Albumin fusion with granulocyte-macrophage colony-stimulating factor acts as an immunotherapy against chronic tuberculosis

A long duration of treatment and emerging drug resistance pose significant challenges for global tuberculosis(TB)eradication ***,there is an urgent need to develop novel strategies to shorten TB treatment regimens and to treat drug-resistant *** an albumin-fusion strategy,we created a novel albumin-fused granulocyte-macrophage colony-stimulating factor(albGM-CSF)molecule that harnesses albumin’s long half-life and targeting abilities to enhance the biostability of GM-CSF and direct it to the lymph nodes,where the effects of GM-CSF can increase dendritic cell populations crucial for eliciting a potent immune *** this study,we demonstrate that albGM-CSF serves as a novel immunotherapy for chronic Mycobacterium tuberculosis(Mtb)infections by enhancing GM-CSF biostability in ***,albumin is very safe,stable,and has a long half-life,thereby enhancing the biostability of *** the lungs and draining lymph nodes,albGM-CSF is able to increase the numbers of dendritic cells,which are crucial for the activation of naive T cells and for eliciting potent immune *** administration of albGM-CSF alone reduced the mean lung bacillary burden in mice with chronic tuberculosis *** GM-CSF administration was associated with IL-1βrelease from Mtb-infected dendritic cells and macrophages,higher IL-1βlevels were observed in albGM-CSF-treated mice with chronic tuberculosis infection than in mice receiving *** fusion with GM-CSF represents a promising strategy for the control of chronic lung tuberculosis infections and serves as a novel therapeutic vaccination platform for other infectious diseases and malignancies.

Identification of key genes regulating the synthesis of quercetin derivatives in Rosa roxburghii through integrated transcriptomics and metabolomics

Rosa roxburghii fruit is rich in flavonoids, but little is known about their biosynthetic pathways. In this study, we employed transcriptomics and metabolomics to study changes related to the flavonoids at five different stages of R. roxburghii fruit development. Flavonoids and the genes related to their biosynthesis were found to undergo significant changes in abundance across different developmental stages, and numerous quercetin derivatives were identified. We found three gene expression modules that were significantly associated with the abundances of the different flavonoids in R. roxburghii and identified three structural UDP-glycosyltransferase genes directly involved in the synthesis of quercetin derivatives within these modules. In addition, we found that RrBEH4, RrLBD1 and RrPIF8could significantly increase the expression of downstream quercetin derivative biosynthesis genes. Taken together,these results provide new insights into the metabolism of flavonoids and the accumulation of quercetin derivatives in R. roxburghii.

Explosion damage effects of aviation kerosene storage tank under strong ignition

In order to study the blast damage effects of aviation kerosene storage tanks,the out-field explosion experiments of 8 m3fixed-roof tanks were carried *** fragments,shock wave and fireball thermal radiation of the tank in the presence of bottom oil,half oil and full oil,as well as empty tank,were investigated under internal explosion by various TNT charge contents(1.8 kg,3.5 kg and 6.2 kg).The results showed that the tank roof was the only fragment produced,and the damage forms could be divided into three *** increase of TNT charge content and oil volume enlarged the deformation of the tank,while the hole ratio presented a trend of increase first and then *** H_r,maxand V_(max)values positively increased as increasing the TNT charge content and oil volume(from empty to half oil),but decreased in full *** Pmaxvalues had a progressive increase with the increment of TNT charge content,but not the case with the increase in oil *** development of fireball was divided into three stages:tank roof‘towed'flame,jet flow flame tumbling and rising,and jet flow flame *** Dmaxand Hf,maxvalues both increased as increasing TNT charge content and oil *** oscillation phenomenon of fireball temperature was observed in the cooling *** average temperature of fireball surface was positively correlated with TNT charge content,and negatively correlated with oil volumes.

