Impact of smoking cannabidiol (CBD)-rich marijuana on driving ability

To investigate effects of smoking cannabidiol(CBD)-rich marijuana on driving ability and determine free CBD andΔ9-tetrahydrocannabinol(THC)concentrations in capillary blood samples,a randomised,double-blind,placebo-controlled,two-way crossover pilot study was conducted with 33 participants.Participants smoked a joint containing 500 mg of tobacco and either 500 mg of CBD-rich marijuana(16.6%total CBD;0.9%total THC)or 500 mg of a placebo substance,then performed three different dimensions of the Vienna Test System TRAFFIC examining reaction time,behaviour under stress,and concentration performance.For further assessment of participants’fitness to drive,three tests of balance and coordination were evaluated and vital signs(blood pressure and pulse)were measured.Dried blood spot samples of capillary blood were taken after smoking and after completion of the tests to determine the cannabinoid concentrations(CBD,THC and THC-metabolites).The results revealed no significant differences between the effects of smoking CBD-rich marijuana and placebo on reaction time,motor time,behaviour under stress,or concentration performance.Maximum free CBD and THC concentrations in capillary blood were detected shortly after smoking,ranging between 2.6-440.0 ng/mL and 6.7-102.0 ng/mL,respectively.After 45 min,capillary blood concentrations had already declined and were in the range of 1.9-135.0 ng/mL(free CBD)and 0.9-38.0 ng/mL(free THC).Although the observed levels of free THC concentrations have been reported to cause symptoms of impairment in previous studies in which THC-rich marijuana was smoked,no signs of impairment were found in the current study.This finding suggests that higher CBD concentrations cause a negative allosteric effect in the endocannabinoid system,preventing the formation of such symptoms.Nevertheless,it is recommended that consumers refrain from driving for several hours after smoking CBD-rich marijuana,as legal THC concentration limits may be exceeded.

The emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)in 2019 prompted scientific,medical,and biotech communities to investigate infection-and vaccine-induced immune responses in the context of this pathogen.B-cell and antibody responses are at the center of these investigations,as neutralizing antibodies(nAbs)are an important correlate of protection(COP)from infection and the primary target of SARS-CoV-2 vaccine modalities.In addition to absolute levels,nAb longevity,neutralization breadth,immunoglobulin isotype and subtype composition,and presence at mucosal sites have become important topics for scientists and health policy makers.The recent pandemic was and still is a unique setting in which to study de novo and memory B-cell(MBC)and antibody responses in the dynamic interplay of infection-and vaccine-induced immunity.It also provided an opportunity to explore new vaccine platforms,such as mRNA or adenoviral vector vaccines,in unprecedented cohort sizes.Combined with the technological advances of recent years,this situation has provided detailed mechanistic insights into the development of B-cell and antibody responses but also revealed some unexpected findings.In this review,we summarize the key findings of the last 2.5 years regarding infection-and vaccine-induced B-cell immunity,which we believe are of significant value not only in the context of SARS-CoV-2 but also for future vaccination approaches in endemic and pandemic settings.

Background Post-weaned piglets suffer from F18+Escherichia coli(E.coli)infections resulting in post-weaning diar-rhoea or oedema disease.Frequently used management strategies,including colistin and zinc oxide,have contrib-uted to the emergence and spread of antimicrobial resistance.Novel antimicrobials capable of directly interacting with pathogens and modulating the host immune responses are being investigated.Lactoferrin has shown promising results against porcine enterotoxigenic E.coli strains,both in vitro and in vivo.Results We investigated the influence of bovine lactoferrin(bLF)on the microbiome of healthy and infected weaned piglets.Additionally,we assessed whether bLF influenced the immune responses upon Shiga toxin-producing E.coli(STEC)infection.Therefore,2 in vivo trials were conducted:a microbiome trial and a challenge infection trial,using an F18+STEC strain.BLF did not affect theα-andβ-diversity.However,bLF groups showed a higher relative abundance(RA)for the Actinobacteria phylum and the Bifidobacterium genus in the ileal mucosa.When analysing the immune response upon infection,the STEC group exhibited a significant increase in F18-specific IgG serum levels,whereas this response was absent in the bLF group.Conclusion Taken together,the oral administration of bLF did not have a notable impact on theα-andβ-diversity of the gut microbiome in weaned piglets.Nevertheless,it did increase the RA of the Actinobacteria phylum and Bifi-dobacterium genus,which have previously been shown to play an important role in maintaining gut homeostasis.Furthermore,bLF administration during STEC infection resulted in the absence of F18-specific serum IgG responses.

