Amplifiable protein identification via residue-resolved barcoding and composition code counting

Ultrasensitive protein identification is of paramount importance in basic research and clinical diagnostics but remains extremely challenging. A key bottleneck in preventing single-molecule protein sequencing is that, unlike the revolutionary nucleic acid sequencing methods that rely on the polymerase chain reaction(PCr) to amplify DNA and rNA molecules, protein molecules cannot be directly amplified. Decoding the proteins via amplification of certain fingerprints rather than the intact protein sequence thus represents an appealing alternative choice to address this formidable challenge. Herein, we report a proof-of-concept method that relies on residue-resolved DNA barcoding and composition code counting for amplifiable protein fingerprinting(AmproCode). In AmproCode, selective types of residues on peptides or proteins are chemically labeled with a DNA barcode, which can be amplified and quantified via quantitative PCr. The operation generates a relative ratio as the residue-resolved ‘composition code’ for each target protein that can be utilized as the fingerprint to determine its identity from the proteome database. We developed a database searching algorithm and applied it to assess the coverage of the whole proteome and secretome via computational simulations, proving the theoretical feasibility of AmproCode. We then designed the residue-specific DNA barcoding and amplification workflow, and identified different synthetic model peptides found in the secretome at as low as the fmol/L level for demonstration. These results build the foundation for an unprecedented amplifiable protein fingerprinting method. We believe that, in the future,AmproCode could ultimately realize single-molecule amplifiable identification of trace complex samples without further purification, and it may open a new avenue in the development of next-generation protein sequencing techniques.

Validation of the Arabic version of the Ocular Surface Disease Index Questionnaire

AIM: To develop an Arabic version of the ocular surface disease index(OSDI) and to assess its reliability and ***: A cross sectional study was carried out to validate the Arabic version of the OSDI questionnaire. Initially, forward-backward translation was used to translate the English version of OSDI into Arabic. The translated questionnaire was tested for equivalence and cultural adaptability. Totally 200 subjects were then recruited from a non-clinical population and asked to complete the Arabic version of the OSDI(ArB-OSDI). The reliability of the questionnaire was assessed using Cronbach’s-α. A subgroup of 30 participants was asked to complete the questionnaire on two occasions to test the test-retest ***: A total of 200 participants were enrolled in the study. The average age of the study participants was 31.21±13.2 y and 57% were male. An acceptable internal consistency level for the ArB-OSDI questionnaire measured by Cronbach’s-α was revealed. All questions showed good internal consistency. Test-retest reliability analysis revealed good stability(interclass correlation coefficient, r=0.832, Pruct validity for the questionnaire was also ***: The ArB-OSDI questionnaire shows very good psychometric properties(acceptable internal consistency and test-retest reliability). That makes the questionnaire a valid potential tool to use in Arabic-speaking countries to monitor symptoms of dry eye disease in a larger population.

The number and variety of applications of artificial intelligence(AI)in gastr-ointestinal(GI)endoscopy is growing *** technologies based on machine learning(ML)and convolutional neural networks(CNNs)are at various stages of development and deployment to assist patients and endoscopists in preparing for endoscopic procedures,in detection,diagnosis and classification of pathology during endoscopy and in confirmation of key performance *** based on ML and CNNs require regulatory approval as medical *** between humans and the technologies we use are complex and are influenced by design,behavioural and psychological *** to the substantial differences between AI and prior technologies,important differences may be expected in how we interact with advice from AI ***-AI interaction(HAII)may be optimised by developing AI algorithms to minimise false positives and designing platform interfaces to maximise *** factors influencing HAII may include automation bias,alarm fatigue,algorithm aversion,learning effect and *** of these areas merits further study in the specific setting of AI applications in GI endoscopy and professional societies should engage to ensure that sufficient emphasis is placed on human-centred design in development of new AI technologies.

Molecular diversity of barley yellow dwarf virus-PAV from China and the Czech republic

Wheat yellow dwarf disease(BYD),caused by different species of barley/cereal yellow dwarf viruses(B/CYDVs),is one of the most serious cereal diseases in China and the Czech *** genetic diversity of the virus directly influences disease epidemiology,the molecular diversity and population structure of 24 Chinese isolates and 16 the Czech republic isolates of BYDV-PAV from different regions in two countries were analyzed by sequencing their coat protein(CP)and readthrough protein(rTP)domain(rTD)genes and comparing the sequences with six CP and 16 rTP sequences of BYDVPAV isolates from the NCBI database based on nucleotide identity position,phylogenetic analysis and nucleotide *** identities between the Chinese and the Czech republic isolates for the CP were 76.6–99.4%,73.9–89.1%for rTD(OrF5),*** Chinese and the other country isolates showed 74.7–99.2%nucleotide identity for rTP(OrF3+OrF5).Phylogenetic analysis of CP sequences showed that 20 Chinese isolates clustered in the same clade,but the other four Chinese isolates clustered in another clade with the isolates from the Czech republic and other *** population of BYDV-PAV in China had greater nucleotide variability and was more divergent than that in the Czech *** and ecological factors but not hosts might contribute to the population differences in the two countries.

Assessment of rainwater Quality in Warri and Environ, Southern Nigeria for Domestic Purposes

The study aimed at assessing the physiochemical characteristics of rainwater in Warri and it environ was investigated. Pb, Zn, Cd, Cu and Cr concentrations in rainwater from roof and non-roof sources were determined using Atomic Absorption Spectrophotometry. Three geospatial locations comprising Jakpa, Udu, and Ubeji were selected based on prevailing anthropogenic activities. The rainwater samples were systematically collected from (aluminum) roof and non-roof sources for the months of April, June, and August and October 2022, treated and analyzed in the laboratory for sixteen physicochemical parameters. results were statistically analyzed using ANOVA, and T-test for the determination of the level of relationships and variations across geospatial locations. Significant correlations (r = 0.72) exist between Cr in rainwater from roof and non-roof sources. Implying point-source contaminations and may be emanating from the influence of roof materials. Furthermore, high concentrations of Cd and Pb in roof source above WHO standards were mostly in Jakpa and Ubeji. Calculated Health risk Index (HrI) for children and adult is greater than 1. The results showed that most samples from the locations are considered not safe (HrI > 1) especially for Cd, which means that there are potential health risks consuming rainwater from Jakpa, Udu and Ubeji. Therefore, there is need for prompt sensitization program to dissuade people from directly drinking rainwater from these locations.
