Bioinformatic Comparison of Bacterial Secretomes

The rapid increasing number of completed bacterial genomes provides a good opportunity to compare their proteomes. This study was undertaken to specifically compare and contrast their secretomes--the fraction of the proteome with predicted N-terminal signal sequences, both type Ⅰ and type Ⅱ. A total of 176 theoretical bacterial proteomes were examined using the ExProt program. Compared with the Gram-positives, the Gram-negative bacteria were found, on average, to contain a larger number of potential Sec-dependent sequences. In the Gram-negative bacteria but not in the others, there was a positive correlation between proteome size and secretome size, while there was no correlation between secretome size and pathogenicity. Within the Gram-negative bacteria, intracellular pathogens were found to have the smallest secretomes. However, the secretomes of certain bacteria did not fit into the observed pattern. Specifically, the secretome of Borrelia burgdoferi has an unusually large number of putative lipoproteins, and the signal peptides of mycoplasmas show closer sequence similarity to those of the Gramnegative bacteria. Our analysis also suggests that even for a theoretical minimal genome of 300 open reading frames, a fraction of this gene pool （up to a maximum of 20%） may code for proteins with Sec-dependent signal sequences.

Mitigation of sulfate reducing Desulfovibrio ferrophilus microbiologically influenced corrosion of X80 using THPS biocide enhanced by Peptide A

Tetrakis hydroxymethyl phosphonium sulfate(THPS) was enhanced by a 14-mer Peptide A, with its core12-mer sequence mimicking part of Equinatoxin II protein, in the mitigation of sulfate reducing Desulfovibrio ferrophilus MIC(microbiologically influenced corrosion) of X80 carbon steel. Results proved that50 ppm(w/w) THPS was sufficient to mitigate the D. ferrophilus biofilm, and its very agressive MIC(19.7mg/cm^(2) in 7 days or 1.31 mm/a), but not 20 ppm THPS. To achieve effective mitigation at a low dosage of THPS, biofilm-dispersing Peptide A was added to 20 ppm THPS in the culture medium. Sessile cell counts were reduced by 2-log and 4-log after enhancement by 10 ppb and 100 ppb Peptide A, respectively. Enhancement efficiency(further reduction in corrosion rate) reached 69% for 10 ppb Peptide A and 83% for100 ppb Peptide A compared with 20 ppm THPS alone treatment, indicating that Peptide A was a good biocide enhancer for THPS.

Cyanobacteria Diversity in Blooms from the Greater Sudbury Area

The Greater Sudbury Area is approximately 400 km north of the city of Toronto and falls within a large number of temperate lakes of various sizes. This area has been mined for nickel and other metals for several decades. These activities have affected the watersheds of Northern Ontario and have influenced the chemistry of a number of lakes. Blooms of cyanobacteria occur yearly in several lakes, mainly in the early and late summer months. Much of the chemistry of these lakes is known but the nature of the cyanobacterial blooms and the factors that may contribute to their sudden appearance are not. We sampled blooms from five Greater Sudbury Area lakes and identified the species present by morphological and molecular methods. The dominant genera present as characterized by morphological examination were Synechocystis, Leptolyngbya, Anabaena, Cyl-indrospermum, Nostoc, Borzia, Phormidium, Pseudoanabaena, Oscillatoria, and Planktothrix. Three of these isolates, Leptolyngbya, Anabaena, and Planktothrix were confirmed by partial rRNA sequence analysis.

Evaluation of the Pathogen Detect<sup>®</sup>System and Anthracene-Based Enzyme Substrates for the Detection and Differentiation of <i>E. coli</i>and Total Coliforms in Water Samples

Indirect detection of Escherichia coli and total coliforms can be based on the enzymatic activities of β-glucuronidase (β-glu) and β-galactosidase (β-gal). These enzymes utilize the substrates anthracene-β-d-glucuronide and pyrene d-galactopyranoside, respectively. Substrate cleavage by the enzyme releases the soluble fluorescent molecules 2-hydroxyanthracene and 1-hydroxypyrene, which can then be detected by a fluorometer. The Pathogen Detect? system is an automated portable unit that can measure fluorescent enzyme products. In this report, we investigated the utility of the Pathogen Detect? system for potential automation of water quality monitoring. The PDS unit has the ability to detect E. coli, mean 14.7 h at a standard deviation of 1.5, when the sample mean is 9.1 cells in 100 mL with a standard deviation of 12.6. Similarly, total coliforms may be detected at mean 14.7 h with a standard deviation of 1.4 when the sample mean is 59.6 cells in 100 mL, with a standard deviation of 144.5. The PDS unit has the ability to detect single cells of either total coliforms or E. coli in 100 mL water sample within 18 hours. Turbidity and color of water samples have no impact on the detection of E. coli and total coliforms.

Total knee arthroplasty:Effect of obesity and other patients' characteristics on operative duration and outcome

AIM: To examine the effects of patients' characteristics mainly obesity on operative duration and other outcomemeasures of knee arthroplasty. METHODS: This is a retrospective chart review of 204 patients who had knee arthroplasty within the past five years(2007-2011) at King Abdulaziz Medical City in Riyadh, Kingdom of Saudi Arabia. The data collection form was developed utilizing the literature review to gather all the needed variables. Data were gathered from admission notes, nursing notes, operative reports and discharge ***: A feasible sample of 204 patients were included in the study. Of those patients, 155(76%) were females. The mean age was 70.1 years for males(SD ± 9.4) and 62.7 years(SD ± 8) for females. Regarding the type of total knee replacement(TKR), 163(79.9%) patients had unilateral TKR and 41(20.1%) had bilateral TKR. Nine patients(4.4%) had a normal body mass index(BMI)(18.5 to < 25). Overweight patients(BMI 25 to < 30) represented 18.1%. Obesity class Ⅰ(BMI 30 to < 35) and obesity class Ⅱ(BMI from 35 to < 40) were present in 23% and 29.9% of the patients, respectively. Morbid obesity(BMI greater than 40) was present in 24.5%. The mean duration of surgery was 126.3 min(SD ± 30.8) for unilateral TKR and 216.6 min(SD ± 55.4) for bilateral *** mean length of stay in the hospital was 12 d(SD ± 4.9). The complications that patients had after the operation included 2 patients(1%) who developed deep venous thrombosis, 2 patients(1%) developed surgical wound infections and none had pulmonary embolism. Patients' characteristics(including age, gender, BMI and co-morbidities) did not have an effect on the operative duration of knee replacement nor the length of hospital stay. CONCLUSION: Our study shows that obesity and other patients' characteristics do not have effect on the operative duration nor the length of hospital stay following TKR.
