Simvastatin Potentiates the Antihyperglycemic, Antidyslipidimic and Antioxidative Effect of Glibenclamide on Alloxan-Induced Diabetic Rats

The aim of the current study is to investigate the effect of combination of glibenclamide;an antidiabetic drug and simvastatin;a HMG-CoA reductase inhibitor on long-term (four weeks) alloxan-induced diabetes rats (ADRs). Methods: Alloxan (120 mg/kg body weight, BW) was injected intra-peritonially (i.p.) in rats. At first alloxan (120 mg/kg BW) induced diabetic rats were treated with single dose of glibenclamide (1.2 mg/70kg BW) and simvastatin (10 mg/70kg BW) for two weeks. Then fixed dose combinations of glibenclamide (0.6 mg/70kg BW) and simvastatin (5 mg/70kg BW) were injected along with those of two drugs for four weeks. Results: At first it was found that glibenclamide reduced significant amount of glucose in blood, but it had no significant effect on lipid profile on short term (two weeks) ADRs. In contrast, simvastatin had no effect on blood glucose level, whereas it significantly reduced total cholesterol (TC), triglycerides (TG) and low density lipoprotein cholesterol (LDL-C) and increased significant amount of high density lipoprotein cholesterol (HDL-C). However, pathological changes of pancreas’s Islets of Langerhans were observed only after long-term (four weeks) induction of alloxan in rats. The inhibitory effect of combination therapy on blood glucose, TC, TG and LDL-C level was higher than those of monotherapy alone on long term ADRs. In addition, treatment with combination therapy on long term ADRs showed higher amount of HDL-C level and super oxide dismutase and catalase enzyme activity than those with monotherapy. They also decreased serum glutamic pyruvic transaminase (SGPT) and Serum glutamic oxaloacetic transaminase (SGOT) level. Administration of simvastatin recovered Langerhans cells from shrinkage whereas glibenclamide displayed slight recovery. In recent years, the popularity of medicinal plants as a remedy has been increased manifold due to having minimal adverse effects. The current study aimed to compare the hypoglycemic, hypolipidemic and hepatoprotective effects of the ethanolic extract of Asparagus racemosus (EEAR) Linn. alone and combinedly with conventional antidiabetic agents (gliclazide and pioglitazone) in alloxan-induced diabetic rats. Diabetes was induced in male Wister albino rats by the administration of single intra-peritoneal injection of alloxan monohydrate (120 mg/kg b.w.). Effect of oral administration of two different doses of EEAR (200 and 400 mg/kg b.w.), gliclazide (10 mg/kg b.w.) and pioglitazone (10 mg/70kg/b.w.) alone for 2 weeks and a combination of EEAR (200 mg/kg b.w.) with either gliclazide (10 mg/kg b.w.) or pioglitazone (10 mg/70kg/b.w.) for 2 weeks were examined on hypoglycemic activity on 0th, 5th, 10th and 14th day of treatment. After 2 weeks of treatment, hypolipidemic and hepatoprotective effects were estimated by serum biochemical markers such as total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), very low density lipoprotein (VLDL), high density lipoprotein (HDL), serum glutamate oxaloacetate transaminases (SGOT), serum glutamate pyruvate trans-aminases (SGPT) and total protein (TP) with the help of commercially available kits. The survival rate, body weight and organ weight were also measured. Alloxan treatment resulted in persistent hyperglycemia, hyperlipidemia and liver dysfunction in rats. Treatment with EEAR at different doses improved hyperglycemia significantly (p th and 14th day of treatment in a dose-dependent mood when compared to the disease control rats, gliclazide treated rats and pioglitazone treated rats. Waste Sneaker Conversion into Liquid Hydrocarbon Fuel

Sneaker is a composition of Thermoplastic Polyurethane (TPU), suede and fabrics. Among three materials, thermoplastic polyurethane is the major component that makes up the sneakers soles and body. It gives total shape of bottom portion of sneakers. Thermoplastics polyurethane is an elastomer that resembles rubber in consistency and feel. TPU has outstanding abrasion resistance, great low temperature flexibility, resistance to oil and a high threshold of supporting weight. It is also very bondable, durable, paintable, and impact resistant and demonstrates good wear resistance. However after use, TPU (sneakers) become a problem to recycle. The objective of the experiment is to convert sneaker materials into liquid hydrocarbon fuel by thermal cracking process. Hydrocarbon groups present in the TPU can be converted using this thermal process. Wastes of the group comprising synthetic plastics and rubber are thermally disposed of by cracking at temperature ranging from 250 degree to 450 degree C. The process of waste sneaker to liquid fuel is to be discussed.