Experimental investigation of radiative heat propagation in a simplified generic packed bed

A novel test rig for the investigation of radiative heat transfer in packed beds has been developed and is introduced with representative experimental results.The individual components and the calibration are discussed.The generic packed bed is realized in a simplified way by an arrangement of parallel rods,which represent particles in pseudo-2D.In this arrangement,electrically heated rods provide the radiation propagating through the rod array to heat the passive counterparts.A sophisticated temperature-control scheme with a large number of thermocouples and infrared-imaging provides in-depth information about heat transfer in the system.Spectral radiation intensities are determined with a Fourier-transform infrared spectroscopy,which has been modified and validated for this specific application.In order to compare the influence of different surface properties of particles on the heat propagation and surface reflections,rod samples made of stainless steel and magnesium oxide are used.The influence of material properties becomes clearly visible by comparing the high radiation intensities resulting from a stainless steel rod array to the same geometry built from magnesium oxide rods.In addition,the influence of the surface properties is particularly evident in the infrared images since the reflections are significantly higher for the stainless steel samples than for the magnesium oxide samples.The experimental results in the current work demonstrate the ability of the test rig to provide data with a well-defined accuracy as a validation base for numerical radiation simulations in packed beds.

A numerical and experimental comparison of heat transfer in a quasi two-dimensional packed bed

Heat transfer plays a major role in many industrial processes taking place in packed beds.An accurate and reliable simulation of the heat exchange between particles is therefore crucial for a reliable operation and to optimize the processes in the bed.The discrete ordinates method(DOM)provides an established numerical technique to model radiative heat transfer in granular media that offers the possibility to consider the directional dependence of the radiation propagation.In this work,DOM is compared with Monte Carlo ray tracing,which provides an alternative method for heat transfer simulations.Geomet-rically simple configurations are used to investigate the influence of the angular discretization on the accuracy of the results and the computation time in both methods.The obtained insights are then transferred to a more complex configuration of a quasi two-dimensional test rig consisting of metal rods for which also experimental results are available.Our results show that both DOM and Monte Carlo ray tracing allow for an accurate simulation of heat transfer in packed beds.Monte Carlo ray tracing requires thereby computation times that are surprisingly competitive(although still somewhat slower)compared to DOM and allows for an easier computation of highly accurate reference solutions.In our preliminary comparison to the experimental test rig,Monte Carlo ray tracing also provides the advantage that it can more easily model highly specular materials such as stainless steel.Both methods are comparable for diffuse materials such as magnesium oxide.

Mapping the clinical practice of traditional,complementary and integrative medicine in oncology in Western countries:A multinational cross-sectional survey

Background Many cancer patients seek adjunctive therapies to biomedical cancer treatments at some point of their disease trajectory.While acupuncture is increasingly recommended by leading oncological associations,limited evidence exists concerning the evidence-informed practice and adherence to current guidelines of traditional complementary and integrative medicine(TCIM)practitioners treating cancer patients.Methods An international online-survey assessed the demographical data,clinical practice,and sources of information used by TCIM practitioners in Austria,Germany,United States of America,Australia,and New Zealand.Results In total,404 respondents completed the survey,of which 254(62.9%)treated cancer patients.Most practitioners were acupuncturists and herbalists(57.1%),had(16.8±9.9)years of clinical experience and see a median of 2(1,4)cancer patients per week.Breast cancer(61.8%)is the most common cancer type seen in TCIM clinics.Adjunctive TCIM treatments are frequently concurrent with the patient’s cancer specific treatment(39.9%),which is also reflected by the main goal of a TCIM treatment to alleviate side effects(52.4%).However,only 28.0%of the respondents are in contact with the treating oncologist.According to the respondents,pain is most effectively treated using acupuncture,while herbal medicine is best for cancer-related fatigue.TCIM practitioners mostly use certified courses(33.1%)or online databases(28.3%)but often believe that experts are more reliable to inform their practice(37.0%)than research publications(32.7%).Conclusion Acupuncturists and herbalists commonly treat cancer patients.Most practitioners use TCIM as an adjunct to biomedicine as supportive care and use it largely in accordance with current oncological guidelines.

