Physical and Numerical Modelling Applied to a Major New Port Development and Impacts of Its Deep Navigational Channel

This paper describes the use of a numerical and physical modelling study in the design of large breakwaters for a new port and dry dock complex on the southern coast of Oman. The numerical modelling was carried out to optimise the entrance channel layout with respect to wave penetration into the port and to refine design conditions for the sizing of the primary armour on the breakwaters. Wave conditions inside and outside of the port have been assessed using the 2-dimensional numerical wave penetration model MIKE21 EMS （Elliptic Mild-Slope）. As part of the design process, 3D physical modelling studies were also undertaken at Delft Hydraulics in the Netherlands to confirm the stability of the armour on the trunk and roundhead of the breakwaters and to verify the influence of the deep approach channel on stability. The opportunity was taken to extend the physical model tests to assess the influence of the deep channel on wave penetration through the port entrance. The paper focuses on the influence of the deep channel on wave conditions in the entrance to the port and compares the results from the numerical and physical modelling.

Preliminary Assessment of Operational Downtime of Sea Ports and Berthing Facilities through Numerical Modelling

Downtime due to adverse wave conditions for vessels at berth is an important commercial aspect in the planning and development of a sea port or a berthing terminal. This paper describes a practical technique for preliminary assessment of operational downtime at a proposed bulk terminal. Time-series wind and wave data at an offshore location was purchased. Numerical modelling was then carried out using the MIKE21 SW （spectral wave） model developed by DHI （Deutsches Hydrographisches Institut） to transform these offshore waves into inshore in order to derive wave conditions at the berth. Both wind-waves and swell-waves were considered. Waves affecting the head and beam of a vessel were considered separately for a wide range of vessel sizes with DT （displacement tonnage） ranging from 5,000 tons to 〉 200,000 tons. The limiting wave height H5% was used. Operational downtime was also calculated using significant wave height, Hs as a criterion with limits ofHs = 1.0 m for beam seas and Hs = 1.5 m for head seas. The methodology and lessons learnt from the study can be applied for the development of any sea port worldwide.

ADAM8 has been identified as the fibronectin-cleaving enzyme in human degenerative intervertebral disc(IVD)tissues.^(1)Increase of the active form of ADAM8 and its cleavage product,fibronectin fragment,correlated with an increased degree of IVD degeneration.1 Furthermore,fibronectin fragments have been shown to accelerate the progression of IVD degeneration.^(2)These findings suggest that ADAM8 may play a role in degenerative disc disease,a clinical problem with tremendous socioeconomic burdens in the United States.

While hypoxic signaling has been shown to play a role in many cellular processes,its role in metabolism-linked extracellular matrix(ECM)organization and downstream processes of cell fate after musculoskeletal injury remains to be ***(HO)is a debilitating condition where abnormal bone formation occurs within extra-skeletal *** andhypoxia-inducible factor 1α(HIF-1α)activation have been shown to promote ***,the underlying molecular mechanisms bywhich the HIF-1αpathway in mesenchymal progenitor cells(MPCs)contributes to pathologic bone formation remain to ***,we used a proven mouse injury-induced HO model to investigate the role of HIF-1αon aberrant cell ***-cell RNA sequencing(scRNA-seq)and spatial transcriptomics analyses of the HO site,we found that collagen ECM organizationis the most highly up-regulated biological process in *** mesenchymal cell-specific deletion of Hif1α(Hoxa11-CreER^(T2);Hif1a^(fl/fl))significantly mitigated HO in ***-seq analysis of these Hoxa11-CreER^(T2);Hif1a^(fl/fl)mice identified the PLOD2/LOXpathway for collagen cross-linking as downstream of the HIF-1αregulation of ***,our scRNA-seq data and mechanisticstudies further uncovered that glucose metabolism in MPCs is most highly impacted by HIF-1α*** a translational aspect,a pan-LOX inhibitor significantly decreased HO.A newly screened compound revealed that the inhibition of PLOD2 activity in MPCssignificantly decreased osteogenic differentiation and glycolytic *** suggests that the HIF-1α/PLOD2/LOX axis linked tometabolism regulates HO-forming MPC *** results suggest that the HIF-1α/PLOD2/LOX pathway represents a promisingstrategy to mitigate HO formation.
