Reference ranges and age-related changes of peripheral blood lymphocyte subsets in Chinese healthy adults

This study was performed to build region-specific reference ranges of peripheral blood lymphocyte subsets for Chinese healthy adults from the young to the elderly and analyze the trends of changes in lymphocyte subsets for evaluating the impact of age on the values.151 healthy adults aged 19-86 were recruited based on the SENIEUR protocol.Three sets of reference ranges were finally built applicable for the healthy young(19-44 years),middle-aged(45-64 years) and elder adults(≥65).Comparisons in parameters among the three cohorts showed that a statistically significant increase in CD16CD56+ NK cell was observed between the middle-aged and elder cohorts,whereas for the majority of the parameters,a significant decline was observed between the young and the middle-aged cohorts.Further results showed that inverse correlations were observed between the age and CD19+ B,CD3+ T,CD3+CD4+ T,CD4+CD45RA+CD62L+ nave T cell and CD4+CD28+/CD4+,while the positive one was identified between the age and the NK cell.These significant changes of the most of immune parameters provided evidence for immunosenescence.Notably,T cell activation markers of CD8+CD38+ and CD8+HLA-DR+ showed reverse trends of association with age,which provides a clue for further researches on the mechanisms underlying the paradoxical clinical presentation of the elder patients.

Translational medicine is a new discipline which aims to eliminate the barrier between preclinical and clinical medicine.Here,Dr.Li discusses the application of translational medicine in the research,teaching and clinical work of Prof.Liu Shih-Hao,the founder of endocrinology in China,who was particularly renowned for his early work in calcium and phosphorus metabolism.This well-known success story can be traced back to an early appreciation of translational medicine by Prof.Liu Shih-Hao,and serves as an important and revelatory lesson for us all.

Introduction Primary liver cancer is a significant health issue in China,where it ranks fourth in morbidity and second in mortality among all malignant tumors.[1-4]The 5-year overall survival rate of Chinese patients with liver cancer from 2012 to 2015 was only 12.2%for males and 13.1%for females.[5]Hepatocellular carcinoma(HCC)accounts for 75-85%of primary liver cancers.[6]Furthermore,most liver cancers are unresectable at diagnosis.To combat this,conversion therapy is used,which involves systematic treatments like anti-angiogenic drugs or molecular targeted therapy combined with immunotherapies,as well as locoregional treatments such as transcatheter arterial chemoembolization(TACE),hepatic artery infusion chemotherapy(HAIC),ablation therapy,and radiation therapy.The goal is to eliminate unresectable status and increase the chances of surgery.The reported 5-year survival rate after salvage surgery following tumor downstaging varied from 24.9-57%.

AIM: To determine the mechanisms of effects of phytoestrogen genistein on the contracted rabbit aortic arteries in vitro, and observe the effect of genistein and 17-β estradiol on mean arterial pressure (MAP) in ovariectomized (OVX) rats. METHODS: (1) Strips of rabbit aortic smooth muscle were suspended in organ baths containing Kreb’s solution, and then isometric tension was measured. (2) Female mature Wistar rats underwent a bilateral ovariectomy (OVX). Sham-operated rats (SHAM) were used as controls. After administration of genistein (0.4 mg·kg-1·d-1, sc), 17-β estradiol (1 mg·kg-1·d-1, sc) or theirvehicle sesame oil for 21 d,MAP was measured. RESULTS: (1) Similar to 17-β estradiol, genistein could dose-dependently relax 40 mmol/L KCl-precontracted arterial strips. Incubation with Nω-L-nitro-arginine (L-NNA), methylene blue (MB), indomethacin, propranolol or endothelium removal did not affect relaxation induced by genistein. In calcium-free solution containing 0.01mmol/L egtazic acid (EGTA), genistein inhibited not only the first phase contraction induced by noradrenaline (NA), but also the second contraction induced by CaCl2. In addition, genistein could reduce the contractile responses of NA, KCl and CaCl2, and shift their cumulative concentration-response curves rightward. (2) MAP in OVX rats was significantlyhigher compared with that of SHAM rats. However,after chronically treatment with genistein or 17-β estradiol for 21 d the baseline MAP in OVX rats was reduced significantly. CONCLUSIONS: (1) The vasodilator effect of genistein in vitro is endothelium independent and not related to the nitric oxide, its mechanisms being probably due to inhibition of Ca2+ influx through calcium channels in a noncompetitive manner and Ca2+ release from intracellular store induced by NA. (2) Administration of genistein or 17-β estradiol can chronically decrease MAP in OVX rats.