BackgroundThe COVID-19 pandemic posed a danger to global public health because of the unprecedented physical,mental,social,and environmental impact affecting quality of life(QoL).The study aimed to find the changes in QoL among COVID-19 recovered individuals and explore the determinants of change more than 1 year after recovery in low-resource ***-19 patients from all eight divisions of Bangladesh who were confirmed positive by reverse transcription-polymerase chain reaction from June 2020 to November 2020 and who subsequently recovered were followed up twice,once immediately after recovery and again 1 year after the first *** follow-up study was conducted from November 2021 to January 2022 among 2438 individuals using the World Health Organization Quality of Life Brief Version(WHOQOL-BREF).After excluding 48 deaths,95 were rejected to participate,618 were inaccessible,and there were 45 cases of incomplete *** statistics,paired-sample analyses,generalized estimating equation(GEE)analysis,and multivariable logistic regression analyses were performed to test the mean difference in participants’QoL scores between the two *** participants(n=1710,70.1%)were male,and one-fourth(24.4%)were older than *** average physical domain score decreased significantly from baseline to follow-up,and the average scores in psychological,social,and environmental domains increased significantly at follow-up(P<0.05).By the GEE equation approach,after adjusting for other factors,we found that older age groups(P<0.001),being female(P<0.001),having hospital admission during COVID-19 illness(P<0.001),and having three or more chronic diseases(P<0.001),were significantly associated with lower physical and psychological QoL *** age and female sex[adjusted odd ratio(aOR)=1.3,95%confidence interval(CI)1.0–1.6]were associated with reduced social domain scores on multivariable logistic regression *** or semi-urban peo

Bioguided Fractionation and Purification of Natural Bioactives Obtained from<i>Alpinia conchigera</i>Water Extract with Melanin Inhibition Activity

Crude extracts of Alpinia conchigera a species from the Malaysian Ginger (Zingiberaceae) family and its fractions obtained from various extraction methods were assayed for melanogenesis inhibition activity and cell viability. The crude extract obtained from the ethanolic extraction and the super critical fluid extraction did not exhibit significant melanin inhibition activity and was shown to be toxic to the melanocyte cells in comparison to the water extract. The crude aqueous extracts displayed melanin inhibition of 96.38 ± 1.60% and cell viability 109.90 ± 8.32% at a concentration of 500 μg/mL. Bioassay guided fractionation was performed on the water extracts to isolate the active compounds. The actives were identified as trans-cinnamaldehyde and chavicol glucopyranoside with both compound showing potent anti-melanogenesis activity. At 4.9 μg/ml, both trans-cinnamaldehyde and chavicol glucopyranoside gave 85% inhibition of melanin formation in vitro with 77% and 97% cell viability respectively. In comparison, kojic acid, a known skin lightening agent showed 90.0% inhibition at 100 μg/mL. The bioactive composition comprising the extract, active fraction, purified compounds or mixture thereof of Alpinia conchigera may be used for cosmetic and pharmaceutical applications, particularly for the purposes of reducing skin pigmentation.

Objective:Pancreatic ductal adenocarcinoma(PDAC)is a deadly malignancy,due in large part to its resistance to conventional therapies,including radiotherapy(RT).Despite RT exerting a modest antitumor response,it has also been shown to promote an immunosuppressive tumor *** studies demonstrated that focal adhesion kinase inhibitors(FAKi)in clinical development inhibit the infiltration of suppressive myeloid cells and T regulatory(T regs)cells,and subsequently enhance effector T cell *** inhibitors in clinical development have not been investigated in combination with RT in preclinical murine models or clinical ***,we investigated the impact of FAK inhibition on RT,its potential as an RT sensitizer and immunomodulator in a murine model of ***:We used a syngeneic orthotopic murine model to study the effect of FAKi on hypofractionated ***:In this study we showed that IN10018,a small molecular FAKi,enhanced antitumor response to *** activity of the combination of FAKi and RT is T cell *** in combination with RT enhanced CD8+T cell infiltration significantly in comparison to the radiation or FAKi treatment alone(P<0.05).FAKi in combination with radiation inhibited the infiltration of granulocytes but enhanced the infiltration of macrophages and T regs in comparison with the radiation or FAKi treatment alone(P<0.01).Conclusions:These results support the clinical development of FAKi as a radiosensitizer for PDAC and combining FAKi with RT to prime the tumor microenvironment of PDAC for immunotherapy.

