In Vitro Antioxidant and Radio Protective Activities of Lycopene from Tomato Extract against Radiation—Induced DNA Aberration

Background: The accumulation of free radicals is linked to a number of diseases. Free radicals can be scavenged by antioxidants and reduce their harmful effects. It is therefore essential to look for naturally occurring antioxidants that come from plants, as synthetic antioxidants are toxic, carcinogenic and problematic for the environment. Lycopene is one of the carotenoids, a pigment that dissolves in fat and has antioxidant properties. Materials and Methods: The antioxidant and free radical scavenging activity were assessed using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. The impact of lycopene on bacteria (E. coli) susceptibility to γ-radiation was examined by radio sensitivity assay. The study also examined the induction of strand breaks in plasmid pUC19 DNA and how lycopene extract protected the DNA from γ-radiation in vitro. Results: At varying concentrations, lycopene demonstrated its ability to scavenge free radicals such as 2, 2-diphenyl-1-picrylhydrazyl (DPPH). IC50 for lycopene was determined at 112 μg/mL which was almost partial to IC50 of standard antioxidant L-ascorbic acid. The D10 value 180 Gy of E. coli was found to be >2-fold higher in the extract-containing lycopene sample than in the extract-free controls. The lycopene extracts inhibited the radiation-induced deterioration of the plasmid pUC19 DNA. At an IC50 concentration, lycopene provided the highest level of protection. Conclusion: Lycopene functions as an efficient free radical scavenger and possible natural antioxidant source. For cancer patients and others who frequently expose themselves to radiation, lycopene may be a useful plant-based pharmaceutical product for treating a variety of diseases caused by free radicals.

Seasonal changes in Porteresia coarctata (Tateoka) beds along a subtropical coast

Shoot density, standing crop （above- and below-ground biomass） and habitat of salt marsh grass Porteresia coarctata were investigated along the coast of Bakkhali estuary, Cox＇s Bazar, Bangladesh from January to December 2006. Shoot density of P. coarctata was influenced by season and was found to be higher （〉2 500 shoots/m2） in post-monsoon and minimal in monsoon season; plants were particularly active in vegetative propagation during pre-monsoon. Above-ground biomass was greater along the protected coast compared with the exposed one in this estuary. Below-ground biomass was higher （7.75-269.53 g DW/m2） than that above ground （2.20-114.75 g DW/m2）. Standing crops ofP. coarctata showed a negative relationship （R=-0.77; P〈0.05） with sedimentation rate, while seasonal activity influenced sedimentation. The recorded sedimentation rate was lower （6.09 mg/（cm2·d）） in pre-monsoon and highest （14.55 mg/（cm2·d）） in monsoon season. The mean value of pore water salinity was higher （34.25±65.05） during post-monsoon and lowest （18.0±3.71） in monsoon season. The soil was sandy clay in this P. coarctata bed; it consisted of 86% sand, 13% clay and 1% silt. Soil organic matter dropped during the monsoon season （0.78%-0.67%） and was highest （（2.17±1.42）%-（2.3±1.47）%） during post-monsoon, probably owing to accumulation of decomposed peat on the marsh surface. The mean pore water NH4-N concentration ranged from 2.444-1.65 to 3.33±1.82 μg/L, with a minimum air temperature of 22.09℃ in post-monsoon and a maximum of 31.16℃ in pre-monsoon. Variations of physico-chemical parameters in the soil, water, and climate governed biological parameters of P. coarctata in the Bakkhali estuary, and were comparable with estuarine environments elsewhere.

Annual pattern of zooplankton communities and their environmental response in a subtropical maritime channel system in the northern Bay of Bengal, Bangladesh

Zooplankton plays an important role in aquatic food webs by fluxing of energy from primary producer to subsequent trophic levels in the food chain. The annual pattern of zooplankton communities and potential environmental drivers were studied in the Kohelia channel, Bangladesh from summer 2014 to spring *** were collected using net at a depth of 1 m. A total of 32 species belonged to 18 orders, 27 families and 15 taxonomic groups were identified. Of these species, 22 distributed in all four seasons of which 8 were dominant and highly contributing to the total communities. Species number peaked in summer next to winter and fall in spring while maximum abundance was in summer and minimum in spring. Multivariate analyses showed that there was a clear annual pattern in the zooplankton communities. Species diversity and evenness peaked in spring but fall in autumn while the high value of species richness was found in winter. Biological-environmental best matching（BIO-ENV） analyses conformed that community pattern of zooplankton was mainly driven by transparency salinity, and temperature individually or combined with water nutrients. These results demonstrate that annual pattern of the zooplankton community shaped by channel environmental factors in subtropical channel ecosystems, thus might be used for community-based subtropical coastal water bioassessment.

