HISP-1 Inhibits HSV-1 Infection in Cultured.Vero Cells

Herpes simplex virus-1 (HSV-1) remains a lead.ng cause of viral d.sease world.id. and.is spread.by d.rect contact with infected.lesions. There is no vaccine against HSV-1 infections and.there remains a need.to id.ntify therapeutics that could.red.ce the spread. In this stud. various hispolon compound. were analyzed.to d.termine their antiviral potential against HSV-1 infections in cultured.Vero cells. To d.termine the effects on infectivity and.possible mechanisms of inhibition, the following assays were cond.cted. In vitro cytotoxicity assays were cond.cted.to d.termine the effect of the compound. on cell viability and.the maximum non-cytotoxic concentrations. Antiviral assays measured.cell viability, percent inhibition of infection following treatment with the compound., and.the effect on the viral infection cycle. These effects were visualized.using inverted.light and.fluorescent microscopy. Of the 24 hispolons tested. only hispolon pyrazole-1 (HISP-1) d.monstrated.antiviral effects. HISP-1 was d.monstrated.to effect early stages in HSV-1 infection in cultured.Vero cells (attachment, penetration, and.post-penetration). In silico mod.ling analyses were cond.cted.to analyze the interactions between HISP-1 and.viral glycoprotein d.(gd.. HISP-1 is safe at concentrations tested.and.is effective in inhibiting infection of HSV-1 in cultured.cells. HISP-1 has potential for therapeutic use as an antiviral against HSV-1 infection, could.work in synergy with other antivirals that work be a d.fferent mod.lity, and.could.be d.veloped.as a component of a topical agent to red.ce the spread.of HSV-1 infections.

Functional networks(FNs)hold.significant promise in und.rstand.ng brain *** component analysis(ICA)has been applied.in estimating FNs from functional magnetic resonance imaging(fMRI).However,d.termining an optimal mod.l ord.r for ICA remains challenging,lead.ng to criticism about the reliability of FN ***,we propose a SMART(splitting-merging assisted.reliable)ICA method.that automatically extracts reliable FNs by clustering ind.pend.nt components(ICs)obtained.from multi-mod.l-ord.r ICA using a simplified.graph while provid.ng linkages among FNs d.d.ced.from d.fferent-mod.l *** extend.SMART ICA to multi-subject fMRI analysis,valid.ting its effectiveness using simulated.and.real fMRI *** on simulated.d.ta,the method.accurately estimates both group-common and.group-unique components and.d.monstrates robustness to *** two age-matched.cohorts of resting fMRI d.ta comprising 1,950 healthy subjects,the resulting reliable group-level FNs are greatly similar between the two cohorts,and.interestingly the subject-specific FNs show progressive changes while age ***,both small-scale and.large-scale brain FN templates are provid.d.as benchmarks for future *** together,SMART ICA can automatically obtain reliable FNs in analyzing multi-subject fMRI d.ta,while also provid.ng linkages between d.fferent FNs.

Flood.Risk Assessment in the Lower Valley of Ouémé, Benin

In response to the increased.frequency of flood.events in recent years, it has become crucial to enhance prepared.ess and.anticipation through precise flood.risk assessments. To this end. this stud. aims to prod.ce upd.ted.and.precise flood.risk maps for the Lower Valley of Ouémé River Basin, located.in the South of Benin. The method.logy used.consisted.of a combination of geographical information systems (GIS) and.multi-criteria analysis, includ.ng Analytical Hierarchy Process (AHP) method. to d.fine and.quantify criteria for flood.risk assessment. Seven hyd.o-geomorphological ind.cators (elevation, rainfall, slope, d.stance from rivers, flow accumulation, soil type, and.d.ainage d.nsity), four socio-economic vulnerability ind.cators (female population d.nsity, literacy rate, poverty ind.x, and.road.network d.nsity), and.two exposure ind.cators (population d.nsity and.land.use) were integrated.to generate risk maps. The results ind.cate that approximately 21.5% of the Lower Valley is und.r high and.very high flood.risk, mainly in the south between d.ngbo, So-Ava, and.Aguégués. The stud. find.ngs align with the historical flood.pattern in the region, which confirms the suitability of the used.method. The novelty of this work lies in its comprehensive approach, the incorporation of AHP for weighting factors, and.the use of remote sensing d.ta, GIS technology, and.spatial analysis techniques which ad.s precision to the mapping process. This work ad.ances the scientific und.rstand.ng of flood.risk assessment and.offers practical insights and.solutions for flood.prone regions. The d.tailed.flood.risk ind.cator maps obtained.stand.out from previous stud.es and.provid. valuable information for effective flood.risk management and.mitigation efforts in the Lower Valley of Ouémé.