Resilience-oriented Transmission Expansion Planning with Optimal Transmission Switching Under Typhoon Weather

This paper presents resilience-oriented transmission expansion planning(RTEP)with optimal transmission switching(OTS)model under typhoon *** proposed model carefully considers the uncertainty of component vulnerability by constructing a typhoon-related box uncertainty set where component failure rate varies within a range closely related with typhoon ***,a min-max-min model is developed to enhance transmission network resilience,where the upper level minimizes transmission lines investment,the middle level searches for the probability distribution of failure status leading to max worst-case expected load-shedding(WCEL)under typhoon,and the lower level optimizes WCEL by economic dispatch(ED)and OTS.A nested decomposition algorithm based on benders decomposition is developed to solve the *** studies of modified IEEE 30-bus and 261-bus system of a Chinese region illustrate that:a)the proposed RTEP method can enhance resilience of transmission network with less investment than widely used RTEP method based on attacker and defender(DAD)model,b)the influence of OTS on RTEP is closely related with contingency severity and system scale and c)the RTEP model can be efficiently solved even in a large-scale system.

A unified pre-training and adaptation framework for combinatorial optimization on graphs

Combinatorial optimization(CO)on graphs is a classic topic that has been extensively studied across many scientific and industrial ***,solving CO problems on graphs through learning methods has attracted great *** deep learning methods,e.g.,graph neural networks(GNNs),have been used to effectively assist the process of solving ***,current frameworks based on GNNs are mainly designed for certain CO problems,thereby failing to consider their transferable and generalizable abilities among different COs on ***,simply using original graphs to model COs only captures the direct correlations among objects,which does not consider the mathematical logicality and properties of *** this paper,we propose a unified pre-training and adaptation framework for COs on graphs with the help of the maximum satisfiability(max-SAT)*** first use max-SAT to bridge different COs on graphs since they can be converted to max-SAT problems represented by standard formulas and clauses with logical *** we further design a pre-training and domain adaptation framework to extract the transferable and generalizable features so that different COs can benefit from *** the pre-training stage,max-SAT instances are generated to initialize the parameters of the *** the fine-tuning stage,instances from CO and max-SAT problems are used for adaptation so that the transferable ability can be further *** experiments on several datasets show that features extracted by our framework exhibit superior transferability and max-SAT can boost the ability to solve COs on graphs.

The configuration of Au^(δ+)-ZrO_(2)^(δ−)species induced activation enhances electrocatalytic CO_(2)to formate conversion

Electrochemical CO_(2)conversion into value-added chemicals is a promising technology to solve the greenhouse effect and recycle chemical ***,the electrochemical CO_(2)reduction reaction(e-CO_(2)RR)is seriously compromised by weak CO_(2)adsorption and a rough CO_(2)activation process based on the chemical inertness of the CO_(2)molecule and the formed fragile metal–C/O *** this paper,we designed and fabricated Au particles embedded in ZrO_(2).The configuration of Au particles being of positive charge and ZrO_(2)with negative charge is induced and generated by metal–support interactions(MSIs).As a result,Au/ZrO_(2)@C presents a big difference in the CO_(2)conversion compared with the known work,affording a formate yield of 112.5μmol·cm^(−2)·h^(−1)at−1.1 V *** hydrogen electrode(RHE),and a max formate Faradaic efficiency of up to 94.1%at−0.9 V *** superior performance was attributed to the activated Au–ZrO_(2)interface to form the Au^(δ+)*** insitu Fourier transform infrared(FTIR)spectroscopy and theoretical calculations show that the MSIs configuration can be inclined to the*OCO intermediate generation on Au^(δ+)species activating CO_(2)molecules and then accelerate the formation of the*OCHO intermediate in e-CO_(2)RR,thereby favoring the CO_(2)conversion to formate.