Liver transplantation as an alternative for the treatment of neuroendocrine liver metastasis: Appraisal of the current evidence

Background:Liver transplantation(LT)for neuroendocrine liver metastases(NELM)is still in debate.Studies comparing LT with liver resection(LR)for NELM are scarce,as patient selection is heterogeneous and experience is limited.The goal of this review was to provide a critical analysis of the evidence on LT versus LR in the treatment of NELM.Data sources:A scoping literature search on LT and LR for NELM was performed with PubMed,including English articles up to March 2023.Results:International guidelines recommend LR for NELM in resectable,well-differentiated tumors in the absence of extrahepatic metastatic disease with superior results of LR compared to systemic or liver-directed therapies.Advanced liver surgery has extended resectability criteria whilst entailing increased perioperative risk and short disease-free survival.In highly selected patients(based on the Milan criteria)with unresectable NELM,oncologic results of LT are promising.Prognostic factors include tumor biology(G1/G2)and burden,waiting time for LT,patient age and extrahepatic spread.Based on low-level evi-dence,LT for low-grade NELM within the Milan criteria resulted in improved disease-free survival and overall survival compared to LR.The benefits of LT were lost in patients beyond the Milan NELM-criteria.Conclusions:With adherence to strict selection criteria especially tumor biology,LT for NELM is becoming a valuable option providing oncologic benefits compared to LR.Recent evidence suggests even stricter selection criteria with regard to tumor biology.

This comprehensive review explores the intricate relationship between nutrition,the gut microbiome,steroid hormones,and Parkinson's disease within the context of the gut-brain axis.The gut-brain axis plays a pivotal role in neurodegenerative diseases like Parkinson's disease,encompassing diverse components such as the gut microbiota,immune system,metabolism,and neural pathways.The gut microbiome,profoundly influenced by dietary factors,emerges as a key player.Nutrition during the first 1000 days of life shapes the gut microbiota composition,influencing immune responses and impacting both child development and adult health.High-fat,high-sugar diets can disrupt this delicate balance,contributing to inflammation and immune dysfunction.Exploring nutritional strategies,the Mediterranean diet's anti-inflammatory and antioxidant properties show promise in reducing Parkinson's disease risk.Microbiome-targeted dietary approaches and the ketogenic diet hold the potential in improving brain disorders.Beyond nutrition,emerging research uncovers potential interactions between steroid hormones,nutrition,and Parkinson's disease.Progesterone,with its anti-inflammatory properties and presence in the nervous system,offers a novel option for Parkinson's disease therapy.Its ability to enhance neuroprotection within the enteric nervous system presents exciting prospects.The review addresses the hypothesis thatα-synuclein aggregates originate from the gut and may enter the brain via the vagus nerve.Gastrointestinal symptoms preceding motor symptoms support this hypothesis.Dysfunctional gut-brain signaling during gut dysbiosis contributes to inflammation and neurotransmitter imbalances,emphasizing the potential of microbiota-based interventions.In summary,this review uncovers the complex web of interactions between nutrition,the gut microbiome,steroid hormones,and Parkinson's disease within the gut-brain axis framework.Understanding these connections not only offers novel therapeutic insights but also illuminates the origins of neurode

A simulation study on spatial and time resolution for a cost-effective positron emission particle tracking system

This work is the second part of a simulation study investigating the processing of densely packed and moving granular assemblies by positron emission particle tracking (PEPT).Since medical positron emission tomography (PET) scanners commonly used for PEPT are very expensive,a PET-like detector system based on cost-effective organic plastic scintillator bars is being developed and tested for its capabilities.In this context,the spatial resolution of a resting positron source,a source moving on a freely designed model path,and a particle motion given by a discrete element method (DEM) simulation is studied using Monte Carlo simulations and the software toolkit Geant4.This not only extended the simulation and reconstruction to a moving source but also significantly improved the spatial resolution compared to previous work by adding oversampling and iteration to the reconstruction algorithm.Furthermore,in the case of a source following a trajectory developed from DEM simulations,a very good resolution of about 1 mm in all three directions and an average 3D deviation between simulated and reconstructed events of 2.3 mm could be determined.Thus,the resolution for realistic particle motion within the generic grate system (which is the test rig for further experimental studies) is well below the smallest particle size.The simulation of the dependence of the reconstruction accuracy on tracer particle location revealed a nearly constant efficiency within the entire detector system,which demonstrates that boundary effects can be neglected.
