Objective:Autophagy is one of the basic cellular mechanism during preimplantation development of mammalian embryos,and it plays crucial role in several physiological *** aim of this study is to research the effects of Interleukin-1 beta(IL-1β) on preimplantation embryonic development and autophagy in ***:We collected the different periods of preimplantation mouse embryos cultured in vitro,detecting the effects of IL-1β on autophagy and blastocyst quality by immunofluorescence staining,RT-qPCR and western ***:Our in vitro culture system analysis shows effect of IL-1β in medium on the development of mouse embryos and it was found to be concentration dependent.A preimplantation embryo culture using medium containing IL-1(3 did not improve cleavage and blastocyst development rates of mouse embryos;however,blastocyst quality was significantly improved by increasing total cell number,especially in supplementary 20 ng/mL IL-1β.Furthermore,autophagy activation mainly occurs in 2 to 4 cell embryo and blastocyst,20 ng/mL IL-1β into culture medium can effectively enhance levels of messenger RNA and protein of autophagy-related-factors in 2 to 4 cell embryos and blastocyst,while these factors reduce in VGX-1027(IL-1β inhibitor) groups that also reduce the quality of *** of IL-1β on the development of embryo reduced in 20 ng/mL IL-1β supplemented group when 5 mM3-methyladenine(3-MA) was also added,which used to inhibit autophagy activation in endogenous PtdIns3 Ks signal ***:Our current results show that exogenous IL-1β can effectively induce autophagy in mouse embryos at stages of 2 to 8 cell and blastocyst,that also help to improve the quality of blastocyst.

Power Series Solution for Strongly Non-linear Conservative Oscillators

This paper compares the accuracy of the power series approach with that of the modified Lindstedt-Poincare method for strongly nonlinear *** free vibration of an undamped Duffing oscillator is considered because it has an exact *** the power series approach,the time variable is transformed into an“oscillating time”which reduces the governing equation to a form well-conditioned by the power series *** results show that the power series approach provides extremely accurate vibration frequencies,even at large values of the nonlinear parameter,compared with errors of up to nine percent for the modified Lindstedt-Poincare method.

Yield Stability and Adaptability of Elite Doubled Haploid (DH) Rice Genotypes in Different Locations in Central Clay Plains of the Sudan

Stability analysis of grain yield is an efficient tool for the selection of varieties adapted to fairly wide cultivation zone. A study aims to determine adaptability and stability for grain yield and agronomic performance of 10 elite Doubled Haploid (DH) rice genotypes generated through another culture technique, along with local cultivar Umgar as a check under irrigated lowland conditions in three different environments in the central clay plains of the Sudan;during two cropping seasons 2019 and 2020. The trials were conducted in a randomized complete block design with three replications. Grain yield stability was studied, using the Additive Main effect and Multiplicative Interaction (AMMI) model. Bi-plots were developed following GGE bi-plot methodology over the six environments. The combined analysis of variance revealed significant to highly significant (P < 0.001) effects of genotypes, environments, and genotype by environment interaction. Moreover, the analysis of variance based on AMMI indicated significant genotypes, environments, and GE interaction with a total variation of 43.16%, 44.26% and 12.58% respectively. A biplot-AMMI analysis and yield stability index incorporating the AMMI stability value and yield in a single non-parametric index were used to identify the genotypes with the highest and stable yield. The overall mean for grain yield was 5.69 t/ha, with a range of 5.77 to 4.12 t/ha. The genotypes k150147, k150870, k150032, k150912, and k150307 out yielded the local check Umgar by 40%, 38%, 34%, 34% and 33% respectively. The most genotypes were mid-early maturities which were harvested at 107, 113, 108, 109 and 112 days after sowing, respectively. These five genotypes also showed grain yield stability along with their high mean yield performance according to the AMMI analysis and widely adaptable to the tested locations. Randomized, double-blinded, placebo-controlled trial evaluating simethicone pretreatment with bowel preparation during colonoscopy