Smartphone-Imaged HIV-1 Reverse-Transcription Loop-Mediated Isothermal Ampliflcation(RT-LAMP) on a Chip from Whole Blood

Viral load measurements are an essential tool for the long-term clinical care of human immunodeficiency virus （HIV）-positive individuals. The gold standards in viral load instrumentation, however, are still too limited by their size, cost, and sophisticated operation for these measurements to be ubiquitous in remote settings with poor healthcare infrastructure, including parts of the world that are disproportionately affected by HIV infection. The challenge of developing a point-of-care platform capable of making viral load more accessible has been frequently approached but no solution has yet emerged that meets the practical requirements of low cost, portability, and ease-of-use. In this paper, we perform reverse-transcription loop-mediated isothermal amplification （RT-LAMP） on minimally processed HIV-spiked whole blood samples with a microfluidic and silicon microchip platform, and perform fluorescence measurements with a consumer smartphone. Our integrated assay shows amplification from as few as three viruses in a - 60 nL RT- LAMP droplet, corresponding to a whole blood concentration of 670 viruses per μL of whole blood. The technology contains greater power in a digital RT-LAMP approach that could be scaled up for the determination of viral load from a finger prick of blood in the clinical care of HIV-positive individuals. We demonstrate that all aspects of this viral load approach, from a drop of blood to imaging the RT-LAMP reaction, are compatible with lab-on-a-chip components and mobile instrumentation.

Background:Having the ability to identify potential health risk factors such as lifestyle,ethnicity,health status,and social determinants associated with oral health status risk,health care providers can take an active role in health screening to discover any need for clinical preventive services,including dental preventive services,and can detect health ***:A descriptive type of cross-sectional study was conducted to explore the practice of common oral diseases among the patients visiting in the selected dental college and hospital in Bangladesh from October 2019 to December *** 100 patients were interviewed through a semi-structured questionnaire,followed by an oral examination through a checklist and a face-to-face interview.A systematically random sampling technique was employed to select the study ***:Among them,the(21-30)year age group shows a maximum 20%,whereas the(71-80)year age group represents the minority with only 2%.Among them,56%were male and 44%were *** represent the majority of 26%,whereas others(day laborers)represent only 4%.Decayed teeth remained in 72%of cases,teeth were missing in 31%of cases,and filled teeth were present in 24%of cases.A majority of 71%clean teeth once daily,whereas only 29%clean teeth twice ***,only 30%brush teeth at night and a maximum of 95%brush teeth in the ***,a maximum of 68%use toothpaste and brushes,while only 4%use meswak for cleaning their *** 4%of cases with no debris,51%of cases with less than 1/3rd of the tooth with debris,32%of cases with 1/3rd-2/3rd of the tooth with debris,and 13%of cases with more than 2/3rd of the tooth with debris were ***,about 30%had no calculus,52%less than 1/3rd of tooth,16%in between 1/3rd and 2/3rd of tooth,and 2%more than 2/3rd of the calculus was *** 47%of mild,22%of moderate,6%of severe and 25%had no ***,for treatment purposes,20.69%was advised for scaling,19.66%for

Objectives:To determine the association of Human leukocyte antigen B(HLA‑B)alleles with ankylosing spondylitis and the allelic association of HLAB27 with endoplasmic reticulum aminopeptidase 1(ERAP1)single-nucleotide polymorphism(SNP)rs30187 in patients with ankylosing spondylitis(AS).Methods:We studied 48 consecutively enrolled well-defined AS patients and 48 healthy controls recruited from the outpatient clinic of the rheumatology department of Bangabandhu Sheikh Mujib Medical University(BSMMU),Dhaka,*** genotype HLA-B alleles from peripheral blood DNA,we designed sequence-specific primers for polymerase chain ***'s sequencing method was used to detect the ERAP1 rs30187 *** HLA-B allele distributions were compared between cases and *** infer an allelic association between HLA-B27 and ERAP1,allele distributions from both loci were compared using ***-squared test or Fisher's exact test was used to quantify the association,and their strength of association was tested by odds ratio(OR)with 95%confidence interval(CI).Results:The frequency of HLA-B27 was found to be higher in AS patients(60.4%)than in healthy controls(8.3%).HLA-B27 allele had an OR of 16.8(95%CI=5.2–54.4)for the development of ***,4%of AS patients had both HLA-B27 and *** significant association was found between ERAP1 rs30187 SNP and HLA-B27 in patients with AS(OR=1.7,95%CI=0.5–5.6).Conclusions:HLA-B27 was significantly associated with AS in patients in ***,no association between ERAP1 rs30187 SNP and HLA-B27 was observed.