Sodium dimercaptopropane sulfonate is used in place of dimercaprol in the treatment of poisoning by heaving metals and metalloids as a well-known specific antidote. It is claimed to be less toxic. In early stage, the pharmacologists and clinical doctors in Wenzhou medical college found this drug as antidote against non-metallic pesticides as nereistoxin insecticides, Chlordimeform, bactercide 402, and tetramine. We did a lot of animal experiments and clinical trials to confirm that this antagonist possesses a strong antidotal effect. Now it is widely used in China. Great break through has been made not only in the theory but also in its practice. Furthermore, the combination use of this antidote and central anticonvulsant as diazepam indicated synergistic antidotal effect, which is probably the best utilization in the treatment of acute poisoning by tetramine.

G protein-coupled receptor kinases(GRKs)mediate agonist-induced phosphorylation anddesensitization of various G protein-coupled receptors(GPCRs).We investigate the role of GRK2 on epidermalgrowth factor(EGF)receptor signaling,including EGF-induced extracellular signal-regulated kinase andmitogen-activated protein kinase(ERK/MAPK)activation and EGFR internalization.Immunoprecipitationand immunofluorescence experiments show that EGF stimulates GRK2 binding to EGFR complex and GRK2translocating from cytoplasm to the plasma membrane in human embryonic kidney 293 cells.Westernblotting assay shows that EGF-induced ERK/MAPK phosphorylation increases 1.9-fold,1.1-fold and 1.5-fold(P<0.05)at time point 30,60 and 120 min,respectively when the cells were transfected with GRK2,suggesting the regulatory role of GRK2 on EGF-induced ERK/MAPK activation.Flow cytometry experimentsshow that GRK2 overexpression has no effect on EGF-induced EGFR internalization,however,it increasesagonist-induced G protein-coupled δ opioid receptor internalization by approximately 40% (P<0.01).Overall,these data suggest that GRK2 has a regulatory role in EGF-induced ERK/MAPK activation,and that themechanisms underlying the modulatory role of GRK2 in EGFR and GPCR signaling pathways are somewhatdifferent at least in receptor internalization.

Monoclonal antibodies(mAb)have been successfullyapplied to the treatment of chronic diseases,such as cancer,inflammation and immune diseases.With the technicaladvances in antibody engineering,development of smallrecombinant antibody fragments as high affinity therapeu-tics with reduced immunogenicity has become under thespotlight.A popular format of the engineered recombinantantibody fragments is the single-chain fixed-variable(scFv)molecules,in which the VH and VL regions of the parentalantibody are jointed together by a polypeptide linker.ThescFv fragment retains the target specificity and antigen-binding affinity of the intact antibody,and can be geneti-cally designed and produced in large quantity by ectopicallyexpressing both VH and VL regions from a single cDNAin cells.Due to its smaller size,the scFv molecule showsimproved pharmacokinetics in tumor penetration and isbetter tolerated by the host immune system.Despite thesepotential advantages of using scFv molecules for immu-notherapy,especially for chronic diseases such as cancers,the treatment efficacy,however,is often compromised bythe rapid blood clearance and insufficient concentration ofthe infused scFv fragments at the target site.To achievehigh local concentrations in the target tissues,large amountof the recombinant scFv proteins(hundreds milligrams or