BACKGROUND The presence of small air bubbles and foam are an impediment to a successful colonoscopy. They impair an endoscopist’s view and diminish the diagnostic accuracy of the study. This has been particularly noted to be of concern with the switch to lower volume polyethylene glycol (PEG) and bisacodyl combination preparation. AIM To evaluate the effect of oral simethicone addition to bowel preparation on intraluminal bubbles reduction during colonoscopy. METHODS Described is a prospective, randomized, multi-center, double-blinded, placebocontrolled study to evaluate the use of premixed simethicone formulation with split-regimen, low-volume PEG-bisacodyl combination bowel preparation for 168 outpatients undergoing screening, surveillance, and diagnostic colonoscopies. Primary outcome includes evaluation of bubbles during colonoscopy graded using the Intraluminal Bubbles Scale. Secondary outcomes include evaluation of the Boston Bowel Preparation Scale (BBPS), total number of polyps, polyp size differentiation, polyp laterality, adenoma detection, mass detection, cecal insertion time, withdrawal time, and patient-reported adverse events. RESULTS Higher Intraluminal Bubbles grades III and IV (less than 75% of the mucosa cleared of bubbles/foam requiring intervention with simethicone infused wash) were detected in the placebo group [Simethicone n = 4/84 vs Placebo n = 20/84 (P = 0.007)]. BBPS total score was 7.42 [standard deviation (SD)=± 1.51] in the simethicone group and 7.28 (SD =± 1.44) in the placebo group (P = 0.542) from a total of 9. Significantly higher number of adenomas were detected in the simethicone group (P = 0.001). CONCLUSION The addition of simethicone to bowel preparation is well advised for its antifoaming properties. The results of this study suggest that addition of oral simethicone can improve bowel wall visibility.

Nitrogenase activity is a very distinct property of Pseudomonas stutzeri A1501. It is carried out in the presence of microaerophilic condition that is in oxygen limited condition. The biofilm formation is at its best in minimal media at nitrogen depletion condition Anr, a global transcription regulator that promotes biofilm formation. Anr absence plays a positive role in nitrogen fixation by regulating nifA, nifH and ntrC. The nitrogenase activity in anr insersion mutant was significantly down regulated. At different concentration of oxygen the nitroagenase activity was substantially differentiated. The qRT-PCR shows down regulation of nif island gene at nitrogen fixation condition. The finding suggests that different oxygen concentration in the environment play differential role against the nitrogenase activity and biofilm formation. The qRT-PCR results for rpoS during biofilm formation was up-regulated showing that low level of oxygen it has a signaling effect causing Anr to expresses which in turns controls not only rsm XYZ but also regulate the biofilm formation. Effect of Oxygen stress shows no significant effect on the survival of pseudomonas stutzeri A1501 which suggests that oxidative stress has no role on anr regulation.

Background and Aims:Pharmaceutical therapy for NASH is associated with lipid modulation,but the consensus on drug treatment is limited and lacks comparative analysis of effectiveness.A network meta-analysis was conducted to compare NASH drug classes in lipid ***:Online databases were searched for randomized controlled trails(RCTs)evaluating NASH treatments in biopsy-proven NASH *** were classified into four groups:(1)inflammation,(2)energy,(3)bile acids,and(4)fibro-sis based on the mechanism of action.A Bayesian network analysis was conducted with outcome measured by mean difference(MD)with credible intervals(Crl)and surface un-der the cumulative ranking curve(SUCRA).Results:Forty-four RCTs were included in the *** acid modulat-ing treatments(MD:0.05,Crl:0.03-0.07)were the best treatment for improvement in high-density lipid(HDL)cho-lesterol,followed by treatments modulating energy(MD:0.03,Crl:0.02-0.04)and fibrosis(MD:0.01,Crl:−0.12 to 0.14)compared with *** top three treatments for reduction in triglycerides were treatments modulating energy(MD:−0.46,Crl:−0.49 to−0.43),bile acids(MD:−0.22,Crl:−0.35 to−0.09),and inflammation(MD:−0.08,Crl:−0.13 to−0.03)compared with *** found treatment modulating fibrosis(MD:−1.27,Crl:−1.76 to−0.79)was the best treatment for reduction in low-density lipid(LDL)cholesterol followed by treatment modulating in-flammation(MD:−1.03,Crl:−1.09 to−0.97)and energy(MD:−0.37,Crl:−0.39 to−0.34)compared with placebo,but LDL cholesterol was worsened by treatments modulat-ing bile ***:Network analysis comparing the class effects of dyslipidemia modulation in NASH found that treatment targets can include optimization of athero-genic *** studies are required to evaluate the cardiovascular outcomes.
