory. In vitro data have suggested that DNA methyltransferase inhibitors can trigger DNA demethylation and subsequent gene expression of the brain-derived neurotrophic factor gene in an activity dependent manner. To examine if these processes occur in vivo, we chronically infused DNMT inhibitors into the hippocampus and examined the impact on behavior. We find that chronic DNMT inhibition in the hippocampus results in increased anxiety-related behavior and deficits in context-dependent fear conditioning accompanied by an increase in BDNF expression. Gene expression changes were blocked by pretreatment with the NMDA receptor antagonist AP5, suggesting that DNMT inhibition enhances gene expression in an activity-dependent manner and that, conversely, the behavior deficits and abnormal gene expression are facilitated by NMDA receptor activity.

Porcine Placenta Extract Enhances Ceramide Synthase 3 Expression through the PPARδ/ILK/Akt/mTOR/STAT3 Pathway

Porcine placenta extract (PPE) is widely accepted as an ingredient in complementary and alternative medicine and previous studies have reported its availability, however, its underlying action mechanism remains unclear. In this study, we investigated the underlying mechanism of PPE-induced ceramide synthase 3 upregulation. PPE enhanced the expression of ceramide synthase 3 at both the mRNA and protein levels in HaCaT cells. Moreover, PPE-induced ceramide synthase3 upregulation was suppressed by the Akt inhibitor, suggesting the involvement of Akt in the underlying mechanism. As the PI3K inhibitor did not affect PPE-induced ceramide synthase 3 upregulation, the factors upstream of Akt were estimated. Inhibition and small interfering RNA experiments demonstrated that the peroxisome proliferator-activated receptors δ (PPARδ) and integrin-linked kinase (ILK) are involved in the phosphorylation of Akt. Next, we explored the factors downstream of Akt and found that PPE induced phosphorylation of the STAT3 while PPE-induced upregulation of ceramide synthase 3 was significantly suppressed by the inhibitor of the mTOR, suggesting that the mTOR/STAT3 pathway is involved in the downstream of Akt. These results demonstrate that PPE upregulates CerS3 expression via the PPARδ/ILK/Akt/mTOR/STAT3 pathway.

Relevance of Visual, Auditory, and Olfactory Cues in Pet Dogs’ Awareness of Humans

Many studies have looked at how dogs respond to human communicative information. Here, we examined which human communicative factors were important in influencing dogs’ responses. Eleven healthy pet dogs with no apparent aggressive behaviour toward people were recruited. Five sensory conditions (all cues presented;either a visual, an auditory, or an olfactory cue presented;no cues presented) were provided three times randomly to each dog during the tests. All tests were video recorded, and both the dogs’ behaviour and time taken to reach the person when she presented each of the sensory cue conditions were observed. Total rates of reaching the person were as follows: 97.0% (all cues), 87.9% (auditory cues), 84.4% (visual cues), 84.4% (olfactory cues), and 69.7% (no cues). The time taken for the dog to notice the person in the box and then obtain a reward from her differed among the five conditions: all cues (6.00 ± 0.32 s) and visual cues (6.02 ± 0.91 s) were significantly faster than auditory cues (18.56 ± 9.57 s) and no cues (26.55 ± 11.72 s). Thus the type of information input was important in recognition of the person by the dogs and influenced the dogs’ response times;visual cues appeared advantageous in confirming the person’s presence.

Contribution of Ground-Based Cloud Observation to Satellite-Based Cloud Discrimination

One of the biggest factors to deteriorate the satellite product quality is cloud coverage. Therefore, cloud masking process is important to improve the quality of various satellite products. However, satellite-based cloud discrimination algorithm has been developing and efficient ground-based cloud observations are necessary to validate satellite-based cloud discrimination. The purpose of this study is to develop the efficient ground-based cloud observation methodology using whole sky camera. This paper deals with methods how to discriminate cloud portions on whole sky image, how to apply the ground-based cloud observation to the validations for satellite products. For the cloud discrimination on whole sky image, we propose SI (sky index) and BI (brightness index) calculated from RGB (red, green and blue) channels. SI shows the extent of the blueness and gray scale and BI indicates the extent of the brightness. Sun, cloud and blue sky portions are divided by SI and BI threshold. As an application of ground-based cloud observation for the validation of satellite products, clouds portions discriminated from whole sky image are projected onto ground surface with map coordinate. We also examine to compare with cloud portions on whole sky images and MODIS (MODerate resolution Imaging Spectroradiometer) image as one of experiments. The proposed ground-based cloud observation method and its extension to satellite-based cloud discrimination should be connected to improve the quality of satellite products.

Novel Treatment of Hepatitis C Virus Infection for Patients with Renal Impairment

Prevalence of hepatitis C virus (HCV) infection is high in patients with end-stage renal dysfunction,including patients undergoing hemodialysis (HD).The HCV infection itself can cause glomerulonephritis and puts individuals at increased risk of developing end-stage renal disease;fortunately,successful HCV eradication sometimes restore HCV-related renal ***,the prognosis of dialysis patients infected with HCV is significantly worse and the renal allograft survival in HCV-infected patients is also worse than in dialysis patients without HCV *** life prognosis is favorable,therefore,anti-HCV therapy is strongly recommended for HCV-infected patients with severe renal *** standard therapy for HCV-infected patients with severe renal dysfunction has historically been interferon-based ***,this therapy remains ineffective in achieving high,sustained viral response rates and the rate of adverse events and treatment discontinuation due to treatment-induced adverse events continues to be high in patients with severe renal *** and effective anti-HCV therapies are urgently needed,and crucial,for patients with severe renal ***,direct-acting antivirals (DAAs) that specifically target viral proteins have been developed,and these targets include the NS3,NS5A,and NS5B of *** trials have revealed high efficacy and safety of the DAA-based therapies,but patients with severe renal dysfunction were not included in the majority of these ***,several recent reports have shown high efficacy and safety for some regimens of DAA combination therapy for HCV-infected patients with severe renal *** this review,we discuss novel treatments for HCV-infected patients with severe renal dysfunction and the pharmacokinetics of these drugs.

Determination of Lanthanoids in Seawater by Inductively Coupled Plasma Mass Spectrometry after Pre-Concentration with a Chelating Resin Disk

A seawater sample was analyzed for its lanthanoids (Ln’s) contents by inductively coupled plasma mass spectrometry (ICP-MS). The analytical procedure included the pre-concentration and partial purification of Ln’s with a chelating resin disk and the employment of the combination of the internal standard and standard addition methods in ICP-MS, which seemed indispensable for the determination of low-level Ln’s by ICP-MS. The recoveries of the Ln’s with the disk were mostly 70% or better with the concentration factor of 200. The measured concentrations of Ln’s were on the order of ng?dm–3 or lower and reasonable except that of neodymium and samarium.