Decaying mangrove detritus plays a significant role in nutrient cycling and fueling in both the forests and aquatic habitats where the detritus stems from estuarine and coastal food *** decay rates partly depend on the type of detritus,but most studies have thus far focused on leaf litter decay and decomposition,whereas other detrital sources(except for roots)have largely been *** compare the decay rates of Rhizophora apiculata and Xylocarpus granatum non-leafy detritus(flowers,propagules,stipules,and twigs)with mangrove leaf litter in field studies,using litter bags during the dry and the wet seasons in the tropical mangrove forest of Sibuti,Sarawak,*** observed higher microbial decay rates of stipules,flowers,propagules,and twigs for both the *** and *** the wet *** contrast,leaf decay rates were higher during the dry *** rates depended on the lignin content of the detrital sources of both species,both during dry and wet ***,the half-life(T0.5)and 95%lifespan(T0.95)of non-leaf materials(flowers,propagules,stipules and twigs)for both species were remarkably longer than those of leaf *** decaying non-leafy detritus may play a significant role in nutrient and carbon cycling over longer time,when leaf litter is either being decomposing rapidly and/or being washed away by tidal flush and river ***,non-leafy(flowers,propagules,stipules and twigs)detritus should be taken into account when budgeting organic matter turnover in mangroves.

In vitro callus induction and plantlet regeneration of Achyranthes aspera L.,a high value medicinal plant

Objective:To study callus induction from different explants(internode,leaf,root)and in vitro plantlets propagation from medicinally important plant Achyranthes aspera ***:Sterilized explants were prepared by uning 0.1%HgCl_2 and 0.5%Bavistin and callus was obtained when cultured onto Murashige Skoog's(MS)medium by using different concentrations and combination of 2,***,IAA,IBA with 3%sucrose and 0.8%*** callus was immediately transferred to MS medium containing at different concentrations of phytohormones for shootlets and rootlets induction ***:Sterilization treatment of 0.1%HgCl_*** 2-3 min and Bavistin 0.5%for 10-12 min showed the highest percentage of asepsis and survival *** induction of callus was obtained from a combination of 2.0 mg/L 2,4-D and 0.5 mg/L NAA from *** shootlets number(4.83±0.l7)and length(3.8±0.16)cm were observed on full strength MS medium when fortified with BAP 4.0 mg/L and KIN 0.5 mg/*** efforts of BAP 10 mg/L and NAA 0.5 mg/L on full strength MS medium showed highest leaf number(6.77±0.94).In vitro raised shoots were allowed to root on different strengths of MS medium fortified with IAA and IBA at different ***,3.0 mg/L IBA was enabled to induce maximum rootlets number(10.0±9.82)on full strength MS ***,regenerated shoots with well developed roots were successfully subjected to hardening process and were *** survived plantlets showed 66.67%survival frequency without any morphological ***:The results demonstrated that different explants were good source of callus induction,morphology analysis as well as indirect plantlets regeneration.

Climate change impacts are visible in Bangladesh which requires adoption of judicious crop management practices for sustainable agriculture. Crop simulation models are effective in this regard and can be used for optimizing water and nitrogen inputs for maximization of wheat yields. Two field studies were conducted at Bangladesh Agricultural Research Institute (BARI) for evaluating the effect of increased temperature on wheat production under variable water and N rates, through the use of DSSAT v4.6 model. Thirty years historic weather data and BARI Gom-26 wheat-cultivar coefficients, as generated through use of GLUE of DSSAT model, were used for evaluating the impact of temperature rise with variable rates of irrigation water and nitrogen. The effects of four levels of nitrogen (0, 40, 80 and 120 kg ha-1) and four irrigation levels [0 (no irrigation), 1, 2 and 3] on wheat yield were evaluated under 0°C, 1°C, 2°C and 3°C rise in temperature. Simulation results indicated that wheat yield increased with increase in nitrogen application rate, but decreased with the temperature rise. Crop growing duration was reduced by about five days for each degree rise in temperature, irrespective of levels of irrigation water and nitrogen-rates. In general, there was yield reduction of 8.13, 16.77 and 24.97 % with increased temperature of 1°C, 2°C and 3°C, respectively, when compared with no temperature rise treatment. For interaction of temperature rise-N dose, the magnitude of yield decline under temperature rise was higher with increased dose of N. Agronomic N use efficiency at different N rates and temperature rise level were calculated. N use efficiency decreases with the increase in temperature rise and the highest (29.95 kg grain kg-1 N) was obtained from 40 kg N ha-1 treated plot with 0°C temperature rise i.e. existing atmospheric temperature. Functioning Hydraulic and Morphological Sustainability of River Training Works in Bangladesh