Efficient synthesis of bottle-brush polymers enabled by selective photoactivation strategy in NIR-induced PET-RAFT polymerization

Bottle-brush polymers possessing densely grafted side chains have special properties that are distinct from linear ***,an efficient method for preparing bottle-brush polymers based on a selective photoactivation strategy in the photoinduced electron/energy transfer reversible addition-fragmentation chain transfer(PET-RAFT)polymerization was developed.A methacrylate monomer 2-(2-(dodecylthiocarbonothioylthio)propionate)ethyl methacrylate(DTPEMA)containing a 2-(dodecylthiocarbonothioylthio)propionate(DTP)unit with a secondary R group was *** the unique selectivity of metal naphthalocyanine photocatalyst zinc 2,11,20,29-tetra-tert-butyl-2,3-naphthalocyanine(ZnTtBNc)toward trithiocarbonate with tertiary R group,the PET-RAFT polymerization of DTPEMA catalyzed by ZnTtBNc was performed under near infrared(NIR)light(λ_(max)=760 nm)irradiation at room temperature,using 4-cyano-4-[(dodecylsulfanylthiocarbonyl)-sulfanyl]pentanoic acid(CDTPA)with a tertiary R group as a chain transfer agent(CTA).Proton nuclear magnetic resonance(1H NMR)and gel permeation chromatography(GPC)results demonstrated that the DTP units in the side chains were inert during the *** grafting poly(methyl acrylate)(PMA)from PDTPEMA via thermal-initiated RAFT polymerization,welldefined P(DTPEMA-g-PMA)bottle-brush polymers could be ***,the film prepared from the bottle-brush polymer exhibited excellent scratch self-healing property due to the interlocking of side chains.

Transferable adversarial slow feature extraction network for few-shot quality prediction in coal-to-ethylene glycol process

In the coal-to-ethylene glycol(CTEG)process,precisely estimating quality variables is crucial for process monitoring,optimization,and control.A significant challenge in this regard is relying on offline laboratory analysis to obtain these variables,which often incurs substantial monetary costs and significant time *** resulting few-shot learning scenarios present a hurdle to the efficient development of predictive *** address this issue,our study introduces the transferable adversarial slow feature extraction network(TASF-Net),an innovative approach designed specifically for few-shot quality prediction in the CTEG ***-Net uniquely integrates the slowness principle with a deep Bayesian framework,effectively capturing the nonlinear and inertial characteristics of the CTEG ***,the model employs a variable attention mechanism to identify quality-related input variables adaptively at each time step.A key strength of TASF-Net lies in its ability to navigate the complex measurement noise,outliers,and system interference typical in CTEG *** learning strategy using a min-max game is adopted to improve its robustness and ability to model irregular industrial data accurately and ***,an incremental refining transfer learning framework is designed to further improve few-shot prediction performance achieved by transferring knowledge from the pretrained model on the source domain to the target *** effectiveness and superiority of TASF-Net have been empirically validated using a real-world CTEG *** with some state-of-the-art methods,TASF-Net demonstrates exceptional capability in addressing the intricate challenges for few-shot quality prediction in the CTEG process.

Rapid Phase Ambiguity Elimination Methods for DOA Estimator via Hybrid Massive MIMO Receive Array

For a sub-connected hybrid multiple-input multiple-output(MIMO) receiver with K subarrays and N antennas, there exists a challenging problem of how to rapidly remove phase ambiguity in only single time-slot. A direction of arrival(DOA) estimator of maximizing received power(max-RP) is proposed to find the maximum value of K-subarray output powers, where each subarray is in charge of one sector, and the center angle of the sector corresponding to the maximum output is the estimated true DOA. To make an enhancement on precision, max-RP plus quadratic interpolation(max-RP-QI) method is designed. In the proposed max-RP-QI, a quadratic interpolation scheme is adopted to interpolate the three DOA values corresponding to the largest three receive powers of *** achieve the Cramer Rao lower bound, a Root-MUSIC plus max-RP-QI scheme is developed. Simulation results show that the proposed three methods eliminate the phase ambiguity during one time-slot and also show low computational complexities. The proposed Root-MUSIC plus max-RP-QI scheme can reach the Cramer Rao lower bound,and the proposed max-RP and max-RP-QI are still some performance losses 2–4 d B compared to the Cramer Rao lower bound.