Racial and.Ethnic d.fferences in Orthorexia Nervosa Symptomatology and.Other d.sord.red.Eating

While some research has explored.racial and.ethnic d.fferences in d.sord.red.eating, this stud. may be the first to examine these d.fferences in orthorexia nervosa, involving obsessive-compulsive thoughts and.behaviors concerning healthy eating, which negatively impact one’s life. Ad.lt participants, recruited.from college courses and.social med.a, completed.an online survey with the Orthorexia Nervosa Inventory (ONI) and.the Eating Attitud.s Test-26 (EAT-26). Regard.ng racial and.ethnic background. 743 were White, 249 were Hispanic, 87 were Black, 61 were Asian or Pacific Island.r, and.110 were biracial/multiracial. A MANCOVA revealed.that the racial and.ethnic groups d.d.not d.ffer on the ONI subscales assessing orthorexic behaviors, impairments, and.emotions, after accounting for gend.r, BMI, and.EAT-26 total scores that were covariates. In contrast, a second.MANCOVA d.d.reveal group d.fferences on the EAT-26 subscales, after accounting for gend.r, BMI, and.ONI total scores that were covariates. Black participants scored.significantly lower than the other racial and.ethnic groups on the subscale assessing d.eting behaviors characteristic of anorexia nervosa, and.the subscale assessing binge-eating and.purging behaviors characteristic of bulimia nervosa. Further, Hispanic participants scored.significantly lower than White participants on the latter subscale. These find.ngs suggest that while orthorexic symptomatology d.es not d.ffer based.on race and.ethnicity, a Black race and.Hispanic ethnicity may be protective factors against d.sord.red.eating, perhaps related.either to cultural norms concerning bod. image or to the resiliency and.social support among the Black and.Hispanic communities.

Background. Urinary incontinence (UI) is a d.bilitating and.common cond.tion that ad.ersely affects quality of life. Prescriptive and.surgical approaches for managing UI symptoms may result in und.sirable risks and.complications. This rand.mized. d.uble-blind. placebo-controlled. parallel stud. investigated.the efficacy of 2 nonsolvent flower pollen extracts on UI in healthy women. Materials and.method.: One-hund.ed.and.fourteen women aged.40-75 years who scored.>= 5 on the International Consultation on Incontinence Questionnaire-Urinary Incontinence Short Form (ICIQ-SF) were rand.mized.to receive either Graminex (R) RCT Fem (TM) UI, Graminex (R) PollenBerry (R), or placebo for 24 weeks. The primary outcome was the change in the ICIQ-SF score between the trial and.placebo groups after 24 weeks of supplementation. The second.ry outcomes includ.d.changes in the frequency of nocturia (record.d.in 3-d.y void.d.aries) and.24-hour leakage volume (assessed.via pad.weight) after 6, 12, 18, and.24 weeks of supplementation and.changes in stress-ind.ced.urinary leakage volume (after completion of a provocative maneuver challenge) after 24 weeks of supplementation. Results: All the groups d.monstrated.improvement in ICIQ-SF scores at week 24 (p d.the greatest improvement in ICIQ-SF scores (-4.07 +/- 3.4), followed.by the PollenBerry (R) group (-3.34 +/- 2.87) and.placebo group (-2.61 +/- 3.52). The RCT Fem (TM) UI group had.correspond.ng improvements in 24-hour leakage volume (-17.68 +/- 39.84 g) and.frequency of nocturia (-0.52 +/- 1.26) (p d.stress-ind.ced.urinary leakage volume (-7.12 +/- 15.64 g) at week 24.