River dynamics play the most vital role in the socio-economic of the country. These rivers are highly dynamic in nature which causes extensive riverbank erosion. Active river bank erosion and bed scouring, is apprehended to change the scenario, leading to loss of limited valuable land of Bangladesh. River training is being practiced in Bangladesh since 1960s but the process is very expensive especially in the Ganges and the Jamuna Rivers. Five types of river training works were selected at different reaches. Hydro-morphological characteristics of the major rivers are being evaluated not only in engineering concepts but also in socio-economic and environmental aspects. PRA （participatory rural appraisal） tools were applied to evaluate protection safety, hydraulic and hydrological connectivity, ecological and environmental characteristics, causes of failure and social acceptability. This comparative study is being carried out for closing the gap between the modem river training works and in country experience through adaptation of research findings with a view to effectively arresting river bank erosion. Sirajganj Hardpoint, Chandpur Town Protection work and Nakalia Revetment are found to be suitable and effective. A technically suitable structure may not have good social acceptance without IWRM （integrated water resource management）.

Management of Sepsis in Intensive Care Units in Rajshahi Medical College, Bangladesh

Background: Sepsis was a life-threatening situation affecting roughly 27 million individuals globally every year. Devoid of proper management, sepsis could develop to austere sepsis and septic shock that reason certain eight million expiries every year. Initial identification and analysis of contaminations and quick introductions of antimicrobial management had long been predictable as essential principles of treatment. Methods: All patients were evaluated thoroughly by history, clinical and laboratory findings. Serum lactate levels were collected at 0 hr and 24 hrs of the total admission in the ICU. Blood culture and sensitivity was performed prior to antibiotic administration in this study. Results: In this research, total sample was 1367 patients. Among these sepsis patients were 305 (nearly 22%). These 305 studied sepsis cases, majority 128 (42%) of the patients were within the age group of 41 - 65 years, followed by 18 - 30 years consisting of 116 (38%) and the least number of sepsis patients were in the age group of >65 years (20%). This study found that 129 (42.3%) out of 305 blood samples screened from assumed sepsis infection were affirmative for the presence of sepsis causing pathogens. Among the 129 culture positive samples, majority of the isolated micro-organisms were gram negative bacteria 58% (75), followed by gram positive bacteria 38% (49), and 3.8% (5) were the fungi Candida albicans. Conclusion: At present, mortality & morbidity of sepsis subject was too big due to late identification, wrong & inadequate management of sepsis in the ward and also in the intensive care unit of Bangladesh.

Amelioration of Saline Soil by the Application of Gypsum, Calcium Chloride, Rice Husk and Cow Dung

A pot experiment was conducted to investigate the effect of cow dung, rice husks, calcium chloride and gypsum on soil reclamation and compare the effect of organic and inorganic amendments on soil reclamation during the period of 5th March to 20th April, 2017. The experiment was laid to fit a completely randomized design (CRD) with seven treatments [Reference soil (T0), Cow dung (T1), Rice husk (T2), Gypsum (T3), Calcium chloride (T4), Cow dung + Rice husk (T5) and Gypsum + Calcium chloride (T6)] each having three replications for this experiment. After incubation (45 days), the laboratory investigation was carried out in the Soil, Water and Environment Discipline, Khulna University, Khulna, Bangladesh. Results indicate that the individual or combined effect of gypsum (T3) was more effective in changing EC and SAR. Gypsum application in combination with calcium chloride (T6) improved the soil chemical properties by reducing the EC. Among the treatment, calcium chloride (T4) had a remarkable effect in reducing sodium adsorption ratio and gypsum had a remarkable effect in reducing pH. Cow dung (T1), rice husk (T2), combination of cow dung and rice husk (T5) were less effective to reduce EC, pH and SAR. It’s measured for soils of different soil amendments varied significantly
